Clinical and biochemical indexes from 2019-nCoV infected patients linked to viral loads and lung injury显示文摘The outbreak of the 2019-nCoV infection began in December 2019 in Wuhan,Hubei province, and rapidly spread to many provinces in China as well as other countries. Here we report the epidemiological, clinical, laboratory, and radiological characteristics,as well as potential biomarkers for predicting disease severity in 2019-nCoV-infected patients in Shenzhen,China. All 12 cases of the 2019-nCoV-infected patients developed pneumonia and half of them developed acute respiratory distress syndrome (ARDS).The most common laboratory abnormalities were hypoalbuminemia, lymphopenia, decreased percentage of lymphocytes (LYM) and neutrophils (NEU),elevated C-reactive protein (CRP) and lactate dehydrogenase (LDH),and decreased CD8 count. The viral load of 2019-nCoV detected from patient respiratory tracts was positively linked to lung disease severity.ALB,LYM,LYM (%),LDH,NEU (%),and CRP were highly correlated to the acute lung injury.Age,viral load, lung injury score, and blood biochemistry indexes, albumin (ALB),CRP,LDH,LYM (%),LYM,and NEU (%),may be predictors of disease severity. Moreover, the Angiotensin Ⅱlevel in the plasma sample from 2019-nCoV infected patients was markedly elevated and linearly associated to viral load and lung injury. Our results suggest a number of potential diagnosis biomarkers and angiotensin receptor blocker (ARB) drugs for potential repurposing treatment of 2019-nCoV infection. | Yingxia Liu Yang Yang Cong Zhang Fengming Huang Fuxiang Wang Jing Yuan Zhaoqin Wang Jinxiu Li Jianming Li Cheng Feng Zheng Zhang Lifei Wang Ling Peng Li Chen Yuhao Qin Dandan Zhao Shuguang Tan Lu Yin Jun Xu Congzhao Zhou Chengyu Jiang Lei Liu | 2020 | Science China(Life Sciences)2020,63,3: | 157
China's wetland change (1990-2000) determined by remote sensing显示文摘Two wetland maps for the entire China have been produced based on Landsat data acquired around 1990 and 2000. Wetlands in China have been divided into 3 broad categories with 15 sub-categories except rice fields. In 1990, the total wetland area in China was 355208 km2 whereas in 2000 it dropped to 304849 km2 with a net loss of 50360 km2. During an approximate 10-year period, inland wetland reduced from 318326 to 257922 km2, coastal wetland dropped from 14335 to 12015 km2, while artificial wetland increased from 22546 to 34911 km2. The greatest natural wetland loss occurred in Heilongjiang, Inner Mon- golia, and Jilin with a total loss of over 57000 km2 of wetland. In western China, over 13000 km2 of wetlands were newly formed in Xinjiang, Tibet, and Qinghai. About 12000 km2 of artificial wetlands were also added for fish farm and reservoir constructions. The newly formed wetlands in western China were caused primarily by climate warming over that region whereas the newly created artificial wetlands were caused by economic developments. China’s wetland loss is caused mainly by human activities. | Peng Gong ZhenGuo Niu Xiao Cheng KuiYi Zhao DeMin Zhou JianHong Guo Lu Liang XiaoFeng Wang DanDan Li HuaBing Huang et al. | 2010 | Science China Earth Sciences2010,53,7: | 28
Additive manufacturing of metals:Microstructure evolution and multistage control显示文摘As a revolutionary industrial technology, additive manufacturing creates objects by adding materials layer by layer and hence can fabricate customized components with an unprecedented degree of freedom. For metallic materials, unique hierarchical microstructures are constructed during additive manufacturing, which endow them with numerous excellent properties. To take full advantage of additive manufacturing,an in-depth understanding of the microstructure evolution mechanism is required. To this end, this review explores the fundamental procedures of additive manufacturing, that is,the formation and binding of melt pools.A comprehensive processing map is proposed that integrates melt pool energy-and geometry-related process parameters together.Based on it,additively manufactured microstructures are developed during and