1Introduction of antisense waxy gene into main parent lines of indica hybrid rice显示文摘Amylose content in rice endosperm is one of the key determinants of rice eating and cooking quality, and the poor quality of indica hybrid rice is closely related to the high amylose level in rice grains. In order to improve the grain quality of the indica hybrid rice by genetic engineering, an antisense fragment of rice waxy gene, driven by the 5’-franking sequences of the rice waxy gene, was successfully introduced into three major parent lines of indica hybrid rice, all contain a high amylose level in the grains, via Agrobacte-rium, and more than 100 hygromycin-resistant plants were regenerated. The analysis of PCR amplification and Southern blots indicated that the T-DNA containing the antisense waxy gene had been integrated into the genome of transgenic rice plants. Most of the primary transgenic rice plants grew normally, and the mature seeds from these transgenic plants were performed for analysis of the amylose content. 2A new class of luminescent nanoprobes based on main-group Sb^(3+) emitters显示文摘Inorganic luminescent nanocrystals(NCs)doped with main-group ns^(2)-metal ions have evoked tremendous interest in many technological fields owing to their superior optical properties.Herein,we report a new class of luminescent nanoprobes based on 5s2-metal Sb3+-doped CaS NCs that are excitable by using a near ultraviolet light-emitting diode.The optical properties and excited-state dynamics of Sb3+in CaS NCs are comprehensively surveyed through temperature-dependent steady-state and transient photoluminescence(PL)spectroscopies.Owing to the strong electron-phonon coupling of Sb^(3+)in CaS NCs,Sb3+ions experience a dynamic Jahn-Taller distortion on the excited state,which results in bright green PL of Sb3+with a broad emission band,a large Stokes shift,and a high PL quantum yield up to 17.3%.By taking advantage of the intense PL of Sb^(3+),we show in proof-of-concept experiments the application of biotinylated CaS:Sb^(3+)NCs as sensitive luminescent nanoprobes for biotin receptor-targeted cancer cell imaging and zebrafish imaging with a high imaging contrast.These findings provide fundamental insights into the excited-state dynamics of Sb^(3+)in CaS NCs,thus laying a foundation for future design of novel and versatile luminescent nanoprobes via main-group ns2-metal doping.Chunguang Zhang Meiran Zhang Wei Zheng Jiaojiao Wei Shiteng Wang Ping Huang Xingwen Cheng Tao Dai Zhuo Chen Xueyuan Chen 2022Nano Research2022,15,1:3
3Unusual dinosaur trackway preservation as clues to paleo-landscape and behavior from the Lower Cretaceous Luohe Formation,Shaanxi Province,China显示文摘Poorly preserved tracks have limited ichnotaxonomic or biotaxon utility,but may reveal useful information about the paleoenvironment,behavior and track taphonomy.Eight mostly parallel to sub parallel trackway segments(T1–T8)were registered on a truncation surface in the Lower Cretaceous Luohe Formation of Shaanxi Province.These attest to the passage of several bipeds,probably all theropods,in a paleo-contour-parallel,south-north direction in an arid setting.Quality of preservation in these trackways is poor,but notably superior in two additional trackways(T9–T10)on a foreset surface.Trackway T9 indicates a didactyl trackmaker,probably a deinonychosaurian,heading north to south.This is the 13th report of deinonychosaurian tracks from the Lower Cretaceous of China.If any or all the eight south-north oriented trackway segments represent continuations of other segments in the same trackways,the total number of individual trackmakers heading in this direction may have been as lowas three.Although the trackway pattern and sedimentological evidence could indicate a physically controlled pathway influencing the direction taken by these trackmakers,the possibility that the trackways also represented small social or gregarious group cannot be ruled out.Lida Xing Martin G.Lockley Yongzhong Tang Hendrik Klein W.Scott Persons IV Miaoyan Wang Xingwen Li Hao Wu 2021Geoscience Frontiers2021,12,2:2

