| 1 | YAP promotes osteogenesis and suppresses adipogenic differentiation by regulatingβ-catenin signaling显示文摘YAP(yes-associated protein) is a transcriptional factor that is negatively regulated by Hippo pathway, a conserved pathway for the development and size control of multiple organs. The exact function of YAP in bone homeostasis remains controversial. Here we provide evidence for YAP's function in promoting osteogenesis, suppressing adipogenesis, and thus maintaining bone homeostasis.YAP is selectively expressed in osteoblast(OB)-lineage cells. Conditionally knocking out Yap in the OB lineage in mice reduces cell proliferation and OB differentiation and increases adipocyte formation, resulting in a trabecular bone loss. Mechanistically, YAP interacts with β-catenin and is necessary for maintenance of nuclear β-catenin level and Wnt/β-catenin signaling. Expression of β-catenin in YAP-deficient BMSCs(bone marrow stromal cells) diminishes the osteogenesis deficit. | 3 | The genome sequence of celery(Apium graveolens L.),an important leaf vegetable crop rich in apigenin in the Apiaceae family显示文摘Celery(Apium graveolens L.)is a vegetable crop in the Apiaceae family that is widely cultivated and consumed because it contains necessary nutrients and multiple biologically active ingredients,such as apigenin and terpenoids.Here,we report the genome sequence of celery based on the use of HiSeq 2000 sequencing technology to obtain 600.8 Gb of data,achieving~189-fold genome coverage,from 68 sequencing libraries with different insert sizes ranging from 180 bp to 10 kb in length.The assembled genome has a total sequence length of 2.21 Gb and consists of 34,277 predicted genes.Repetitive DNA sequences represent 68.88%of the genome sequences,and LTR retrotransposons are the main components of the repetitive sequences.Evolutionary analysis showed that a recent whole-genome duplication event may have occurred in celery,which could have contributed to its large genome size.The genome sequence of celery allowed us to identify agronomically important genes involved in disease resistance,flavonoid biosynthesis,terpenoid metabolism,and other important cellular processes.The comparative analysis of apigenin biosynthesis genes among species might explain the high apigenin content of celery.The whole-genome sequences of celery have been deposited at CeleryDB(http://gffzz7798edb5f31c4ba3hk5q59bnk6v9p665c.ffgz.tsg.suse.edu.cn/celerydb).The availability of the celery genome data advances our knowledge of the genetic evolution of celery and will contribute to further biological research and breeding in celery as well as other Apiaceae plants. | Meng-Yao Li Kai Feng Xi-Lin Hou Qian Jiang Zhi-Sheng Xu Guang-Long Wang Jie-Xia Liu Feng Wang Ai-Sheng Xiong | 2020 | Horticulture Research2020,7,1: | 16 |
| 4 | Immunohistochemical study of hepatic oval cells in human chronic viral hepatitis显示文摘AIM To detect immunohistochemically the presence of oval cells in chronic viral hepatitis with antibody against c-kit.METHODS We detected oval cells in paraffin-embedded liver sections of 3 normal controls and 26 liver samples from patients with chronic viral hepatitis, using immunohistochemistry with antibodies against c-kit, π-class glutathione Stransferase ( Tr-GST ) and cytokeratins 19(CK19).RESULTS Oval cells were not observed in normal livers. In chronic viral hepatitis, hepatic oval cells were located predominantly in the periportal . region and fibrosis septa,characterized by an ovoid nucleus, small size,and scant cytoplasm. Antibody against stem cell factor receptor, c-kit, had higher sensitivity and specificity than π-GST and CK19. | 5 | Coupled dynamic model of state estimation for hypersonic glide vehicle显示文摘Aiming at handling complicated maneuvers or other unpredicted emergencies for hypersonic glide vehicle tracking,three coupled dynamic models of state estimation based on the priori information between guidance variables and aerodynamics are presented. Firstly, the aerodynamic acceleration acting on the target is analyzed to reveal the essence of the target's motion.Then three coupled structures for modeling aerodynamic parameters are developed by different ideas: the spiral model with a harmonic oscillator, the bank model with trigonometric functions of the bank angle and the guide model with the changing rule of guidance variables. Meanwhile, the comparison discussion is concluded to show the novelty and advantage of these models.Finally, a performance assessment in different simulation cases is presented and detailed analysis is revealed. The results show that the proposed models perform excellent properties. | 7 | Diagnostic value of elevated serum carbohydrate antigen 199 level in acute cholangitis secondary to choledocholithiasis显示文摘AIM To investigate the diagnostic value of abnormal serum carbohydrate antigen 199(CA199) level in acute cholangitis secondary to choledocholithiasis.METHODS In this retrospective cohort study, the clinical data of 727 patients with choledocholithiasis admitted to the Third Affiliated Hospital of Zunyi Medical College from June 2011 to June 2017 were collected. Among these patients, 258 patients had secondary acute cholangitis and served as observation group, and the remaining 569 choledocholithiasis patients served as the control group. Serum liver function indexes and tumor markers were detected in both groups, and the receiver operating characteristic(ROC) curves were constructed for markers showing statistical significances. The cutoff value, sensitivity, and specificity of each marker were calculated according to the ROC curves. RESULTS The results of liver function tests showed no significant differences between the two groups(P > 0.05). Tumor markers including serum CA125, CA153, carcinoembryonic antigen, and alpha fetoprotein levels were also not significantly different(P > 0.05); however, the serum CA199 level was significantly higher in the observation group than in the control group(P < 0.05). The ROC curve analysis showed that the area under the curve was 0.885(95%CI: 0.841-0.929) for CA199, and the cutoff value of 52.5 kU/L had the highest diagnostic accuracy, with a sensitivity of 86.8% and a specificity of 81.6%.CONCLUSION Abnormally elevated serum CA199 level has an important value in the diagnosis of acute cholangitis secondary to choledocholithiasis. | 9 | Improving anode performances of lithium-ion capacitors employing carbon-Si composites显示文摘The lithium-ion capacitor is a promising energy storage system with a higher energy density than traditional supercapacitors.However,its cycling and rate performances,which depend on the electrochemical properties of the anode,are still required to be improved.In this work,soft carbon anodes reinforced using carbon-Si composites of various compositions were fabricated to investigate their beneficial influences on the performance of lithium-ion capacitors.The results showed that the specific capacities of the anodes increased significantly by 16.6 mAh g^(-1) with 1.0 wt% carbon-Si composite,while the initial discharge efficiency barely changed.The specific capacity of the anode with a 10.0 wt% carbon-Si composite reached 513.1 mAh g^(-1),and the initial discharge efficiency was 83.79%.Furthermore,the anodes with 7.5 wt% or lower amounts of carbon-Si composite demonstrated reduced charge transfer resistances,which caused an improvement in the rate performance of the lithium-ion capacitors.Moreover,the use of the optimized amount(7.5 wt%) of carbon-Si composite in the anode could significantly improve the cycling performance of the lithium-ion capacitor by compensating the consumption of active lithium.The capacity retention of the lithium-ion capacitor reached 95.14% at 20 C after 10,000 cycles,while the anode potential remained below 0.412 V,which is much lower than that of a soft carbon anode. | Ya-Bin An Si Chen Min-Min Zou Lin-Bin Geng Xian-Zhong Sun Xiong Zhang Kai Wang Yan-Wei Ma | 2019 | Rare Metals2019,38,12: | 10 |