after the solidification of constituent melt pool. The solidification structures are composed of primary columnar grains and fine secondary phases that form along the grain boundaries. The post-solidification structures include submicron scale dislocation cells stemming from internal residual stress and nanoscale precipitates induced by intrinsic heat treatment during cyclic heating of adjacent melt pool.Based on solidification and dislocation theories, the formation mechanisms of the multistage microstructures are thoroughly analyzed, and accordingly, multistage control methods are proposed. In addition, the underlying atomic scale structural features are briefly discussed.Furthermore, microstructure design for additive manufacturing through adjustment of process parameters and alloy composition is addressed to fulfill the great potential of the technique. This review not only builds a solid microstructural framework for metallic materials produced by additive manufacturing but also provides a promising guideline to adjust their mechanical properties. | Zhiyuan Liu Dandan Zhao Pei Wang Ming Yan Can Yang Zhangwei Chen Jian Lu Zhaoping Lu | 2022 | Journal of Materials Science & Technology2022,,5: | 27
Herbal decoctosome is a novel form of medicine显示文摘Traditionally, herbal medicine is consumed by drinking decoctions produced by boiling herbs with water. The functional components of the decoction are heat stable. Small RNAs(sRNAs) were reported as a new class of functional components in decoctions. However, the mechanisms by which sRNAs survive heat treatment of the decoction and enter cells are unclear.Previous studies showed that plant-derived exosome-like nanoparticles(ELNs), which we call botanosomes, could deliver therapeutic reagents in vivo. Here, we report that heat-stable decoctosomes(ELNs) from decoctions have more therapeutic effects than the decoctions in vitro and demonstrate therapeutic efficacy in vivo. Furthermore, sRNAs, such as HJT-sRNA-m7 and PGY-sRNA-6, in the decoctosome exhibit potent anti-fibrosis and anti-inflammatory effects, respectively. Decoctosome is comprised of lipids, chemical compounds, proteins, and s RNAs. A medical decoctosome mimic is called bencaosome. A single lipid sphinganine(d22:0) identified in the decoctosome was mixed and heated with the synthesized sRNAs to form the simplest bencaosome. This simple bencaosome structure was identified by critical micelle concentration(cmc) assay that sRNAs coassembled with sphinganine(d22:0) to form the lipid layers of vesicles. The heating process facilitates co-assembly of sRNAs and sphinganine(d22:0) until a steady state is reached. The artificially produced sphinganine-HJT-sRNA-m7 and sphinganinePGY-sRNA-6 bencaosomes could ameliorate bleomycin-induced lung fibrosis and poly(I:C)-induced lung inflammation, respectively, following oral administration in mice. Our study not only demonstrates that the herbal decoctosome may represent a combinatory remedy in precision medicine but also provides an effective oral delivery route for nucleic acid therapy. | Xiaoyun Li Zhu Liang Jianchao Du Zhiqing Wang Song Mei Zhiqing Li Yan Zhao DANDan Zhao Yiming Ma Jun Ye Jiantao Xu Yu Zhao Jiahui Chang Yuhao Qin Lanlan Yu Chenxuan Wang Chengyu Jiang | 2019 | Science China(Life Sciences)2019,62,3: | 26
Plant-derived phosphocholine facilitates cellular uptake of anti-pulmonary fibrotic HJT-sRNA-m7显示文摘Pulmonary fibrosis, a progressive chronic disease with a high mortality rate, has limited treatment options. Currently, lung transplantation remains the only effective treatment. Here we report that a small RNA, HJT-sRNA-m7, from a Chinese herbal medicine Hong Jing Tian(HJT, RHODIOHAE CRENULATAE RADIX ET RHIZOMA, Rhodiola crenulata) can effectively reduce the expressions of fibrotic hallmark genes and proteins both in alveolar in vitro and in mouse lung tissues in vivo. We also discovered over one hundred oil-soluble chemicals from HJT decoctions, most of which are found in lipid extracts from other Chinese herbals decoctions, including Pu Gong Ying(PGY, TARAXACI