6Pseudo-beam method for compressive buckling characteristics analysis of space inflatable load-carrying structures显示文摘This paper extends Le van's work to the case ofnonlinear problem and the complicated configuration. Thewrinkling stress distribution and the pressure effects are alsoincluded in our analysis. Pseudo-beam method is presentedbased on the inflatable beam theory to model the inflatablestructures as a set of inflatable beam elements with a pre-stressedstate. In this method, the discretized nonlinearequations are given based upon the virtual work principlewith a 3-node Timoshenko's beam model. Finite elementsimulation is performed by using a 3-node BEAM 189 elementincorporating ANS YS nonlinear program. The pressureeffect is equivalent included in our method by modifyingbeam element cross-section parameters related to pressure.A benchmark example, the bending case of an inflatable cantileverbeam, is performed to verify the accuracy of our proposedmethod. The comparisons reveal that the numericalresults obtained with our method are close to open publishedanalytical and membrane finite element results. The methodis then used to evaluate the whole buckling and the load-carryingcharacteristics of an inflatable support frame subjectedto a compression force. The wrinkling stress and regioncharacteristics are also shown in the end. This method givesbetter convergence characteristics, and requires much lesscomputation time. 15Pangolin HKU4-related coronaviruses found in greater bamboo bats from southern China显示文摘Coronavirus(CoV)spillover originating from game animals,particularly pangolins,is currently a significant concern.Meanwhile,vigilance is urgently needed for coronaviruses carried by bats,which are known as natural reservoirs of many coronaviruses.In this study,we collected 729 anal swabs of 20 different bat species from nine locations in Yunnan and Guangdong provinces,southern China,in 2016 and 2017,and described the molecular characteristics and genetic diversity of alphacoronaviruses(αCoVs)and betacoronaviruses(βCoVs)found in these bats.Using RT-PCR,we identified 58(8.0%)bat CoVs in nine bat species from six locations.Furthermore,using the Illumina platform,we obtained two representative full-length genomes of the bat CoVs,namely TyRo-CoV-162275 and TyRo-CoV-162269.Sequence analysis showed that TyRo-CoV-162275 shared the highest identity with Malayan pangolin(Manis javanica)HKU4-related coronaviruses(MjHKU4r-CoVs)from Guangxi Province,whereas TyRo-CoV-162269 was closely related to HKU33-CoV discovered in a greater bamboo bat(Tylonycteris robustula)from Guizhou Province.Notably,TyRo-CoV-162275 has a putative furin protease cleavage site in its S protein and is likely to utilize human dipeptidyl peptidase-4(hDPP4)as a cell-entry receptor,similar to MERSCoV.To the best of our knowledge,this is the first report of a bat HKU4r-CoV strain containing a furin protease cleavage site.These findings expand our understanding of coronavirus geographic and host distributions.Min Guo Kai Zhao Xingwen Peng Xiangyang He Jin Deng Bo Wang Xinglou Yang Libiao Zhang 2023Virologica Sinica2023,38,6:0
16Reinforcement Effect Evaluation on Dynamic Characteristics of an Arch Bridge Based on Vehicle-Bridge Coupled Vibration Analysis显示文摘To numerically evaluate the reinforcement effect on dynamic characteristics of a concrete-filled steel tube arch bridge with vibration problems,a 12-degree-of-freedom sprung-mass dynamic vehicle model and a 3D finite element bridge model were established.Then,the coupled equations of vehicle-bridge interaction were derived and a computer program was developed using the FORTRAN language.This program can accurately simulate vehicle-bridge coupled vibration considering the bumping effect and road surface irregularity during motion of the vehicle.The simulated results were compared with those of relevant literatures to verify the correctness of the self-developed program.Then,three reinforcement schemes for the bridge(Addition of longitudinal beams,Reinforcement of bridge decks,and Replacement of suspenders)were proposed and numerically simulated,and the vibration reduction effects of the three schemes were evaluated based on the numerical results to find effective ones.It is confirmed that the reinforcement scheme of Addition of longitudinal beams shows the most significant vibration reduction effect.It is recommended in the engineering practice that the combination of the reinforcement schemes of Addition of longitudinal beams and Replacement of bridge deck can be used to solve the excessive vibration problem.Yanbin Tan Xingwen He Lei Shi Shi Zheng Zhe Zhang Xinshan Wang 2022Computer Modeling in Engineering & Sciences2022,,5:0
