| 2 | 快速木质纤维素分解菌复合系MC1对秸秆的分解能力及稳定性显示文摘以天然水稻秸秆为材料研究了快速降解木质纤维素的细菌复合系MC1对木质纤维素的分解能力;并在不同条件保藏、高温处理以及利用变性梯度胶电泳(DGGE)技术研究了复合系的稳定性.结果表明,复合系MC1在50℃液体静止培养条件下8~10d,把培养液2%干重的水稻秸秆完全分解溶化;经过9d的培养,水稻秸秆的总干重减少81%,其中纤维素减少99%,半纤维素减少74%,木质素减少51%.连续继代培养4a、常温干燥保存4a、-20℃冷冻藏4a、培养液直接在室温和4℃保存1a、90℃处理30min仍具旺盛的分解能力并稳定传代.平板培养基培养证明MC1全部由细菌组成,16S rDNA变性梯度胶电泳(DGGE)检测结果,在6个月内主要条带几乎没有变化,说明MC1的菌种组成相当稳定.MC1对纤维素的分解利用具重要前景. | 王伟东 崔宗均 王小芬 牛俊玲 刘建斌 Igarashi Yasuo | 2005 | 环境科学2005,26,5: | 37 |
| 3 | Characterization of the effective cellulose degrading strain CTL-6显示文摘An efficient cellulose degrading bacteria exists in the thermophilic wheat straw-degrading community, WDC2. However, this strain cannot be isolated and cultured using conventional separation techniques under strict anaerobic conditions. We successfully isolated a strain of effective cellulose degrading bacteria CTL-6 using a wash, heat shock, and solid-liquid alternating process. Analysis of its properties revealed that, although the community containing the strain CTL-6 grew under aerobic conditions, the purified strain CTL-6 only grew under anaerobic culture conditions. The strain CTL-6 had a striking capability of degrading cellulose (80.9% weight loss after 9 days of culture). The highest efficiency value of the endocellulase (CMCase activity) was 0.404 μmol/(min·mL), cellulose degradation efficiency by CTL-6 was remarkably high at 50-65°C with the highest degradation efficiency observed at 60°C. The 16S rRNA gene sequence analysis indicated that the closest relative to strain CTL-6 belonged to the genus Clostridium thermocellum. Strain CTL-6 was capable of utilizing cellulose, cellobiose, and glucose. | 4 | 汞在微生物中的跨膜运输机制研究进展显示文摘甲基汞是一种强亲脂性、高神经毒性的有机汞化合物,可以通过生物富集或生物放大造成人类甲基汞暴露。环境中甲基汞的产生主要是厌氧微生物所调控的无机汞的甲基化。主流观点认为厌氧微生物对汞的甲基化是一种细胞内反应,因此,甲基汞的产生速率不仅与环境中具有汞甲基化能力的厌氧微生物的存在与活性相关,同时也与无机汞在微生物细胞中的跨膜运输过程有着重要联系。要明确无机汞经微生物甲基化的机制,就必须了解无机汞被微生物细胞生物吸收的过程,即无机汞在微生物中的跨膜运输路径。目前研究认为该过程主要有Mer抗汞操纵子转运体系、被动扩散、促进扩散和主动运输4种路径。本综述主要围绕无机汞被微生物细胞生物吸收的这4种路径展开,将系统介绍科学界对这4种路径的最新研究进展,并对相关研究进行展望,指出无机汞经促进扩散或主动运输进入到微生物细胞内将是未来研究的重点。 | 杜红霞 Yasuo Igarashi 王定勇 | 2014 | 微生物学报2014,54,10: | 5 |
| 5 | Y chromosome microdeletion screening using a new molecular diagnostic method in 1030 Japanese males with infertility显示文摘The azoospermia factor(AZF)region is important for spermatogenesis,and deletions within these regions are a common cause of oligozoospermia and azoospermia.Although several studies have reported this cause,the present research,to the best of our knowledge,is the first large-scale study assessing this factor in Japan.In this study,1030 male patients with infertility who were examined for Y chromosome microdeletion using the polymerase chain reaction-reverse sequence-specific oligonucleotide(PCR-rSSO)method,a newly developed method for Y chromosome microdeletion screening,were included.The study enrolled 250 patients with severe oligospermia and 717 patients with azoospermia.Among the 1030 patients,4,4,10,and 52 had AZFa,AZFb,AZFb+c,and AZFc deletions,respectively.The sperm recovery rate(SRR)of microdissection testicular sperm extraction in patients with AZFc deletions was significantly higher than that in those without AZF deletions(60.0%vs 28.7%,P=0.04).In patients with gr/gr deletion,SRR was 18.7%,which was lower than that in those without gr/gr deletion,but was not statistically significant.In conclusion,our study showed that the frequency of Y chromosome microdeletion in male patients in Japan was similar to that reported in patients from other countries,and SRR was higher in patients with AZFc deletion. | Masashi Iijima Kazuyoshi Shigehara Hideki Igarashi Koichi Kyono Yasuo Suzuki Yuji Tsuji Yoshitomo Kobori Hideyuki Kobayashi Atsushi Mizokami | 2020 | Asian Journal of Andrology2020,22,4: | 4 |
| 6 | 显示出与肝炎 B 病毒感染联系的微分表示的细胞的基因的鉴定显示文摘 AIM: To investigate the impact of hepatitis B virus (HBV) infection on cellular gene expression, by conducting both in vitro and in vivo studies. METHODS: Knockdown of HBV was targeted by stable expression of short hairpin RNA (shRNA) in huH-1 cells. Cellular gene expression was compared using a human 30K cDNA microarray in the cells and quantified by real-time reverse transcription-polymerase chain reaction (RT-PCR) (qRT-PCR) in the cells, hepatocellular carcinoma (HCC) and surrounding non-cancerous liver tissues (SL). RESULTS: The expressions of HBsAg and HBx protein were markedly suppressed in the cells and in HBx transgenic mouse liver, respectively, after introduction of shRNA. Of the 30K genes studied, 135 and 103 genes were identified as being down- and up-regulated, respectively, by at least twofold in the knockdown cells. Functional annotation revealed that 85 and 62 genes were classified into four up-regulated and five down-regulated functional categories, respectively. When gene expression levels were compared between HCC and SL, eight candidate genes that were confirmed to be up- or down-regulated in the knockdown cells by both microarray and qRT-PCR analyses were not expressed as expected from HBV reduction in HCC, but had similar expression patterns in HBV- and hepatitis C virus-associated cases.
