1Haze insights and mitigation in China: An overview显示文摘The present article provides an overview of the chemical and physical features of haze in China, focusing on the relationship between haze and atmospheric fine particles, and the formation mechanism of haze. It also summarizes several of control technologies and strategies to mitigate the occurrence of haze. The development of instruments and the analysis of measurements of ambient particles and precursor concentrations have provided important information about haze formation. Indeed, the use of new instruments has greatly facilitated current haze research in China. Examples of insightful results include the relationship between fine particles and haze, the chemical compositions and sources of particles, the impacts of the aging process on haze formation, and the application of technologies that control the formation of haze. 2High‑Performance Flexible Microneedle Array as a Low‑Impedance Surface Biopotential Dry Electrode for Wearable Electrophysiological Recording and Polysomnography显示文摘Microneedle array(MNA)electrodes are an effective solution to achieve high-quality surface biopotential recording without the coordination of conductive gel and are thus very suitable for long-term wearable applications.Existing schemes are limited by flexibility,biosafety,and manufacturing costs,which create large barriers for wider applications.Here,we present a novel flexible MNA electrode that can simultaneously achieve flexibility of the substrate to fit a curved body surface,robustness of microneedles to penetrate the skin without fracture,and a simplified process to allow mass production.The compatibility with wearable wireless systems and the short preparation time of the electrodes significantly improves the comfort and convenience of electrophysiological recording.The normalized electrode–skin contact impedance reaches 0.98 kΩcm^(2)at 1 kHz and 1.50 kΩcm^(2)at 10 Hz,a record low value compared to previous reports and approximately 1/250 of the standard electrodes.The morphology,biosafety,and electrical/mechanical properties are fully characterized,and wearable recordings with a high signal-to-noise ratio and low motion artifacts are realized.The first reported clinical study of microneedle electrodes for surface electrophysiological monitoring was conducted in tens of healthy and sleep-disordered subjects with 44 nights of recording(over 8 h per night),providing substantial evidence that the electrodes can be leveraged to substitute for clinical standard electrodes.Junshi Li Yundong Ma Dong Huang Zhongyan Wang Zhitong Zhang Yingjie Ren Mengyue Hong Yufeng Chen Tingyu Li Xiaoyi Shi Lu Cao Jiayan Zhang Bingli Jiao Junhua Liu Hongqiang Sun Zhihong Li 2022Nano-Micro Letters2022,14,8:1
3RIGID GIGAPOROUS CHROMATOGRAPHIC MEDIA AND THEIR POTENTIAL IMPACT ON DOWNSTREAM PROCESSING显示文摘More and more biomolecules are being produced by the biotechnology industry for applications ranging from medicine and food to engineering materials. Liquid chromatography plays a center-stage role in a typical downstream process producing biomolecules such as recombinant proteins. Rigid gigaporous media are porous particles possessing large transecting through-pores with a pore-to-particle diameter ratio of dpore/dparticle> 0.01. They allow convective flow in the large through-pores, while the smaller diffusion-pores (typically several hundred angstroms in size) supply the needed surface areas. Because of the transecting gigapores, a portion of the mobile phase flows through the pores in addition to fluid flow in the interstitial spaces between the particles in a packed-bed column. This considerably lowers the operating column pressure drop. This lower pressure drop makes axial-direction scale-up of chromatographic columns possible to avoid pancake columns that invariably degrade separation resolution. The large gigapores also make the binding sites on the diffusion pore surfaces more accessible, thus increasing the loading capacity of large protein molecules that can be hindered sterically if only diffusion pores are present. 4Promoter variations in DBR2-like affect artemisinin production in different chemotypes of Artemisia annua显示文摘Artemisia annua is the only known plant source of the potent antimalarial artemisinin,which occurs as the low-and high-artemisinin producing(LAP and HAP)chemotypes.Nevertheless,the different mechanisms of artemisinin producing between these two chemotypes were still not fully understood.Here,we performed a comprehensive analysis of genome resequencing,metabolome,and transcriptome data to systematically compare the difference in the LAP chemotype JL and HAP chemotype HAN.Metabolites analysis revealed that 72.18%of sesquiterpenes was highly accumulated in HAN compared to JL.Integrated omics analysis found a DBR2-Like(DBR2L)gene may be involved in artemisinin biosynthesis.DBR2L was highly homologous with DBR2,belonged to ORR3 family,and had the DBR2 activity of catalyzing artemisinic aldehyde to dihydroartemisinic aldehyde.Genome resequencing and promoter cloning revealed that complicated variations existed in DBR2L promoters among different varieties of A.annua