17Promoter variations in DBR2-like affect artemisinin production in different chemotypes of Artemisia annua显示文摘Artemisia annua is the only known plant source of the potent antimalarial artemisinin,which occurs as the low-and high-artemisinin producing(LAP and HAP)chemotypes.Nevertheless,the different mechanisms of artemisinin producing between these two chemotypes were still not fully understood.Here,we performed a comprehensive analysis of genome resequencing,metabolome,and transcriptome data to systematically compare the difference in the LAP chemotype JL and HAP chemotype HAN.Metabolites analysis revealed that 72.18%of sesquiterpenes was highly accumulated in HAN compared to JL.Integrated omics analysis found a DBR2-Like(DBR2L)gene may be involved in artemisinin biosynthesis.DBR2L was highly homologous with DBR2,belonged to ORR3 family,and had the DBR2 activity of catalyzing artemisinic aldehyde to dihydroartemisinic aldehyde.Genome resequencing and promoter cloning revealed that complicated variations existed in DBR2L promoters among different varieties of A.annua and were clustered into three variation types.The promoter activity of diverse variant types showed obvious differences.Furthermore,the core region(-625 to 0)of the DBR2L promoter was identified and candidate transcription factors involved in DBR2L regulation were screened.Thus,the result indicates that DBR2L is another key enzyme involved in artemisinin biosynthesis.The promoter variation in DBR2L affects its expression level,and thereby may result in the different yield of artemisinin in varieties of A.annua.It provides a novel insight into the mechanism of artemisinin-producing difference in LAP and HAP chemotypes of A.annua,and will assist in a high yield of artemisinin in A.annua.Xingwen Wang Lan Wu Li Xiang Ranran Gao Qinggang Yin Mengyue Wang Zhaoyu Liu Liang Leng Yanyan Su Huihua Wan Tingyu Ma Shilin Chen Yuhua Shi 2023Horticulture Research2023,10,9:0
18Experimental Investigation of Folding Damage for the Rigidizable Carbon-Epoxy Composites显示文摘This paper focused on the folding damage behavior of the space rigidizable materials in terms of 3-layer composite membranes. An experimental scheme was presented. The composite membranes were folded between the two plates for a short time, and then the unfolded composite membranes were compressively cured in an oven. By adjusting the displacement of one plate, the folding radius was changed. As expected, the strength and effective modulus of the cured composite membranes drop with decreasing the folding radius. When the strain controlled failure rule is applied to the composite membranes, a minimal folding radius can be reached, beyond which the membranes will keep intact.Furthermore, folding damage due to folding and unfolding processes was evaluated by a simplified model. Compared with the measured residual strength and effective modulus, calculated results have the same trend. 20lncRNA HITT Inhibits Lactate Production by Repressing PKM2 Oligomerization to Reduce Tumor Growth and Macrophage Polarization显示文摘Lactic acid acidifies the tumor microenvironment and promotes multiple critical oncogenic processes,including immune evasion.Pyruvate kinase M2(PKM2)is a dominant form of pyruvate kinase(PK)expressed in cancers that plays essential roles in metabolic reprograming and lactate production,rendering it as an attractive therapeutic target of cancer.However,the mechanism underlying PKM2 regulation remains unclear.Here,we show that long noncoding RNA(lncRNA)HIF-1αinhibitor at transcription level(HITT)inhibits lactate production in a PKM2-dependent manner.Mechanistically,it physically interacts with PKM2 mapped to a region that has been involved in both dimer(less-active)and tetramer(more-active)formation,inhibiting PKM2 oligomerization and leading to dramatic reduction of PK activity.Under glucose starvation,HITT was reduced as a result of miR-106 induction,which subsequently facilitates PKM2 oligomerization and increases vulnerability to apoptosis under glucose starvation stress.In addition,the interaction also reduces lactate secretion from cancer cells,which subsequently polarizes macrophages toward an M2-like anti-inflammatory phenotype and thus possibly contributes to immune escape in vivo.This study highlights an important role of an lncRNA in regulating PKM2 activity and also reveals a metabolic regulatory effect of PKM2 on macrophage polarization.Kunming Zhao Xingwen Wang Dong Zhao Qingyu Lin Yi Zhang Ying Hu 2022Research2022,,3:0
