2Meningeal lymphatic vessels regulate brain tumor drainage and immunity显示文摘Recent studies have shown that meningeal lymphatic vessels(MLVs),which are located both dorsally and basally beneath the skull,provide a route for draining macromolecules and trafficking immune cells from the central nervous system(CNS)into cervical lymph nodes(CLNs),and thus represent a potential therapeutic target for treating neurodegenerative and neuroinflammatory diseases.However,the roles of MLVs in brain tumor drainage and immunity remain unexplored.Here we show that dorsal MLVs undergo extensive remodeling in mice with intracranial gliomas or metastatic melanomas.RNA-seq analysis of MLV endothelial cells revealed changes in the gene sets involved in lymphatic remodeling,fluid drainage,as well as inflammatory and immunological responses.Disruption of dorsal MLVs alone impaired intratumor fluid drainage and the dissemination of brain tumor cells to deep CLNs(dCLNs).Notably,the dendritic cell(DC)trafficking from intracranial tumor tissues to dCLNs decreased in mice with defective dorsal MLVs,and increased in mice with enhanced dorsal meningeal lymphangiogenesis.Strikingly,disruption of dorsal MLVs alone,without affecting basal MLVs or nasal LVs,significantly reduced the efficacy of combined anti-PD-1/CTLA-4 checkpoint therapy in striatal tumor models.Furthermore,mice bearing tumors overexpressing VEGF-C displayed a better response to anti-PD-1/CTLA-4 combination therapy,and this was abolished by CCL21/CCR7 blockade,suggesting that VEGF-C potentiates checkpoint therapy via the CCL21/CCR7 pathway.Together,the results of our study not only demonstrate the functional aspects of MLVs as classic lymphatic vasculature,but also highlight that they are essential in generating an efficient immune response against brain tumors.Xueting Hu Qiuping Deng Lu Ma Qingqing Li Yidong Chen Yuhan Liao Fan Zhou Chen Zhang Linlin Shao Jun Feng Tubao He Weihai Ning Yan Kong Yingqing Huo Aibin He Bing Liu Jingjing Zhang Ralf Adams Yulong He Fuchou Tang Xiuwu Bian Jincai Luo 2020Cell Research2020,30,3:11
3Selenium deficiency induces spleen pathological changes in pigs by decreasing selenoprotein expression,evoking oxidative stress,and activating inflammation and apoptosis显示文摘Background:The immune system is one aspect of health that is affected by dietary selenium(Se)levels and selenoprotein expression.Spleen is an important immune organ of the body,which is directly involved in cellular immunity.However,there are limited reports on Se levels and spleen health.Therefore,this study established a Se-deficient pig model to investigate the mechanism of Se deficiency-induced splenic pathogenesis.Methods:Twenty-four pure line castrated male Yorkshire pigs(45 days old,12.50±1.32 kg,12 full-sibling pairs)were divided into two equal groups and fed Se-deficient diet(0.007 mg Se/kg)or Se-adequate diet(0.3 mg Se/kg)for 16 weeks.At the end of the trial,blood and spleen were collected to assay for erythroid parameters,the osmotic fragility of erythrocytes,the spleen index,histology,terminal deoxynucleotidyl transferase nick-end labeling(TUNEL)staining,Se concentrations,the selenogenome,redox status,and signaling related inflammation and apoptosis.Results:Dietary Se deficiency decreased the erythroid parameters and increased the number of osmotically fragile erythrocytes(P<0.05).The spleen index did not change,but hematoxylin and eosin and TUNEL staining indicated that the white pulp decreased,the red pulp increased,and splenocyte apoptosis occurred in the Se deficient group.Se deficiency decreased the Se concentration and selenoprotein expression in the spleen(P<0.05),blocked the glutathione and thioredoxin antioxidant systems,and led to redox imbalance.Se deficiency activated the NF-κB and HIF-1αtranscription factors,thus increasing pro-inflammatory cytokines(IL-1β,IL-6,IL-8,IL-17,and TNF-α),decreasing anti-inflammatory cytokines(IL-10,IL-13,and TGF-β)and increasing expression of the downstream genes COX-2 and iNOS(P<0.05),which in turn induced inflammation.In addition,Se-deficiency induced apoptosis through the mitochondrial pathway,upregulated apoptotic genes(Caspase3,Caspase8,and Bak),and downregulated antiapoptotic genes(Bcl-2)(P<0.05)at the mRNA level,thus verifying the results of TUNEL staining.Conclusions:These results indicated that Se deficiency induces spleen injury through the regulation of selenoproteins,oxidative stress,inflammation and apoptosis.Shuang Li Wenjuan Sun Kai Zhang Jiawei Zhu Xueting Jia Xiaoqing Guo Qingyu Zhao Chaohua Tang Jingdong Yin Junmin Zhang 2021Journal of Animal Science and Biotechnology2021,12,4:4

