1Electro-acupuncture regulates the cholinergic anti-inflammatory pathway in a rat model of chronic obstructive pulmonary disease显示文摘Objective: Acupuncture has a definite therapeutic effect on chronic obstructive pulmonary disease(COPD), and the cholinergic anti-inflammatory pathway(CAP) has been shown to be involved in regulation of inflammation. In this study, we investigated whether electro-acupuncture(EA) affects the CAP in COPD.Methods: Sprague-Dawley rats were induced into COPD through exposure to cigarette smoke combined with lipopolysaccharide. EA treatment was applied at Zusanli(ST36) and Feishu(BL13) points for 30 min/d for 7 d. Seventy-two rats were randomly divided into six study groups, including normal, normal + EA,normal + α-bungarotoxin(a-BGT)(the antagonist of the nicotinic acetylcholine receptor a7 subunit(a7 nAChR)) + EA, COPD, COPD + EA, and COPD + α-BGT + EA. Lung function, pathology and vagus nerve discharge were tested. The levels of acetylcholine(ACh), acetylcholinesterase(AChE), interleukin-6(IL-6) and tumor necrosis factor-a(TNF-a) in bronchoalveolar lavage fluid(BALF) and lung tissue were measured by enzyme-linked immunosorbent assay. The mRNA and protein expression and immunoreactivity of a7 nAChR and its postreceptor inflammation signal pathway, including janus kinase 2(JAK2), signal transducers and activators of transcription 3(STAT3), nuclear factor-KB(NF-κB), were observed by quantitative reverse transcription-polymerase chain reaction, Western blot and immunohistochemistry.Results: Compared with normal rats, there were a significant decline in lung function and discharge of the vagus nerve(P < 0.01), a marked sign of lung inflammation and an increase of ACh, AChE, IL-6 and TNF-a level in BALF or lung tissue(P < 0.05, P < 0.01) and higher expression of a7 nAChR, JAK2, STAT3 and NF-κB(P<0.05, P<0.01) in the COPD rats. In rats receiving EA, the lung function and vagal discharge were enhanced(P < 0.01), lung inflammation was improved and the levels of ACh, AChE, IL-6 and TNF-a were decreased(P<0.01). Further, the expression of a7 nAChR, JAK2, STAT3 and NF-κB was downregulated(P < 0.05, P < 0.01). 2Pyroptosis: mechanisms and diseases显示文摘Currently,pyroptosis has received more and more attention because of its association with innate immunity and disease.The research scope of pyroptosis has expanded with the discovery of the gasdermin family.A great deal of evidence shows that pyroptosis can affect the development of tumors.The relationship between pyroptosis and tumors is diverse in different tissues and genetic backgrounds.In this review,we provide basic knowledge of pyroptosis,explain the relationship between pyroptosis and tumors,and focus on the significance of pyroptosis in tumor treatment.In addition,we further summarize the possibility of pyroptosis as a potential tumor treatment strategy and describe the side effects of radiotherapy and chemotherapy caused by pyroptosis.In brief,pyroptosis is a double-edged sword for tumors.The rational use of this dual effect will help us further explore the formation and development of tumors,and provide ideas for patients to develop new drugs based on pyroptosis.Pian Yu Xu Zhang Nian Liu Ling Tang Cong Peng Xiang Chen 2021Signal Transduction and Targeted Therapy2021,6,4:26

4KRAS mutation testing in metastatic colorectal cancer显示文摘The KRAS oncogene is mutated in approximately 35%-45% of colorectal cancers, and KRAS mutational status testing has been highlighted in recent years. The most frequent mutations in this gene, point substitutions in codons 12 and 13, were validated as negative predictors of response to anti-epidermal growth factor receptor antibodies. Therefore, determining the KRAS mutational status of tumor samples has become an essential tool for managing patients with colorectal cancers. Currently, a variety of detection methods have been established to analyze the mutation status in the key regions of the KRAS gene; however, several challenges remain related to standardized and uniform testing, including the selection of tumor samples, tumor sample processing and optimal testing methods. Moreover, new testing strategies, in combination with the mutation analysis of BRAF , PIK3CA and loss of PTEN proposed by many researchers and pathologists, should be promoted. In addition, we recommend that microsatellite instability, a prognostic factor, be added to the abovementioned concomitant analysis. This review provides an overview of KRAS biology and the recent advances in KRAS mutation testing. 