1Single-cell transcriptome analysis of the novel coronavirus (SARS-CoV-2) associated gene ACE2 expression in normal and non-obstructive azoospermia (NOA) human male testes显示文摘Being infected by SARS-CoV-2 may cause damage to multiple organs in patients, such as the lung, liver and heart. Angiotensin-converting enzyme 2(ACE2), reported as a SARS-CoV-2 receptor, is also expressed in human male testes. This suggests a potential risk in human male reproductive system. However, the characteristics of ACE2-positive cells and the expression of other SARS-CoV-2 process-related genes are still worthy of further investigation. Here, we performed singlecell RNA seq(scRNA-seq) analysis on 853 male embryo primordial germ cells(PGCs) and 2,854 normal testis cells to assess the effects of the SARS-CoV-2 virus on the male reproductive system from embryonic stage to adulthood. We also collected and constructed the scRNA-seq library on 228 Sertoli cells from three non-obstructive azoospermia(NOA) patients to assess the effects at disease state. We found that ACE2 expressing cells existed in almost all testis cell types and Sertoli cells had highest expression level and positive cells ratio. Moreover, ACE2 was also expressed in human male PGCs. In adulthood, the level of ACE2 expression decreased with the increase of age. We also found that ACE2 positive cells had high expressions of stress response and immune activation-related genes. Interestingly, some potential SARS-CoV-2 process-related genes such as TMPRSS2, BSG, CTSL and CTSB had different expression patterns in the same cell type. Furthermore, ACE2 expression level in NOA donors’ Sertoli cells was significantly decreased. 2Xa7, a new executor R gene that confers durable and broad-spectrum resistance to bacterial blight disease in rice显示文摘Bacterial blight(BB)is a globally devastating rice disease caused by Xanthomonas oryzae pv.oryzae(Xoo).The use of disease resistance(R)genes in rice breeding is an effective and economical strategy for the control of this disease.Nevertheless,a majority of R genes lack durable resistance for long-term use under global warming conditions.Here,we report the isolation of a novel executor R gene,Xa7,that confers extremely durable,broad-spectrum,and heat-tolerant resistance to Xoo.The expression of Xa7 was induced by incompatible Xoo strains that secreted the transcription activator-like effector(TALE)AvrXa7 or PthXo3,which recognized effector binding elements(EBEs)in the Xa7 promoter.Furthermore,Xa7 induction was faster and stronger under high temperatures.Overexpression of Xa7 or co-transformation of Xa7 with avrXa7 triggered a hypersensitive response in plants.Constitutive expression of Xa7 activated a defense response in the absence of Xoo but inhibited the growth of transgenic rice plants.In addition,analysis of over 3000 rice varieties showed that the Xa7 locuswas found primarily in the indica and aus subgroups.A variation consisting of an 11-bp insertion and a base substitution(G to T)was found in EBEAvrXa7 in the tested varieties,resulting in a loss of Xa7 BB resistance.Through a decade of effort,we have identified an important BB resistance gene and characterized its distinctive interaction with Xoo strains;these findings will greatly facilitate research on the molecular mechanism of Xa7-mediated resistance and promote the use of this valuable gene in breeding.Xifeng Chen Pengcheng Liu Le Mei Xiaoling He Long Chen Hui Liu Shurong Shen Zhandong Ji Xixi Zheng Yuchen Zhang Zhenyu Gao Dali Zeng Qian Qian Bojun Ma 2021Plant Communications2021,2,3:9
3Cancer-associated fibroblasts in breast cancer:Challenges and opportunities显示文摘The tumor microenvironment is proposed to contribute substantially to the progression of cancers,including breast cancer.Cancer-associated fibroblasts(CAFs)are the most abundant components of the tumor microenvironment.Studies have revealed that CAFs in breast cancer originate from several types of cells and promote breast cancer malignancy by secreting factors,generating exosomes,releasing nutrients,reshaping the extracellular matrix,and suppressing the function of immune cells.CAFs are also becoming therapeutic targets for breast cancer due to their specific distribution in tumors and their unique biomarkers.Agents interrupting the effect of CAFs on surrounding cells have been developed and applied in clinical trials.Here,we reviewed studies examining the heterogeneity of CAFs in breast cancer and expression patterns of CAF markers in different subtypes of breast cancer.We hope that summarizing CAFrelated studies from a historical perspective will help to accelerate the development of CAF-targeted therapeutic strategies for breast cancer.Dengdi Hu Zhaoqing Li Bin Zheng Xixi Lin Yuehong Pan Peirong Gong Wenying Zhuo Yujie Hu Cong Chen Lini Chen Jichun Zhou Linbo Wang 2022Cancer Communications2022,42,5:9
