1ILF3 is a substrate of SPOP for regulating serine biosynthesis in colorectal cancer显示文摘The Serine-Glycien-One-Carbon(SGOC)pathway is pivotal in multiple anabolic processes.Expression levels of SGOC genes are deregulated under tumoeigenic condltions,suggesting panicipation of oncogenes in deregulating the SGOC biosynthetic pathway.However,the underlying mechanism remains elusive.Here,we identifed that Interleukin enfiarccer-binding factor 3(ILF3)is overexpressed in primary CRC patient specimens and correlates with poor progroosis.ILF3 is critical in regulating the SGOC pathway by directly regulating the mRNA stability of SGOC genes,thereby increasing SGOC genes expression and facilitating tumor growth.Mehhanktic studies showed that the EGF-MIEK-ERK pathway nnediates ILF3 phosphorylation,which hin ders E3 ligase speckle-type POZ protein(SPOP)-mediated poly-ubiquitination and degradation of ILF3.Sign iftca ntly,combination of SGOC inhibitor and the anti-EGFR monoclonal antibody cetuximab can hinder the growth of patient-derived xenografts that sustain high ERK-ILF3 levels.Taken together,deregutation of ILF3 via the EGF-ERK signaling plays an important role in systemic serine metabolic reprogramming and confers a predilection toward CRC development.Our findings indicate that clinical evaluation of SCOC inhibitor is warranted for CRC patients with ILF3 overexpression.Kai Li Jian-lin Wu Baifu Qin Zongmin Fan Qin Tang Weisi Lu Haipeng Zhang Fan Xing Manqi Meng Shaomin Zou Wenxia Wei Honglei Chen Jian Cai Huaiming Wang Hui Zhang Jiayue Cai Ling Fang Xiqing Bian Chuangqi Chen Ping Lan Bart Ghesquiere Lekun Fang Mong-Hong Lee 2020Cell Research2020,30,2:6
2Detection of quantitative trait loci and heterotic loci for plant height using an immortalized F_2 population in maize显示文摘A set of recombinant inbred lines (RIL) derived from Yuyu22, an elite hybrid widespread in China, was used to construct an immortalized F2 (IF2) population comprising 441 different crosses. Genetic linkage maps were constructed containing 10 linkages groups with 263 simple sequence repeat (SSR) molecular markers. Twelve and ten quantitative trait loci (QTL) were detected for plant height in the IF2 and RIL populations respectively, using the composite interval mapping method, and six same QTL were identified in the two populations. In addition, ten unique heterotic loci (HL) located on seven different chromosomes were revealed for plant height using the mid-parent heterosis as the input data. These HL explained 1.26%―8.41% of the genotypic variance in plant height heterosis and most expressed overdominant effects. 3Surface magnetization of siderite mineral显示文摘Surface self-magnetization of siderite is achieved by generating ferromagnetic substance on the surface of siderite by adjusting slurry temperature,pH value,stirring rate and reaction time.No addition of any iron-containing reagent is required.The temperature of 60 ℃,NaOH concentration of 0.10 mol/L;stirring rate of 900 r/min and the reaction time of 10 min are the optimal conditions.The results show that the siderite recovery in magnetic separation increased from 26.9% to 88.8% after surface magnetization.Magnetization kinetic equation is expressed as 1 [1(e0.269)]1/3 = Kt.Activation energy for the magnetization reaction is 4.30 kJ/mol.VSM,SEM and XPS were used to characterize the siderite,and results show that the saturated magnetization(rs) of siderite increased from 0.652 to 2.569Am2 /kg,the magnetic hysteresis was detected with a coercive force of 0.976 A/m after magnetization;Fe2P3/2 electron binding energy changed which reflects the valence alteration in iron on the surface and the formation of ferromagnetic Fe3O4.Wu Xiqing Xu Pengyun Duan Yunfeng Hu Cong Li Guoping 2012International Journal of Mining Science and Technology2012,22,6:5

11Ultrasensitive quantification of trace amines based on N-phosphorylation labeling chip 2D LC-QQQ/MS显示文摘Trace amines(TAs)are metabolically related to catecholamine and associated with cancer and neurological disorders.Comprehensive measurement of TAs is essential for understanding pathological processes and providing proper drug intervention.However,the trace amounts and chemical instability of TAs challenge quantification.Here,diisopropyl phosphite coupled with chip two-dimensional(2D)liquid chromatography tandem triple-quadrupole mass spectrometry(LC-QQQ/MS)was developed to simultaneously determine TAs and associated metabolites.The results showed that the sensitivities of TAs increased up to 5520 times compared with those using nonderivatized LC-QQQ/MS.This sensitive method was utilized to investigate their alterations in hepatoma cells after treatment with sorafenib.The significantly altered TAs and associated metabolites suggested that phenylalanine and tyrosine metabolic pathways were related to sorafenib treatment in Hep3B cells.This sensitive method has great potential to elucidate the mechanism and diagnose diseases considering that an increasing number of physiological functions of TAs have been discovered in recent decades.Xiqing Bian Yida Zhang Na Li Menglin Shi Xiaolin Chen Hui-Lu Zhang Jie Liu Jian-Lin Wu 2023Journal of Pharmaceutical Analysis2023,13,3:0