5Characteristics and pathological mechanism on magnetic resonance diffusion-weighted imaging after chemoembolization in rabbit liver VX-2 tumor model显示文摘AIM: To investigate dynamic characteristics and pathological mechanism of signal in rabbit VX-2 tumor model on diffusion-weighted imaging (DWI) after chemoembolization. METHODS: Forty New Zealand rabbits were included in the study and forty-seven rabbit VX-2 tumor models were raised by implanting directly and intrahepatically after abdominal cavity opened. Forty VX-2 tumor models from them were divided into four groups. DWI was performed periodically and respectively for each group after chemoembolization. All VX-2 tumor samples of each group were studied by pathology. The distinction of VX-2 tumors on DWI was assessed by their apparent diffusion coefficient (ADC) values. The statistical significance between different time groups, different area groups or different b-value groups was calculated by using SPSS12.0 software. RESULTS: Under b-value of 100 s/mm2, ADC values were lowest at 16 h after chemoembolization in area of VX-2 tumor periphery, central, and normal liver parenchyma around tumor, but turned to increase with further elongation of chemoembolization treatment. The distinction of ADC between different time groups was significant respectively (F = 7.325, P < 0.001; F = 2.496, P < 0.048; F = 6.856, P < 0.001). Cellular edema in the area of VX-2 tumor periphery or normal liver parenchyma around tumor, increased quickly in sixteen h after chemoembolization but, from the 16th h to the 48th h, cellular edema in the area of normal liver parenchyma around tumor decreased gradually and that in the area of VX-2 tumor periphery decreased lightly at, and then increased continually. After chemoembolization, Cellular necrosis in the area of VX-2 tumor periphery was more significantly high than that before chemoembolization. The areas of dead cells in VX-2 tumors manifested low signal and high ADC value, while the areas of viable cells manifested high signal and low ADC value. CONCLUSION: DWI is able to detect and differentiate tumor necrotic areas from viable cellular areas before and after chemoembolization. 6Economic burden and its associated factors of hospitalized patients infected with A (H7N9) virus: a retrospective study in Eastern China, 2013–2014显示文摘Background:H7N9 continues to cause human infections and remains a pandemic concern.Understanding the economic impacts of this novel disease is important for making decisions on health resource allocation,including infectious disease prevention and control investment.However,there are limited data on such impacts.Methods:Hospitalized laboratory-confirmed H7N9 patients or their families in Jiangsu Province of China were interviewed.Patients’direct medical costs of hospitalization were derived from their hospital bills.A generalized linear model was employed to estimate the mean direct medical costs of patients with different characteristics.Results:The mean direct cost of hospitalization for H7N9 was estimated to be¥71060(95%CI,48180–104820),i.e.,US$10996(95%CI,7455–16220),and was¥12060(US$1861),¥136120(US$21001)and¥218610(US$33728)for those who had mild or severe symptoms or who died,respectively.The principal components of the total fees differed among patients with different disease severity,although medication fees were always the largest contributors.Disease severity,proportion of reimbursement and family member monthly average income were identified as the key factors that contributed to a patient’s direct medical cost of hospitalization.Conclusions:The direct medical costs of hospitalized patients with H7N9 are significant,and far surpass the annual per capita income of Jiangsu Province,China.The influencing factors identified should be taken into account when developing related health insurance policies and making health resource allocation.Trial registration:Not applicable.This is a survey study with no health care intervention implemented on human participants.Xiang Huo Li-Ling Chen Lei Hong Lun-Hui Xiang Fen-Yang Tang Shan-Hui Chen Qiang Gao Cong Chen Qi-gang Dai Chuan-Wu Sun Ke Xu Wen-Jun Dai Xian Qi Chang-Cheng Li Hui-Yan Yu Yin Zhou Hao-Di Huang Xing-Yang Pan Chang-sha Xu Ming-Hao Zhou Chang-Jun Bao 2016Infectious Diseases of Poverty2016,5,1:10