5GSH-induced chemotaxis nanomotors for cancer treatment by ferroptosis strategy显示文摘Overexpression of glutathione(GSH) in tumor cells greatly inhibits the therapy effect of traditional ferroptosis inducers;thus,control of the GSH level is an important way to improve the efficacy of ferroptosis.Herein,a kind of nanomotor based on metal organic framework material NH_(2)-MIL-101 is constructed,in which polyethylene glycol(PEG) and glutathione hydrolase γ-glutamyltransferase(GGT) are asymmetrically modified to obtain mPEG@MIL-101@GGT nanomotors(PMG NMs).The nanomotor proposed in this article can be induced by overexpressing GSH in tumors to form chemotactic effects through the specific affinity between enzymes and substrates.Results indicate that the tail structure provided by PEG and the affinity between GGT and GSH can enable the stable chemotaxis behavior of nanomotors in a complex environment,thus enriching and penetrating deeply at the tumor site.In addition,after loading the ferroptosis inducer Erastin,the system shows a highly effective induction effect of tumor ferroptosis.Erastin in the system can effectively inhibit the synthesis of GSH,and PMG NMS can react with GSH through Fe^(3+)and GGT to promote GSH depletion.The produced Fe^(2+)can generate excessive reactive oxygen species through Fenton reaction,which further promotes the death of tumor cells.Meantime,the chemotaxis behavior of the nanomotors based on the endogenous biochemical reaction of GGT-catalyzed GSH hydrolysis can endow nanomotors with the enhanced delivery and penetration ability in tumors,thus collaboratively enhancing the ferroptosis effect.This strategy designed according to the physiological characteristics of tumors has good biosafety and treatment effect,providing new perspectives for micro/nanomotor and tumor treatment.Zhiyong Liu Ting Li Nan Li Yujing Wang Lin Chen Xueting Tang Mimi Wan Chun Mao 2022Science China Chemistry2022,65,5:1
6Selective substitution induced anomalous phonon stiffening within quasi-one-dimensional P–P chains in SiP2显示文摘Light-matter interactions in low-dimensional quantum-confined structures can dominate the optical properties of the materials and lead to optoelectronic applications.In anisotropic layered silicon diphosphide(SiP2)crystal,the embedded quasi-onedimensional(1D)phosphorus–phosphorus(P–P)chains directly result in an unconventional quasi-1D excitonic state,and a special phonon mode vibrating along the P–P chains,establishing a unique 1D quantum-confined system.Alloying SiP2 with the homologous element serves as an effective way to study the properties of these excitons and phonons associated with the quasi-1D P–P chains,as well as the strong interaction between these quasiparticles.However,the experimental observation and the related optical spectral understanding of SiP2 with isoelectronic dopants remain elusive.Herein,with the photoluminescence and Raman spectroscopy measurements,we demonstrate the redshift of the confined excitonic peak and the stiffening of the phonon vibration mode■of a series of Si(P1−xAsx)2 alloys with increasing arsenic(As)compositions.This anomalous stiffening of■is attributed to the selective substitution of As atoms for P atoms within the P–P chains,which is confirmed via our scanning transmission electron microscopy investigation.Such optical spectra evolutions with selective substitution pave a new way to understand the 1D quantum confinement in semiconductors,offering opportunities to explore quasi-1D characteristics in SiP2 and the resulting photonic device application.Xueting Dai Feng Qin Caiyu Qiu Ling Zhou Junwei Huang Fanghua Cheng Xiangyu Bi Caorong Zhang Zeya Li Ming Tang Shengqiang Wu Xiaoxu Zhao Yangfan Lu Huiyang Gou Hongtao Yuan 2023Nano Research2023,16,1:0
7Strain engineering of anisotropic light-matter interactions in onedimensional P-P chain of SiP_(2)显示文摘Strain engineering can serve as a powerful technique for modulating the exotic properties arising from the atomic structure of materials.Examples have been demonstrated that one-dimensional(1D)structure can serve as a great platform for modulating electronic band structure and phonon dispersion via strain control.Particularly,in a van der Waals material silicon diphosphide(SiP_(2)),quasi-1D zigzag phosphorus–phosphorus(P–P)chains are embedded inside the crystal structure,and can show unique phonon vibration modes and realize quasi-1D excitons.Manipulating those optical properties