| 10 | Effect of heparin on apoptosis in human nasopharyngeal carcinoma CNE2 cells显示文摘In order to study the mechanism of the effect of heparin on apoptosis in carcinoma cells, the nasopharyngeal carcinoma cell line CNE2 was used to identify the effect of heparin on apoptosis associated with the expression of c-myc, bax, bcl-2 proteins by use of Hoechst 33258 staining, terminal deoxynucleotidyl transferase-mediated dUTP nick-end labeling (TUNEL), agarose gel electrophoresis, and flow cytometry, as well as Western blot analysis. The results showed that heparin induced apoptosis of CNE2 cells including the morphologic changes such as reduction in the volume, and the nuclear chromatin condensation, as well as the “ladder pattern” revealed by agarose gel electrophoresis of DNA in a concentration-dependent manner.The number of TUNEL-positive cells was dramatically increased to 33.6±1.2% from 2.8±0.3% by treat ment with heparin in different concentrations (10~40 kU/L). The apoptotic index was increased to 32.5% from 3.5% by detecting SubG1 peaks on flow cytometry. | 12 | Dual-comb spectroscopy resolved three-degreeof-freedom sensing显示文摘Precise and fast determination of position and orientation, which is normally achieved by distance and angle measurements, has broad applications in academia and industry. We propose a dynamic three-degree-of-freedom measurement technique based on dual-comb interferometry and a self-designed grating-corner-cube(GCC) combined sensor. Benefiting from its unique combination of diffraction and reflection characteristics, the absolute distance, pitch, and yaw of the GCC sensor can be determined simultaneously by resolving the phase spectra of the corresponding diffracted beams. We experimentally demonstrate that the method exhibits a ranging precision(Allan deviation) of 13.7 nm and an angular precision of 0.088 arcsec, alongside a 1 ms reaction time. The proposed technique is capable of precise and fast measurement of distances and two-dimensional angles over long stand-off distances. | 13 | Combination of serum gamma-glutamyltransferase and alkaline phosphatase in predicting the diagnosis of asymptomatic choledocholithiasis secondary to cholecystolithiasis显示文摘BACKGROUND Gamma-glutamyltransferase(GGT) is one of the most important laboratory tests for the evaluation of liver damage. Through a long-term clinical observation of patients with secondary asymptomatic choledocholithiasis, we found that most patients had abnormal GGT serum levels.AIM To investigate the combination of serum GGT and alkaline phosphatase(ALP) in predicting the diagnosis of asymptomatic choledocholithiasis secondary to cholecystolithiasis.METHODS In this retrospective cohort study, the clinical data of 829 patients with cholecystolithiasis admitted to the Third Affiliated Hospital of Zunyi Medical College from August 2014 to August 2017 were collected. Among these patients,151 patients had secondary asymptomatic choledocholithiasis and served as the observation group, and the remaining 678 cholecystolithiasis patients served as the control group. Serum liver function indexes were detected in both groups,and the receiver operating characteristic(commonly known as ROC) curves were constructed for markers showing statistical significances. The cutoff value,sensitivity, and specificity of each marker were calculated according to the ROC curves.RESULTS The overall incidence of asymptomatic choledocholithiasis secondary to cholecystolithiasis was 18.2%. The results of liver function indexes including serum aspartate aminotransferase, alanine aminotransferase, direct bilirubin and total bilirubin levels showed no significant differences between the two groups(P> 0.05). However, the serum GGT and ALP levels were significantly higher in the observation group than in the control group(P < 0.05). The ROC curve analysis showed that the area under the curve was 0.881(95%CI: 0.830-0.932), 0.647(95%CI: 0.583-0.711) and 0.923(0.892-0.953) for GGT, ALP, and GGT + ALP,respectively. The corresponding cut-off values of GGT and ALP were 95.5 U/L and 151.5 U/L, sensitivity were 90.8% and 65.1%, and specificity were 83.6% and59.8%, respectively. The sensitivity and specificity of GGT + ALP were 93.5% and85.1%, respectively.CONCLUSION An abnormally elevated serum GGT level has an important value in the diagnosis of asymptomatic choledocholithiasis secondary to cholecystolithiasis.The combination of serum GGT and ALP has better diagnostic performance. The results indicate that second phase of the as-extruded 7136 aluminum alloy mainly consists