7Characteristics of liver on magnetic resonance diffusion-weighted imaging:Dynamic and image pathological investigation in rabbit liver VX-2 tumor model显示文摘AIM: To investigate dynamical and image pathological characteristics of the liver on magnetic resonance (MR) diffusion-weighted imaging (DWI) in the rabbit VX-2 tumor model. METHODS: Forty New Zealand rabbits were included in the study and VX-2 tumor piece was implanted intrahepatically. Fifteen animals received two intrahepatic implantations while 25 had one intrahepatical implantation. DWI, T1- and T2-weighted of magnetic resonance imaging (MRI) were carried out on the 7th and the 14th d after implantation and DWI was conducted, respectively on the 21th d. Ten VX-2 tumor samples were studied pathologically. RESULTS: The rate of lump detected by DWI, T1WI and T2WI was 78.7%, 10.7% and 53.5% (χ2 = 32.61,P < 0.001) on the 7th d after implantation and 95.8%, 54.3% and 82.9% (χ2 = 21.50, P < 0.001) on the 14th d. The signal of most VX-2 tumors on DWI was uniform and it was equal on the map of apparent diffusion coefficient (ADC). The signal of VX tumors did not decrease on the 7th d after implantation, most of them slowly growing during the week following implantation without significant cell dying within the tumor. VX-2 tumors grew increasingly within 14 d after implantation but the signal of most VX-2 tumors on DWI or on the map of ADC was uniform or uneven and ADC of VX tumors decreased obscurely or slightly because tumor necrosis was still not obvious. On the 21th d after implantation, the signal of most VX-2 tumors on DWI or on the map of ADC was uneven because tumor necrosis was evident and ADC of VX-2 tumor necrotic areas decreased. The areas of viable cells in VX-2 tumors manifested a high signal on DWI and a low signal on the map of ADC. The areas of dead cells or necrosis in VX-2 tumors manifested low signals on DWI and low, equal or high signals on the map of ADC but they manifested high signals on DWI and on the map of ADC at the same time when the areas of necrotic tumor became liquefi ed or cystic. The border of tumors on DWI appeared gradually distinct and internal signals of tumor became progressively uneven. 10Curcumenol triggered ferroptosis in lung cancer cells via lncRNA H19/miR-19b-3p/FTH1 axis显示文摘Curcumenol,an effective ingredient of Wenyujin,has been reported that exerted its antitumor potential in a few cancer types.However,the effect and molecular mechanism of curcumenol in lung cancer are largely unknown.Here,we found that curcumenol induced cell death and suppressed cell proliferation in lung cancer cells.Next,we demonstrated that ferroptosis was the predominant method that contributed to curcumenol-induced cell death of lung cancer in vitro and vivo for the first time.Subsequently,using RNA sequencing,we found that the long non-coding RNA H19(lncRNA H19)was significantly downregulated in lung cancer cells treated with curcumenol,when compared to untreated controls.Overexpression of lncRNA H19 eliminated the anticancer effect of curcumenol,while lncRNA H19 knockdown promoted ferroptosis induced by curcumenol treatment.Mechanistically,we showed that lncRNA H19 functioned as a competing endogenous RNA to bind to miR-19b-3p,thereby enhanced the transcription activity of its endogenous target,ferritin heavy chain 1(FTH1),a marker of ferroptosis.In conclusion,our data show that the natural product curcumenol exerted its antitumor effects on lung cancer by triggering ferroptosis,and the lncRNA H19/miR-19b-3p/FTH1 axis plays an essential role in curcumenol-induced ferroptotic cell death.Therefore,our findings will hopefully provide a valuable drug for treating lung cancer patients.Ruonan Zhang Ting Pan Yu Xiang Mingming Zhang Han Xie Zimao Liang Bi Chen Cong Xu Jing Wang Xingxing Huang Qianru Zhu Ziming Zhao Quan Gao Chengyong Wen Wencheng Liu Weirui Ma Jiao Feng Xueni Sun Ting Duan Elaine Lai-Han Leung Tian Xie Qibiao Wu Xinbing Sui 2022Bioactive Materials2022,7,7:8
