2Performance of the Seasonal Forecasting of the Asian Summer Monsoon by BCC_CSM1.1(m)显示文摘This paper provides a comprehensive assessment of Asian summer monsoon prediction skill as a function of lead time and its relationship to sea surface temperature prediction using the seasonal hindcasts of the Beijing Climate Center Climate System Model, BCC CSM1.1(m). For the South and Southeast Asian summer monsoon, reasonable skill is found in the model's forecasting of certain aspects of monsoon climatology and spatiotemporal variability. Nevertheless, deficiencies such as significant forecast errors over the tropical western North Pacific and the eastern equatorial Indian Ocean are also found. In particular, overestimation of the connections of some dynamical monsoon indices with large-scale circulation and precipitation patterns exists in most ensemble mean forecasts, even for short lead-time forecasts.Variations of SST, measured by the first mode over the tropical Pacific and Indian oceans, as well as the spatiotemporal features over the Ni no3.4 region, are overall well predicted. However, this does not necessarily translate into successful forecasts of the Asian summer monsoon by the model. Diagnostics of the relationships between monsoon and SST show that difficulties in predicting the South Asian monsoon can be mainly attributed to the limited regional response of monsoon in observations but the extensive and exaggerated response in predictions due partially to the application of ensemble average forecasting methods. 3Herbal decoctosome is a novel form of medicine显示文摘Traditionally, herbal medicine is consumed by drinking decoctions produced by boiling herbs with water. The functional components of the decoction are heat stable. Small RNAs(sRNAs) were reported as a new class of functional components in decoctions. However, the mechanisms by which sRNAs survive heat treatment of the decoction and enter cells are unclear.Previous studies showed that plant-derived exosome-like nanoparticles(ELNs), which we call botanosomes, could deliver therapeutic reagents in vivo. Here, we report that heat-stable decoctosomes(ELNs) from decoctions have more therapeutic effects than the decoctions in vitro and demonstrate therapeutic efficacy in vivo. Furthermore, sRNAs, such as HJT-sRNA-m7 and PGY-sRNA-6, in the decoctosome exhibit potent anti-fibrosis and anti-inflammatory effects, respectively. Decoctosome is comprised of lipids, chemical compounds, proteins, and s RNAs. A medical decoctosome mimic is called bencaosome. A single lipid sphinganine(d22:0) identified in the decoctosome was mixed and heated with the synthesized sRNAs to form the simplest bencaosome. This simple bencaosome structure was identified by critical micelle concentration(cmc) assay that sRNAs coassembled with sphinganine(d22:0) to form the lipid layers of vesicles. The heating process facilitates co-assembly of sRNAs and sphinganine(d22:0) until a steady state is reached. The artificially produced sphinganine-HJT-sRNA-m7 and sphinganinePGY-sRNA-6 bencaosomes could ameliorate bleomycin-induced lung fibrosis and poly(I:C)-induced lung inflammation, respectively, following oral administration in mice. 4Plant-derived phosphocholine facilitates cellular uptake of anti-pulmonary fibrotic HJT-sRNA-m7显示文摘Pulmonary fibrosis, a progressive chronic disease with a high mortality rate, has limited treatment options. Currently, lung transplantation remains the only effective treatment. Here we report that a small RNA, HJT-sRNA-m7, from a Chinese herbal medicine Hong Jing Tian(HJT, RHODIOHAE CRENULATAE RADIX ET RHIZOMA, Rhodiola crenulata) can effectively reduce the expressions of fibrotic hallmark genes and proteins both in alveolar in vitro and in mouse lung tissues in vivo. We also discovered over one hundred oil-soluble chemicals from HJT decoctions, most of which are found in lipid extracts from other Chinese herbals decoctions, including Pu Gong Ying(PGY, TARAXACI HERBA, Taraxacum mongolicum), Chuan Xin Lian(CXL, changed to 'ANDROGRAPHIS HERBA, Andrographis paniculata'), and Jin Yin Hua(JYH, lonicera japonica or Honeysuckle). We identified the active component in these decoctions as two forms of phosphocholines, PC(18:0/18:2) and PC(16:0/18:2). These PCs potentially could form liposomes with small RNAs to enter human alveolar and gastric cells. 