4Recent advances in mammalian reproductive biology显示文摘Reproductive biology is a uniquely important topic since it is about germ cells, which are central for transmitting genetic information from generation to generation. In this review, we discuss recent advances in mammalian germ cell development,including preimplantation development, fetal germ cell development and postnatal development of oocytes and sperm. 5Effect of electroacupuncture on arginine vasopressin-induced endolymphatic hydrops显示文摘OBJECTIVE: To investigate the influence of electroacupuncture(EA) on experimentally induced endolymphatic hydrops(EH) in guinea pigs, and elucidate the association between the dehydrating effect of EA and changes in stria vascularis ultrastructure and expression of vasopressin type 2 receptor(V2R), cyclic adenosine monophosphate(cAMP),and aquaporin 2(AQP2) in the endolymphatic sac(ES).METHODS: The EH model was established by intraperitoneal injection of arginine vasopressin(AVP).As a treatment, EA was delivered to Baihui(GV 20)and Tinggong(SI 19) acupoints, once daily for 10 consecutive days. For histomorphological studies,degree of cochlear hydrops was evaluated by hematoxylin-eosin staining, and the ratio of scala media(SM) area to SM + scala vestibuli area was calculated. In mechanical studies, ultrastructural changes in stria vascularis tissue were examined by transmission electron microscopy. In addition, cAMP levels and mRNA expression levels of V2 R and AQP2 in the ES were compared among groups.RESULTS: EA treatment significantly reduced cochlear hydrops compared with hydropic guinea pigs(P = 0.015). Furthermore, EA attenuated ultrastructural changes in the stria vascularis tissue following EH, significantly upregulated the expression of V2 R(P = 0.016), and attenuated AVP-induced upregulation of both cAMP(P = 0.038) and AQP2 expression(P = 0.017) in the ES.CONCLUSION: Collectively, the results of the present study suggest that the dehydrating effect of EA is associated with improvement of stria vascularis ultrastructure and V2 R-cAMP-AQP2 signaling pathway regulation in the ES.Jiang Liyuan He Jiaojun Chen Xixi Chen Huade 2019Journal of Traditional Chinese Medicine2019,39,2:5
6Subwavelength imaging and detection using adjustable and movable droplet microlenses显示文摘We developed adjustable and movable droplet microlenses consisting of a liquid with a high refractive index.The microlenses were prepared via ultrasonic shaking in deionized water,and the diameter of the microlenses ranged from 1 to 50μm.By stretching the microlenses,the focal length can be adjusted from 13 to 25μm.With the assistance of an optical tweezer,controllable assembly and movement of microlens arrays were also realized.The results showed that an imaging system combined with droplet microlenses could image 80 nm beads under white light illumination.Using the droplet microlenses,fluorescence emission at 550 nm from Cd Se@Zn S quantum dots was efficiently excited and collected.Moreover,Raman scattering signals from a silicon wafer were enhanced by^19 times.The presented droplet microlenses may offer new opportunities for flexible liquid devices in subwavelength imaging and detection.XIXI CHEN TIANLI WU ZHIYONG GONG YUCHAO LI YAO ZHANG BAOJUN LI 2020Photonics Research2020,8,3:5

10Single-layer Rh nanosheets with ultrahigh peroxidase-like activity for colorimetric biosensing显示文摘When the dimensionality of layered compounds decreases to the physical limit, ultimate two-dimensional (2D)anisotropy and/or quantum confinement effects may lead to extraordinary physicochemical attributes.Here,we report single-layer Rh nanosheets (NSs)exhibiting ultrahigh peroxidase-like activity,far exceeding that of horseradish peroxidase (HRP)and of most known layered nanomaterialbased peroxidase mimics.Considering per NS as an active subunit,the Rh NSs displayed a catalytic rate constant (Kcat)as high as 4.45×10^5 s^-1to H2O2,two orders of magnitude higher than those of HRP and Rh nanoparticles.The high atom efficiency of the Rh NSs can be attributed to the full exposure of surface-active Rh atoms,which greatly facilitates electron transfer and formation of superoxide anions,representing reactive oxygen species in the catalytic process.As a proof- of-concept application,the Rh NSs were successfully used as peroxidase mimics for the colorimetric detection of H2O2 and xanthine,with high sensitivity and selectivity.Moreover,a simple,rapid,and sensitive Rh-based paper sensor for ascorbic acid was also developed.In summar^this work provides a novel example of single-layer metallic NSs for biosensing.Shuangfei Cai Wei Xiao Haohong Duan Xixi Liang Chen Wang Rong Yang Yadong Li 2018Nano Research2018,11,12:3

