1Hantavirus infection:a global zoonotic challenge显示文摘Hantaviruses are comprised of tri-segmented negative sense single-stranded RNA, and are members of the Bunyaviridae family. Hantaviruses are distributed worldwide and are important zoonotic pathogens that can have severe adverse effects in humans. They are naturally maintained in specific reservoir hosts without inducing symptomatic infection. In humans, however,hantaviruses often cause two acute febrile diseases, hemorrhagic fever with renal syndrome(HFRS) and hantavirus cardiopulmonary syndrome(HCPS). 2Stereotactic aspiration for hypertensive intracerebral haemorrhage in a Chinese population:a retrospective cohort study显示文摘Objective We aimed to compare the therapeutic effects of stereotactic aspiration and best medical management in patients who developed supratentorial hypertensive intracerebral haemorrhage(HICH)with a volume of haemorrhage between 20 and 40 mL.Methods The clinical data of 220 patients with supratentorial HICH with a volume between 20 and 40 mL were retrospectively analysed.Among them,142 received stereotactic aspiration surgery(stereotactic aspiration group)and 78 received best medical management(conservative group).All were followed up for 6 months.Multivariate logistic regression and Kaplan-Meier survival curves were used to compare the outcome between the two groups.results The rebleeding rate was lower in the group that had stereotactic aspiration when compared with the group with medical treatment(6[4.2%]vs 9[11.5%],χ2=4.364,p=0.037).After 6 months,although the mortality rate did not differ significantly between the two groups(8 cases[5.6%]vs 10 cases[12.8%],χ2=3.461,p=0.063),the rate of a favourable outcome was higher in the group who received stereotactic aspiration(χ2=15.870,p=0.000).Logistic regression identified that medical treatment(OR=1.64,p=0.000)was an independent risk factor for an unfavourable outcome.The Kaplan-Meier curves indicated that the median favourable outcome time in the stereotactic aspiration group was 59.5 days compared with that in the medically treated group(87.0 days).The log-rank test indicated that the prognosis at 6 months was better for those treated with stereotactic haematoma aspiration(χ2=29.866,p=0.000).However,the 6-month survival rate was similar between the two groups(χ2=3.253,p=0.068).Conclusions Stereotactic haematoma aspiration significantly improved the quality of life,although did not effectively reduce the rate of mortality.When selected appropriately,patients with HICH may benefit from this type of surgical intervention.Xuyang Zhang Shaolong Zhou Qiang Zhang Xudong Fu Yuehui Wu Jiasheng Liu Bo Liang Zhuo Yang Xinjun Wang 2019Stroke & Vascular Neurology2019,4,1:19

4Hepatitis B virus X protein induces hepatic stem cell-like features in hepatocellular carcinoma by activating KDM5B显示文摘AIM To determine the role of hepatitis B virus X protein(HBx), HBx in regulating hepatic progenitor cell(HPC)-like features in hepatocellular carcinoma(HCC) and the underlying molecular mechanisms.METHODS We used a retrovirus vector to introduce wild type HBx or empty vector into Hep G2 cells. We then used these cells to analyze cell proliferation, senescence, transformation, and stem-like features. Gene expression profiling was carried out on Affymetrix GeneC hip Human U133A2.0 ver.2 arrays according to the manufacturer's protocol. Unsupervised hierarchical clustering analysis and Class Comparison analysis were performed by BRB-Array Tools software Version 4.2.2. A total of 238 hepatitis B virus(HBV)-related HCC patients' array data were used for analyzing clinical features.RESULTS The histone demethylase KDM5 B was significantlyhighly expressed in HBV-related HCC cases(P < 0.01). In HBV proteins, only HBx up-regulated KDM5 B by activating c-myc. Hepatic stem cell(Hp SC) markers(EpC AM, AFP, PROM1, and NANOG) were significantly highly expressed in KDM5B-high HCC cases(P < 0.01). KDM5 B played an important role in maintaining HpS Clike features and was associated with a poor prognosis. Moreover, inhibition of KDM5 B suppressed spheroid formation and cell invasion in vitro.CONCLUSION HBx activates the histone demethylase KDM5 B and induces HPC-like features in HCC. 5A