| 2 | miR-188-5p regulates proliferation and invasion via PI3K/Akt/MMP-2/9 signaling in keloids显示文摘Keloids(KDs)and hypertrophic scars(HSs),two forms of pathological scars,seriously affect the physical and psychological health of patients.Despite many similarities with HSs,KDs are characterized by invasion and a high rate of recurrence after surgery,features they share in common with tumors.The underlying molecular mechanisms of this phenomenon have not been fully elucidated.In this study,we used microRNA(miRNA)array analysis to search for invasion-associated miRNAs in KDs.The expression of miR-188-5p in KDs,HSs,normal skin(NS)tissues,and cell lines was measured by quantitative real-time polymerase chain reaction.Furthermore,cell proliferation,migration,and invasion were detected in KD fibroblasts(KFs)and HS fibroblasts(HSFs),and interrelated proteins were ascertained by western blot analysis.It was found that miR-188-5p was significantly decreased in KD tissue compared with HS and NS tissues.Upregulated expression of miR-188-5p suppressed KF proliferation,migration,and invasion;and decreased expression of miR-188-5p also promoted HSF proliferation,migration,and invasion.The protein levels of MMP-2,MMP-9,PI3K,and p-Akt in miR-188-5p mimic-transfected KFs were repressed.In contrast,after transfection with miR-188-5p inhibitor,the protein levels of MMP-2,MMP-9,PI3K,and p-Akt were higher than the control in HSFs.Treatment with PI3K/Akt inhibitor LY294002 in KFs with miR-188-5p inhibitor did not further reduce their proliferation,migration,and invasion.The upregulation of MMP-2 and MMP-9 by miR-188-5p inhibitor could be abolished by LY294002.These findings together demonstrate a tumor-suppressive role of miR-188-5p in KD proliferation and invasion via PI3K/Akt/MMP-2/9 signaling,indicating that miR-188-5p may be a potential prognostic marker and therapeutic target for KDs. | Wenyan Zhu Xiaoyan Wu Bo Yang Xiaodong Yao Xiaomei Cui Pan Xu Xiaodong Chen | 2019 | Acta Biochimica et Biophysica Sinica2019,51,2: | 9 |
| 3 | Fractional Fourier domain hopped communication method based on chirp modulation for underwater acoustic channels显示文摘To improve the data rate of underwater acoustic frequency-hopped communications, frequency hopping is applied to different orders of fractional Fourier domain(Fr FD), to enable non-intrusive, bandwidth-limited acoustic communications.An Fr FD frequency-hopped communication method based on chirp modulation, namely multiple chirp shift keying-Fr FD hopping(MCSK-Fr FDH), is proposed for underwater acoustic channels. Validated by both simulations and experimental results, this method can reach a bandwidth efficiency twice more than conventional frequency-hopped methods with the same data rate and anti-multipath capability, suggesting that the proposed method achieves a better performance than the traditional frequencyhopped communication in underwater acoustic communication channels. Visceral hypersensitivity is closely associated with irritable bowel syndrome. Mild moxibustion can relieve chronic visceral hyperalgesia in rats with irritable bowel syndrome. We hypothesized that prokineticin-1 and prokineticin receptor-1 is the key target in the mechanism. This study established chronic visceral hyperalgesia rat models by colorectal distention. | 6 | Electroacupuncture at Tianshu (ST 25) for diarrhea-predominant irritable bowel syndrome using positron emission tomography Changes in visceral sensation center显示文摘Previous studies have demonstrated that electroacupuncture therapy is effective in the treatment of irritable bowel syndrome. However, the precise mechanism of this therapy is unknown. The present study served to investigate the effects of electroacupuncture therapy on treatment of patients with diarrhea-predominant irritable bowel syndrome (IBS). We compared brain activation maps based on the changes of cerebral glucose metabolism obtained by 18-fluorodeoxyglucose positron emission tomography scanning under three conditions: resting, rectal balloon distension and rectal balloon