1The YDA-MKK4/MKK5-MPK3/MPK6 Cascade Functions Downstream of the RGF1-RGI Ligand-Receptor Pair in Regulating Mitotic Activity in Root Apical Meristem显示文摘The mitotic activity of root apical meristem(RAM)is critical to primary root growth and development.Previous studies have identified the roles of ROOT GROWTH FACTOR 1(RGF1),a peptide ligand,and its receptors,RGF1 INSENSITIVEs(RGIs),a clade of five leucine-rich-repeat receptor-like kinases,in promoting cell division in the RAM,which determines the primary root length.However,the downstream signaling components remain elusive.In this study,we identify a complete mitogen-activated protein kinase(MAPK or MPK)cascade,composed of YDA,MKK4/MKK5,and MPK3/MPK6,that functions downstream of the RGF1-RGI ligand-receptor pair.Similar to the rgi1/2/3/4/5 quintuple mutant,loss-of-function mutants of MPK3 and MPK6,MKK4 and MKK5,or YDA show a short-root phenotype,which is associated with reduced mitotic activity and lower expression of PLETHORA 1(PLT1)/PLT2 in the RAM.Furthermore,MPK3/MPK6 activation in response to exogenous RGF1 treatment is impaired in the rgi1/2/3/4/5 quintuple,yda single,and mkk4 m kk5 double mutants.Epistatic analyses demonstrated that the expression of constitutively active MKK4,MKK5,or YDA driven by the RGI2 promoter can rescue the short-root phenotype of the rgi1/2/3/4/5 mutant.Taken together,these results suggest that the YDA-MKK4/MKK5-MPK3/MPK6 cascade functions downstream of the RGF1-RGI ligand-receptor pair and upstream of PLT1/PLT2 to modulate the stem cell population and primary root growth in Arabidopsis.Yiming Shao Xinxing Yu Xuwen Xu Yong Li Wenxin Yuan Yan Xu Chuanzao Mao Shuqun Zhang Juan Xu 2020Molecular Plant2020,13,11:6
2Rod-shape inorganic biomimetic mutual-reinforcing MnO2-Au nanozymes for catalysis-enhanced hypoxic tumor therapy显示文摘Biomimetic nanozymes possessing natural enzyme-mimetic activities have been extensively applied in nanocatalytic tumor therapy.However,engineering hybrid biomimetic nanozymes to achieve superior nanozyme activity remained to be an intractable challenge in hypoxic tumors.Herein,a rod-like biomimetic hybrid inorganic MnO2-Au nanozymes are developed,where MnO2 and ultrasmall Au nanoparticles(NPs)are successively deposited on the mesoporous silica nanorod to cooperatively improve the O2 content and thermal sensitivity of hypoxic solid tumors guided by multi-modal imaging.Under the catalyzing of MnO2,the intratumoral H2O2 is decomposed to greatly accelerate O2 generation,which could boost the curative effect of radiation therapy(RT)and further enhance the Au-catalyzed glucose oxidation.Mutually,the Au NPs can steadily and efficiently catalyze the oxidation of glucose in harsh tumor microenvironment,thus sensitizing tumor cells to thermal ablation for mild photothermal therapy and further promoting the catalytic efficiency of MnO2 with the self-supplied H2O2/H+.As a result,this mutual-reinforcing cycle can endow the nanoplatform with accelerated O2 generation,thus alleviating hypoxic environment and further boosting RT effect.Furthermore,acute glucose consuming can induce downregulation expression of heat shock protein(HSP),achieving starvation-promoted mild photothermal therapy.This synthesized hybrid nanozymes proves to be a versatile theranostic agent for nanocatalytic cancer therapy.Lifang Yang Chuchu Ren Min Xu Yilin Song Qianglan Lu Yule Wang Yan Zhu Xinxing Wang Nan Li 2020Nano Research2020,13,8:5

4Short-working-distance optical imaging system and method for surface detection of underwater structures显示文摘In the surface imaging of underwater structures, long working distance will reduce image quality due to the turbidity of water. To acquire high definition and large field of view(FOV) images for surface detection, a short-working-distance underwater imaging system is proposed based on camera array. A multi-view calibration and rectification method is developed. A look-up table(LUT) method and a multi-resolution spline(MRS) method are applied to stitch array images real-time and seamlessly.Experiments both in the air and in the water are conducted. 