| 1 | Prioritizing natural-selection signals from the deep-sequencing genomic data suggests multi-variant adaptation in Tibetan highlanders显示文摘Human genetic adaptation to high altitudes(>2500 m)has been extensively studied over the last few years,but few functional adaptive genetic variants have been identified,largely owing to the lack of deep-genome sequencing data available to previous studies.Here,w e build a list of putative adaptive variants,including 63 missense,7 loss-of-function;1,298 evolutionarily conserved variants and 509 expression quantitative traits loci.Notably,the top signal of selection is located in TMEM 247,a transmembrane protein-coding gene.The Tibetan version of TMEM 247 harbors one high-frequency(76.3%)missense variant,rsl 16983452(c.248C>T;p.Ala83Val);with the T allele derived from archaic ancestry and carried by>94%of Tibetans but absent or in low frequencies(<3%)in non-Tibetan populations.The rsl 16983452-T is strongly and positively correlated with altitude and significantly associated with reduced hemoglobin concentration(p=5.78×10^-5),red blood cell count(p=5.72×10^-7)and hematocrit(p=2.57×10^-6).In particular,TMEM247-rs 116983452 shows greater effect size and better predicts the phenotypic outcome than any E P A S1 variants in association with adaptive traits in Tibetans.Modeling the interaction between TM£M247-rsl 16983452 and EPAS1 variants indicates weak but statistically significant epistatic effects.Our results support that multiple variants may jointly deliver the fitness of the Tibetans on the plateau,where a complex model is needed to elucidate the adaptive evolution mechanism. | Lian Deng Chao Zhang Kai Yuan Yang Gao Yuwen Pan Xueling Ge Yaoxi He Yuan Yuan Yan Lu Xiaoxi Zhang Hao Chen Haiyi Lou Xiaoji Wang Dongsheng Lu Jiaojiao Liu Lei Tian Qidi Feng Asifullah Khan Yajun Yang Zi-Bing Jin Jian Yang Fan Lu Jia Qu Longli Kang Bing Su Shuhua Xu | 2019 | National Science Review2019,6,6: | 9 |
| 2 | Follistatin-like protein 1 plays a tumor suppressor role in clear-cell renal cell carcinoma显示文摘Background:We previously showed that the expression of follistatin-like protein 1(FSTL1)was significantly down-regulated in metastatic clear-cell renal cell carcinoma(ccRCC).In this study,we aimed to characterize the role of FSTL1 in the development of ccRCC.Methods:The effects of FSTL1 on cell activity and cell cycle were investigated in ccRCC cell lines with altered FSTL1 expression.Gene expression microarray assays were performed to identify the major signaling pathways affected by FSTL1 knockdown.The expression of FSTL1 in ccRCC and its effect on postoperative prognosis were estimated in a cohort with 89 patients.Results:FSTL1 knockdown promoted anchorage-independent growth,migration,invasion,and cell cycle of ccRCC cell lines,whereas FSTL1 overexpression attenuated cell migration.FSTL1 knockdown up-regulated nuclear factor-κB(NF-κB)and hypoxia-inducible factor(HIF)signaling pathways,increased epithelial-to-mesenchymal transition,up-regulated interleukin-6 expression,and promoted tumor necrosis factor-α-induced degradation of NF-κB inhibitor(IκBα)in ccRCC cell lines.FSTL1 immunostaining was selectively positive in epithelial cytoplasm in the loop of Henle,and positive rate of FSTL1 was significantly lower in ccRCC tissues than in adjacent renal tissues(P<0.001).The mul-tivariate Cox regression analysis showed that the intratumoral FSTL1 expression conferred a favorable independent prognosis with a hazard ratio of 0.325(95%confidence interval 0.118-0.894).HIF-2αexpression was negatively cor-related with FSTL1 expression in ccRCC specimens(r=−0.229,P=0.044).Intratumoral expression of HIF-2α,rather than HIF-1α,significantly predicted an unfavorable prognosis in ccRCC(log-rank,P=0.038).Conclusions:FSTL1 plays a tumor suppression role possibly via repressing the NF-κB and HIF-2αsignaling pathways.To increase FSTL1 expression might be a candidate therapeutic strategy for metastatic ccRCC. | Yan Liu Xiaojie Tan Wenbin Liu Xi Chen Xiaomei Hou Dan Shen Yibo Ding Jianhua Yin Ling Wang Hongwei Zhang Yongwei Yu Jianguo Hou Timothy C.Thompson Guangwen Cao | 2018 | Cancer Communications2018,38,1: | 5 |