UV-B-responsive glycosyltransferase,OsUGT706C2,modulates flavonoid metabolism in rice显示文摘Although natural variations in rice flavonoids exist, and biochemical characterization of a few flavonoid glycosyltransferases has been reported, few studies focused on natural variations in tricin-lignan-glycosides and their underlying genetic basis. In this study, we carried out metabolic profiling of tricin-lignan-glycosides and identified a major quantitative gene annotated as a UDPdependent glycosyltransferase OsUGT706C2 by metabolite-based genome-wide association analysis. The putative flavonoid glycosyltransferase OsUGT706C2 was characterized as a flavonoid 7-O-glycosyltransferas in vitro and in vivo. Although the in vitro enzyme activity of OsUGT706C2 was similar to that of OsUGT706D1, the expression pattern and induced expression profile of OsUGT706C2 were very different from those of OsUGT706D1. Besides, OsUGT706C2 was specifically induced by UV-B. Constitutive expression of OsUGT706C2 in rice may modulate phenylpropanoid metabolism at both the transcript and metabolite levels. Furthermore, overexpressing OsUGT706C2 can enhance UV-B tolerance by promoting ROS scavenging in rice. 7Personalized neoantigen pulsed dendritic cell vaccine for advanced lung cancer显示文摘Neoantigens are considered to be ultimate target of tumor immunotherapy due to their high tumor specificity and immunogenicity.Dendritic cell(DCs)vaccines based on neoantigens have exciting effects in treatment of some malignant tumors and are a promising therapeutic modality.Lung cancer is a lethal disease with the highest morbidity and mortality rate in the world.Despite the rapid development of targeted therapy and immune checkpoint inhibitors for lung cancer in recent years,their efficacy is still unsatisfactory overall.Therefore,there is an urgent unmet clinical need for lung cancer treatment.Here,we attempted to treat lung cancer using a personalized neoantigen peptide-pulsed autologous DC vaccine and conducted a single-arm,2 medical centers,pilot study initiated by the investigator(ChiCTR-ONC 16009100,NCT02956551).The patients enrolled were patients with heavily treated metastatic lung cancer.Candidate neoantigens were derived from whole-exome sequencing and RNA sequencing of fresh biopsy tissues as well as bioinformatics analysis.A total of 12 patients were enrolled in this study.A total of 85 vaccine treatments were administered with a median value of 5 doses/person(range:3-14 doses/person).In total,12-30 peptide-based neoantigens were selected for each patient.AIl treatment-related adverse events were grade 1-2 and there were no delays in dosing due to toxic effects.The objective effectiveness rate was 25%;the disease control rate was 75%;the median progression-free survival was 5.5 months and the median overall survival was 7.9 months.This study provides new evidence for neoantigen vaccine therapy and new therapeutic opportunities for lung cancer treatment.Zhenyu Ding Qing Li Rui Zhang Li Xie Yang shu Song Gao Peipei Wang Xiaoqing Su Yun Qin Yuelan Wang Juemin Fang Zhongzheng Zhu Xuyang Xia Guochao Wei Hui Wang Hong Qian Xianling Guo Zhibo Gao Yu Wang Yuquan Wei Qing Xu Heng Xu Li Yang 2021Signal Transduction and Targeted Therapy2021,6,2:3

11Machine Learning Approach to Enhance the Performance of MNP?Labeled Lateral Flow Immunoassay显示文摘The use of magnetic nanoparticle(MNP)-labeled immunochromatography test strips(ICTSs) is very important for point-ofcare testing(POCT). However, common diagnostic methods cannot accurately analyze the weak magnetic signal from ICTSs, limiting the applications of POCT. In this study, an ultrasensitive multiplex biosensor was designed to overcome the limitations of capturing and normalization of the weak magnetic signal from MNPs on ICTSs. A machine learning model for sandwich assays was constructed and used to classify weakly positive and negative samples, which significantly enhanced the specificity and sensitivity. The potential clinical application was evaluated by detecting 50 human chorionic gonadotropin(HCG) samples and 59 myocardial infarction serum samples. The quantitative range for HCG was 1–1000 mIU mL^(-1) and the ideal detection limit was 0.014 mIU mL^(-1), which was well below the clinical threshold. Quantitative detection results of multiplex cardiac markers showed good linear correlations with standard values. 