of Mg(Zn, Cu, Al)_2and Fe-rich phases. The Mg(Zn, Cu, Al)_2phase directly dissolves into the matrix during solution treatment with various solution temperatures. After solution treated at 475 °C for 1 h,Mg(Zn, Cu, Al)_2phases are dissolved into the matrix,while Fe-rich phases still exist. Fe-rich phases could not dissolve into the matrix by prolonging solution time. The mechanical property test and EBSD observation show that two-stage solution treatment makes no significant improvement in mechanical properties and recrystallization of the alloy. | 17 | Clinical features and the traditional Chinese medicine therapeutic characteristics of 293 COVID-19 inpatient cases显示文摘Coronavirus disease 2019 (COVID-19) is now pandemic worldwide and has heavily overloaded hospitals in Wuhan City, China during the time between late January and February. We reported the clinical features and therapeutic characteristics of moderate COVID-19 cases in Wuhan that were treated via the integration of traditional Chinese medicine (TCM) and Western medicine. We collected electronic medical record (EMR) data, which included the full clinical profiles of patients, from a designated TCM hospital in Wuhan. The structured data of symptoms and drugs from admission notes were obtained through an information extraction process. Other key clinical entities were also confirmed and normalized to obtain information on the diagnosis, clinical treatments, laboratory tests, and outcomes of the patients. A total of 293 COVID-19 inpatient cases, including 207 moderate and 86 (29.3%) severe cases, were included in our research. Among these cases, 238 were discharged, 31 were transferred, and 24 (all severe cases) died in the hospital. Our COVID-19 cases involved elderly patients with advanced ages (57 years on average) and high comorbidity rates (61%). Our results reconfirmed several well-recognized risk factors, such as age, gender (male), and comorbidities, as well as provided novel laboratory indications (e.g., cholesterol) and TCM-specific phenotype markers (e.g., dull tongue) that were relevant to COVID-19 infections and prognosis. In addition to antiviral/antibiotics and standard supportive therapies, TCM herbal prescriptions incorporating 290 distinct herbs were used in 273 (93%) cases. The cases that received TCM treatment had lower death rates than those that did not receive TCM treatment (17/273= 6.2% vs. 7/20= 35%, P = 0.0004 for all cases;17/77= 22% vs. 7/9= 77.7%, P = 0.002 for severe cases). The TCM herbal prescriptions used for the treatment of COVID-19 infections mainly consisted of Pericarpium Citri Reticulatae, Radix Scutellariae, Rhizoma Pinellia, and their combinations, which reflected the practical TCM principles (e.g., clearing heat and dampening phlegm). Lastly, 59% of the patients received treatment, including antiviral, antibiotics, and Chinese patent medicine, before admission. This situation might have some effects on symptoms, such as fever and dry cough. By using EMR data, we described the clinical features and therapeutic characteristics of 293 COVID-19 cases treated via the integration of TCM herbal prescriptions and Western medicine. Clinical manifestations and treatments before admission and in the hospital were investigated. | 18 | Transformation and dissolution of second phases during solution treatment of an Al-Zn-Mg-Cu alloy containing high zinc显示文摘The transformation and dissolution of Mg(Zn,Cu, Al)_2 phase during solution treatment of an Al-Zn-MgCu alloy containing high zinc were investigated by means of optical microscopy(OM), scanning electron microscopy(SEM), energy-dispersive X-ray spectrometry(EDX) and X-ray diffraction(XRD). The results show that solution temperature is the main factor influencing phase dissolution. With solution temperature increasing, the content of residual phases decreases. Phase transformation from Mg(Zn, Cu, Al)2 to S(Al_2 CuMg) occurs under solution temperature of 450,460 and 465 ℃. Mg(Zn,Cu,Al)_2 phase is directly dissolved into the matrix under solution temperature of 470 and 475 ℃,and no S(Al_2 CuMg) phase transformed from Mg(Zn, Cu, Al)_2 phase is observed. The formation of S(Al_2 CuMg) phase is mainly