1Neutrophil extracellular traps mediate the crosstalk between glioma progression and the tumor microenvironment via the HMGB1/RAGE/IL-8 axis显示文摘Objective:Neutrophil extracellular traps(NETs)produced by tumor-infiltrating neutrophils(TINs)are associated with poor prognosis in patients with several types of cancer.However,the mechanisms underlying the involvement of NETs in glioma progression remain largely unknown.This study aimed to elucidate the roles of NETs in biological processes that drive the crosstalk between glioma progression and the tumor microenvironment.Methods:Neutrophil infiltration and NETs formation were investigated in glioma tissue through immunohistochemistry,and their relationships with clinicopathological features and outcomes were statistically evaluated.The effects of NETs on glioma cell progression were studied in a co-culture system.In vivo and in vitro experiments validated the reactive oxygen species activity and cytokine production of TINs,as well as the ERK signaling pathway activation and the metastasis of gliomas.Results:Neutrophil infiltration and NETs formation were induced in high-grade glioma compared with low-grade glioma.NETs induced by TINs were determined to be an oncogenic marker of high-grade gliomas and to be involved in cell proliferation and invasion.NETs overproduction promoted glioma cell proliferation,migration,and invasion.Furthermore,HMGB1 was found to bind to RAGE and activate the NF-κB signaling pathway in vitro.In addition,NETs stimulated the NF-κB signaling pathway,thus promoting IL-8 secretion in glioblastoma.Subsequently,IL-8 recruited neutrophils which in turn mediated NETs formation via the PI3 K/AKT/ROS axis in TINs.Conclusions:Our results suggest that NETs produced by TINs mediate the crosstalk between glioma progression and the tumor microenvironment by regulating the HMGB1/RAGE/IL-8 axis.Targeting NETs formation or IL-8 secretion may be an effective approach to inhibit glioma progression.Caijun Zha Xiangqi Meng Lulu Li Shan Mi Da Qian Ziwei Li Pengfei Wu Shaoshan Hu Shihong Zhao Jinquan Cai Yanhong Liu 2020Cancer Biology & Medicine2020,17,1:19
2Prognostic value of tertiary lymphoid structure and tumour infiltrating lymphocytes in oral squamous cell carcinoma显示文摘Tertiary lymphoid structures(TLS)are ectopic lymphoid structures in cancers that are largely associated with favourable prognosis.However,the prognostic value of TLSs in oral squamous cell carcinoma(OSCC)is largely unknown,and the association between tumour infiltrating lymphocytes(TILs)and TLSs has been rarely explored in OSCC.In this study,associated markers of TLS,including peripheral node address(PNAd)in high endothelial venules,CD20 in B cells and CD3 in T cells,were examined in 168 OSCC patients,and survival analysis was performed between TLS-positive and TLS-negative cohorts.We detected the presence of TILs by staining CD8+cytotoxic T cells and CD57+NK cells as well.TLSs appeared as highly organized structures in 45(26.8%)cases.TLSpositive patients had a better 5-year overall survival(OS)rate(88.9%vs.56.1%,P<0.001)and relapse-free survival(RFS)rate(88.9%vs.63.4%,P=0.002).Moreover,the presence of TLS was an independent prognostic factor for both the 5-year OS rate(hazard ratio[HR]=3.784;95%confidence interval[CI],1.498–9.562)and RFS rate(HR=3.296;95%CI,1.279–8.490)in multivariate analysis.Furthermore,a higher density of CD8+T cells and CD57+NK cells was found in TLS-positive sections than in TLS-negative counterparts(P<0.001),and their combination provided a higher predictive accuracy(AUC=0.730;95%CI,0.654–0.805).In conclusion,our results suggest that TLS is an independent positive prognostic factor for OSCC patients.These findings provide a theoretical basis for the future diagnostic and therapeutic value of TLSs in OSCC treatment.Qunxing Li Xiangqi Liu Dikan Wang Yanqiong Wang Huanzi Lu Shuqiong Wen Juan Fang Bin Cheng Zhi Wang 2020International Journal of Oral Science2020,12,3:12