| 3 | OsPKpa_1 encodes a plastidic pyruvate kinase that affects starch biosynthesis in the rice endosperm显示文摘Pyruvate kinase(PK) is a key enzyme in glycolysis and carbon metabolism. Here, we isolated a rice(Oryza sativa) mutant, w59, with a white-core floury endosperm. Map-based cloning of w59 identified a mutation in OsPKpa_1, which encodes a plastidic isoform of PK(PKp). OsPKpa_1 localizes to the amyloplast stroma in the developing endosperm, and the mutation of OsPKpa_1 in w59 decreases the plastidic PK activity, resulting in dramatic changes to the lipid biosynthesis in seeds. The w59 grains were also characterized by a marked decrease in starch content. Consistent with a decrease in number and size of the w59 amyloplasts, large empty spaces were observed in the central region of the w59 endosperm, atthe early grain-filling stage. Moreover, a phylogenetic analysis revealed four potential rice isoforms of OsPKp. We validated the in vitro PK activity of these OsPKps through reconstituting active PKp complexes derived from inactive individual OsPKps, revealing the heteromeric structure of rice PKps, which was further confirmed using a protein–protein interaction analysis. | 4 | YR36/WKS1-Mediated Phosphorylation of PsbO,an Extrinsic Member of Photosystem Ⅱ,Inhibits Photosynthesis and Confers Stripe Rust Resistance in Wheat显示文摘Wheat stripe rust,due to infection by Puccinia striiformis f.sp.tritici(Pst),is a devastating disease that causes significant global grain yield losses.Yr36,which encodes Wheat Kinase START1(WKS1),is an effec-tive high-temperature adult-plant resistance gene and confers resistance to a broad spectrum of Pst races.We previously showed that WKS1 phosphorylates the thylakoid ascorbate peroxidase protein and reduces its ability to detoxify peroxides,which may contribute to the accumulation of reactive oxygen species(ROS).WKS1-mediated Pst resistance is accompanied by leaf chlorosis in Pst-infected regions,but the un-derlying mechanisms remain elusive.Here,we show that WKS1 interacts with and phosphorylates PsbO,an extrinsic member of photosystem Ⅱ(PS Ⅱ),to reduce photosynthesis,regulate leaf chlorosis,and confer Pst resistance.A point mutation in PsbO-A1 or reduction in its transcript levels by RNA interference resulted in chlorosis and reduced Pst sporulation.Biochemical analyses revealed that WKS1 phosphorylates PsbO at two conserved amino acids involved in physical interactions with PS Ⅱ and reduces the binding affinity of PsbO with PS Ⅱ.Presumably,phosphorylated PsbO proteins dissociate from the PS Ⅱ complex and then un-dergo rapid degradation by cysteine and aspartic proteases.Taken together,these results demonstrate that perturbations of wheat PsbO by point mutation or phosphorylation by WKS1 reduce the rate of photo-synthesis and delay the growth of Pst pathogen before the induction of ROS. | Shuai Wang Qiu-Ping Li Jianfeng Wang Yan Yan Guo-Liang Zhang Yan Yan Huifei Zhang Jiajie Wu Feng Chen Xiaojie Wang Zhensheng Kang Jorge Dubcovsky Jin-Ying Gou | 2019 | Molecular Plant2019,12,12: | 3 |