by the atom displacements via strain engineering is of great interest in understanding underlying mechanism of such P–P chains,however,which remains elusive.Herein,we demonstrate the strain engineering of Raman and photoluminescence(PL)spectra in quasi-1D P–P chains and resulting in anisotropic manipulation in SiP_(2).We find that the phonon frequencies of SiP_(2)in Raman spectra linearly evolve with a uniaxial strain along/perpendicular to the quasi-1D P–P chain directions.Interestingly,by applying tensile strain along the P–P chains,the band gap energy of strained SiP_(2)can significantly decrease with a tunable value of~55 meV.Based on arsenic(As)element doping into SiP_(2),the strain-induced redshifts of phonon frequencies decrease,indicating the stiffening of the phonon vibration with the increased arsenic doping level.Such results provide an opportunity for strain engineering of the light–matter interactions in the quasi-1D P–P chains of SiP_(2)crystal for potential optical applications.Fanghua Cheng Junwei Huang Feng Qin Ling Zhou Xueting Dai Xiangyu Bi Caorong Zhang Zeya Li Ming Tang Caiyu Qiu Yangfan Lu Huiyang Gou Hongtao Yuan 2022Nano Research2022,15,8:0
8Experimental Study on the Effect of Bed Material Amount and Fuel Particle Size on Load Change of Circulating Fluidized Bed显示文摘The large-scale integration of new energy generation has put forward higher requirements for the peak-shaving capability of thermal power.The circulating fluidized bed(CFB)depends on the advantages of a wide load adjustment range and low cost of pollutant control to become a good peak shaving power supply.However,the large delay and inertia caused by its unique combustion mode make it very difficult to change the load quickly.To further understand the factors that affect the load change of CFB,and explore the method of increasing CFB load change rate,the load change experiment on the combustion side was carried out in the 0.1 MW CFB experiment platform.The influence law of bed material amount and fuel particle size on load change of CFB combustion side was revealed for the first time.The results indicated that the increase of bed material amount was beneficial to improve the load change rate on the combustion side of CFB and reduce the carbon content of fly ash,but had no obvious effect on NO_(x)emission.When the bed height at rest increased from 200 mm to 400 mm,the load change rate of the CFB combustion side load from 50%to 75%increased from 0.78%/min to 1.14%/min,and the carbon content of fly ash at 75%load decreased from 26.6%to 24.9%.In addition,the reduction of fuel particle size positively improved the load change rate on the combustion side of the CFB and reduced NO_(x)emission but had a negative effect on reducing the carbon content of fly ash.When the fuel particle size decreased from 0-1 mm to 0-0.12 mm,the load change rate of CFB combustion side load from 50%to 75%increased from 0.78%/min to 1.09%/min,and the NO_(x)emission and carbon content of fly ash at 75%load decreased from 349.5 mg/m^(3)to 194.1 mg/m^(3)and increased from 26.6%to 31.8%,respectively.TANG Zihua SONG Guoliang JIANG Yu YANG Xueting JI Zengcai SUN Liwei 2023Journal of Thermal Science2023,32,5:0
9Roles of small RNAs in crop disease resistance显示文摘Small RNAs(sRNAs)are a class of short,non-coding regulatory RNAs that have emerged as critical components of defense regulatory networks across plant kingdoms.Many sRNA-based technologies,such as host-induced gene silencing(HIGS),spray-induced gene silencing(SIGS),virus-induced gene silencing(VIGS),artificial microRNA(amiRNA)and synthetic trans-acting siRNA(syn-tasiRNA)-mediated RNA interference(RNAi),have been developed as disease control strategies in both monocot and dicot plants,particularly in crops.This review aims to highlight our current understanding of the roles of sRNAs including miRNAs,heterochromatic siRNAs(hc-siRNAs),phased,secondary siRNAs(phasiRNAs)and natural antisense siRNAs(nat-siRNAs)in disease resistance,and sRNAs-mediated trade-offs between defense and growth in crops.In particular,we focus on the diverse functions of sRNAs in defense responses to bacterial and fungal pathogens,oomycete and virus in crops.Further,we highlight the application of sRNA-based technologies in protecting crops from pathogens.Further research perspectives are proposed to develop new sRNAs-based efficient strategies to breed non-genetically modified(GMO),diseasetolerant crops for sustainable agriculture.Jun Tang Xueting Gu Junzhong Liu Zuhua He 2021Stress Biology2021,1,1:0