and were clustered into three variation types.The promoter activity of diverse variant types showed obvious differences.Furthermore,the core region(-625 to 0)of the DBR2L promoter was identified and candidate transcription factors involved in DBR2L regulation were screened.Thus,the result indicates that DBR2L is another key enzyme involved in artemisinin biosynthesis.The promoter variation in DBR2L affects its expression level,and thereby may result in the different yield of artemisinin in varieties of A.annua.It provides a novel insight into the mechanism of artemisinin-producing difference in LAP and HAP chemotypes of A.annua,and will assist in a high yield of artemisinin in A.annua.Xingwen Wang Lan Wu Li Xiang Ranran Gao Qinggang Yin Mengyue Wang Zhaoyu Liu Liang Leng Yanyan Su Huihua Wan Tingyu Ma Shilin Chen Yuhua Shi 2023Horticulture Research2023,10,9:0
5A lncRNA from an inflammatory bowel disease risk locus maintains intestinal host-commensal homeostasis显示文摘Inflammatory bowel diseases(IBD)are known to have complex,genetically influenced etiologies,involving dysfunctional interactions between the intestinal immune system and the microbiome.Here,we characterized how the RNA transcript from an IBD-associated long non-coding RNA locus(“CARINH-Colitis Associated IRF1 antisense Regulator of Intestinal Homeostasis”)protects against IBD.We show that CARINH and its neighboring gene coding for the transcription factor IRF1 together form a feedforward loop in host myeloid cells.The loop activation is sustained by microbial factors,and functions to maintain the intestinal host-commensal homeostasis via the induction of the anti-inflammatory factor IL-18BP and anti-microbial factors called guanylate-binding proteins(GBPs).Extending these mechanistic insights back to humans,we demonstrate that the function of the CARINH/IRF1 loop is conserved between mice and humans.Genetically,the T allele of rs2188962,the most probable causal variant of IBD within the CARINH locus from the human genetics study,impairs the inducible expression of the CARINH/IRF1 loop and thus increases genetic predisposition to IBD.Our study thus illustrates how an IBD-associated lncRNA maintains intestinal homeostasis and protects the host against colitis.Hongdi Ma Taidou Hu Wanyin Tao Jiyu Tong Zili Han Dietmar Herndler-Brandstetter Zheng Wei Ruize Liu Tingyue Zhou Qiuyuan Liu Xuemei Xu Kaiguang Zhang Rongbin Zhou Judy H.Cho Hua-Bing Li Hailiang Huang Richard A.Flavell Shu Zhu 2023Cell Research2023,33,5:0

8Photobiomodulation inhibits the expression of chondroitin sulfate proteoglycans after spinal cord injury via the Sox9 pathway显示文摘Both glial cells and glia scar greatly affect the development of spinal cord injury and have become hot spots in research on spinal cord injury treatment.The cellular deposition of dense extracellular matrix proteins such as chondroitin sulfate proteoglycans inside and around the glial scar is known to affect axonal growth and be a major obstacle to autogenous repair.These proteins are thus candidate targets for spinal cord injury therapy.Our previous studies demonstrated that 810 nm photo biomodulation inhibited the formation of chondroitin sulfate proteoglycans after spinal cord injury and greatly improved motor function in model animals.However,the specific mechanism and potential targets involved remain to be clarified.In this study,to investigate the therapeutic effect of photo biomodulation,we established a mouse model of spinal cord injury by T9 clamping and irradiated the injury site at a power density of 50 mW/cm~2 for 50 minutes once a day for 7 consecutive days.We found that photobiomodulation greatly restored motor function in mice and down regulated chondroitin sulfate proteoglycan expression in the injured spinal cord.Bioinformatics analysis revealed that photobiomodulation inhibited the expression of proteoglycan-related genes induced by spinal cord injury,and versican,a type of proteoglycan,was one of the most markedly changed molecules.Immunofluorescence staining showed that after spinal cord injury,versican was present in astrocytes in spinal cord tissue.The expression of versican in primary astrocytes cultured in vitro increased after inflammation induction,whereas photobiomodulation inhibited the expression of ve rsican.Furthermore,we found that the increased levels of p-Smad3,p-P38 and p-Erk in inflammatory astrocytes were reduced after photobiomodulation treatment and after delivery of inhibitors including FR 180204,(E)-SIS3,and SB 202190.This suggests that Sma d 3/Sox9 and MAP K/Sox9 pathways may be involved in the effects of photobiomodulation.In summary,our findings show that photobiomodulation modulates the expression of chondroitin sulfate proteoglycans,and versican is one of the key target molecules of photo biomodulation.MAPK/Sox9 and Smad3/Sox9 pathways may play a role in the effects of photo biomodulation on chondroitin sulfate proteoglycan accumulation after spinal cord injury.Zhihao Zhang Zhiwen Song Liang Luo Zhijie Zhu Xiaoshuang Zuo Cheng Ju Xuankang Wang Yangguang Ma Tingyu Wu Zhou Yao Jie Zhou Beiyu Chen Tan Ding Zhe Wang Xueyu Hu 2024Neural Regeneration Research2024,19,1:0
