2Andrographolide sulfonate ameliorates lipopolysaccharide-induced acute lung injury in mice by down-regulating MAPK and NF-κB pathways显示文摘Acute lung injury(ALI) or acute respiratory distress syndrome(ARDS) is a severe, lifethreatening medical condition characterized by widespread inflammation in the lungs, and is a significant source of morbidity and mortality in the patient population. New therapies for the treatment of ALI are desperately needed. In the present study, we examined the effect of andrographolide sulfonate, a water-soluble form of andrographolide(trade name: Xi-Yan-Ping Injection), on lipopolysaccharide(LPS)-induced ALI and inflammation. Andrographolide sulfonate was administered by intraperitoneal injection to mice with LPS-induced ALI. LPS-induced airway inflammatory cell recruitment and lung histological alterations were significantly ameliorated by andrographolide sulfonate. Protein levels of pro-inflammatory cytokines in bronchoalveolar lavage fluid(BALF) and serum were reduced by andrographolide sulfonate administration. m RNA levels of pro-inflammatory cytokines in lung tissue were also suppressed. Moreover, andrographolide sulfonate markedly suppressed the activation of mitogen-activated protein kinase(MAPK) as well as p65 subunit of nuclear factor-κB(NF-κB). In summary, these results suggest that andrographolide sulfonate ameliorated LPS-induced ALI in mice by inhibiting NF-κB and MAPK-mediated inflammatory responses. 4Neuronal apoptosis in cerebral ischemia/reperfusion area following electrical stimulation of fastigial nucleus显示文摘Previous studies have indicated that electrical stimulation of the cerebellar fastigial nucleus in rats may reduce brain infarct size, increase the expression of Ku70 in cerebral ischemia/ reperfusion area, and decrease the number of apoptotic neurons. However, the anti-apoptotic mechanism of Ku70 remains unclear. In this study, fastigial nucleus stimulation was given to rats 24, 48, and 72 hours before cerebral ischemia/reperfusion injury. Results from the electrical stimulation group revealed that rats exhibited a reduction in brain infarct size, a significant increase in the expression of Ku70 in cerebral ischemia/reperfusion regions, and a decreased number of terminal deoxynucleotidyl transferase dUTP nick end labeling(TUNEL)-positive cells. Double immunofluorescence staining revealed no co-localization of Ku70 with TUNEL-positive cells. However, Ku70 partly co-localized with Bax protein in the cytoplasm of rats with cerebral ischemia/reperfusion injury. 6Time-dependent categorization of volatile aroma compound formation in stewed Chinese spicy beef using electron nose profile coupled with thermal desorption GC–MS detection显示文摘In the present study,flavor profiles of Chinese spiced beef in the cooking process were comparatively analyzed by electronic nose,gas chromatography–mass spectrometry(GC–MS)with a thermal desorption system(TDS),and solid-phase microextraction(SPME).A total of 82 volatile compounds were identified,and 3-methyl-butanal,pentanal,hexanal,-xylene,heptanal,limonene,terpinene,octanal,linalool,4-terpinenol,-terpineol,and(E)-anethole were identified as the characteristic flavor compounds in Chinese spiced beef.Variation in the content of volatile components produced by different cooking processes was observed.In general,a cooking time of 4 h resulted in optimal flavor quality and stability.Results indicated that the electronic nose could profile and rapidly distinguish variation among different cooking time.The volatile profiling by TDS-GC–MS and responses from the electronic nose,in combination with multivariate statistical analysis,are a promising tool for control the cooking process of spiced beef.Hui Gong Zhen Yang Meng Liu Zhijia Shi Jiapeng Li Wenhua Chen Xiaoling Qiao 2017Food Science and Human Wellness2017,6,3:21