distension plus electroacupuncture. Under the resting condition, compared with healthy controls, IBS patients displayed an increasing regional cerebral metabolic rate of glucose over a wide range: bilateral superior temporal gyrus, right middle occipital gyrus, superior frontal gyrus and bilateral middle frontal gyrus. However, there was no significant activity in the visceral pain center. Compared with the resting condition, under the rectal balloon distension condition, patients with IBS had a greater regional cerebral metabolic rate of glucose in the prefrontal cortex, left anterior cingulate cortex, postcentral gyrus, precentral gyrus and temporal gyrus. Under the rectal balloon distension plus electroacupuncture condition, stimulation by electroacupuncture at Tianshu (ST 25) manifested a decreased regional cerebral metabolic rate of glucose in the left cingulate gyrus, right insula, right caudate nucleus, fusiform gyrus and hippocampal gyrus. | 7 | Effects of electroacupuncture on c-Fos expression in the spinal cord and brain of rats with chronic visceral hypersensitivity显示文摘BACKGROUND:Visceral hypersensitivity is the main cause of irritable bowel syndrome.c-Fos is a marker of visceral hypersensitivity in the central nervous system.Electroacupuncture can relieve chronic visceral hypersensitivity in rats,but the mechanism is still unknown. OBJECTIVE:To identify c-Fos expression in the spinal cord and cerebral cortex of rats with chronic visceral hypersensitivity,and to test the effects of electroacupuncture on pain sensitivity in rats with chronic visceral hypersensitivity. DESIGN,TIME AND SETTING:A randomized controlled animal experiment was performed at the Animal Experimental Center,Shanghai University of Traditional Chinese Medicine,from January to April,2007. MATERIALS:A total of 24 neonatal,male,Sprague Dawley rats,aged five days old,were equally and randomly assigned into a normal group,a model group,and an electroacupuncture group. Rabbit anti-rat c-Fos antibody and Evision secondary antibody kits(Sigma,USA),diaminobenzidine kit(Dako,Denmark),and an LD202H electroacupuncture apparatus(Huawei,Beijing,China) were used in this study. METHODS:Neonatal rats from the model and electroacupuncture groups were used to establish rat models of chronic visceral hypersensitivity by the saccule stimulation method.After model establishment,0.25 mm diameter electric needles were inserted into Tianshu(ST 25) and Shangjuxu(ST37) at a depth of approximately 0.5 cm,with an square wave(alternating current frequency at 100/20 Hz,amplitude ranged 0.2-0.6 ms,intensity at 1 mA) once for 20 minutes,once a day,for seven days.Rats in the normal and model groups were not treated. MAIN OUTCOME MEASURES:Following 7 days of treatment,c-Fos expression in the spinal cord and cerebral cortex was detected by immunohistochemistry.After the first electroacupuncture treatment,abdominal withdrawal reflex scores were investigated to evaluate the pain threshold for chronic visceral hypersensitivity in rats. RESULTS:Visceral hypersensitivity increased c-Fos staining(P<0.05),and electroacupuncture significantly decreased the number of these cells to near normal levels(P>0.05).Abdominal withdrawal reflex scores were significantly lower in the electroacupuncture and normal groups than in the model group(P<0.05) and were similar between the electroacupuncture and normal groups (P>0.05). | 8 | Evaluation of human epidermal growth factor receptor 2 status of breast cancer using preoperative multidetector computed tomography with deep learning and handcrafted radiomics features显示文摘Objective:To evaluate the human epidermal growth factor receptor 2(HER2)status in patients with breast cancer using multidetector computed tomography(MDCT)-based handcrafted and deep radiomics features.Methods:This retrospective study enrolled 339 female patients(primary cohort,n=177;validation cohort,n=162)with pathologically confirmed invasive breast cancer.Handcrafted and deep radiomics features were extracted from the MDCT images during the arterial phase.After the