controlled by Zn elemental diffusion. The mechanism of transformation and dissolution of second phases was investigated. At low temperature, the dissolution of Zn is faster than that of Mg and Cu, resulting in an appropriate condition to form S(Al_2 CuMg) phase. | 19 | Aging behavior and fatigue crack propagation of high Zn-containing Al-Zn-Mg-Cu alloys with zinc variation显示文摘In the present work, the influence of two-step aging treatments on hardness, electrical conductivity and mechanical properties of two high Zn-containing Al-Zn-Mg-Cu alloys with zinc content variation was investigated and the detailed T76 aging parameters were proposed. The microstructure of the precipitates were studied by transmission electron microscopy(TEM) and high-resolution transmission electron microscopy(HREM) and then quantitatively investigated with the aid of an image analysis. The fatigue performances were researched by the fatigue crack propagation(FCP) rate test and fracture morphology was observed with scanning electron microscopy(SEM). The results show that the matrix precipitate size distributions of both alloys had significant difference, so as to fatigue crack propagation rates and fracture appearance. The shear and bypass mechanisms of dislocation-precipitate interactions were employed to explain the difference. Among the shearable precipitates, the proportion of larger size precipitates for the higher zinc content alloy is bigger than that for the lower zinc content alloy. The coarse shearable precipitates hinder the propagation of the fatigue cracks, leading to inferior FCP rate. | 20 | Characteristics and outcomes following transcatheter aortic valve replacement in China:a report from China aortic valve transcatheter replacement registry(CARRY)显示文摘Background:The past decade has witnessed an ever-increasing momentum of transcatheter aortic valve replacement(TAVR)and a subsequent paradigm shift in the contemporary management of severe aortic stenosis(AS).We conducted a multi-centric TAVR registry based on Chinese patients(the China Aortic valve tRanscatheter Replacement registrY[CARRY])to delineate the clinical characteristics and outcomes of Chinese patients who underwent TAVR and compare the results between different valve types in different Chinese regions.Methods:CARRY is an all-comer registry of aortic valve disease patients undergoing TAVR across China and was designed as an observational study that retrospectively included all TAVR patients at each participating site.Seven hospitals in China participated in the CARRY,and 1204 patients from April 2012 to November 2020 were included.Categorical variables were compared using the chi-squared test,and continuous variables were analyzed using a t test or analysis of variance(ANOVA)test.The Kaplan–Meier curve was used to estimate the risk of adverse events during follow-up.Results:The mean age of the patients was 73.8±6.5 years and 57.2%were male.The median Society of Thoracic Surgeon-Predicted Risk of Mortality score was 6.0(3.7–8.9).Regarding the aortic valve,the proportion of bicuspid aortic valve(BAV)was 48.5%.During the hospital stay,the stroke rate was 0.7%,and the incidence of high-degree atrioventricular block indicating permanent pacemaker implantation was 11.0%.The in-hospital all-cause mortality rate was 2.2%.After 1 year,the overall mortality rate was 4.5%.Compared to patients with tricuspid aortic valve(TAV),those with BAV had similar in-hospital complication rates,but a lower incidence of in-hospital mortality(1.4%vs.3.3%)and 1 year mortality(2.3%vs.5.8%).Conclusions:TAVR candidates in China were younger,higher proportion of BAV,and had lower rates of post-procedural complications and mortality than other international all-comer registries.Given the use of early generation valves in the majority of the population,patients with BAV had similar rates of complications,but lower mortality than those with TAV.These findings further propel the extension of TAVR in low-risk patients. | Yi-Ming Li Tian-Yuan Xiong Kai Xu Zhen-Fei Fang Lei Jiang Jun Jin Sheng-Hu He Yi-Ning Yang Jing-Jing He Yu-Heng Jia Yi Zhang Yong Peng Yuan Feng Mao Chen | 2021 | Chinese Medical Journal2021,,22: | 6 |