7CMIP6 Evaluation and Projection of Temperature and Precipitation over China显示文摘This article evaluates the performance of 20 Coupled Model Intercomparison Project phase 6(CMIP6)models in simulating temperature and precipitation over China through comparisons with gridded observation data for the period of 1995–2014,with a focus on spatial patterns and interannual variability.The evaluations show that the CMIP6 models perform well in reproducing the climatological spatial distribution of temperature and precipitation,with better performance for temperature than for precipitation.Their interannual variability can also be reasonably captured by most models,however,poor performance is noted regarding the interannual variability of winter precipitation.Based on the comprehensive performance for the above two factors,the“highest-ranked”models are selected as an ensemble(BMME).The BMME outperforms the ensemble of all models(AMME)in simulating annual and winter temperature and precipitation,particularly for those subregions with complex terrain but it shows little improvement for summer temperature and precipitation.The AMME and BMME projections indicate annual increases for both temperature and precipitation across China by the end of the 21st century,with larger increases under the scenario of the Shared Socioeconomic Pathway 5/Representative Concentration Pathway 8.5(SSP585)than under scenario of the Shared Socioeconomic Pathway 2/Representative Concentration Pathway 4.5(SSP245).The greatest increases of annual temperature are projected for higher latitudes and higher elevations and the largest percentage-based increases in annual precipitation are projected to occur in northern and western China,especially under SSP585.However,the BMME,which generally performs better in these regions,projects lower changes in annual temperature and larger variations in annual precipitation when compared to the AMME projections.Xiaoling YANG Botao ZHOU Ying XU Zhenyu HAN 2021Advances in Atmospheric Sciences2021,38,5:21

9Fault zone structures of northern and southern portions of the main central fault generated by the 2008 Wenchuan earthquake using fault zone trapped waves显示文摘The rupture process of the May 12,2008 MS8.0 Wenchuan earthquake was very complex. To study the rupture zones generated by this earthquake,four dense temporary seismic arrays across the two surface breaking traces of the main-shock were deployed in July and recorded a great amount of aftershocks. This paper focuses on the data interpretation of two arrays across the central main fault,the northern array line 1 and southern array line 3. The fault zone trapped waves recorded by the two arrays were used to study the structure of the central main fault and the difference between the northern and southern portions. The results show that the widths of the rupture zone are about 170?200 m and 200?230 m for northern and southern portions respectively. And the corresponding dip angles are 80° and 70°. The seismic velocity inside the fracture zone is about one half of the host rock. By comparison,the northern portion of the rupture zone is slightly narrower and steeper than the southern portion. Besides these differences,one more interesting and important difference is the positions of the rupture zone with respect to surface breaking traces. At the northern portion,the rupture zone is centered at the surface breaking trace,while at the southern portion it is not but is shifted to the northwest. This difference reflects the difference of rupture behaviors between two portions of the central main fault. The width of the rupture zone is smaller than that of M8.1 Kunlun earthquake though these two earthquakes have almost the same magnitudes. 11Intervention effect and dose-dependent response of Tanreqing injection on airway inflammation in lipopolysaccharide-induced rats显示文摘OBJECTIVE: To assess the effect of Tanreqing injection on airway inflammation in rats. METHODS: A rat model of airway inflammation was generated with lipopolysaccharide (LPS). Tanreqing injection was given by intratracheal instillation, and bronchoalveolar lavage fluid (BALF) from the right lung was collected. BALF total cell and neutrophil counts were then determined. In addition, BALF levels of inflammatory cytokines interleukin-1β, cytokine-induced neutrophil chemoattractant-1, and tumor necrosis factor-α were measured using enzyme linked immunosorbent assay.The middle lobe of the right lung was stained with hematoxylin-eosin and histological changes examined. RESULTS: LPS increased airway inflammation, decreased BALF inflammatory cell count, inflammatory cytokine levels, and suppressed leukocyte influx of the lung. The LPS-induced airway