5Dendritic Cells Transduced with SOCS1 Gene Exhibit Regulatory DC Properties and Prolong Allograft Survival显示文摘SOCS1 is a key regulator of cytokine signaling and is important for maintaining balance in the immune system. It is thought to participate in negative feedback loops in cytokine signaling and may be an important signal for the regulation of dendritic cell (DC) maturation. However, it remains unclear whether DCs transduced with SOCS1 exhibit characteristics of regulatory DCs and induce allogeneic T-cell hyporesponsiveness. In this study, we constructed adenoviral vector coding SOCS1 (Ad-SOCS1) that can efficiently increase SOCS1 gene expression in bone marrow-derived dendritic cells. DCs transduced with Ad-SOCS1 (DC-SOCS1) expressed low levels of costimulatory and MHC molecules, were resistant to maturation and activation stimulation, induced allogeneic T-cell hyporesponsiveness, and promoted the generation of regulatory-like T cells in vitro. DC-SOCS1 pretreatment significantly prolonged the survival of allografts and led to a substantial increase in the generation of regulatory T cells. 6Relationships between Interannual and Intraseasonal Variations of the Asian–Western Pacific Summer Monsoon Hindcasted by BCC CSM 1.1(m)显示文摘Using hindcasts of the Beijing Climate Center Climate System Model, the relationships between interannual variability(IAV) and intraseasonal variability(ISV) of the Asian–western Pacific summer monsoon are diagnosed. Predictions show reasonable skill with respect to some basic characteristics of the ISV and IAV of the western North Pacific summer monsoon(WNPSM) and the Indian summer monsoon(ISM). However, the links between the seasonally averaged ISV(SAISV) and seasonal mean of ISM are overestimated by the model. This deficiency may be partially attributable to the overestimated frequency of long breaks and underestimated frequency of long active spells of ISV in normal ISM years, although the model is capable of capturing the impact of ISV on the seasonal mean by its shift in the probability of phases.Furthermore, the interannual relationships of seasonal mean, SAISV, and seasonally averaged long-wave variability(SALWV; i.e., the part with periods longer than the intraseasonal scale) of the WNPSM and ISM with SST and low-level circulation are examined. The observed seasonal mean, SAISV, and SALWV show similar correlation patterns with SST and atmospheric circulation, but with different details. 8Advancement in separation materials for blood purification therapy显示文摘Blood purification refers to the extra corporeal therapies of removing potentially toxic substances, in which blood is circulated through an adsorption system loading separation materials. High-efficient inexpensive separation materials are critical to success. In this review, separation materials such as polymers and nanomaterials are summarized and compared. Combining the advantages of the adsorptive membranes and nanomaterials, organic–inorganic hybrid/blend membranes have been developed explosively. These hybrid/blend membranes have both the characteristics of high permeability, easy fabrication, good biocompatibility of adsorptive membranes, and characteristics of fast adsorption rate and high adsorption capacity of nanomaterials. The preparation and modification methodology of the separation materials is reviewed. For affinity separation materials, the relationship of ligand chemistry, ligand density and pores of the matrix is discussed. 9TCP7 functions redundantly with several Class Ⅰ TCPs and regulates endoreplication in Arabidopsis显示文摘TCP(TEOSINTE BRANCHED1/CYCLOIDEA/PROLIFERATING CELL FACTOR) proteins, a family of plantspecific transcription factors, play important roles in many developmental processes. However, genetic and functional redundancy among class I TCP limits the analysis of their biological roles. Here, we identified a dominantnegative mutant of Arabidopsis thaliana TCP7 named leaf curling-upward(lcu), which exhibits smaller leaf cells and shorter hypocotyls than the wild type, due to defective endoreplication. A septuple loss-of-function mutant of TCP7, TCP8, TCP14, TCP15, TCP21, TCP22, and TCP23 displayed similar developmental defects to those of lcu.Genome-wide RNA-sequencing showed that lcu and the septuple mutant share many misexpressed genes.Intriguingly, TCP7 directly targets the CYCLIN D1;1 (CYCD1;1) locus and activates its transcription. We determined that the C-terminus of TCP7 accounts for its transcriptional activation activity. Furthermore, the mutant protein LCU exhibited reduced transcriptional activation activity due to the introduction of an EAR-like repressive domain at its C-terminus. 