12Handling OOV Words in Mandarin Spoken Term Detection with an Hierarchical n-Gram Language Model显示文摘In this paper, an hierarchical n-gram Language model(LM) combining words and characters is explored to improve the detection of Out-of-vocabulary(OOV) words in Mandarin Spoken term detection(STD).The hierarchical LM is based on a word-level LM, with a character-level LM estimating probabilities of OOV words in a class-based way. The region containing OOV words in the sentence to be decoded is detected with the help of the word-level LM and the probabilities of OOV words are derived from the character-level LM. The implementation of the proposed approach is based on a dynamic decoder. The proposed approach is evaluated in terms of Actual term weighted value(ATWV) on two Mandarin data sets. Experiment results show that more than 10% relative improvement for OOV word detection is achieved on both sets. 14A writable lithium metal ink显示文摘The recent revival of Li metal anodes(LMA)leads to a renewed interest in LMA as the ultimate choice for rechargeable lithium batteries towards high energy density.However,multiple challenges stand in the way of using LMA,of which high reactivity,dendrite growth,the difficulty of fabricating Li thin foils,and the flammability of organic liquid electrolytes are typical.Here,a writable Li metal ink(LMI)prepared by introducing biomass-derived carbon particles into molten Li is presented.Due to the significantly decreased surface tension,LMI is able to directly write on copper foils or other substrates that ultrathin Li foils with a remarkably small thickness(<10μm)can be achieved.The versatility of LMI is further demonstrated in addressing the interface issue between LMA and garnet-type solid-state electrolytes,where directly writing LMI on the garnet offers a perfect contact and enables an extremely low interfacial resistance of 6Ωcm^2,in sharp contrast to 939Ωcm^2 between the pure Li and the garnet.Due to the successful partnership with non-flammable solid-state electrolytes,ink-based technology may have a chance to bring us very close to the use of solid-state lithium metal batteries(SSLMBs)with high safety and high energy density.Wangyan Wu Jian Duan Jiayun Wen Yuwei Chen Xuyang Liu Liqiang Huang Zhengfeng Wang Shengyuan Deng Yunhui Huang Wei Luo 2020Science China Chemistry2020,63,10:2
15High-efficiency Gaussian key reconciliation in continuous variable quantum key distribution显示文摘Efficient reconciliation is a crucial step in continuous variable quantum key distribution. The progressive-edge-growth(PEG) algorithm is an efficient method to construct relatively short block length low-density parity-check(LDPC) codes. The qua-sicyclic construction method can extend short block length codes and further eliminate the shortest cycle. In this paper, by combining the PEG algorithm and quasi-cyclic construction method, we design long block length irregular LDPC codes with high error-correcting capacity. 16Modifed Pre-stretching Assembly Method for Cable-Driven Systems显示文摘Soft cable-driven systems have been employed in many assembled mechanisms, such as industrial robots, parallel kinematic mechanism machines, medical devices, and humaniform hands. A pre-stretching process is necessary to guarantee the quality of cable-driven systems during the assembly process. However, the stress relaxation of cables becomes a critical concern during long-term operation. This study investigates the effects of non-uniform deformation and long-term stress relaxation of the driven cables owing to moving parts in the system. A simple closed-loop cable-driven system is built and an alternating load is applied to it to replicate the operation of transmission cables. Under different experimental conditions, the cable tension is recorded and the boundary data are selected to be curve-fitted. Based on the fitted results, a formula is presented to estimate the stress relaxation of cables to evaluate the assembly performance. Further experimental results show that the stress relaxation is mainly caused by cable creep and the assembly procedure. To remove the influence of the assembly procedure, a modified pre-stretching assembly method based on the stress relaxation theory is proposed and verification experiments are performed. Finally, the assembly performance is optimized using a cable-driven surgical robot as an example. 17The effects of circadian rhythm on catechin accumulation in tea leaves显示文摘As the major bitter-and astringent-tasting components in tea leaves,catechins play an essential role in determining flavor characteristics and health benefits of tea products.This study analyzed the circadian rhythm effects on the accumulation pattern of catechins in tender leaves of three tea cultivars,'Huangkui'(HK),'Longjing43'(LJ43)and'Fuding Dabaicha'(FD).There were two major factors involved in the catechin regulation in tea plants:genetic background of the cultivar-specific regulation,and temporal effects of the circadian rhythm.Etiolated leaves of'HK'contained lower levels of catechins than'LJ43'and'FD'.The composition of EGCG in total catechins was significantly higher in'LJ43'leaves(65%)when compared with that in'HK'and'FD'(45%).The accumulation of individual catechins increased during the daytime but decreased overnight,especially over the time period of 22:00−02:00.The correlation analysis between catechins and environmental factors indicated catechin accumulation in tea leaves potentially resulted from a combined regulation of light and temperature.These findings provide new insights into our understanding of circadian rhythm regulation on flavor-important secondary metabolites in tea leaves.Xiaohui Wang Wenfeng Zhu Xin Cheng Zhiqiang Lu Xuyang Liu Xiaochun Wan Chuankui Song Linlin Liu 2021Beverage Plant Research2021,1,1:2

19Effects of blade aspect ratio and taper ratio on hovering performance of cycloidal rotor with large blade pitching amplitude显示文摘In recent years, a lot of research work has been carried out on the cycloidal rotors. However, it lacks thorough understanding about the effects of the blade platform shape on the hover efficiency of the cycloidal rotor, and the knowledge of how to design the platform shape of the blades. This paper presents a numerical simulation model based on Unsteady ReynoldsAveraged Navier–Stokes equations(URANSs), which is further validated by the experimental results. The effects of blade aspect ratio and taper ratio are analyzed, which shows that the cycloidal rotors with the same chord length have quite similar performance even though the blade aspect ratio varies from a very small value to a large one. By comparing the cycloidal rotors with different taper ratios, it is found that the rotors with large blade taper ratio outperform those with small taper ratio. This is due to the fact that the blade with larger taper ratio has longer chord and hence better efficiency. The analysis results show that the unsteady aerodynamic effects due to blade pitching motion play a more important role in the efficiency than the blade platform shape. 20DEPENDENCE OF TROPICAL CYCLONE INTENSIFICATION ON THE CORIOLIS PARAMETER显示文摘The dependence of tropical cyclone(TC) intensification on the Coriolis parameter was investigated in an idealized hurricane model. By specifying an initial balanced vortex on an f-plane, we observed faster TC development under lower planetary vorticity environment than under higher planetary vorticity environment. The diagnosis of the model outputs indicates that the distinctive evolution characteristics arise from the extent to which the boundary layer imbalance is formed and maintained in the presence of surface friction. Under lower planetary vorticity environment, stronger and deeper subgradient inflow develops due to Ekman pumping effect, which leads to greater boundary layer moisture convergence and condensational heating. The strengthened heating further accelerates the inflow by lowing central pressure further. This positive feedback loop eventually leads to distinctive evolution characteristics.The outer size(represented by the radius of gale-force wind) and the eye of the final TC state also depend on the Coriolis parameter. The TC tends to have larger(smaller) outer size and eye under higher(lower) planetary vorticity environment.
