| 1 | Sound Source Localisation for a High-Speed Train and Its Transfer Path to Interior Noise显示文摘Noise is one of the key issues in the operation of high-speed railways, with sound source localisation and its transfer path as the two major aspects. This study investigates both the exterior and interior sound source distribution of a high-speed train and presents a method for performing the contribution analysis of airborne sound with regard to the interior noise. First, both exterior and interior sound source locations of the high-speed train are identified through in-situ measurements. Second, the sound source contribution for di erent regions of the train and the relationships between the exterior and interior noises are analysed. Third, a method for conducting the contribution analysis of airborne sound with regard to the interior noise of the high-speed train is described. Lastly, a case study on the sidewall area is carried out, and the contribution of airborne sound to the interior noise of this area is obtained. The results show that, when the high-speed train runs at 310 km/h, dominant exterior sound sources are located in the bogie and pantograph regions, while main interior sound sources are located at the sidewall and roof. The interior noise, the bogie area noise and the sound source at the middle of the coach exhibit very similar rates of increase with increasing train speed. | 2 | A LysM Receptor Heteromer Mediates Perception of Arbuscular Mycorrhizal Symbiotic Signal in Rice显示文摘Symbiotic microorganisms improve nutrient uptake by plants.To initiate mutualistic symbiosis with arbus-cular mycorrhizal(AM)fungi,plants perceive Myc factors,including lipochitooligosaccharides(LCOs)and short-chain chitooligosaccharides(CO4/CO5),secreted by AM fungi.However,the molecular mechanism of Myc factor perception remains elusive.In this study,we identified a heteromer of LysM receptor-like kinases consisting of OsMYR1/OsLYK2 and OsCERK1 that mediates the perception of AM fungi in rice.CO4 directly binds to OsMYR1,promoting the dimerization and phosphorylation of this receptor complex.Compared with control plants,Osmyr1 and Oscerk1 mutant rice plants are less sensitive to Myc factors and show decreased AM colonization.We engineered transgenic rice by expressing chimeric receptors that respectively replaced the ectodomains of OsMYR1 and OsCERK1 with those from the homologous Nod factor receptors MtNFP and MtL YK3 of Medicago truncatula.Transgenic plants displayed increased cal-cium oscillations in response to Nod factors compared with control rice.Our study provides significant mechanistic insights into AM symbiotic signal perception in rice.Expression of chimeric Nod/Myc recep-tors achieves a potentially important step toward generating cereals that host nitrogen-fixing bacteria. | Jiangman He Chi Zhang Huiling Dai Huan Liu Xiaowei Zhang Jun Yang Xi Chen Yayun Zhu Dapeng Wang Xiaofeng Qi Weichao Li Zhihui Wang Guoyong An Nan YU Zuhua He Yong-Fei Wang Youli Xiao Peng Zhang Ertao Wang | 2019 | Molecular Plant2019,12,12: | 9 |
| 3 | Adapting rice anther culture to gene transformation and RNA interference显示文摘Anther culture offers a rapid method of generating homozygous lines for breeding pro- gram and genetic analysis.To produce homozygous transgenic lines of rice(Oryza sativa L.)in one step,we developed an efficient protocol of anther-callus-based transformation mediated by Agro- bacterium after optimizing several factors influencing efficient transformation,including callus induc- tion and Agrobacterium density for co-cultivation.Using this protocol,we obtained 145 independent green transformants from five cultivars of japonica rice by transformation with a binary vector pCXK1301 bearing the rice gene,Xa21 for resistance to bacterial blight,of which 140 were further confirmed by PCR and Southern hybridization analysis,including haploids(32.1%),diploids(62.1%) and mixoploids(7.5%).Fifteen diploids were found to be doubled haploids,which accounted for 10.7%of the total positive lines.Finally,by including 28 from colchicine induced or spontaneous dip- loidization of haploids later after transformation,a total of 43 doubled haploids(30.7%)of Xa21 transgenic lines were obtained.We also generated two RNAi transgenic haploids of the rice Os- MADS2 gene,a putative redundant gene of OsMADS4 based on their sequence similarity,to inves- tigate its possible roles in rice flower development by this method.Flowers from the two OsMADS2 RNAi transgenic haploids displayed