11Fundamental and Technical Challenges for a Compatible Design Scheme of Oxyfuel Combustion Technology显示文摘Oxyfuel combustion with carbon capture and sequestration(CCS) is a carbon-reduction technology for use in large-scale coal-fired power plants. Significant progress has been achieved in the research and development of this technology during its scaling up from 0.4 MWth to 3 MWth and 35 MWth by the combined efforts of universities and industries in China. A prefeasibility study on a 200 MWe large-scale demonstration has progressed well, and is ready for implementation. The overall research development and demonstration(RD&D) roadmap for oxyfuel combustion in China has become a critical component of the global RD&D roadmap for oxyfuel combustion. An air combustion/oxyfuel combustion compatible design philosophy was developed during the RD&D process. In this paper, we briefly address fundamental research and technology innovation efforts regarding several technical challenges, including combustion stability, heat transfer, system operation, mineral impurities, and corrosion. 14Activation of STAT3 stimulates AHSP expression in K562 cells显示文摘Studies on the chaperone proteinα-hemoglobin stabilizing protein(AHSP)reveal that abundant AHSP in erythroid cells enhance the cells’tolerance to oxidative stress imposed by excessα-hemoglobin in pathological conditions.However,the potential intracellular modulation of AHSP expression itself in response to oxidative stress is still unknown.The present study examined the effect and molecular mechanism of STAT3,an oxidative regulator,on the expression of AHSP.AHSP expression increased in K562 cells upon cytokine IL-6-induced STAT3 activation and decreased in STAT3 knock-down K562 cells.Regulation of AHSP in oxidative circumstance was then examined inα-globin-overloaded K562 cells,and real-time PCR showed strengthened expression of both AHSP and STAT3.ChIP analysis showed binding of STAT3 to AHSP promoter and binding was significantly augmented with IL6 stimulation and uponα-globin overexpression.Dual luciferase reporter assays of the wildtype and mutated SB3 element,an IL-6RE site,in the AHSP promoter in K562 cells highlighted the direct regulatory effect of STAT3 on AHSP gene.Finally,direct binding of STAT3 to SB3 site of AHSP promoter was confirmed with EMSA assays.Our work reveals an adaptive AHSP regulation mediated by the redox-sensitive STAT3 signaling pathway,and provides clues to the therapeutic strategy for AHSP enhancement.CAO Cong ZHAO GuoWei YU Wei XIE XueMin WANG WenTian YANG RuiFeng LV Xiang LIU DePei 2014Science China(Life Sciences)2014,57,5:5

16Aquaporin 5 is degraded by autophagy in diabetic submandibular gland显示文摘Autophagy is a catabolic process which is involved in the development of many diseases including diabetes mellitus and its complications. Hyposalivation is a common complication of diabetes mellitus, whereas its mechanism remains unclear. Here, we observed that the stimulated salivary flow rate of SMG was significantly decreased in db/db mice, a diabetic mice model. The expressions of aquaporin 5(AQP5), a water channel protein, were decreased, whereas the m RNA level of AQP5 was increased in SMGs of both diabetic patients and mice. Under transmission electron microcope, more autophagosomes were detected in diabetic SMGs. Expressions of autophagy related proteins LC3 II, Beclin-1 and ATG5 were increased, meanwhile autophagy substrate p62 was decreased in SMGs of diabetic patients and mice, indicating that autophagy was activated in diabetic SMG.Double immunofluorescence staining showed that the colocalization of AQP5 and LC3 was increased in SMGs of diabetic mice.In cultured SMG-C6 cells, high glucose(HG), but not high osmotic pressure, reduced AQP5 protein expression and induced autophagy. Moreover, inhibition of autophagy by 3-methyladenin, an autophagy inhibitor, or by autophagy-related gene 5 siRNA, decreased HG-induced AQP5 reduction in SMG-C6 cells. Additionally, the expression of p-p85, p-Akt and p-mTOR were decreased in HG-treated SMG-C6 cells. Pretreatment with 740 Y-P, a PI3 K agonist, significantly suppressed HG-induced autophagy and AQP5 degradation. Taken together, these results indicate that autophagy plays a crucial role in AQP5 degradation in diabetic SMG via PI3 K/Akt/mTOR signaling pathway, which contributes to the dysfunction of diabetic SMG. 17Effective and biocompatible antibacterial surfaces via facile synthesis and surface