10Alzheimer's disease treated with combined therapy based on nourishing marrow and reinforcing Qi显示文摘OBJECTIVE:To study the efficacy and safety of combined Traditional Chinese Medicine(TCM) therapy based on nourishing marrow to improve intellect and reinforcing Qi to activate bloodon mild to moderate Alzheimer's disease(AD).METHODS:Sixty-six patients with AD,whoseMini-Mental State Examination(MMSE) score were from 10-24,were randomized equally into an intervention group and a control group.The control group was given Aricept(5 mg,once daily).The intervention group was further divided into Yang-Qi deficiency(n = 18) and of Yin-Qi deficiency(n = 15)subgroups.Patients in the Yang-Qi deficiency group were intravenously administered Shenfu injection,60 mL,and deproteinized calf blood injection(DCBl),1.2 g,once daily.The Yin-Qi deficiency group was given Shenmai injection,60 mL,and DCBl,1.2 g,once daily.Each course lasted 21 days.RESULTS:Compared with the control group and with pre-treatment in the same group,MMSE,clinical dementia rating,and activities of daily living scale scores in the intervention group were significantly improved(all P<0.05).These metrics mildly improved in the control group compared with before treatment(P>0.05).No adverse effects were observed in any group during treatment.CONCLUSION:We found that combined TCM therapy is effective and safe for managing mild to moderate AD.Chen Songlin Yao Xiaoli Liang Yinying Mei Weiyi Liu Xiaoyun Zhang Changran 2015Journal of Traditional Chinese Medicine2015,35,3:4

14An Expression for Gravity Generated by an Anomalous Geological Body and Its Application in Bathymetry Inversion显示文摘The gravity anomaly and vertical gravity gradient due to anomalous geological bodies are mainly computed by numerical methods,so it is difficult and time-consuming to use the gravity-geological method to invert seafloor topography.This paper addresses this issue by deriving an expression for gravity generated by a cylinder based on a series expansion.The choice of number for terms in the series is estimated by comparing with the numerical method,especially when the depth H=4000m,the accuracy of 1 mGal(1 Gal=10^(-2)m/s^(2))can be achieved when the series are 9.The expressions can be used to establish the relationships between the shape of an anomalous body and the generated vertical gravity and vertical gravity gradient,respectively.Finally,the potential applications of the expressions in inverting seafloor topography are illustrated by synthetic examples.Huan XU Jinhai YU Xiaoyun WAN Lei LIANG 2021Journal of Geodesy and Geoinformation Science2021,4,4:2
15CUL4B facilitates HBV replication by promoting HBx stabilization显示文摘Objective:Hepatitis B virus(HBV)infection is a major public health problem worldwide.However,the regulatory mechanisms underlying HBV replication remain unclear.Cullin 4 B-RING ubiquitin E3 ligase(CRL4 B)is involved in regulating diverse physiological and pathophysiological processes.In our study,we aimed to explain the role of CUL4 B in HBV infection.Methods:Cul4 b transgenic mice or conditional knockout mice,as well as liver cell lines with CUL4 B overexpression or knockdown,were used to assess the role of CUL4 B in HBV replication.Immunoprecipitation assays and immunofluorescence staining were performed to study the interaction between CUL4 B and HBx.Cycloheximide chase assays and in vivo ubiquitination assays were performed to evaluate the half-life and the ubiquitination status of HBx.Results:The hydrodynamics-based hepatitis B model in Cul4 b transgenic or conditional knockout mice indicated that CUL4 B promoted HBV replication(P<0.05).Moreover,the overexpression or knockdown system in human liver cell lines validated that CUL4 B increased HBV replication in an HBx-dependent manner.Importantly,immunoprecipitation assays and immunofluorescence staining showed an interaction between CUL4 B and HBx.Furthermore,CUL4 B upregulated HBx protein levels by inhibiting HBx ubiquitination and proteasomal degradation(P<0.05).Finally,a positive correlation between CUL4 B expression and HBV pg RNA level was observed in liver tissues from HBV-positive patients and HBV transgenic mice.Conclusions:CUL4 B enhances HBV replication by interacting with HBx and disrupting its ubiquitin-dependent proteasomal degradation.CUL4 B may therefore be a potential target for anti-HBV therapy.Haixia Shan Bo Wang Xiaodong Zhang Hui Song Xi Li Yongxin Zou Baichun Jiang Huili Hu Hao Dou Changshun Shao Lifen Gao Chunhong Ma Xiaoyun Yang Xiaohong Liang Yaoqin Gong 2022Cancer Biology & Medicine2022,19,1:2
