| 4 | Alternations in the gut microbiota and metabolome with newly diagnosed unstable angina显示文摘Gut microbiota plays an important role in coronary heart disease, but its compositional and functional changes in unstable angina(UA) remain unexplored. We performed metagenomic sequencing of 133 newly diagnosed UA patients and 133 sex-and age-matched controls, and profiled the fecal and plasma metabolomes in 30 case-control pairs. The alpha diversity of gut microbiota was increased in UA patients:the adjusted odds ratios(ORs) per standard deviation increase in Shannon and Simpson indices were 1.30(95% confidence interval, 1.01-1.70) and 1.36(1.05-1.81), respectively. Two common species(depleted Klebsiella pneumoniae and enriched Streptococcus parasanguinis;P ≤ 0.002) and three rare species(depleted Weissella confusa, enriched Granulicatella adiacens and Erysipelotrichaceae bacterium 6_1_45;P ≤ 0.005) were associated with UA. The UA-associated gut microbiota was depleted in the pathway of Lphenylalanine degradation(P = 0.001), primarily contributed by Klebsiella pneumoniae. Consistently, we found increased circulating phenylalanine in UA patients(OR = 2.76 [1.17-8.16]). Moreover, Streptococcus parasanguinis was negatively correlated with fecal citrulline(Spearman's r_(s)=-0.470, P = 0.009), a metabolite depleted in UA patients(OR = 0.26 [0.08-0.63]). | 5 | Construction of a universal recombinant expression vector that regulates the expression of human lysozyme in milk显示文摘The mammary gland provides a novel method for producing recombinant proteins in milk of transgenic animals. A key component in the technology is the construction of an efficient milk expression vector. Here,we established a simple method to construct a milk expression vector, by a combination of homologous recombination and digestion-ligation. Our methodology is expected to have the advantages of both plasmid and bacterial artificial chromosome(BAC) vectors. The BAC of mouse whey acidic protein gene(mWAP) was modified twice by homologous recombination to produce a universal expression vector, and the human lysozyme gene(hLZ) was then inserted into the vector by a digestionligation method. The final vector containing the 8.5 kb mWAP 5′ promoter, 4.8 kb h LZ genomic DNA, and 8.0 kb m WAP 3′ genomic DNA was microinjected into pronuclei of fertilized mouse embryos, to successfully generate two transgenic mouse lines that expressed recombinant human lysozyme(rhLZ) in milk. The highest expression level of rhLZ was 0.45 g$L–1, and rhLZ exhibited the same antibacterial activity as native h LZ. | 6 | Bivariate genome-wide association study suggests that the DARC gene influences lean body mass and age at menarche显示文摘Lean body mass(LBM) and age at menarche(AAM) are two important complex traits for human health.The aim of this study was to identify pleiotropic genes for both traits using a powerful bivariate genome-wide association study(GWAS).Two studies,a discovery study and a replication study,were performed.In the discovery study,909622 single nucleotide polymorphisms(SNPs) were genotyped in 801 unrelated female Han Chinese subjects using the Affymetrix human genome-wide SNP array 6.0 platform.Then,a bivariate GWAS was performed to identify the SNPs that may be important for LBM and AAM.In the replication study,significant findings from the discovery study were validated in 1692 unrelated Caucasian female subjects.One SNP rs3027009 that was bivariately associated with left arm lean mass and AAM in the discovery samples(P=7.26×10 6) and in the replication samples(P=0.005) was identified.The SNP is located at the upstream of DARC(Duffy antigen receptor for chemokines) gene,suggesting that DARC may play an important role in regulating the metabolisms of both LBM and AAM. | HAI Rong ZHANG Lei PEI YuFang ZHAO LanJuan RAN Shu HAN YingYing ZHU XueZhen SHEN Hui TIAN Qing DENG HongWen | 2012 | Science China(Life Sciences)2012,55,6: | 0 |