modification of peptide polymers显示文摘It is an urgent need to tackle drug-resistance microbial infections that are associated with implantable biomedical devices.Host defense peptide-mimicking polymers have been actively explored in recent years to fight against drug-resistant microbes.Our recent report on lithium hexamethyldisilazide-initiated superfast polymerization on amino acid N-carboxyanhydrides enables the quick synthesis of host defense peptide-mimicking peptide polymers.Here we reported a facile and cost-effective thermoplastic polyurethane(TPU)surface modification of peptide polymer(DLL:BLG=90:10)using plasma surface activation and substitution reaction between thiol and bromide groups.The peptide polymer-modified TPU surfaces exhibited board-spectrum antibacterial property as well as effective contact-killing ability in vitro.Furthermore,the peptide polymer-modified TPU surfaces showed excellent biocompatibility,displaying no hemolysis and cytotoxicity.In vivo study using methicillin-resistant Staphylococcus aureus(MRSA)for subcutaneous implantation infectious model showed that peptide polymer-modified TPU surfaces revealed obvious suppression of infection and great histocompatibility,compared to bare TPU surfaces.We further explored the antimicrobial mechanism of the peptide polymer-modified TPU surfaces,which revealed a surface contact-killing mechanism by disrupting the bacterial membrane.These results demonstrated great potential of the peptide-modified TPU surfaces for practical application to combat bacterial infections that are associated with implantable materials and devices.Ziyi Lu Yueming Wu Zihao Cong Yuxin Qian Xue Wu Ning Shao Zhongqian Qiao Haodong Zhang Yunrui She Kang Chen Hengxue Xiang Bin Sun Qian Yu Yuan Yuan Haodong Lin Meifang Zhu Runhui Liu 2021Bioactive Materials2021,6,12:5
18Gene expression and MR diffusion-weighted imaging after chemoembolization in rabbit liver VX-2 tumor model显示文摘AIM: To investigate the dynamic characteristics and the correlation between PCNA, Bax, nm23, E-cadherin expression and apparent diffusion coefficient (ADC) on MR diffusion-weighted imaging (DWI) after chemoembolization in rabbit liver VX-2 tumor model. METHODS: Forty New Zealand rabbit liver VX-2 tumor models were included in the study. DWI was carried out periodically after chemoembolization. All VX-2 tumor samples in each group were examined by histopathology and Strept Avidin-Biotin Complex (SABC) immunohistochemical staining. RESULTS: The PCNA expression index in VX-2 tumors was higher than in the normal parenchyma around the tumor (P < 0.001). Nm23, Bax or E-caderin expression index in VX-2 tumors were lower than in the normal parenchyma around the tumor (all P < 0.001). PCNAand nm23 expression in the VX-2 tumor periphery first increased and then decreased (P < 0.001 and P = 0.03, respectively), while the expression of Bax and E-cadherin before and after chemoembolization was insignificant. When b-value was 100 s/mm2, there was a linear correlation between PCNA expression and ADC in the area of VX-2 tumor periphery (P < 0.001), and PCNA expression in VX-2 tumor periphery influenced the ADC. CONCLUSION: The potential of VX-2 tumor infiltrating and metastasizing decreases, while its ability to proliferate increases for a short time after chemoembolization. 19Characterization of particle size distribution of mainstream cigarette smoke generated by smoking machine with an electrical low pressure impactor显示文摘Cigarette smoking is a particle-related exposure. Studying the characteristics of the particle size distribution of cigarette smoke can aid in providing knowledge of smoke aerosol attributes. We used an electrical low pressure impactor(ELPI) to measure the particle size distribution of mainstream cigarette smoke generated by a smoking machine and provided a continuum of particle sizes of cigarette smoke from a whole cigarette. The results showed that the aerodynamic diameters(D, geometric mean of a channel) of particles ranged from 0.021 to 1.956 μm, and the number concentrations were on the order of 105–109cm-3for different sizes of particles. The particle number of the size category below 0.1 μm approximated that of the category 0.1–2.0 μm, and the particles in the size category of 0.1–2.0 μm contributed extremely heavily to total particulate mass.