feature selection procedures,handcrafted and deep radiomics signatures and the combined model were built using multivariate logistic regression analysis.Performance was assessed by measures of discrimination,calibration,and clinical usefulness in the primary cohort and validated in the validation cohort.Results:The handcrafted radiomics signature had a discriminative ability with a C-index of 0.739[95%confidence interval(95%CI):0.661-0.818]in the primary cohort and 0.695(95%CI:0.609-0.781)in the validation cohort.The deep radiomics signature also had a discriminative ability with a C-index of 0.760(95%CI:0.690-0.831)in the primary cohort and 0.777(95%CI:0.696-0.857)in the validation cohort.The combined model,which incorporated both the handcrafted and deep radiomics signatures,showed good discriminative ability with a C-index of 0.829(95%CI:0.767-0.890)in the primary cohort and 0.809(95%CI:0.740-0.879)in the validation cohort.Conclusions:Handcrafted and deep radiomics features from MDCT images were associated with HER2 status in patients with breast cancer.Thus,these features could provide complementary aid for the radiological evaluation of HER2 status in breast cancer. | Xiaojun Yang Lei Wu Ke Zhao Weitao Ye Weixiao Liu Yingyi Wang Jiao Li Hanxiao Li Xiaomei Huang Wen Zhang Yanqi Huang Xin Chen Su Yao Zaiyi Liu Changhong Liang | 2020 | Chinese Journal of Cancer Research2020,32,2: | 4 |
| 9 | Testosterone attenuates pulmonary epithelial inflammation in male rats of COPD model through preventing NRF1-derived NF-κB signaling显示文摘Testosterone deficiency is common in male patients with chronic obstructive pulmonary disease (COPD) and may correlate with the deterioration of COPD. Clinical research suggests that testosterone replacement therapy may slow the COPD progression, but the specific biological pathway remains unclear. In this study, we explored the effect of testosterone on pulmonary inflammation in male COPD rats. The animals were co-treated with lipopolysaccharide (LPS) and cigarette to induce COPD. In COPD rats, nuclear respiratory factor 1 (NRF1) and NF-κB p65 were upregulated. In cigarette smoke extract (CSE)-, LPS-, or the combination of CSE and LPS-treated L132 cells, NRF1 and p65 were also upregulated. Silencing NRF1 resulted in the downregulation of p65. ChIP‒seq, ChIP‒qPCR, and luciferase results showed that NRF1 transcriptionally regulated p65. Both male and female COPD rats showed an upregulated NRF1 level and similar pulmonary morphology. But NRF1 was further upregulated in male castrated rats. Further supplementing testosterone in castrated male rats significantly reduced NRF1, pulmonary lesions, and inflammation. Supplementation of testosterone also reduced the phosphorylation of p65 and IKKβ induced by LPS or CSE in L132 cells. | 13 | Negative association of time in range and urinary albumin excretion rate in patients with type 2 diabetes mellitus:a retrospective study of inpatients显示文摘Background:Time in range(TIR)refers to the time an individual spends within their target glucose range,which now has been popularized as an important metric to classify glycemic management and also recognized as an important outcome of current diabetes therapies.This study aimed to investigate the association between TIR and the severity of the urinary albumin excretion rate(UAER)in patients with type 2 diabetes mellitus(T2DM).Methods:We retrospectively analyzed the data of 1014 inpatients with T2DM at the Department of Endocrinology and Metabolism of Peking University International Hospital,China.TIR was defined as the percentage of blood glucose within the target range of 3.90-10.00 mmol/L.Urine samples for assessment of UAER were collected for 3 consecutive days from the start of hospitalization.Results:The TIR values for patients with normal urine levels of albumin,microalbuminuria,and macroalbuminuria were 70%±20%,50%±20%,and 30%±20%,respectively(allP<0.001).The patients were stratified according to quartiles of TIR as follows:quartile(Q)1,<55%;Q2,55%-72%;Q3,73%-83%;and Q4,>83%.The incidences of