5High level architecture evolved modular federation object model显示文摘To improve the agility,dynamics,composability,reusability,and development efficiency restricted by monolithic federation object model(FOM),a modular FOM is proposed by high level architecture(HLA)evolved product development group.This paper reviews the state-of-the-art of HLA evolved modular FOM.In particular, related concepts,the overall impact on HLA standards,extension principles,and merging processes are discussed. 6Effect of Dietary Vitamin E on the Sperm Quality of Turbot(Scophthalmus maximus)显示文摘A 3-month feeding experiment was conducted in an in-door seawater system to investigate the effect of dietary vitamin E(Ve) on the sperm quality of turbot(Scophthalmus maximus). D-α-tocopherol acetate was supplemented to the basal(control) diet(65.14 mg kg–1 Ve) to obtain low and high levels of dietary Ve(244.60 mg kg–1, LVe; 721.60 mg kg–1, HVe). Compared with the control, sperm concentration was significantly increased in Ve-supplemented groups(LVe and HVe); while relative sperm volume and testis-somatic index were significantly increased in group HVe only. Sperm motility duration was significantly longer in group HVe than in the control, but no significant difference was observed in percent motility among groups. Sperm size, the uniformity of mitochondrial size, and the integrity of mitochondria cristae and plasma membrane were improved by dietary Ve, especially in HVe. The content of Ve in testis and liver as well as polyunsaturated fatty acids in sperm increased with dietary Ve. 8Classification of facial nerve aberration in congenital malformation of middle ear:Implications for surgery of hearing restoration显示文摘Objectives: Facial nerve aberration is the most troublesome situation in congenital malformations of middle ear.The aim of our study is to investigate its imaging and clinical features as well as relevant choice of surgical techniques for hearing improvement.Methods: A retrospective study involving review of clinical data of 227 patients(256 ears) with congenital middle ear anomaly was undertaken, including preoperative computed tomography(CT) data, surgical records and videos.Results: Aberration involving intratemporal facial nerve was found in 82/256 ears(32.03%) with congenital middle ear anomaly. The most common forms of aberration included overhanging over the oval window(50/82 ears, 60.98%), bifurcation(3/82 ears, 3.66%) and transverse over the promontory(3/82 ears, 3.66%), counting for 68.29%(56/82) of the cases with facial nerve aberration. Concomitant stapes malformation was found in 76/82 ears(92.68%) and atresia or stenosis of the oval window in 27/82 ears(32.93%). In 9/82 ears(10.98%) both stapes and oval window was absent. Elective surgeries for the purpose of hearing improvement included stapodotomy + piston implantation, labyrinthotomy, labyrinthotomy + total ossicular replacement prosthesis(TORP) implantation and Vibrant Soundbridge(VSB) implantation.Conclusion: The majority of facial nerve aberration in congenital malformation of middle ear involves displacement of facial nerve, in addition to concomitant malformations of the stapes and/or oval window, which may influence the choice of surgery for hearing improvement. 10Sporophytic control of anther development and male fertility by glucose-6-phosphate/phosphate translocator 1(OsGPT1) in rice显示文摘Coordination between the sporophytic tissue and the gametic pollen within anthers is tightly controlled to achieve the optimal pollen fitness. Glucose-6-phosphate/phosphate translocator(GPT) transports glucose-6-phosphate, a key precursor of starch and/or fatty acid biosynthesis, into plastids. Here, we report the functional characterization of Os GPT1 in the rice anther development and pollen fertility. Pollen grains from homozygous osgpt1 mutant plants fail to accumulate starch granules, resulting in pollen sterility. Genetic analyses reveal a sporophytic effect for this mutation. Os GPT1 is highly expressed in the tapetal layer of rice anther. Degeneration of the tapetum, an important process to provide cellular contents to support pollen development, is impeded in osgpt1 plants. In addition, defective intine and exine are observed in the pollen from osgpt1 plants. Expression levels of multiple genes that are important to tapetum degeneration or pollen wall formation are significantly decreased in osgpt1 anthers. Previously, we reported that At GPT1 plays a gametic function in the accumulation of lipid bodies in Arabidopsis pollen. This report highlights a sporophytic role of Os GPT1 in the tapetum degeneration and pollen development. 