HERBA, Taraxacum mongolicum), Chuan Xin Lian(CXL, changed to 'ANDROGRAPHIS HERBA, Andrographis paniculata'), and Jin Yin Hua(JYH, lonicera japonica or Honeysuckle). We identified the active component in these decoctions as two forms of phosphocholines, PC(18:0/18:2) and PC(16:0/18:2). These PCs potentially could form liposomes with small RNAs to enter human alveolar and gastric cells. Our experimental results suggest an unprecendent lipid complex route through which botanic small RNA can enter human bodies. Our results provide an innovative treatment strategy for oral delivery of siRNAs as therapeutic medication. | Jianchao Du Zhu Liang Jiantao Xu Yan Zhao Xiaoyun Li Yanli Zhang DANDan Zhao Ruxuan Chen Yang Liu Trupti Joshi Jiahui Chang Zhiqing Wang Yanxu Zhang Jindong Zhu Qiang Liu Dong Xu Chengyu Jiang | 2019 | Science China(Life Sciences)2019,62,3: | 21

Development and application of mechanized maize harvesters显示文摘Maize harvesting is one of the most important filed operations of maize production. As the accelerating development of maize industry, mechanized maize harvesting is widely accepted and used by farmers in the world.According to the harvesting methods, maize harvesters could be classified into two types, one is maize-for-grain harvesters, including pickers and grain harvesters, the other is whole plant harvesters, including forage harvesters and combined grain-stover harvesters.Structure characteristics, appropriate areas and relative technologies of those harvesters are described in this paper,i.e.,pickers are suitable for multi-crop areas,Grain harvesters are mainly for one-crop areas when grain moisture content is lower than 25%,combined grain-stover harvesters are applicable in agro-pastoral ecotone, and forage harvesters are capable in large-scaled animal husbandry farming areas where large amount of silage are required.Meanwhile, the future development trend of the different harvesters is predicted.Big horsepower engines, enhanced working efficiency, automation and intelligence are required by large scaled farms. As maize breeding development, more varieties that are suitable for grain harvesting are put into practice,so grain combine harvesters will be popular in future. | Yang Li Cui Tao Qu Zhe Li Kehong Yin Xiaowei Han Dandan Yan Bingxin Zhao Dongyue Zhang Dongxing | 2016 | International Journal of Agricultural and Biological Engineering2016,9,3: | 13
Using borehole core analysis to reveal Late Quaternary paleoearthquakes along the Nankou-Sunhe Fault,Beijing显示文摘The Nankou-Sunhe Fault is a buried active normal fault that traverses the urban area of Beijing. Its seismic risks have caused considerable concerns. This paper studies paleoearthquakes along this fault by analyzing and correlating bore-hole cores obtained from triple-tube coring, incorporating experience acquired from trenching. As a result,a model for identifying earthquake-derived colluvium by sediment-core analysis is proposed.Triple-tube coring technique is useful to collect continuous undis-turbed soil core near the Nankou-Sunhe Fault.By identifying fault-scarp colluviums, determining cumulative displacement, and analysing stratum thickening on the hanging wall,we are able to establish a preliminary paleoearthquake sequence consisting of 13 surface-rupturing events since 60 ka.The seismic history can be divided into three periods based on different recurrence intervals.Between 60 and 40 ka,three earthquakes occurred with recurrence interval of ~10 ka.From 40 to 25 ka,there were six earthquakes with the recurrence interval of about 2.5 ka.In the last 25 ka,four earthquakes have taken place with the recurrence interval varying considerably. The recurrence interval between the last three events is ~5 ka.Smaller recurrence intervals correspond to stages of faster fault slip. The coseismic displacement of a single event is 0.8 to 2.2 m,average 1.4 m,largely equivalent to moment magnitudes 6.7-7.1.This study demonstrates the feasibility of bore-hole drilling in investigating paleoearthquakes along normal faults. It also suggests that closely spaced boreholes with continuous undisturbed cores are essential for reconstructing the complete paleoearthquake sequence. | ZHANG ShiMin WANG DanDan LIU XuDong ZHANG GuoHong ZHAO JunXiang LUO MingHui REN JunJie WANG Rui ZHANG YingLi | 2008 | Science China Earth Sciences2008,51,8: | 11