16Lowing the energy loss of organic solar cells by molecular packing engineering via multiple molecular conjugation extension显示文摘The central unit(benzo[c][1,2,5]thiadiazole) in Y6 series of molecules plays a determining role in their unique intermolecular packing for a three-dimensionally(3D) network, largely endowing their organic solar cells(OSCs) with so far the best power conversion efficiencies(PCEs) and also largely suppressed energy losses(Eloss). Despite its vital role in molecular packing, very few explorations for central unit have been conducted due to possibly the constructing challenge of central heterocyclic units.Herein, a highly efficient acceptor-donor-acceptor(A-D-A) type electron acceptor, CH17, has been designed and constructed,featured with a prominent π extension in both directions of the central and end units with respect to Y6 series. Such a multiple and much enhanced conjugation extension in CH17 enables a much more effective and compact 3D molecular packing compared with that of Y6 supported by X-ray single crystal and other analysis, mainly caused by a newly observed distinctive dual “end unit to central unit” packing mode. This much favorable molecular packing, also kept in its blends with donor materials, leads a larger electron and hole transfer integrals and hence much improved charge transport, and reduced energetic disorders in CH17blends. More importantly, the observed upshifted charge transfer(CT) state of CH17 blends compared with that of Y6, due to its increased molecular conjugation extension in both directions, further enhances the hybridization between its CT and local exciton(LE) states, resulting in higher luminescence efficiency, much suppressed non-radiative recombination loss and smaller Elosswith respect to that of Y6. Consequently, an excellent PCE of 17.84% is achieved with PM6 as the donor in a binary device compared with a PCE of 16.27% for the controlled Y6 device. Furthermore, a further improved PCE of 18.13% is achieved by CH17-based ternary single-junction OSCs along with a markedly reduced Elossof 0.49 e V and larger open-circuit voltage(Voc) of0.89 V, compared with that(16.27% of PCE, 0.85 V of Voc, and 0.53 e V of Eloss) of the control device using Y6. 17New techniques and strategies in drug discovery显示文摘Great success has been witnessed in last decades,some new techniques and strategies have been widely used in drug discovery.In this roadmap,several representative techniques and strategies are highlighted to show recent advances in this filed.(A)A DOX protocol has been developed for accurate protein-ligand binding structure prediction,in which first principle method was used to rank the binding poses.Validation against crystal structures have found that DOX prediction achieved an impressive success rate of 99%,indicating significant improvement over molecular docking method.(B)Virtual target profiling is a compound-centric strategy enabling a parallel implementation of interrogating compounds against various targets in a single screen,which has been used in hit/lead identification,drug repositioning,and mechanism-of-action studies.Current and emerging methods for virtual target profiling are briefly summarized herein.(C)Research on targeted autophagy to treat diseases has received encouraging progress.However,due to the complexity of autophagy and disease,experimental and in silico methods should be performed synergistically for the entire process.This part focuses on in silico methods in autophagy research to promote their use in medicinal research.(D)Histone deacetylases(HDACs)play important roles in various biological functions through the deacetylation of lysine residues.Recent studies demonstrated that HDACs,which possess low deacetylase activities,exhibited more efficient defatty-acylase activities.Here,we review the defatty-acylase activity of HDACs and describe examples for the design of isoform selective HDAC inhibitor.(E)The FDA approval of three kinase allosteric inhibitors and some others entering clinical study has spurred considerable interests in this targeted drug discovery area.(F)Recent advances are reviewed in structure-based design of novel antiviral agents to combat drug resistance.(G)Since nitric oxide(NO)exerts anticancer activity depending on its concentration,optimal levels of NO in cancer cells is desirable.In this minireview,we briefly describe recent advances in the research of NO-based anticancer agents by our group and present some opinions on the future development of these agents.(H)The field of photoactivation strategies have been extensively developed for controlling chemical and biological processes with light.This review will summarize and provide insight into recent research advances in the understanding of photoactivatable molecules including photoactivatable caged prodrugs and photoswitchable molecules.Jintong Du Jing Guo Dongwei Kang Zhihong Li Guan Wang Jianbing Wu Zhen Zhang Hao Fang Xuben Hou Zhangjian Huang Guobo Li Xiaoyun Lu Xinyong Liu Liang Ouyang Li Rao Peng Zhan Xiaojin Zhang Yihua Zhang 2020Chinese Chemical Letters2020,31,7:1
