| 1 | Mathematic principles of interrupted-sampling repeater jamming (ISRJ)显示文摘Coherent jamming is one of the important trends in modern radar electronic war- fare. High-speed sampling of wideband radio frequency (RF) signals and high iso- lation of two receive-transmit antennas are key technologies for the realization of coherent jamming. However, these technologies present significant challenges to engineering application. In this paper, a novel repeater jamming based on inter- rupted sampling technique is presented. For a jammer with a receive-transmit time-sharing antenna, a radar signal is sampled with a low rate by the jammer. Then, a train of false targets will be achieved after the jamming signal feed the matched filter of a pulse compression radar. | 4 | Lx2-32c,a novel semi-synthetic taxane,exerts antitumor activity against prostate cancer cells in vitro and in vivo显示文摘Tubulin has been shown to be an effective target for the development of cytotoxic agents against prostate cancer. Previously, we reported that Lx2-32 c is an anti-tubulin agent with high binding affinity to tubulin. In this study, we investigated the potential of Lx2-32 c to act as an effective cytotoxic agent in the treatment of prostate cancer. MTT assays showed that Lx2-32 c was cytotoxic to all tested prostate cancer cell lines. The Lx2-32c-treated cells typically exhibited a rounded morphology associated with the onset of apoptosis, as evidenced by immunocytochemical staining. Human prostate cancer cell lines treated with Lx2-32 c arrest in the G2/M phase of the cell cycle and the treatment is associated with an increased ratio of cells in the sub-G0/G1 phase as determined by flow cytometry. Furthermore, expression of the cleaved form of poly(ADP-ribose) polymerase in prostate cancer cell lines treated with Lx2-32 c was shown by Western blotting assay. Xenograft implants of LNCa P and PC3-derived tumors in nude mice showed that Lx2-32 c treatment significant inhibited tumor growth with effects equivalent to those of docetaxel. | 6 | Regulation of phenylpropanoid biosynthesis by MdMYB88 and MdMYB124 contributes to pathogen and drought resistance in apple显示文摘MdMYB88 and MdMYB124 have been demonstrated to be responsible for lignin accumulation in apple under drought stress.In this study,using a metabolomic approach,we identified differentially accumulated phenylpropanoid and flavonoid metabolites in MdMYB88/124 transgenic RNAi plants under control and long-term drought stress conditions in apple roots.We confirmed the regulation of phenylalanine by MdMYB88 and MdMYB124 via UPLC-MS in apple roots under both control and drought conditions.Using Electrophoretic Mobility Shift Assay(EMSA)and ChIPquantitative PCR(qPCR)analyses,we found that MdMYB88 positively regulates the MdCM2 gene,which is responsible for phenylalanine biosynthesis,through binding to its promoter region.Under long-term drought conditions,MdMYB88/124 RNAi plants consistently accumulated increased amounts of H2O2 and MDA,while MdMYB88 and MdMYB124 overexpression plants accumulated decreased amounts of H2O2 and MDA.We also examined the accumulation of metabolites in the phenylpropanoid biosynthesis pathway in the leaves of MdMYB88 and MdMYB124 transgenic apple plants after long-term drought stress.We found that metabolites responsible for plant defense,including phenylpropanoids and flavonoids,accumulated less in the RNAi plants but more in the overexpression plants under both control and drought conditions.We further demonstrated that MdMYB88/124 RNAi plants were more sensitive to Alternaria alternata f.sp.mali and Valsa mali,two pathogens that currently severely threaten apple production.In contrast,MdMYB88 and MdMYB124 overexpression plants were more tolerant to these pathogens.The cumulative results of this study provided evidence for secondary metabolite regulation by MdMYB88 and MdMYB124,further explained the molecular roles of MdMYB88 and MdMYB124 in drought resistance,and provided information concerning molecular aspects of their roles in disease resistance. | Dali Geng Xiaoxia Shen Yinpeng Xie Yusen Yang Ruiling Bian Yuqi Gao Pengmin Li Liying Sun Hao Feng Fengwang Ma Qingmei Guan | 2020 | Horticulture Research2020,7,1: | 5 |

| 8 | Omics-based integrated analysis identified ATRX as a biomarker associated with glioma diagnosis and prognosis显示文摘Objective:ATRX is a multifunctional protein that is tightly regulated by and implicated in transcriptional regulation and chromatin remodeling.Numerous studies have shown that genetic alterations in ATRX play a significant role in gliomas.This study aims to further determine the relationship between ATRX and glioma prognosis and identify possible mechanisms for exploring the biological significance of ATRX using large data sets.Methods:We used The Cancer Genome Atlas(TCGA)database and 130 immunohistochemical results to confirm the difference in ATRX mutations in high-and low-grade gliomas.An online analysis of the TCGA glioma datasets using the cBioPortal platform was performed to study the relationship between ATRX mutations and IDH1,TP53,CDKN2 A and CDKN2 B mutations in the corresponding TCGA glioma dataset.In combination with clinical pathology data,the biological significance of the relationships were analyzed.