microalbuminuria in Q1,Q2,Q3,and Q4 were 41.1%,21.6%,7.1%,and 5.5%(allP<0.001),respectively.The respective incidences of macroalbuminuria were 24.2%,1.1%,1.4%,and 0%(allP<0.001).In multinomial logistic regression analyses,TIR was significantly correlated with microalbuminuria(odds ratio[OR]0.58,95%confidence interval[CI]:0.52-0.65,P<0.001)and macroalbuminuria(OR 0.26,95%CI:0.18-0.38,P<0.001)after adjusting for age,sex,body mass index,diabetes duration,systolic blood pressure,and levels of triglycerides,glycosylated hemoglobin A1c,and creatinine.Conclusion:The proportion of blood glucose in TIR is closely related to the severity of UAER in patients with T2DM. | Sanbao Chai Shanshan Wu Sixu Xin Ning Yuan Jianbin Sun Xiaomei Zhang Linong Ji | 2022 | Chinese Medical Journal2022,,9: | 3 |

| 17 | Immobilization of acetylcholinesterase on functionalized SBA-15 mesoporous molecular sieve for detection of organophosphorus and carbamate pesticide显示文摘The immobilization of acetylcholinesterase(AChE) on amino functionalized SBA-15 mesoporous sieves(NH_2-SBA-15) was explored for detection of organophosphorus and carbamate pesticides. Several methods have been studied and the 'adsorption-crosslinking' is found to be the best one. Firstly, AChE and bovine serum albumin(BSA) were adsorbed in turn on the surface of NH_2-SBA-15 by electrostatic adsorption, and then glutaraldehyde was added for cross-linking the amino group of AChE. The NH_2-SBA-15 showed around 95% immobilization efficiency and the specific activity of immobilized AChE(ACh ENH_2-SBA-15) reached 130% relative to free AChE under optimal immobilization conditions. It was found ACh E-NH_2-SBA-15 retained 94.0% and 82.8% of its initial activity after 60 days at 4℃ and 25℃,respectively. It remained 55.0% of its initial activity after 6 times recycling. During the detection of pesticide, the AChE-NH_2-SBA-15 showed wider linear range and much lower limit of detection compared with free AChE. The current method showed good recovery(93.8%-109.3%) and low RSD(<5%) for both standard solution and real vegetable samples of Carbaryl and Trichlorfon. | 18 | Effect of Jiaji electroacupuncture in transected rat spinal cord显示文摘We evaluated the effect of Jiaji electroacupuncture on cell proliferation and the expression of markers of endogenous neural stem cell activation after complete spinal cord transection. Female Wistar rats were assigned to 4 groups (n = 24 each): a sham-operated group, a control group, a Jiaji electroacupuncture group, and a Jiaji electroacupuncture preconditioning group. Motor function was significantly improved in the acupuncture groups compared to the control group at 7 and 14 d. Numbers of bromodeoxyuridine (BrdU)-, nestin-, and glial fibrillary acidic protein (GFAP)-positive cells were significantly greater in the acupuncture groups than in the controls at each time point. Expression of nestin and GFAP mRNA was significantly higher in the acupuncture groups than in the controls at each time point. | 19 | Performance of CO_2 absorption in a diameter-varying spray tower显示文摘The application of spray towers for CO_2 capture is a development trend in recent years. However, most of the previous jobs were conducted in a cylindrical tower by using a single spray nozzle, whose configuration and performance is not good enough for industrial application. To solve this problem, the present work proposed a diameter-varying spray tower and a new spray mode of dual-nozzle opposed impinging spray to enhance the heat and mass transfer of CO_2 absorption process. Experiments were performed to investigate the mass transfer performance (in terms of the CO_ 2 removal rate (η) and the overall mass transfer coefficient (K_Ga_e)) of the improved spray tower under various operating conditions. Experimental results showed that the liquid to gas ratio and mole ratio of MEA to CO_2 are major factors, which affect the absorption performance and the maximums of η and (K_Ga_e that are 94.0% and 0.574 kmol·m^(-3)·h^(-1)·kP a^(-1), respectively, under the experimental conditions.
