| 2 | High-strength wrought magnesium alloy with dense nano-scale spherical precipitate显示文摘This paper reported the influences of Yb addition on the precipitate and mechanical properties of wrought magnesium alloy ZK60. The ingots of ZK60-1.78Yb (wt%, 0.26 at%) alloys were cast using permanent mould and extruded at 370℃ . By means of TEM and HRTEM, it was observed that Yb affected the precipitate and precipitation of ZK60-1.78Yb alloys significantly. Dynamic precipitation occurred in the as-extruded alloy and spherical nano-scale precipitate with high density and homoge-neity exhibited in the aged alloys. The precipitate particles were about 5-20 nm in diameter, 10-30 nm in average space length. | 3 | Single-cell transcriptomes of peripheral blood cells indicate and elucidate severity of COVID-19显示文摘The blood and immune system of coronavirus disease 2019(COVID-19)infected patients are dysfunctional,and numerous studies have been conducted to resolve their characteristics and pathogenic mechanisms.Nevertheless,the variations of immune responses along with disease severity have not been comprehensively documented.Here,we profiled the single-cell transcriptomes of 96,313 peripheral blood mononuclear cells(PBMCs)derived from 12 COVID-19 patients(including four moderate,four severe and four critical cases)and three healthy donors.We showed that proliferative CD8 effector T cells with declined immune functions and cytotoxicity accumulated in the critical stage.By contrast,the quantity of natural killer(NK)cells was significantly reduced,while they exhibited enhanced immune activities.Notably,a gradually attenuated responseto COVID-19 along with disease severity was observed in monocytes,in terms of cellular composition,transcriptional discrepancy and transcription factor regulatory network.Furthermore,we identified immune cell-type dependent cytokine signatures distinguishing the severity of COVID-19 patients.In addition,cell interactions between CD8 effector T/NK cells and monocytes mediated by inflammatory cytokines were enhanced in moderate and severe stages,but weakened in critical cases.Collectively,our work uncovers the cellular and molecular players underlying the disordered and heterogeneous immune responses associated with COVID-19 severity,which could provide valuable insights for the treatment of critical COVID-19 patients. | Xiaowei Xie Xuelian Cheng Gaoxiang Wang Biao Zhang Mengyao Liu Liting Chen Hui Cheng Sha Hao Jianfeng Zhou Ping Zhu Tao Cheng | 2021 | Science China(Life Sciences)2021,64,10: | 3 |
| 4 | Whole exome sequencing identifies an AMBN missense mutation causing severe autosomal-dominant amelogenesis imperfecta and dentin disorders显示文摘Tooth development is a complex process that involves precise and time-dependent orchestration of multiple genetic, molecular,and cellular interactions. Ameloblastin(AMBN, also named 'amelin' or 'sheathlin') is the second most abundant enamel matrix protein known to have a key role in amelogenesis. Amelogenesis imperfecta(AI [MIM: 104500]) refers to a genetically and phenotypically heterogeneous group of conditions characterized by inherited developmental enamel defects. The hereditary dentin disorders comprise a variety of autosomal-dominant genetic symptoms characterized by abnormal dentin structure affecting either the primary or both the primary and secondary teeth. The vital role of Ambn in amelogenesis has been confirmed experimentally using mouse models. Only two cases have been reported of mutations of AMBN associated with non-syndromic human AI. However, no AMBN missense mutations have been reported to be associated with both human AI and dentin disorders.We recruited one kindred with autosomal-dominant amelogenesis imperfecta(ADAI) and dentinogenesis imperfecta/dysplasia characterized by generalized severe enamel and dentin defects. Whole exome sequencing of the proband identified a novel heterozygous C-T point mutation at nucleotide position 1069 of the AMBN gene, causing a Pro to Ser mutation at the conserved amino acid position 357 of the protein. Exfoliated third molar teeth from the affected family members were found to have enamel and dentin of lower mineral density than control teeth, with thinner and easily fractured enamel, short and thick roots, and pulp obliteration. | 5 | Excretion of manure-borne estrogens and androgens and their potential risk estimation in the Yangtze River Basin显示文摘The Yangtze River