12Onset location of scintillation-producing spread-F plume over Sanya显示文摘Equatorial spread-F (ESF) backscatter plumes are often observed in radar range-time-intensity (RTI) maps at low latitude. Except case studies, few statistical investigations on the onset locations of scintillation-producing ESF plumes at given sites have been conducted. In this study, a statistical analysis is carried out on onset locations of ESF backscatter plumes observed at a low-latitude location Sanya (18.4°N, 109.6°E; dip lat 12.8°N) during equinoctial months of 2013. By employing a tracing method to locate backscatter plumes, we estimate the onset longitudes of periodic plumes obtained from the Sanya VHF radar five-beam steering measurements. The results show that the inter-plume distances (in longitude) are mostly confined within 200–600 km, and the ESF plumes producing ionospheric scintillations over Sanya are almost exclusively generated at the longitudes of 94°–110°E. 13Prediction of line-spectrum noise induced by high speed vehicle counter-rotation propellers in water显示文摘Line-Spectrum noise of counter-rotation propellers has constructed the main part of the radiated noise of high speed vehicles in water. The line-spectrum noise of the counter-rotation propellers is due to the interaction between fore or aft propeller and wake of the vehicle,and the interaction between fore and aft propeller. Based on a combination of the lifting surface theory and acoustic method, the prediction of line-spectrum noise is presented in this paper.Theoretical calculation method, characteristics and numerical prediction of the line-spectrum noise are detailed too. The effect of different wake and different distance between fore and aft propeller on the propeller noise is also studied by numerical method. 16Characterization of the core microflora and nutrient composition in packaged pasteurized milk products during storage显示文摘Pasteurized milk contains complex microbial communities affected by sterilization and storage conditions.This complex microflora may be the possible reason that pasteurized dairy products are highly prone to spoilage.In this study,packaged pasteurized milk products collected from dairy processing factories in China were stored at 0,4,10,15,and 25℃ for 0−15 days and subjected to microbial identification using high-throughput sequencing.Accordingly,6 phyla and 44 genera were identified as the dominant microbiota.Moreover,the changes in nutritional composition of the pasteurized milk,including in 16 free amino acids,7 taste values,and 8 chemical constituents,were analyzed using principal component and multi-factor analyses.The Pearson correlation analysis identified Pseudomonas,Aeromonas,Paenibacillus,and Serratia genera as the core functional microbiota that significantly affects the nutritional composition of pasteurized milk.Hence,the results provide a comprehensive understanding of the safety and shelf-life of stored pasteurized milk.Ruixue Ding Shanshan Yang Lijuan Geng Yumeng Liu Baoping He Liyun Liu Xiqing Yue Rina Wu Junrui Wu 2023Food Science and Human Wellness2023,12,4:0
17A prospective cohort study of the presence of SARS-CoV-2 in clinical samples from multiple bodily sites:implications for transmission routes of COVID-19显示文摘Objective: The coronavirus disease 2019 (COVID-19) epidemic resulting from severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has still spread globally. The occurrence of the Delta variant, which is more infectious and spreads faster than earlier forms of the virus that causes COVID-19, makes infection prevention more challenging. Therefore, this study aimed to gain a comprehensive insight into the transmission routes of SARS-CoV-2 for curbing the propagation of SARS-CoV-2 in human populations.Methods: We studied a prospective cohort of 576 patients admitted consecutively to the First Affiliated Hospital of Guangzhou Medical University from January 21 to June 8, 2020. These patients were chosen based on their similar clinical phenotypes or imaging findings. There were 21 (3.6%) laboratory-confirmed COVID-19 patients (16 severe and 5 mild cases) and 555 non-COVID-19 patients. The antibody response and routes and duration of viral shedding were systematically evaluated in serial clinical specimens. Moreover, SARS-CoV-2 RNA was also detected in a mouth rinse, urine, and tear samples. This study was approved by the Medical Ethical Committee of The First Affiliated Hospital of Guangzhou Medical University (approval No. 2020-77).Results: SARS-CoV-2 mainly existed in sputum, nasal and throat swabs, and feces samples. Virus latency was longer in sputum and feces samples than in nasopharyngeal samples. IgG antibody response in respiratory samples was related to disease severity. 