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analyses and annotations of all adjacent genes in the network were performedin the Database for Annotation,Visualization and Integrated Discovery(DAVID)and R language.A protein-protein interaction(PPI)network was constructed,and the interactions of all adjacent nodes were analyzed by the String database and using Cytoscape software.Results:In the selected TCGA glioma datasets,a total of 2,228 patients were queried,21%of whom had ATRX alterations,which co-occurred frequently with TP53 and IDH1 mutations.ATRX alterations are associated with multiple critical molecular events,which results in a significantly improved overall survival(OS)rate.In low-grade gliomas,ATRX mutations are significantly associated with multiple important molecular events,such as ZNF274 and FDXR at mRNA and protein levels.A functional cluster analysis revealed that these genes played a role in chromatin binding and P53,and a link was observed between ATRX and IDH1 and TP53 in the interaction network.ATRX and TP53 are important nodes in the network and have potential links with the blood oxygen imbalance.Conclusions:ATRX mutations have clinical implications for the molecular diagnosis of gliomas and can provide diagnostic and prognostic information for gliomas.ATRX is expected to serve as a new therapeutic target. | Yingbin Xie Yanli Tan Chao Yang Xuehao Zhang Can Xu Xiaoxia Qiao Jianglong Xu Shaohui Tian Chuan Fang Chunsheng Kang | 2019 | Cancer Biology & Medicine2019,16,4: | 5 |

| 14 | Astrocytic effect of low molecular weight heparin-superoxide dismutase conjugate in interleukin-6 overexpressing mice following local cerebral ischemia显示文摘BACKGROUND:Studies have shown that low molecular weight heparin-superoxide dismutase conjugate exhibits a remarkable neuroprotective effect. OBJECTIVE:To investigate the effect of low molecular weight heparin-superoxide dismutase conjugate on astrocytes in an interleukin-6(IL-6) overexpressing mice following local cerebral ischemia. DESIGN,TIME AND SETTING:Randomized,cytological,controlled,animal study was performed in the Department of Physiology and Neuroscience,Neurology and Biochemistry and Molecular Biology,Medical University of South Carolina from January 2005 to March 2005. MATERIALS:Nine IL-6 transgenic mice,irrespective of gender,were randomly divided into three groups:sham-operated,model,and treatment,with three mice in each group.With exception of the sham-operated group,right middle cerebral artery occlusion was induced in the mice. Expression of glial fibrillary acidic protein,an astrocyte marker,was determined by immunohistochemistry.Low molecular weight heparin-superoxide dismutase conjugate was purchased from Biochemistry and Biotechnique Institute,Shandong University. METHODS:Two minutes prior to ischemia induction,0.5 mL/kg saline or 20 000 U/kg low molecular weight heparin-superoxide dismutase conjugate were administrated via the femoral artery in the model group and treatment group,respectively.The sham-operated group underwent the same protocols,with the exception of occlusion and treatment. MAIN OUTCOME MEASURES:The number of glial fibrillary acidic protein-positive cells was quantified under light microscopy(x200). RESULTS:In the sham-operated group,there were a large number of astrocytes in the IL-6 transgenic mice.However,the cell bodies were small,and the branches were few and thin.The number of astrocytes in the model group was remarkably less than the sham-operated group. Compared to the model and sham-operated groups,the number of astrocytes significantly increased,and the cell body became larger,following treatment with low molecular weight heparin-superoxide dismutase conjugate.Astrocytes exhibited hypertrophy and hyperplasia,and the processes became longer and thicker. | 18 | Synthesis and evaluation of ^(64)Cu-radiolabeled NOTA-cetuximab(^(64)Cu-NOTA-C225) for immuno-PET imaging of EGFR expression显示文摘Objective: Epidermal growth factor receptor(EGFR) is overexpressed in a wide variety of solid tumors, serving as a well-characterized target for cancer imaging or therapy. In this study, we aimed to design and synthesize a radiotracer, ^(64) Cu-NOTA-C225, targeting EGFR for tumor positron emission tomography(PET) imaging.Methods: Cetuximab(C225) was conjugated to a bifunctional chelator, p-isothiocyanatobenzyl-1,4,7-triazacyclononane-1,4,7-triacetic acid(NOTA), and further radiolabeled with copper-^(64) for PET imaging. ^(64) CuNOTA-IgG and Cy5.5-C225 were also synthesized as control probes. A431 and A549 mouse models were established for micro-PET and/or near-infrared fluorescence(NIRF) imaging.Results: ^(64) Cu-NOTA-C225 exhibited stability in vivo and in vitro up to 24 h and 50 h post-injection,respectively. A431 tumors with average standard uptake values(SUVs) of 5.61±0.69, 6.68±1.14, 7.80±1.51 at 6, 18 and 36 h post-injection, respectively, which were significantly higher than that of moderate EGFR expressing tumors(A549), with SUVs of 0.89±0.16, 4.70±0.81, 2.01±0.50 at 6, 18 and 36 h post-injection, respectively. The expression levels of A431 and A549 were confirmed by western blotting. Additionally, the tracer uptake in A431 tumors can be blocked by unlabeled cetuximab, suggesting that tracer uptake by tumors was receptor-mediated.Furthermore, NIRF imaging using Cy5.5-C225 showed that the fluorescence intensity in tumors increased with time, with a maximal intensity of 8.17 E+10(p/s/cm^2/sr)/(μW/cm^2) at 48 h post-injection, which is consistent with the paradigm from micro-PET imaging in A431 tumor-bearing mice.Conclusions: The ^(64) Cu-NOTA-C225 PET imaging may be able to specifically and sensitively differentiate tumor models with different EGFR expression levels.