4Transcriptomic and genetic approaches reveal an essential role of the NAC transcription factor SlNAP1 in the growth and defense response of tomato显示文摘With global climate change,plants are frequently being exposed to various stresses,such as pathogen attack,drought,and extreme temperatures.Transcription factors(TFs)play crucial roles in numerous plant biological processes;however,the functions of many tomato(Solanum lycopersicum L.)TFs that regulate plant responses to multiple stresses are largely unknown.Here,using an RNA-seq approach,we identified SlNAP1,a NAC TF-encoding gene,which was strongly induced by various stresses.By generating SlNAP1 transgenic lines and evaluating their responses to biotic and abiotic stresses in tomato,we found that SlNAP1-overexpressing plants showed significantly enhanced defense against two widespread bacterial diseases,leaf speck disease,caused by Pseudomonas syringae pv.tomato(Pst)DC3000,and root-borne bacterial wilt disease,caused by Ralstonia solanacearum.In addition,SlNAP1 overexpression dramatically improved drought tolerance in tomato.Although the SlNAP1-overexpressing plants were shorter than the wild-type plants during the early vegetative stage,eventually,their fruit yield increased by 10.7%.Analysis of different hormone contents revealed a reduced level of physiologically active gibberellins(GAs)and an increased level of salicylic acid(SA)and abscisic acid(ABA)in the SlNAP1-overexpressing plants.Moreover,EMSAs and ChIP-qPCR assays showed that SlNAP1 directly activated the transcription of multiple genes involved in GA deactivation and both SA and ABA biosynthesis.Our findings reveal that SlNAP1 is a positive regulator of the tomato defense response against multiple stresses and thus may be a potential breeding target for improving crop yield and stress resistance.Jiao Wang Chenfei Zheng Xiangqi Shao Zhangjian Hu Jianxin Li Ping Wang Anran Wang Jingquan Yu Kai Shi 2020Horticulture Research2020,7,1:9

8Neck observation versus elective neck dissection in management of clinical T1/2N0 oral squamous cell carcinoma: a retrospective study of 232 patients显示文摘Objective: The management of early-stage(cT1/2N0) oral squamous cell carcinoma(OSCC) remains a controversial issue. The aim of this study was to compare the clinical outcomes of neck observation(OBS) and elective neck dissection(END) in treating patients with cT1/2N0 OSCC.Methods: A total of 232 patients with cT1/2N0 OSCC were included in this retrospective study. Of these patients, 181 were treated with END and 51 with OBS. The survival curves of 5-year overall survival(OS), diseasespecific survival(DSS), and recurrence-free survival(RFS) rates were plotted using the Kaplan-Meier method for each group, and compared using the Log-rank test.Results: There was no significant difference in 5-year OS and DSS rates between END and OBS groups(OS:89.0% vs. 88.2%, P=0.906; DSS: 92.3% vs. 92.2%, P=0.998). However, the END group had a higher 5-year RFS rate than the OBS group(90.1% vs. 76.5%, P=0.009). Patients with occult metastases in OBS group(7/51) had similar 5-year OS rate(57.1% vs. 64.1%, P=0.839) and DSS rate(71.4% vs. 74.4%, P=0.982) to those in END group(39/181). In the regional recurrence patients, the 5-year OS rate(57.1% vs. 11.1%, P=0.011) and DSS rate(71.4% vs. 22.2%, P=0.022) in OBS group(7/51) were higher than those in END group(9/181).Conclusions: The results indicated that OBS policy could obtain the same 5-year OS and DSS as END. It has been described as a critical link between feathered dinosaurs and birds. Recent studies, including those of Anchiornis, Xiaotingia, Eosinopteryx and Aurornis, challenged Archaeopteryx as the most basal bird. 11白细胞介素-18抑制Lewis肺癌小鼠移植瘤生长与诱导移植瘤细胞凋亡的研究(英文)显示文摘Objective:The aim of our study was to investigate the effects of interleukin-18(IL-18) on implanted Lewis lung cancer in suppressing tumor growth and inducing tumor cells apoptosis.Methods:One week after hypodermic inoculation of Lewis cells, sixteen tumor-bearing syngeneic mice were randomly divided into two groups.The mice in the treatment group were intraperitoneal injected with the IL-18, in the control group were intraperitoneal injected with normal saline.The health conditions of the mice were measured, the morphology of the tumors was studied and the apoptosis of implanted lung cancer cells was detected by TUNEL to evaluated the effects of IL-18.Results:IL-18 had no significant effects on the health conditions of the mice, but it could suppress the implanted tumor growth(inhibition rate was 75%) and induce tumor cells apoptosis.Conclusion:IL-18 can suppress the implanted Lewis lung cancer cells by way of inducing tumor cells apoptosis.It could be used as a new strategy for lung cancer.Sheng Yang Huishan Lu Xiangqi Chen Tingyan Lin Zhiying Li Mingqiang Kang 2010The Chinese-German Journal of Clinical Oncology2010,9,3:1

