1Collagen-binding basic fibroblast growth factor improves functional remodeling of scarred endometrium in uterine infertile women: a pilot study显示文摘Intrauterine adhesion(IUA) is a common cause of uterine infertility and one of the most severe clinical features is endometrial fibrosis namely endometrial scarring for which there are few cures currently. Blocked angiogenesis is the main pathological change in the scarred endometrium. The fibroblast growth factor 2(b FGF), a member of FGF family, is usually applied to promote healing of refractory ulcer and contributes to angiogenesis of tissues. In this study, the sustained-release system of b FGF100 μg was administrated around scarred endometrium guiding by ultrasound every 4 weeks in 18 patients(2–4 times). Results showed that after treatment, the menstrual blood volume, endometrial thickness and the scarred endometrial area were improved.Histological study showed blood vessel density increased obviously. Three patients(3/18) achieved pregnancy over 20 gestational weeks. 2Adaptive robust control of nonholonomic systems with stochastic disturbances显示文摘This paper deals with nonholonomic systems in chained form with unknown covariance stochastic disturbances. The objective is to design the almost global adaptive asymptotical controllers in probability u0 and u1 for the systems by using discontinuous control. A switching control law u0 is designed to almost globally asymptotically stabilize the state x0 in both the singular x0 (t0)=0 case and the non-singular x0 (t0)≠0 case. Then the state scaling technique is introduced for the discontinuous feedback into the (x1, x2, …, xn)-subsystem. Thereby, by using backstepping technique the global adaptive asymptotical control law u1 has been presented for (x1, x2, …, xn) -subsystem for both different u0 in non-singular x0 (t0)≠0 case and the singular case x0 (t0)=0. 3Numerical simulation of cavitation turbulence in Francis turbine runner with splitter blades显示文摘Cavitation will reduce the turbine performance and even damage the turbine components.To verify the effects of splitter blades on improving the cavitation performance,the cavitation flow inside a Francis turbine runner with splitter blades was numerically simulated by using the Singhal cavitation model and the standard k-ε turbulence model.The distributions of static pressure and gas volume fractions on the surface of the runner blades were predicated under different conditions,and the cavitation in the flow field of the runner was analyzed.The results show that the static pressure and gas volume fractions are more uniformly distributed on the short blades than those on the long blades in Francis turbines with splitter blades,and there is almost no cavitation on the short blades;their distributions are more uniform under small flow conditions than those under large flow conditions;and large gas volume fractions are concentrated at the outlet tip near the band on the suction side of the long blade.The installation of splitter blades can improve the cavitation performance of conventional Francis turbines.WANG Huiyan LIU Xiaobing JIANG Qifeng HUA Hong OU Shunbing 2020排灌机械工程学报2020,38,1:2

14Gene regulatory network inference based on causal discovery integrating with graph neural network显示文摘Gene regulatory network (GRN) inference from gene expression data is asignificant approach to understanding aspects of the biological system.Compared with generalized correlation-based methods, causality-inspiredones seem more rational to infer regulatory relationships. We proposeGRINCD, a novel GRN inference framework empowered by graph representationlearning and causal asymmetric learning, considering both linearand non-linear regulatory relationships. First, high-quality representation ofeach gene is generated using graph neural network. Then, we apply theadditive noise model to predict the causal regulation of each regulator-targetpair. Additionally, we design two channels and finally assemble them forrobust prediction. Through comprehensive comparisons of our frameworkwith state-of-the-art methods based on different principles on numerousdatasets of diverse types and scales, the experimental results show that ourframework achieves superior or comparable performance under variousevaluation metrics. 17Zhenxin Anshen formula(镇心安神方)ameliorates atopic der-matitis-like skin dysfunction in mice and in vitro via regulation of transient receptor potential vanilloid 1 and transient receptor potential ankyrin 1 in Neural pathways显示文摘OBJECTIVE:To investigate the efficacy of Zhenxin Anshen formula(镇心安神方,ZXAS)on atopic dermatitis(AD)by transient receptor potential vanilloid 1(TRPV1)and transient receptor potential ankyrin 1(TRPA1)signalling pathway in mice and in vitro.METHODS:AD-like lesions were induced by 1-chloro-2,4-dinitrobenzene(DNCB)to the shaved dorsal skin of BALB/c mice.BALB/c mice were divided into five groups:normal control,model control,cetirizine,low-,medium-,and high-dose of ZXAS.After ZXAS in-tervention,the skin lesions and blood samples were collected for hematoxylin and eosin-stained and measuring the concentrations of inflammatory cytokines.Immunoglobulin E(IgE),interleukin(IL)-4,IL-5,IL-13,and thymic stromal lymphopoietin(TSLP)were de-tected by Enzyme-linked immunosorbent assay(ELISA).The spinal cords were collected for measuring the expression of gastrin-releasing peptide receptor(GRPR),TRPV1,and TRPA1 by using immunohistochemistry,western blotting,and quantitative real-time polymerase chain reaction(qR T-PCR)analyses.In addition,3-(4,5-dimethylthiazolyl-2)-2,5-diphenyltetrazolium bromide(MTT)assay,flow cytometry,ELISA,and Western blotting were conducted for analysis of primary dorsal root ganglia(DRG)neurons in vitro.RESULTS:ZXAS treatment improved DNCB-induced AD-like lesions through reducing dermatitis score,number of scratching and epidermal thickness,accompanied by the de-creased IgE and Th2 inflammatory cytokines.ZXAS also supressed the mRNA and protein expression of GRPR,TRPV1,and TRPA1 in the spinal cord.The medicated sera of ZXAS decreased capsaicin-induced Ca^(2+)influx and downregulated the expression of TRPV1,TRPA1,and phospholipase C in DRG neurons.CONCLUSIONS:The therapeutic effect of ZXAS on AD may be related to the regulation of TRPV1 and TRPA1 and inhibition of Ca^(2+)signals in neurons.ZHAO Yiding YAN Xiaoning JIANG Shanshan LIU Yong DONG Chun CHI Huiyan MAO Chaoyi 2023Journal of Traditional Chinese Medicine2023,43,5:0
18Expression of Fibroblast Growth Factor 21 in Escherichia coli Induced by Lactose显示文摘[Objective] This study aimed to investigate the expression of recombinant protein of fibroblast growth factor 21 (FGF21) in Escherichia coli induced by lactose and explore the possibility of engineering production.[Method] The effects of lactose concentration, induction duration, induction starting time, and induction temperature on expression of the recombinant product were compared, and the optimal condition for lactose induction was determined with IPTG induction as a control.[Result] As an induction agent, lactose plays an ideal role in the production of recombinant FGF21. In addition, the expression level of lactose induction product is consistent with that of IPTG induction product. Induction in a 250ml flask containing 50ml of medium at A600 0.8-1.0 using 0.5g/L lactose at 35℃ for 6h are the optimal condition.[Conclusion] Lactose can induce the expression of FGF21 gene and achieve ideal results.
