| 2 | Suitable internal control genes for qRT-PCR normalization in cotton fiber development and somatic embryogenesis显示文摘The mechanisms of cotton fiber development and somatic embryogenesis have been explored sys-tematically with microarray and suppression subtractive hybridization. Real-time RT-PCR provides the simultaneous measurement of gene expression in many different samples,with which the data from microarray or others can be confirmed in detail. To achieve accurate and reliable gene expression re-sults,normalization of real-time PCR data against one or several internal control genes is required,which should not fluctuate in different tissues during various stages of development. We assessed the gene expression of 7 frequently used housekeeping genes,including 18S rRNA,Histone3,UBQ7,Actin,Cyclophilin,Gbpolyubiquitin-1 and Gbpolyubiquitin-2,in a diverse set of 21 cotton samples. For fiber developmental series the expression of all housekeeping genes had the same down tendency after 17 DPA. But the expression of the AGP gene(arabinogalactan protein) that has high expression level at the later fiber development stage was up-regulated from 15 to 27 DPA. So the relative absolute quanti-fication should be an efficient and convenient method for the fiber developmental series. The expres-sion of nonfiber tissues series varied not so much against the fiber developmental series. | 4 | Zeolite P synthesis based on fly ash and its removal of Cu(Ⅱ) and Ni(Ⅱ) ions显示文摘Zeolite P was synthesized through hydrothermal method based on a kind of Class C fly ash(FA). X-ray diffraction(XRD), scanning electron microscopy(SEM), and Brunauer–Emmett–Teller(BET) were used to analyze and characterize the synthetic sample. The kinetics and thermodynamics of copper and nickel ions removed by the zeolite samples were experimentally explored in detail. The results of kinetic treatment showed the second-order exchange second-order saturation model(SESSM) can well describe the removal process of copper ions, while the first-order empirical kinetic model(FEKM) is the best kinetic model for nickel ions. Langmuir and Freundlich isotherms were used to fit the equilibrium concentration of Cu(Ⅱ) or Ni(Ⅱ) under certain conditions. | 7 | Regeneration and identification of interspecific asymmetric somatic hybrids obtained by donor-recipient fusion in cotton显示文摘Asymmetric hybrids between Gossypium hirsutum (YZ-1) and G. davidsonii were obtained by donor-recipient fusion. YZ-1 was considered the recipient and was pretreated with iodoacetamide (IOA), while G. davidsonii was considered the donor and was irradiated with ultraviolet (UV) before fusion. YZ-1 protoplasts stopped growth when treated with 0.5 mmol/L IOA for 20 min, and G. davidsonii protoplasts stopped growth when irradiated with 38.7 J/cm2 UV for 30 s. Asymmetric somatic hybrids were obtained by electrofusion between the separately treated protoplasts of the 2 species. The regenerated plants were identified by morphological, cytological, and molecular analysis. Most regenerated plants derived from fused protoplasts displayed new morphology; some were intermediate between the two parents and a few displayed recipient-like morphology. Chromosome numbers in these somatic hybrids mostly ranged from 40 to 73. The hybridity was confirmed by random amplified polymorphic DNA and simple sequence repeat analysis. Organelle DNA inheritance of the YZ-1 and G. davidsonii somatic hybrid was investigated by cleaved amplified polymorphism sequence and chloroplast simple sequence repeat analysis, which indicated that recombination and rearrangements might have occurred in some regions of mitochondrial and chloroplastic DNA. | 8 | Evaluation of channel-like porous-structured titanium in mechanical properties and osseointegration显示文摘The porous titanium with a channel-like pore structure fabricated by infiltration casting followed by selectively dissolving the precursor woven three dimensional(3D)structure technique was comprehensively investigated by means of mechanical tests,in vitro and in vivo evaluation.Such porous structure exhibited superiority in compressive,tensile strength and osseointegration.At 40%porosity,the average compressive and tensile strength reached about 145MPa and 85 MPa,which was superior to that of other porous titanium,e.g.,Selective Laser Melting or powder sintered ones,and was comparable to that of the human cortical bone.Without any bioactive surface treatment,this porous titanium exhibited good cell adhesion,rapid cell proliferation and excellent osseointegration.Based on the study,the 0.4mm pore size resulted in the most rapid cell proliferation and the maximal BV/TV ratio and trabecular bone number of the new bone that ingrew into the porous titanium.To balance the excellent osseointegration and adequate mechanical properties,the optimal structural parameters were 0.4mmpore size with 40%porosity.This porous titanium is very promising for orthopedic applications where compressive and tensile load-bearing is extremely important. | Dong Wang Guo He Ye Tian Ning Ren Jiahua Ni Wei Liu Xianlong Zhang | 2020 | Journal of Materials Science & Technology2020,44,9: | 2 |