16Aldosterone may induce visfatin expression via the GR-ERK1/2 signal pathway in 3T3-L1 adipocytes显示文摘The main function of aldosterone is to regulate blood pressure and electrolytic balance;however,recent studies have also focused on the effect of aldosterone on glucose metabolism especially in adipose tissue.Aldosterone was found to inhibit insulin-induced glucose uptake in 3T3-L1 adipocytes [1] and aldosterone deficiency was found to prevent high-fat-feeding-induced hyperglycemia and adipocyte dysfunction in mice [2].Visfatin,an adipokine discovered by the team of Fukuhara,is mainly released by adipose tissue [3].Treatment of 3T3-L1 adipocytes and L6 myocytes with visfatin increased basal glucose uptake,and glucose release was suppressed in H4IIEC3 hepatocytes after visfatin treatment [3].It has also been suggested that visfatin may act as a proinflammatory cytokine [4] and produce chronic inflammation in adipocytes [5].Eyileten et al.[6] reported that treatment with angiotensin-converting enzyme inhibitor,which can reduce plasma aldosterone concentration,decreased circulating visfatin level in diabetic nephropathy patients.These findings suggest a possible interaction between the renin–angiotensin–aldosterone system(RAAS)and visfatin.Considering the important roles of aldosterone and visfatin and the association between RAAS and visfatin,we hypothesize that there may be a direct regulatory interaction between aldosterone and visfatin.To test this hypothesis,3T3-L1 adipocytes were used as the cell model and the expression of visfatin was measured by qRT-PCR and western blot analysis.Junhua Ma Xuemei Zhao Jichen Zhang Fei Sun Jiancan Lu Hongling Zhu Mingjun Gu Jingnan Wang Xiangqi Li 2019Acta Biochimica et Biophysica Sinica2019,51,3:1
17Collective and Individual Assessment of the Risk of Death from COVID-19 for the Elderly,2020–2022显示文摘Introduction:Coronavirus disease 2019(COVID-19)has had profound disruptions worldwide.For a population or individual,it is critical to assess the risk of death for making preventative decisions.Methods:In this study,clinical data from approximately 100 million cases were statistically analyzed.A software and an online assessment tool were developed in Python to evaluate the risk of mortality.Results:Our analysis revealed that 76.51%of COVID-19-related fatalities occurred among individuals aged over 65 years,with frailty-associated deaths accounting for more than 80%of these cases.Furthermore,over 80%of the reported deaths involved unvaccinated individuals.A notable overlap was observed between aging and frailty-associated deaths,both of which were connected to underlying health conditions.For those with at least two comorbidities,the proportion of frailty and the proportion of COVID-19-related death were both close to 75 percent.Subsequently,we established a formula to calculate the number of deaths,which was validated using data from twenty countries and regions.Using this formula,we developed and verified an intelligent software designed to predict the death risk for a given population.To facilitate rapid risk screening on an individual level,we also introduced a six-question online assessment tool.Conclusions:This study examined the impact of underlying diseases,frailty,age,and vaccination history on COVID-19-related mortality,resulting in a sophisticated software and a user-friendly online scale to assess mortality risk.These tools offer valuable assistance in informed decision-making.Chaobao Zhang Hongzhi Wang Zilu Wen Zhijun Bao Xiangqi Li 2023China CDC weekly2023,5,18:1