obvious homeotic alternations,in which lodicules were trans- formed into palea/lemma-like tissues,whereas identities of other floral organs were maintained.The phenotypic alternations were proved to result from specific transcriptional suppression of OsMADS2 gene by the introduced RNAi transgene.The results confirmed that OsMADS2 is involved in lodicule development of rice flower and functionally redundant with OsMADS4 gene.Our results demonstrated that rice anther culture could be adapted to gene transformation and RNAi analysis in rice. | CHEN Caiyan 1,2* ,XIAO Han 1,2* ,ZHANG Wenli 1,2 ,WANG Aiju 1,2 ,XIA Zhihui 1 ,LI Xiaobing 1 , ZHAI Wenxue 1 ,CHENG Zhukuan 1 &ZHU Lihuang 1 1.State Key Laboratory of Plant Genomics and National Center for Plant Gene Research,Institute of Genetics and Develop- mental Biology,Chinese Academy of Sciences,Beijing 100101,China 2.Graduate School of the Chinese Academy of Sciences,Beijing 100039,China | 2006 | Science China(Life Sciences)2006,49,5: | 5 |
| 4 | Combustion instability of pilot flame in a pilot bluff body stabilized combustor显示文摘Combustion instability of pilot flame has been investigated in a model pilot bluff body stabilized combustor by running the pilot flame only. The primary objectives are to investigate the pilot flame dynamics and to provide bases for the study of the interaction mechanisms between the pilot flame and the main flame. Dynamic pressures are measured by dynamic pressure transducers. A high speed camera with CH*bandpass filter is used to capture the pilot flame dynamics. The proper orthogonal decomposition(POD) is used to further analyze the high speed images. With the increase of the pilot fuel mass flow rate, the pilot flame changes from stable to unstable state gradually. The combustion instability frequency is 136 Hz when the pilot flame is unstable. Numerical simulation results show that the equivalence ratios in both the shear layer and the recirculation zone increase as the pilot fuel mass flow rate increases. The mechanism of the instability of the pilot flame can be attributed to the coupling between the second order acoustic mode and the unsteady heat release due to symmetric vortex shedding. | 5 | Patient-derived non-small cell lung cancer xenograft mirrors complex tumor heterogeneity显示文摘Objective:Patient-derived xenograft(PDX)models have shown great promise in preclinical and translational applications,but their consistency with primary tumors in phenotypic,genetic,and pharmacodynamic heterogeneity has not been well-studied.This study aimed to establish a PDX repository for non-small cell lung cancer(NSCLC)and to further elucidate whether it could preserve the heterogeneity within and between tumors in patients.Methods:A total of 75 surgically resected NSCLC specimens were implanted into immunodeficient NOD/SCID mice.Based on the successful establishment of the NSCLC PDX model,we compared the expressions of vimentin,Ki67,EGFR,and PD-L1 proteins between cancer tissues and PDX models using hematoxylin and eosin staining and immunohistochemical staining.In addition,we detected whole gene expression profiling between primary tumors and PDX generations.We also performed whole exome sequencing(WES)analysis in 17 first generation xenografts to further assess whether PDXs retained the patient heterogeneities.Finally,paclitaxel,cisplatin,doxorubicin,atezolizumab,afatininb,and AZD4547 were used to evaluate the responses of PDX models to the standard-of-care agents.Results:A large collection of serially transplantable PDX models for NSCLC were successfully developed.The histology and pathological immunohistochemistry of PDX xenografts were consistent with the patients’tumor samples.WES and RNA-seq further confirmed that PDX accurately replicated the molecular heterogeneities of primary tumors.Similar to clinical patients,PDX models responded differentially to the standard-of-care treatment,including chemo-,targeted-and immuno-therapeutics.Conclusions:Our established PDX models of NSCLC faithfully reproduced the molecular,histopathological,and therapeutic characteristics,as well as the corresponding tumor heterogeneities,which provides a clinically relevant platform for drug screening,biomarker discovery,and translational research. | Xuanming Chen Cheng Shen Zhe Wei Rui Zhang Yongsheng Wang Lili Jiang Ke Chen Shuang Qiu Yuanli Zhang Ting Zhang Bin Chen Yanjun Xu Qiyi Feng Jinxing Huang Zhihui Zhong Hongxia Li Guowei Che Kai Xiao | 2021 | Cancer Biology & Medicine2021,18,1: | 5 |