14Quantitative proteomic analysis of S-nitrosated proteins in diabetic mouse liver with ICAT switch method显示文摘In this study we developed a quantitative proteomic method named ICATswitch by introducing isotope-coded affinity tag(ICAT)reagents into the biotin-switch method,and used it to investigate S-nitrosation in the liver of normal control C57BL/6J mice and type 2 diabetic KK-Ay mice.We got fifty-eight S-nitrosated peptides with quantitative information in our research,among which thirty-seven had changed S-nitrosation levels in diabetic mouse liver.The S-nitrosated peptides belonged to fortyeight proteins(twenty-eight were new S-nitrosated proteins),some of which were new targets of S-nitrosation and known to be related with diabetes.S-nitrosation patterns were different between diabetic and normal mice.Gene ontology enrichment results suggested that S-nitrosated proteins are more abundant in amino acid metabolic processes.The network constructed for Snitrosated proteins by text-mining technology provided clues about the relationship between S-nitrosation and type 2 diabetes.Our work provides a new approach for quantifying S-nitrosated proteins and suggests that the integrative functions of S-nitrosation may take part in pathophysiological processes of type 2 diabetes.Xu Zhang Bo Huang Xixi Zhou Chang Chen 2010Protein & Cell2010,1,7:1

16Risk Prevention and Control for Agricultural Non-Point Source Pollution Based on the Process of Pressure-Transformation-Absorption in Chongqing, China显示文摘According to China’s second national survey of pollution sources, the contribution of agricultural non-point sources(ANS) to water pollution is still high. Risk prevention and control are the main means to control costs and improve the efficiency of ANS, but most studies directly take pollution load as the risk standard, leading to a considerable misjudgment of the actual pollution risk. To objectively reflect the risk of agricultural non-point source pollution(ANSP) in Chongqing, China, we investigated the influences of initial source input, intermediate transformation, and terminal absorption of pollutants via literature research and the Delphi method and built a PTA(pressure kinetic energy, transformation kinetic energy, and absorption kinetic energy) model that covers 12 factors, with the support of geographical information system(GIS) technology. The terrain factor calculation results and the calculation results of other factors were optimized by Python tools to reduce human error and workload. Via centroid migration analysis and Kernel density analysis, the risk level, spatial aggregation degree, and key prevention and control regions could be accurately determined. There was a positive correlation between the water quality of the rivers in Chongqing and the risk assessment results of different periods, indirectly reflecting the reliability of the assessment results by the proposed model. There was an obvious tendency for the low-risk regions transforming into high-risk regions. The proportion of high-risk regions and extremely high-risk regions increased from 17.82% and 16.63%in 2000 to 18.10% and 16.76% in 2015, respectively. And the risk level in the main urban areas was significantly higher than that in the southeastern and northeastern areas of Chongqing. The centroids of all grades of risky areas presented a successive distribution from west to east, and the centroids of high-risk and extremely high-risk regions shifted eastward. From 2000 to 2015, the centroids of highrisk and extremely high-risk regions moved 4.63 km(1.68°) and 4.48 km(12.08°) east by north, respectively. The kernel density analysis results showed that the high-risk regions were mainly concentrated in the main urban areas and that the distribution of agglomeration areas overall displayed a transition trend from contiguous distribution to decentralized concentration. The risk levels of the regions with a high proportion of cultivated land and artificial surface were significantly increased, and the occupation of cultivated land in the process of urbanization promoted the movement of the centroids of high-risk and extremely high-risk regions.