12Identifying the turbulent flow developing inside and around the bottom trawl by Electromagnetic Current Velocity Meter approach in the flume tank显示文摘The Knowledge of turbulent flow developing inside and around the bottom trawl net is of great importance not only for improving the hydrodynamic performance of the gear but also for the selectivity via the fish response,such as the herding response or escape behavior.The 3-D Electromagnetic Current Velocity Meter(ECVM)measurements were performed to investigate the effect of turbulent flow on the bottom trawl net performance and to analyze the turbulence intensity and velocity ratio inside and around different parts of the trawl net.Proper orthogonal decomposition(POD)method was applied in order to extract the phase averaged mean velocity field of turbulent flow from each available ECVM instantaneous velocity.The results demonstrated the existence of turbulence flow,consisting of turbulent boundary layer flow and the turbulence due to the trawl wake developing all inside and around the bottom trawl net.Increasing input streamwise velocity results in faster trawl movement and a significant turbulent flow.The maximum turbulence intensity inside and around trawl wing,square part,first belly,second belly,third belly,cod-end is 0.95%,1.34%,3.40%,4.10%,4.25%and 3.80%,respectively.It was found that the mean velocity field in a turbulent flow inside and around trawl net cod-end recovered on the average was~77.58%of the input streamwise velocity.It is~12.92%,~13.07%,~11.40%,~13.00%and~0.45%less than that inside and around trawl wing,square part,first belly,second belly,and third belly of the bottom trawl net,respectively.The turbulent flow behavior depends strongly on the structure oscillation,input streamwise velocity and,porosity of the net structure.It is necessary to take into account the velocity reduction inside and around a different part of the trawl net to improve the entire drag force determination,cod-end design,and further selectivity control of the fishing gear.Nyatchouba Nsangue Bruno Thierry Hao Tang Liu-xiong Xu Fuxiang Hu Xinxing You Micah Adekunle David Njomoue Pandong Achille 2021Journal of Hydrodynamics2021,33,3:1

15LAMC2 regulates proliferation, migration, and invasion mediated by the Pl3K/AKT/mTOR pathway in oral显示文摘Background:Oral squamous cell carcinoma(OSCC)is a common malignant tumor.Recently,Laminin Gamma 2(LAMC2)has been shown to be abnormally expressed in OSCC;however,how LAMC2 signaling contributes to the occurrence and development of OSCC and the role of autophagy in OSCC has not been fully explored.This study aimed to analyze the role and mechanism of LAMC2 signaling in OSCC and the involvement of autophagy in OSCC.Methods:To explore the mechanism by which LAMC2 is highly expressed in OSCC,we used small interfering RNA(siRNA)to knock down LAMC2 to further observe the changes in the signaling pathway.Furthermore,we used cell proliferation assays,Transwell invasion assays,and wound-healing assays to observe the changes in OSCC proliferation,invasion,and metastasis.RFP-LC3 was used to detect the level of autophagy intensity.A cell line-derived xenograft(CDX)model was used to detect the effect of LAMC2 on tumor growth in vivo.Results:This study found that the level of autophagy was correlated with the biological behavior of OSCC.The downregulation of LAMC2 activated autophagy and inhibited OSCC proliferation,invasion,and metastasis via inhibiting the PI3K/AKT/mTOR pathway.Moreover,autophagy has a dual effect on OSCC,and the synergistic downregulation of LAMC2 and autophagy can inhibit OSCC metastasis,invasion,and proliferation via the PI3K/AKT/mTOR pathway.Conclusions:LAMC2 interacts with autophagy to regulate OSCC metastasis,invasion,and proliferation via the PI3K/AKT/mTOR pathway.LAMC2 down-regulation can synergistically modulate autophagy to inhibit OSCC migration,invasion,and proliferation.FAYU SHAN LANLAN LIANG CHONG FENG HONGBAO XU ZIROU WANG WEILI LIU LINGLING PU ZHAOLI CHEN GANG CHEN XINXING WANG 2023Oncology Research2023,31,4:1