18Near-infrared light(NIR)-responsive nanoliposomes combining photodynamic therapy and chemotherapy for breast tumor control显示文摘Light-responsive carriers have been used for the controlled release of antitumor drugs in recent years.However, most light-responsive vectors require high-energy ultraviolet or visible light to achieve local drug release, and ultraviolet light would cause cellular damage. Near-infrared light has a deeper tissuepenetration depths and minimal harm to tissues, but it is difficult to cleave the chemical bond directly.The aim of this study is to develop a novel near-infrared light-responsive carrier for local release of antitumor drugs. Unsaturated phospholipids can be oxidized by singlet oxygen to achieve liposomal drug release, and singlet oxygen can be produced by photosensitizer under light irradiation. A new near-infrared light-responsive nanoliposome was designed that imparts light-triggered local drug release. Nanoliposomes, which were composed of matrix phospholipids and unsaturated phospholipids, were prepared by ammonium sulfate gradient method, and loaded with antitumor drug doxorubicin(DOX) and photosensitizer 1,4,8,11,15,18,22,25-octabutoxypalladium phthalocyanine. Under near-infrared light, photosensitizers could produce singlet oxygen and damage tumor cells by photodynamic therapy. Simultaneously, the unsaturated phospholipids were oxidized by singlet oxygen and result in DOX release, causing sustained cell damage by chemotherapy. Near-infrared light-responsive nanoliposomes exhibit enhanced anticancer activity owing to combined treatment of photodynamic therapy and chemotherapy. 19Polarity-regulated derivatization-assisted LC-MS method for amino-containing metabolites profiling in gastric cancer显示文摘Amino-containing compounds,including amino acids,aliphatic amines,aromatic amines,small peptides and catecholamines,are involved in various biological processes and play vital roles in multiple metabolic pathways.Previous studies indicated that some amino-containing metabolites are significant diagnostic and prognostic biomarkers of gastric cancer.However,the discovery of precise biomarkers for the preoperative diagnosis of gastric cancer is still in an urgent need.Herein,we established a polarityregulated derivatization method coupled with liquid chromatography-mass spectrometry(LC-MS)for amino-containing metabolites profiling in the serum samples of patients with gastric cancer and healthy controls,based on our newly designed and synthesized derivatization reagent(S)-3-(1-(diisopropoxyphosphoryl)pyrrolidine-2-carboxamido)-N-hydroxysuccinimidyl ester(3-DP-NHS).Enhanced separation efficiency and detection sensitivity for amino-containing metabolites were achieved after derivatization.This method exhibited good linearity,recovery,intra-and inter-day precision and accuracy.Only 5 mL serum is needed for untargeted analysis,enabling 202 amino-containing metabolites to be detected.Statistical analysis revealed altered amino acid metabolisms in patients with gastric cancer.Furthermore,ultra high performance liquid chromatography coupled with mass spectrometry(UHPLCMS/MS)analysis quantification revealed increased serum levels of tryptamine and decreased concentrations of arginine and tryptophan in patients with gastric cancer.Receiver operating characteristic(ROC)curves indicated that an increased tryptamine/tryptophan ratio could serve as a potential biomarker for gastric cancer diagnosis.This study demostrated the possibility of using serum amino acid biomarkers for gastric cancer diagnosis,providing new avenues for the treatment of gastric cancer.Jie Han Shilin Gong Xiqing Bian Yun Qian Guilan Wang Na Li Jian-Lin Wu 2023Journal of Pharmaceutical Analysis2023,13,11:0
20Tailoring carbon chains for repairing graphite from spent lithium-ion battery toward closed-circuit recycling显示文摘Graphite, as a strategic mineral resource, the recycling from spent lithium-ion batteries(LIBs) has attracted considerable attention for meeting considerable economic value. However, closed-circuit recycling still suffers from the lack of effective repair methods. Considering the existing defects, a series of Cchain length carbons have been successfully introduced to repair spent graphite. Obviously, with the evolution of carbon resources, the thickness and pores of the coating layer were tailored with the functional groups. Benefitting from the increased active sites and created fold structure, their coulombic efficiency is obviously restored from 14% to 86.89%, while the stable capacity is kept at approximately 384.9 mAh gafter 100 cycles. Moreover, their excellent rate properties are kept about approximately 200 mAh gat2 C, meeting the standard of commercial materials. Supported by the detailed kinetic behaviors, the enhanced rate is mainly dominated by pseudocapacitive behaviors, accompanied by deepening redox reactions. Meanwhile, the cost of the proposed approach for recycling spent graphite is 894.87 $ t^(-1),and the recycling profit for regenerating graphite is approximately 7000 $ t^(-1).