Potassium 2-(l-hydroxypentyl)-benzoate attenuates neuroinflammatory responses and upregulates heme oxygenase-1 in systemic lipopolysaccharide-induced inflammation in mice显示文摘A neuroinflammatory response is commonly involved in the progression of many neurodegenerative diseases. Potassium 2-(1-hydroxypentyl)-benzoate(PHPB), a novel neuroprotective compound, has shown promising effects in the treatment of ischemic stroke and Alzheimer's disease(AD). In the present study, the anti-inflammatory effects of PHPB were investigated in the plasma and brain of C57BL/6 mice administered a single intraperitoneal(i.p.) injection of lipopolysaccharide(LPS). Levels of i NOS and the cytokines TNFα, IL-1β and IL-10 were elevated in plasma, cerebral cortex and hippocampus after LPS injection and the number of microglia and astrocytes in cortex and hippocampus were increased. LPS also upregulated the expression of heme oxygenase-1(HO-1) in the cortex and hippocampus. PHPB reduced the levels of i NOS and cytokines in the plasma and brain, decreased the number of microglia and astrocytes and further enhanced the upregulation of HO-1. In addition, PHPB inhibited the LPS-induced phosphorylation of ERK, P38 and JNK. These results suggest that PHPB is a potential candidate in the treatment of neurodegenerative diseases through inhibiting neuroinflammation. | Chunyang Zhao Weizhen Hou Hui Lei Longjian Huang Shan Wang Dandan Cui Changhong Xing Xiaoliang Wang Ying Peng | 2017 | Acta Pharmaceutica Sinica B2017,7,4: | 9
High-Performance Li-ion Batteries and Super-capacitors Based on Prospective 1-D Nanomaterials显示文摘One-dimensional(1-D) nanomaterials with superior specific capacity, higher rate capability, better cycling peroperties have demonstrated significant advantages for high-performance Li-ion batteries and supercapacitors. This review describes some recent developments on the rechargeable electrodes by using 1-D nanomaterials(such as Li Mn2O4 nanowires, carbon nanofibers, Ni Mo O4 · n H2O nanorods, V2O5 nanoribbons, carbon nanotubes, etc.). New preparation methods and superior electrochemical properties of the 1-D nanomaterials including carbon nanotube(CNT), some oxides, transition metal compounds and polymers, and their composites are emphatically introduced. The VGCF/Li Fe PO4/C triaxial nanowire cathodes for Li-ion battery present a positive cycling performance without any degradation in almost theoretical capacity(160 m Ah/g).The Si nanowire anodes for Li-ion battery show the highest known theoretical charge capacity(4277 m Ah/g),that is about 11 times lager than that of the commercial graphite(372 m Ah/g). The SWCNT/Ni foam electrodes for supercapacitor display small equivalent series resistance(ESR, 52 m?) and impressive high power density(20 k W/kg). The advantages and challenges associated with the application of these materials for energy conversion and storage devices are highlighted. | Dandan Zhao Ying Wang Yafei Zhang | 2011 | Nano-Micro Letters2011,3,1: | 9
The Hippo pathway regulates stem cell proliferation,self-renewal,and differentiation显示文摘Stem cells and progenitor cells are the cells of origin for multi-cellular organisms and organs. They play key roles during development and their dysregulation gives rise to human diseases such as cancer. The recent de-velopment of induced pluripotent stem cell(iPSC)technology which converts somatic cells to stem-like cells holds great promise for regenerative medicine.Nevertheless, the understanding of proliferation,dif-ferentiation,and self-renewal of stem cells and or-gan-specific progenitor cells is far from clear.Recently, the Hippo pathway was demonstrated to play important roles in these processes. The Hippo pathway is a newly established signaling pathway with critical functions in limiting organ size and suppressing tumorigenesis. This pathway was first found to inhibit cell proliferation and promote apoptosis, therefore regulating cell num-ber and organ size in both Drosophila and mammals. However,in several organs, disturbance of the pathway leads to specific expansion of the progenitor cell com-partment and manipulation of the pathway in embryonic stem cells strongly affects their self-renewal and dif-ferentiation.In this review,we summarize current ob-servations on roles of the Hippo pathway in different types of stem cells and discuss how these findings changed our view on the Hippo pathway in organ de-velopment and tumorigenesis. | Huan Liu Dandan Jiang Fangtao Chi Bin Zhao | 2012 | Protein & Cell2012,3,4: | 8
Atomic mobilities and diffusivities in Al alloys显示文摘Knowledge of diffusivity is a prerequisite for understanding many scientific and technological disciplines. In this paper, firstly major experimental methods, which are employed to provide various diffusivity data, are briefly described.Secondly, the fundamentals of various computational methods, including first-principles method, embedded atomic method/molecular dynamic simulation,semi-empirical approaches, and phenomenological DICTRA technique, are demonstrated.Diffusion models recently developed for order/disorder transitions and stoichiometric compounds are also briefly depicted.Thirdly,a newly established diffusivity database for liquid,fcc_A1,L12,bcc_A2,bcc_B2,and intermetallic phases in the multicomponent Al alloys is presented via a few case studies in binary, ternary and quaternary systems. And the integration of various computational techniques and experimental methods is highlighted. The reliability of this diffusivity database is validated by comparing the calculated and measured concentration profiles, diffusion paths, and Kirkendall shifts in various binary, ternary and quaternary diffusion couples.Next, the established diffusivity databases along with thermodynamic and other thermo-physical properties are utilized to simulate the microstructural evolution for Al alloys during solidification, interdiffusion and precipitation.A special discussion is presented on the phase-field simulation of interdiffusion microstructures in a series of Ni-Al diffusion couples composed of γ,γ',and β phases under the effects of both coherent strain and external compressive force.Future orientations in the establishment of next generation of diffusivity database are finally addressed. | DU Yong ZHANG LiJun CUI SenLin ZHAO DongDong LIU DanDan ZHANG WeiBin SUN WeiHua JIE WanQi | 2012 | Science China(Technological Sciences)2012,55,2: | 6

Development of the triazole-fused pyrimidine derivatives as highly potent and reversible inhibitors of histone lysine specific demethylase1(LSD1/KDM1A)显示文摘Histone lysine specific demethylase 1(LSD1) has been recognized as an important modulator in post-translational process in epigenetics. Dysregulation of LSD1 has been implicated in the development of various cancers. Herein, we report the discovery of the hit compound 8 a(IC50=3.93 μmol/L) and further medicinal chemistry efforts, leading to the generation of compound 15 u(IC50=49 nmol/L, and Ki= 16 nmol/L), which inhibited LSD1 reversibly and competitively with H3 K4 me2, and was selective to LSD1 over MAO-A/B. Docking studies were performed to rationalize the potency ofcompound 15 u. Compound 15 u also showed strong antiproliferative activity against four leukemia cell lines(OCL-AML3, K562, THP-1 and U937) as well as the lymphoma cell line Raji with the IC50 values of 1.79, 1.30, 0.45, 1.22 and 1.40 μmol/L, respectively. In THP-1 cell line, 15 u significantly inhibited colony formation and caused remarkable morphological changes. | 15 | Elevated plasma levels of selective cytokines in COVID-19 patients reflect viral load and lung injury显示文摘A recent outbreak of pneumonia in Wuhan,China was found to be caused by a 2019 novel coronavirus(2019-nCoV or SARS-CoV-2 or HCoV-19).We previously reported the clinical features of 12 patients with2019-n Co V infections in Shenzhen,China. To