| 5 | The Effect of Unannounced Inspection on Prevention of Drug Fraud显示文摘Alarge number of incidents related to fake/inferior-quality drugs have occurred in China in recent years. In order to reduce drug crimes, the China Food and Drug Administration (CFDA) has exploited the new supervision approach, i.e., unannounced inspection (UI), since September 2014. However, the effectiveness of UI in driving drug producers to put more effort into meeting the requirements of Good Manufacturing Practice (GMP) is yet to be ascertained. In this paper we apply game theory to study the effects of UI on the prevention of drug fraud, drug producers’ profits, and social welfare under scenarios of complete and incomplete information. We show that UI is not always an effective way of supervision. Specifically, if the upper bound on the punishment is not very large, UI can drive the firmto make the largest self-supervision effort only when the firm’s technical level is high and the supervision cost is low. Otherwise, UI cannot drive the firm to make the largest self-supervision effort. Under incomplete information, firms with a high technical level would adopt more self-supervision and make more profit than those with a low technical level. | 7 | Bioactivity and constituents of several common seaweeds显示文摘Bioactivity and constituents of 8 common seaweeds from Dalian intertidal zone of northern Yellow Sea were investigated. In the anti-methicillin-resistant Staphylococcus aureus (MRSA) test, Symphyocladia latiuscula and Enteromorpha intestinalis showed obvious activities with MICs much lower than 1.0 mg·mL-1 . In the DNA damage repair test (DDRT), Chondrus ocellatu showed selective inhibitory activity against the DNA repair-defective E. coli strain vs. the wild-type E. coli strain; while Sym. latiuscula, Enteromorpha intestinalis and Sar. kjellmanianum showed significant anti-E. coli activity with MICs of 64-128 g·mL-1 . In the anti-Pyricularia oryzae test, Sym. latiuscula and Rh. confervoides strongly inhibited the germination of the spores of P. oryzae on agar plate. In the brine shrimp larvicidal test, Sym. latiuscula, Rh. confervoides and Sar. kjellmanianum exhibited potent toxicity against brine shrimp larvae, with LC50 much lower than 1 mg·mL-1 . The HPTLC analysis revealed their diversified secondary metabolites. The HPLC-DAD-MS analysis of the strongest species Sym. latiuscula and database searching showed that it can produce quite diversified metabolites, including halogenated ones, some of which may be new natural products. | 19 | Rational design of robust iridium-ceria oxide-carbon nanofibers to boost oxygen evolution reaction in both alkaline and acidic media显示文摘Anodic oxygen evolution reaction(OER)is essential to participate in diverse renewable energy conversion and storage processes,while most OER electrocatalysts present satisfactory catalytic performance in only alkaline or acidic medium,limiting their practical applications in many aspects.Herein,we have designed and prepared Ir-CeO_(2)-C nanofibers(NFs)via an electrospinning and a relatively low-temperature calcination strategy for OER application in both alkaline and acidic conditions.Density functional theory(DFT)simulations demonstrate the high catalytic active sites of Ir atoms for OER,that the formation of Ir–O bonds at the interface between Ir and CeO_(2)can modulate the electron density of the relevant Ir atoms to promote the OER activity.In addition,the unique nanofibrous heterostructure increases the exposed active sites and promotes the electrical conductivity.Therefore,the prepared Ir-CeO_(2)-C nanofibrous catalyst delivers an excellent OER property in both alkaline and acidic solutions.Impressively,the overpotentials to reach 10 mA·cm^(−2)are only 279 and 283 mV in the alkaline and acidic electrolyte,respectively,with favorable long-term stabilities.In addition,the two-electrode overall water splitting set-ups equipped with Ir-CeO_(2)-C NFs as anode and commercial Pt/C as cathode provide a cell voltage of 1.54 and 1.53 V to drive 10 mA·cm^(−2)in the alkaline and acidic electrolyte,respectively,which are much lower than Pt/C||IrO_(2)and lots of transition metal oxides-based electrolyzers.This research presents an efficient means to design OER catalysts with superior properties in both alkaline and acidic solutions. | Xiaojie Chen Wenying Liao Mengxiao Zhong Junjie Chen Su Yan Weimo Li Ce Wang Wei Chen Xiaofeng Lu | 2023 | Nano Research2023,16,5: | 1 |
