| 1 | Cell cycle-related kinase reprograms the liver immune microenvironment to promote cancer metastasis显示文摘The liver is an immunologically tolerant organ and a common metastatic site of multiple cancer types.Although a role for cancer cell invasion programs has been well characterized,whether and how liver-intrinsic factors drive metastatic spread is incompletely understood.Here,we show that aberrantly activated hepatocyte-intrinsic cell cycle-related kinase(CCRK)signaling in chronic liver diseases is critical for cancer metastasis by reprogramming an immunosuppressive microenvironment.Using an inducible liverspecific transgenic model,we found that CCRK overexpression dramatically increased both B16F10 melanoma and MC38 colorectal cancer(CRC)metastasis to the liver,which was highly infiltrated by polymorphonuclear-myeloid-derived suppressor cells(PMNMDSCs)and lacking natural killer T(NKT)cells.Depletion of PMN-MDSCs in CCRK transgenic mice restored NKT cell levels and their interferon gamma production and reduced liver metastasis to 2.7% and 0.7%(metastatic tumor weights)in the melanoma and CRC models,respectively.Mechanistically,CCRK activated nuclear factor-kappa B(NF-κB)signaling to increase the PMN-MDSC trafficking chemokine C-X-C motif ligand 1(CXCL1),which was positively correlated with liver-infiltrating PMN-MDSC levels in CCRK transgenic mice.Accordingly,CRC liver metastasis patients exhibited hyperaaivation of hepatic CCRK/NF-κB/CXCL1 signaling,which was associated with accumulation of PMN-MDSCs and paucity of NKT cells compared to healthy liver transplantation donors.In summary,this study demonstrates that immunosuppressive reprogramming by hepatic CCRK signaling undermines antimetastatic immunosurveillance.Our findings offer new mechanistic insights and therapeutic targets for liver metastasis intervention. | Xuezhen Zeng Jingying Zhou Zhewen Xiong Hanyong Sun Weiqin Yang Myth T.S.Mok Jing Wang Jingqing Li Man Liu Wenshu Tang Yu Feng Hector Kwong-Sang W ang Shun-Wa Tsang King-Lau Chow Philip Chun Yeung John Wong Paul Bo-San Lai Anthony Wing-Hung Chan Ka Fai To Stephen Lam Chan Qiang Xia Jing Xue Xiao Chen Jun Yu Sui Peng Joseph Jao-Yiu Sung Ming Kuang Alfred Sze-Lok Cheng | 2021 | Cellular & Molecular Immunology2021,18,4: | 5 |

| 4 | Evaluation of multidecadal and longer-term temperature changes since 850 CE based on Northern Hemisphere proxy-based reconstructions and model simulations显示文摘The temperature variability over multidecadal and longer timescales(e.g., the cold epochs in the late 15 th, 17 th, and early 19 th centuries) is significant and dominant in the millennium-long, large-scale reconstructions and model simulations;however, their temporal patterns in the reconstructed and simulated temperature series are not well understood and require a detailed assessment and comparison. Here, we compare the reconstructed and simulated temperature series for the Northern Hemisphere(NH) at multidecadal and longer-term timescales(>30 years) by evaluating their covariance, climate sensitivity and amplitude of temperature changes. We found that covariances between different reconstructions or between reconstructions and simulations are generally high for the whole period of 850–1999 CE, due to their similar long-term temporal patterns. However,covariances between different reconstructions or between reconstructions and simulations steadily decline as time series extends further back in time, becoming particularly small during Medieval times. This is related to the large uncetainties in the reconstructions caused by the decreased number of proxy records and sample duplication during the pre-instrumental periods.Reconstructions based solely on tree-ring data show higher skill than multiproxy reconstructions in capturing the amplitude of volcanic cooling simulated by models. Meanwhile, climate models have a shorter recovery(i.e., lag) in response to the cooling caused by volcanic eruptions and solar activity minima, implying the lack of some important feedback mechanisms between external forcing and internal climate processes in climate models. Amplitudes of temperature variations in the latest published tree-ring reconstructions are comparable to those of the multiproxy reconstructions. | 7 | A genome wide association study between copy number variation (CNV) and human height in Chinese population显示文摘Copy number variation (CNV) is a type of genetic variation which may have important roles in phenotypic variability and disease susceptibility. To hunt for genetic variants underlying human height variation, we performed a genome wide CNV association study for human height in 618 Chinese unrelated subjects using Affymetrix 500K array set. After adjusting for age and sex, we found that four CNVs at 6p21.3, 8p23.3-23.2, 9p23 and 16p12.1 were associated with human height (with borderline significant p value: 0.013, 0.011, 0.024, 0.049; respectively). However, after multiple tests correction, none of them was associated with human height. We observed that the gain of copy number (more than 2 copies) at 8p23.3-23.2 was associated with lower height (normal copy number vs. gain of copy number: 161.2 cm vs. 153.7 cm, p = 0.011), which accounted for 0.9% of height variation. Loss of copy number (less than 2 copies) at 6p21.3 was associated with 0.8% lower height (loss of copy number vs. normal copy number: 154.5 cm vs. 161.1 cm, p = 0.013). Since no important genes influencing height located in CNVs at loci of 8p23.3-23.2 and 6p21.3, the two CNVs may cause the structural rear- rangements of neighbored important candidate genes, thus regulates the variation of height. | 15 | Peroxymonosulfate activation based on Co_(9)S_(8)@N-C:A new strategy for highly efficient hydrogen production and synchronous formaldehyde removal in wastewater显示文摘Formaldehyde(FA),as an important chemical raw material,has been widely used in many fields.However,the discharge of a large amount of FA-containing wastewater poses a serious threat to the environment and human health.Recently,the in-situ hydrogen energy release technology of hydrogen-containing stable liquid has been extensively explored due to its safe storage.Exploring a robust method to achieve FA removal and synchronous in-situ hydrogen release from FA containing wastewater is of great significant for environmental protection and energy crisis alleviation.Here,we have innovatively introduced peroxymonosulfate(PMS)activation technology into FA removal and hydrogen production simultaneously.The composite of nitrogen doped carbon coating Co_(9)S_(8)nanotubes(Co_(9)S_(8)@N-C)is employed as a proof of concept for FA decomposition and simultaneously hydrogen production based on PMS activation system.As expected,the Co_(9)S_(8)@N-C/PMS system presents much superior hydrogen production efficiency and satisfactory FA removal rate towards FA wastewater than those of common catalysis,photocatalysis and Fenton reaction in the basic condition in a wide range of FA concentration.The hydrogen yield reaches a value as high as 471μmol within 60 min,corresponding to a FA degradation rate of 30%with an initial FA concentration of 0.722 mol L^(-1).Characterizations and density functional theory(DFT)calculations suggest that the free radical process dominated by superoxide radical(O_(2)·^(-))and nonradical process dominated by singlet oxygen(^(1)O_(2)),which are induced by Co_(9)S_(8)@N-C/PMS system,are responsible for highly efficient hydrogen production via FA degradation.These generated O_(2)·^(-)and ^(1)O_(2)can extract·H from FA to form·OOH intermediate,which can further combine with the·H from water to produce hydrogen.This study provides an applicable technique for environmental purification and new energy development based on FA containing wastewater. | Caiyan Gao Xuezhen Feng Lian Yi Xiaoyong Wu Renji Zheng Gaoke Zhang Yubiao Li | 2022 | Journal of Materials Science & Technology2022,,32: | 0 |