further understand the pathogenesis of COVID-19 and find better ways to monitor and treat the disease caused by 2019-n Co V,we measured the levels of 48 cytokines in the blood plasma of those 12 COVID-19 patients.Thirty-eight out of the 48 measured cytokines in the plasma of 2019-n Co V-infected patients were significantly elevated compared to healthy individuals.Seventeen cytokines were linked to 2019-nCoV loads.Fifteen cytokines, namely M-CSF,IL-10,IFN-α2,IL-17,IL-4,IP-10,IL-7,IL-1 ra,G-CSF,IL-12,IFN-γ,IL-1α,IL-2,HGF and PDGF-BB,were strongly associated with the lung-injury Murray score and could be used to predict the disease severity of2019-n Co V infections by calculating the area under the curve of the receiver-operating characteristics. Our results suggest that 2019-nCoV infections trigger extensive changes in a wide array of cytokines, some of which could be potential biomarkers of disease severity of 2019-n Co V infections. These findings will likely improve our understanding of the immunopathologic mechanisms of this emerging disease. Our results also suggest that modulators of cytokine responses may play a therapeutic role in combating the disease once the functions of these elevated cytokines have been characterized. | Yingxia Liu Cong Zhang Fengming Huang Yang Yang Fuxiang Wang Jing Yuan Zheng Zhang Yuhao Qin Xiaoyun Li Dandan Zhao Shunwang Li Shuguang Tan Zhaoqin Wang Jinxiu Li Chenguang Shen Jianming Li Ling Peng Weibo Wu Mengli Cao Li Xing Zhixiang Xu Li Chen Congzhao Zhou William J Liu Lei Liu Chengyu Jiang | 2020 | National Science Review2020,7,6: | 6
Analysis on Traditional Chinese Medicine syndrome elements and relevant factors for senile diabetes显示文摘OBJECTIVE: To explore the laws governing the distribution ofTraditional Chinese Medicine (TCM) syndrome elements (SEs) of senile diabetes (SD) and their relationship to relevant factors. METHODS: An investigation of patients who met the inclusion criteria was conducted by trained doctors, using case report forms. All related data were collected, including body mass index, glycated hemoglobin, illness course, complications, symptoms, and tongue and pulse manifestation. The SEs of each patient were judged by three qualified associate chief physicians independently. RESULTS: The main SEs of SD are Yin deficiency, Qi deficiency, blood stasis, and phlegm turbidity. Yindeficiency, Qi deficiency, blood stasis, and phlegm turbidity are most commonly seen among 4-SE combinations. Yang deficiency is typically related to illness course and BMI, phlegm turbidity to hypertension and hyperlipidemia, excessive heat to diabetic microangiopathy, and blood stasis to illness course and diabetic macroangiopathy. CONCLUSION: SD pathogenesis has a deficiency in origin and excess in superficiality. Deficiency syndrome mainly manifests as deficiency of both Qi and Yin, and concurrently in Yang deficiency. Excess syndrome is characterized by blood stasis and phlegm turbidity. SEs analysis provides a basis for the prevention and treatment of SD with TCM and lays the foundation for objectively evaluating multicentric clinical research for SD inTCM. | Junping Wei Rui Wu Dandan Zhao | 2013 | Journal of Traditional Chinese Medicine2013,33,4: | 5
A detached petal disc assay and virus-induced gene silencing facilitate the study of Botrytis cinerea resistance in rose flowers显示文摘Fresh-cut roses(Rosa hybrida)are one of the most important ornamental crops worldwide, with annual trade in the billions of dollars.Gray mold disease caused by the pathogen Botrytis cinerea is the most serious fungal threat to cut roses, causing extensive postharvest losses. In this study,we optimized a detached petal disc assay(DPDA)for artificial B.cinerea inoculation and quantification of disease symptoms in rose petals.Furthermore,as the identification of rose genes involved in B.cinerea resistance could provide useful genetic and genomic resources,we devised a virusinduced gene silencing(VIGS)procedure for the functional analysis of B.cinerea resistance genes in rose petals. We used RhPR10.1 as a