is the longest river in China, and the river basin spans one fifth of the area of the whole country. Based on statistical data, the excretion of manure-borne steroid hormones, including steroid estrogens(SEs) and steroid androgens(SAs), in 10 provinces of China within the region has been estimated. The potential environmental and ecological risk of manure-borne steroid estrogens to the surface water in this region was also assessed. The manure-borne SE and SA excretions in the 10 provinces and municipalities vary in the order: Sichuan > Hunan > Hubei > Yunnan > Jiangsu > Anhui > Jiangxi >Chongqing > Qinghai > Shanghai. The highest increase of manure-borne SEs(1434.3 kg)and SAs(408.5 kg) was found in Hunan and Hubei provinces, respectively, and the total excretion in 2013 was 65% more than 15 years earlier in these two provinces. However, the emissions in Anhui and Shanghai decreased in this 15 year period of time. Swine urine,chicken feces, cattle urine, and laying hen feces were considered the dominant sources of manure-borne E1, βE2, αE2, and SAs, respectively. Although Jiangsu province did not have the largest excretion of manure-borne SEs, it had the highest level of predicted17β-estradiol equivalency(EEQs) value of 16.65 ng/L in surface water because of the limited surface water resources. | 13 | Clinical efficacy and safety of linezolid in intensive care unit patients显示文摘Background:To characterize the population of critically ill patients and infections treated with linezolid in the intensive care unit(ICU),and to evaluate the clinical efficacy and safety of linezolid therapy.Methods::This multi-center,observational,real-world study was conducted across 52 hospitals between June 9,2018,and December 28,2019.Patients who met the following inclusion criteria were included:(1)admitted to the ICU,(2)of any age group,and(3)having a clinical or laboratory diagnosis of a Gram-positive bacterial infection.Clinical efficacy was categorized as success(cured or improved),failed,or non-evaluable.Adverse events and serious adverse events were recorded during treatment.Results::A total of 366 ICU patients who met the inclusion criteria were evaluated.Linezolid was used as second-and first-line treatment in 232(63.4%)and 134(36.6%)patients,respectively.The most common isolated strain was Staphylococcus aureus(methicillin-resistant Staphylococcus aureus:n=37/119,31.1%;methicillin-susceptible Staphylococcus aureus:n=15/119,12.6%);this was followed by Enterococci(vancomycin-resistant Enterococci:n=8/119,6.7%;vancomycin-susceptible Enterococci:n=11/119,9.2%)and Streptococcus pneumoniae(multidrug-resistant:n=4/119,3.4%;non-multidrug resistant:n=2/119,1.7%).The main infection sites where pathogens were detected included the lung(n=216/366,59.6%),skin and soft tissue(n=104/366,28.4%),and blood(n=50/366,13.7%).Clinical success was achieved in 301(82.2%)patients;34(9.3%)were cured and 267(73.0%)improved;treatment failure and non-evaluable outcomes were observed in 29(7.9%)in 36(9.8%)patients,respectively.Linezolid-related adverse events were reported in 8(2.2%)patients.No treatment-related serious adverse events were reported.Conclusions::Based on real-world results,linezolid was found to be effective and safe in the treatment of Gram-positive bacterial infections in critically ill patients. | Aijia Ma Meiling Dong Jiangli Cheng Xuelian Liao Wei Dong Chang Liu Chenggong Hu Jing Yang Yan Kang | 2023 | Journal of Intensive Medicine2023,3,1: | 0 |

| 15 | Study on an electrochemical biosensor for thrombin recognition based on aptamers and nano particles显示文摘This paper presents a high specific, sensitive electrochemical biosensor for recognition of protein such as thrombin based on aptamers and nano particles. Two different aptamers were chosen to construct a sandwich manner for detecting thrombin. Aptamer I was immobilized on nano magnetic particle for capturing thrombin, and aptamer II labled with nano gold was used for detection. The electrical current generated from gold after the formation of the complex of magnetic particle, thrombin and nano gold, and then an electrochemical cell designed by ourselves was used for separating, gathering, and electrochemical detecting. Through magnetic separation, high specific and sensitive detection of the target protein, thrombin, was achieved. Linear response was observed over the range 5.6×10-12―1.12×10-9 mol/L, with a detection limit of 1.42×10-12 mol/L.