| 10 | PM_(2.5)and water-soluble inorganic ion concentrations decreased faster in urban than rural areas in China显示文摘We investigated variations of PM_(2.5)and water-soluble inorganic ions chemical characteristics at nine urban and rural sites in China using ground-based observations.From 2015 to 2019,mean PM_(2.5)concentration across all sites decreased by 41.9μg/m~3with a decline of 46%at urban sites and 28%at rural sites,where secondary inorganic aerosol(SIAs)contributed to 21%(urban sites)and 17%(rural sites)of the decreased PM_(2.5).SIAs concentrations underwent a decline at urban locations,while sulfate(SO_(4)^(2–)),nitrate(NO_(3)^(–)),and ammonium(NH_(4)^(+))decreased by 49.5%,31.3%and 31.6%,respectively.However,only SO_(4)^(2–)decreased at rural sites,NO_(3)^(–)increased by 21%and NH_(4)^(+)decreased slightly.Those changes contributed to an overall SIAs increase in 2019.Higher molar ratios of NO_(3)^(–)to SO_(4)^(2–)and NH_(4)^(+)to SO_(4)^(2–)were observed at urban sites than rural sites,being highest in the heavily polluted days.Mean molar ratios of NH_(3)/NH_xwere higher in 2019 than 2015 at both urban and rural sites,implying increasing NH_xremained as free NH_(3).Our observations indicated a slower transition from sulfate-driven to nitrate-driven aerosol pollution and less efficient control of NO_(x)than SO_(2)related aerosol formation in rural regions than urban regions.Moreover,the common factor at urban and rural sites appears to be a combination of lower SO_(4)^(2–)levels and an increasing fraction of NO_(3)^(–)to PM_(2.5)under NH_(4)^(+)-rich conditions.Our findings imply that synchronous reduction in NO_(x)and NH_(3)emissions especially rural areas would be effective to mitigate NO_(3)^(–)-driven aerosol pollution. | Yangyang Zhang Aohan Tang Chen Wang Xin Ma Yunzhe Li Wen Xu Xiaoping Xia Aihua Zheng Wenqing Li Zengguo Fang Xiufen Zhao Xianlong Peng Yuping Zhang Jian Han Lijuan Zhang Jeffrey L.Collett Jr Xuejun Liu | 2022 | Journal of Environmental Sciences2022,34,12: | 2 |
| 11 | Growth mechanism of primary Ti_(5)Si_(3) phases in special brasses and their effect on wear resistance显示文摘In the present work, in-situ Ti_(5)Si_(3) reinforced special brasses were prepared by melt reaction method. The synthesized Ti_(5)Si_(3) phase shows various morphologies in brasses with different Ti_(5)Si_(3) content, and the3 D morphological evolution of primary Ti_(5)Si_(3)and its growth mechanism were investigated. The Ti_(5)Si_(3) crystal, which bears D8_(8) hexagonal crystal structure, grows along <0001> direction and is revealed by{1010} faces during growth. With the increase of Ti_(5)Si_(3) content in the brasses, the morphology of primary Ti_(5)Si_(3)significantly changes from fibers to hexagonal prisms to short-rods with hollow. In addition,the influence of Ti_(5)Si_(3) volume fraction and morphology on the wear behavior of special brass was also revealed. It was substantiated that the wear resistance increases with the increasing volume fraction of Ti_(5)Si_(3), and the corresponding wear mechanism changes from delamination to slight adhesive wear and abrasive wear. However, the friction coefficient shows an abnormal increase when most of the Ti_(5)Si_(3) containing hollows appears in the brass. | 13 | Effect of cyclic mechanical loading on immunoinflammatory microenvironment in biofabricating hydroxyapatite scaffold for bone regeneration显示文摘It has been proven that the mechanical microenvironment can impact the differentiation of mesenchymal stem cells(MSCs).However,the effect of mechanical stimuli in biofabricating hydroxyapatite scaffolds on the inflammatory response of MSCs remains unclear.This study aimed to investigate the effect of mechanical loading on the inflammatory response of MSCs seeded on scaffolds.Cyclic mechanical loading was applied to biofabricate the cell-scaffold composite for 15 min/day over 7,14,or 21 days.At the predetermined time points,culture supernatant was collected for inflammatory mediator detection,and gene expression was analyzed by qRT-PCR.The results showed that the expression of inflammatory mediators(IL1B and IL8)was downregulated(p<0.05)and the expression of ALP(p<0.01)and COL1A1(p<0.05)was upregulated under mechanical loading.The cell-scaffold composites biofabricated with or without mechanical loading were freeze-dried to prepare extracellular matrix-based scaffolds(ECM-based scaffolds).Murine macrophages were seeded on the ECM-based scaffolds to evaluate their polarization.The ECM-based scaffolds that were biofabricated with mechanical loading before freeze-drying enhanced the expression of M2 polarization-related biomarkers(Arginase 1 and Mrc1,p<0.05)of macrophages in vitro and increased bone volume/total volume ratio in vivo.Overall,these findings demonstrated that mechanical loading could dually modulate the inflammatory responses and osteogenic differentiation of MSCs.Besides,the ECM-based scaffolds that were biofabricated with mechanical loading before freeze-drying facilitated the M2 polarization of macrophages in vitro and bone regeneration in vivo.Mechanical loading may be a promising biofabrication strategy for bone biomaterials. | Penghui Zhang Xizhe Liu Peng Guo Xianlong Li Zhongyuan He Zhen Li Martin J.Stoddart Sibylle Grad Wei Tian Dafu Chen Xuenong Zou Zhiyu Zhou Shaoyu Liu | 2021 | Bioactive Materials2021,6,10: | 2 |