reporter of silencing efficiency and found that the rose cultivar‘Samantha’showed the greatest decrease in RhPR10.1 expression among the cultivars tested. To determine whether jasmonic acid and ethylene are required for B.cinerea resistance in rose petals,we used VIGS to silence the expression of RhLOX5 and RhEIN3(encoding a jasmonic acid biosynthesis pathway protein and an ethylene regulatory protein, respectively)and found that petal susceptibility to B.cinerea was affected. Finally,a VIGS screen of B.cinerea-induced rose transcription factors demonstrated the potential benefits of this method for the high-throughput identification of gene function in B.cinerea resistance.Collectively, our data show that the combination of the DPDA and VIGS is a reliable and highthroughput method for studying B.cinerea resistance in rose. | Xiaoqian Cao Huijun Yan Xintong Liu Dandan Li Mengjie Sui Jie Wu Hongqiang Yu Zhao Zhang | 2019 | Horticulture Research2019,6,1: | 5

Rational development of a human antibody cocktail that deploys multiple functions to confer Pan-SARS-CoVs protection显示文摘Structural principles underlying the composition and synergistic mechanisms of protective monoclonal antibody cocktails are poorly defined. Here,we exploited antibody cooperativity to develop a therapeutic antibody cocktail against SARS-CoV-2.On the basis of our previously identified humanized cross-neutralizing antibody H014,we systematically an a lyzed a fully human n aive antibody library and rationally identified a potent neutralizing antibody partner,P17,which confers effective protection in animal model.Cryo-EM studies dissected the nature of the P17 epitope, which is SARS-CoV-2 specific and distinctly different from that of HOI4.High-resolution structure of the SARS-CoV-2 spike in complex with HOM and P17,together with functional investigations revealed that in a two-antibody cocktail, synergistic neutralization was achieved by S1 shielding and conformational locking, thereby blocking receptor attachment and viral membrane fusion, conferring high potency as well as robustness against viral mutation escape.Furthermore, cluster analysis identified a hypothetical 3rd antibody partner for further reinforcing the cocktail as pan-SARS-CoVs therapeutics. | Hangping Yao Yao Sun Yong-Qiang Deng Nan Wang Yongcong Tan Na-Na Zhang Xiao-Feng Li Chao Kong Yan-Peng Xuc Qi Chen Tian-Shu Cao Hui Zhao Xintian Yan Lei Cao Zhe Lv Dandan Zhu Rui Feng Nanping Wu Wenhai Zhang Yuhao Hu Keda Chen Rong-Rong Zhang Qingyu Lv Shihui Sun Yunhua Zhou Run Yan Guan Yang Xinglu Chanjuan Liu Xiangyun Lu Linfang Cheng Hongying Qiu Xing-Yao Huang Tianhao Weng Danrong Shi Weidong Jiang Junbin Shao Lei Wang Jie Zhang Tao Jiang Guojun Lang Cheng-Feng Qinc Lanjuan Li Xiangxi Wang | 2021 | Cell Research2021,31,1: | 5
TMEM43-S358L mutation enhances NF-κBTGFp signal cascade in arrhythmogenic right ventricular dysplasia/cardiomyopathy显示文摘Arrhythmogenic right ventricular dysplasia/cardiomyopathy(ARVD/C)is a genetic cardiac muscle disease that accounts for approximately 30%sudden cardiac death in young adults. The Ser358Leu mutation of transmembrane protein 43(TMEM43)was commonly identified in the patients of highly lethal and fully penetrant ARVD subtype,ARVD5.Here,we generated TMEM43 S358L mouse to explore the underlying mechanism. This mouse strain showed the classic patholo.gies of ARVD patients, including structural abnormalities and cardiac fibrofatty.TMEM43 S358L mutation led to hyper-activated nuclear factor kB(NFkB)activation in heart tissues and primary cardiomy.ocyte cells.Importantly, this hyper activation of NF-κB directly drove the expression of pro-fibrotic gene, transforming growth factor beta(TGFβ),and enhanced downstream signal, indicating that TMEM43 S358L mutation up-regulates NF-κB-TGFβ signal cascade during ARVD cardiac fibrosis. Our study partially reveals the regulatory mechanism of ARVD development. | Guoxing Zheng Changying Jiang Yulin Li Dandan Yang Youcai Ma Bing Zhang Xuan Li Pei Zhang Xiaoyu Hu Xueqiang Zhao Jie Du Xin Lin | 2019 | Protein & Cell2019,10,2: | 5