inflammation peaked at 24 h, decreased beginning at 48 h, and had decreased markedly by 96 h. 13Improvement of oxytetracycline production mediated via cooperation of resistance genes in Streptomyces rimosus显示文摘Increasing the self-resistance levels of Streptomyces is an effective strategy to improve the production of antibiotics.To increase the oxytetracycline(OTC) production in Streptomyces rimosus,we investigated the cooperative effect of three co-overexpressing OTC resistance genes:one gene encodes a ribosomal protection protein(otrA) and the other two express efflux proteins(otrB and otrC).Results indicated that combinational overexpression of otrA,otrB,and otrC(MKABC) exerted a synergetic effect.OTC production increased by 179%in the recombinant strain compared with that of the wild-type strain M4018.The resistance level to OTC was increased by approximately two-fold relative to the parental strain,thereby indicating that applying the cooperative effect of self-resistance genes is useful to improve OTC production.Furthermore,the previously identified cluster-situated activator OtcR was overexpressed in MKABC in constructing the recombinant strain MKRABC;such strain can produce OTC of approximately7.49 g L^((-1)),which represents an increase of 19%in comparison with that of the OtcR-overexpressing strain alone.Our work showed that the cooperative overexpression of self-resistance genes is a promising strategy to enhance the antibiotics production in Streptomyces.Shouliang Yin Xuefeng Wang Mingxin Shi Fang Yuan Huizhuan Wang Xiaole Jia Fang Yuan Jinliang Sun Tiejun Liu Keqian Yang Yuxiu Zhang Keqiang Fan Zilong Li 2017Science China(Life Sciences)2017,60,9:10
14Increased serum IL-10 in lupus patients promotes apoptosis of T cell subsets via the caspase 8 pathway initiated by Fas signaling显示文摘We sought to investigate the expression of Fas and FasL on T cell surface and caspase 8 involvement in T cell apoptosis promoted by serum IL-10 in systemic lupus erythematosus(SLE) patients.Cells and sera were obtained from 35 SLE patients.Apoptosis of T cells in patients with SLE was increased and associated with the SLE disease activity index(SLEDAI).Elevated expression of Fas and FasL on T cell surface contributed to increased apoptosis of T cells.Increased IL-10 in the sera of SLE patients was capable of inducing Fas and FasL expression on CD4^+T cell surface,promoting apoptosis of this cell subset.Decreased IL-10 serum levels and low expression of Fas were found in 5 patients of the first follow-up group after 2-month treatment.In another group with one-year treatment,the SLEDAI declined to inactive scores.Serum IL-10 was decreased significantly,and expression of Fas and FasL on T cells was also reduced.Declined apoptosis was predominant only in CD4^+T cell subset.When sera with high level of IL-10 were used to culture PBMCs from healthy controls,activated caspase 8 was elevated in CD3^+T,CD4^+T and CD8^+T cells.The study showed that serum IL-10 induced apoptosis of T cell subsets via the caspase8 pathway initiated by Fas signaling.Increased apoptosis of T cells contributes to autoantigen burden,which is pathogenic in the development of SLE.Xiaofan Yang Bin Sun Huijuan Wang Cheng Yin Xiaole Wang Xiaohui Ji 2015The Journal of Biomedical Research2015,29,3:9
15LONG-TERM CREEP ASSESSMENT OF VISCOELASTIC POLYMER BY TIME-TEMPERATURE-STRESS SUPERPOSITION显示文摘Nonlinear viscoelastic creep properties of poly (methyl methacrylate) at various temperatures and stress levels were measured in short-term tests to check the applicability of time-temperature-stress superposition principle, which is the combined form of time-temperature superposition principle and time-stress superposition principle. A unified master creep compliance curve was constructed from the short-term tests by joint application of time-temperature superposition and time-stress superposition. The unified master curve establishes the creep compliance over two years, which is 4.2 decades longer than the test duration. 16Endovascular therapy for Acute ischemic Stroke Trial (EAST): study protocol for a prospective, multicentre control trial in China显示文摘Background and purpose:5 recent trials have shown the benefit of endovascular treatment for acute ischaemic stroke(AIS)due to large vessel occlusion of the anterior circulation.This study aims to evaluate the safety and efficacy of Solitaire thrombectomy in patients with moderate-to-severe stroke in the Chinese population,which has a high prevalence of intracranial atherosclerosis.Methods and analysis:This multicentre prospective control study will involve 17 stroke centres in China,and plans to recruit 150 patients in the intervention group,and 150 patients in the medical group,in which patients meet enrolment criteria but refuse intervention.Patients with AIS due to large vessel occlusion indicated for treatment with Solitaire stent retriever within 12 hours of symptom onset,and who meet the inclusion and exclusion criteria,will be enrolled in this study.The primary efficacy endpoint is functional independence as defined by a modified Rankin Scale(mRS)score≤2 at 90 days or by functional improvement as defined by mRS,using shift analysis.The procedural efficacy endpoint is arterial recanalisation of the occluded target vessel measured by a modified Thrombolysis in Cerebral Infarction(mTICI)score equal or superior to 2b right following the use of the study device.The primary safety endpoint is symptomatic intracranial haemorrhage(sICH)within 24±3 hours postprocedure.Ethics and dissemination:The protocol was approved by the Ethics Committee at the coordinating centre and by the local Institutional Review Board of each participating centre.Trial registration number:NCT02350283.Zhongrong Miao Xiaochuan Huo Feng Gao Xiaoling Liao Chunjuan Wang Ya Peng Yibin Cao Shengli Chen Meng Zhang Changchun Jiang Xiaoxiang Peng Cunfeng Song Liping Wei Qiyi Zhu Zaiyu Guo Li Liu Hang Lin Hua Yang Wei Wu Hui Liang Anding Xu Kangning Chen Xingquan Zhao Yuesong Pan Hao Li Liping Liu Yilong Wang Yongjun Wang EAST investigators 2016Stroke & Vascular Neurology2016,1,2:8

18New technology to examine and improve the purity of hybrid rice with herbicide resistant gene显示文摘The herbicide resistant bar gene has been widely used as selectable marker genes in the study on plant genetic transformation. Owing to the integration of the gene into rice genome, transgenic rice was resistant to the herbicide Basta. Therefore, selection of transgenic plant and genetic analysis became easier. In the studies, bar gene was introduced as a genetic marker gene into rice restore line variety of two-line or three-line. Combined with conventional breeding method, the good herbicide resistant individual plant was derived as the new restore line for hybrid combination. After sprayed with Basta at seedling phase, the real hybrid rice that had herbicide resistance could grow normally, while the fake hybrid rice and weeds, having no bar gene, were killed by herbicide. 20Respective IL-17A production by γδ T and Th17 cells and its implication in host defense against chlamydial lung infection显示文摘The role of IL-17A is important in protection against lung infection with Chlamydiae,an obligate intracellular bacterial pathogen.In this study,we explored the producers of IL-17A in chlamydial lung infection and specifically tested the role of major IL-17A producers in protective immunity.We found thatγδT cells and Th17 cells are the major producers of IL-17A at the early and later stages of chlamydial infection,respectively.Depletion ofγδT cells in vivo at the early postinfection(p.i.)stage,when mostγδT cells produce IL-17A,failed to alter Th1 responses and bacterial clearance.In contrast,the blockade of IL-17A at the time when IL-17A was mainly produced by Th17(day 7 p.i.)markedly reduced the Th1 response and increased chlamydial growth.The data suggest that theγδT cell is the highest producer of IL-17A in the very early stages of infection,but the protection conferred by IL-17A is mainly mediated by Th17 cells.In addition,we found that depletion ofγδT cells reduced IL-1αproduction by dendritic cells,which was associated with a reduced Th17 response.This finding is helpful to understand the variable role of IL-17A in different infections and to develop preventive and therapeutic approaches against infectious diseases by targeting IL-17A.Hong Bai Xiaoling Gao Lei Zhao Ying Peng Jie Yang Sai Qiao Huili Zhao Shuhe Wang YiJun Fan Antony George Joyee Zhi Yao Xi Yang 2017Cellular & Molecular Immunology2017,14,10:7
