| 8 | Detection of genomic signatures for pig hairlessness using high-density SNP data显示文摘Hair provides thermal regulation for mammals and protects the skin from wounds,bites and ultraviolet(UV)radiation,and is important in adaptation to volatile environments.Pigs in nature are divided into hairy and hairless,which provide a good model for deciphering the molecular mechanisms of hairlessness.We conducted a genomic scan for genetically differentiated regions between hairy and hairless pigs using 60K SNP data,with the aim to better understand the genetic basis for the hairless phenotype in pigs.A total of 38405 SNPs in 498 animals from 36 diverse breeds were used to detect genomic signatures for pig hairlessness by estimating between-population(FST)values.Seven diversifying signatures between Yucatan hairless pig and hairy pigs were identified on pig chromosomes(SSC)1,3,7,8,10,11 and 16,and the biological functions of two notable genes,RGS17 and RB1,were revealed.When Mexican hairless pigs were contrasted with hairypigs,strong signatures were detected on SSC1 and SSC10,which harbor two functionally plausible genes,REV3L and BAMBI.KEGG pathway analysis showed a subset of overrepresented genes involved in the T cell receptor signaling pathway,MAPK signaling pathway and the tight junction pathways.All of these pathways may be important in local adaptability of hairless pigs.The potential mechanisms underlying the hairless phenotype in pigs are reported for the first time.RB1 and BAMBI are interesting candidate genes for the hairless phenotype in Yucatan hairless and Mexico hairless pigs,respectively.RGS17,REV3L,ICOS and RASGRP1 as well as other genes involved in the MAPK and T cell receptor signaling pathways may be important in environmental adaption by improved tolerance to UV damage in hairless pigs.These findings improve our understanding of the genetic basis for inherited hairlessness in pigs. | Ying SU Yi LONG Xinjun LIAO Huashui AI Zhiyan ZHANG Bin YANG Shijun XIAO Jianhong TANG Wenshui XIN Lusheng HUANG Jun REN Nengshui DING | 2014 | Frontiers of Agricultural Science and Engineering2014,1,4: | 0 |
| 9 | Bariatric surgery induces alterations in effective connectivity between the orbitofrontal cortex and limbic regions in obese patients显示文摘Obese subjects show enhanced brain responses in motivation and reward neurocircuitry encompassing sensory and somatic integration-interception,motivation-reward(striatal),emotion,and memory processes,which attenuate frontal region activation during food cues.Bariatric surgery(BS) is the only reliable treatment for morbid obesity.Unfortunately,it is unknown how BS affects neurocircuitry after weight loss.We aimed to examine effects of BS on the basal activity of brain areas involved in reward and motivation processing,emotion,memory,and gut-brain interaction.We combined resting-state fMRI with amplitude of low-frequency fluctuation(ALFF) and Granger causality analysis(GCA) to assess interactions between regions within the frontal-mesolimbic circuitry in 16 obese subjects(OB) and 22 normal-weight(NW) subjects.The OB group was studied at baseline and 1 month post BS.Comparisons between OB and NW,and preand post BS showed significant differences in ALFF in areas involved in drive(caudate,orbitofrontal cortex(OFC)),arousal(thalamus),and conditioning/memory(amygdala,hippocampus)(P < 0.05,FDR correction).GCA revealed that in the OB group,the OFC had greater connectivity to limbic regions(amygdala,hippocampus,and medial thalamus) and the caudate.Post BS,the connectivity of the OFC to limbic regions decreased,whereas the connectivity from the amygdala and hippocampus to the caudate and thalamus was enhanced,particularly in subjects with lower body mass index(BMI).OFC activation in the OB group was associated with BMI prior to surgery,and changes in OFC post surgery were associated with alterations in BMI.Overall,the functional connectivity of the OFC was significantly decreased.As it is important for salience attribution and connected to limbic brain regions involved with emotional reactivity and conditioning after BS,its significant association with BMI changes indicates the contribution of OFC changes to the improved control of eating behavior after surgery. | Shijun DUAN Gang JI Guanya LI Yang HU Wenchao ZHANG Jia WANG Dardo TOMASI Nora DVOLKOW Yongzhan NIE Guangbin CUI Gene-Jack WANG Yi ZHANG | 2020 | Science China(Information Sciences)2020,63,7: | 0 |

| 11 | Collagen triple helix repeat containing-1 promotes functional recovery of sweat glands by inducing adjacent microvascular network reconstruction in vivo显示文摘Background:Sweat glands(SGs)have low regenerative potential after severe burns or trauma and their regeneration or functional recovery still faces many obstacles.In practice,restoring SG function requires not only the structural integrity of the gland itself,but also its neighboring tissues,especially blood vessels.Collagen triple helix repeat containing-1(CTHRC1)was first identified in vascular repair,and increasing reports showed a close correlation between cutaneous appendage specification,patterning and regeneration.The purpose of the present study was to clarify the role of CTHRC1 in SGs and their adjacent microvessels and find therapeutic strategies to restore SG function.Methods:The SGs and their adjacent microvascular network of Cthrc^(1−/−)mice were first inves-tigated using sweat test,laser Doppler imaging,tissue clearing technique and transcriptome analysis.The effects of CTHRC1 on dermal microvascular endothelial cells(DMECs)were further explored with cell proliferation,DiI-labeled acetylated low-density lipoprotein uptake,tube for-mation and intercellular junction establishment assays.The effects of CTHRC1 on SG function restoration were finally confirmed by replenishing the protein into the paws of Cthrc(1−/−)mice.Results:CTHRC1 is a key regulator of SG function in mice.At the tissue level,Cthrc1 deletion resulted in the disorder and reduction of the microvascular network around SGs.At the molecular level,the knockout of Cthrc1 reduced the expression of vascular development genes and functional proteins in the dermal tissues.Furthermore,CTHRC1 administration considerably enhanced SG function by inducing adjacent vascular network reconstruction.Conclusions:CTHRC1 promotes the development,morphogenesis and function execution of SGs and their neighboring vasculature.Our study provides a novel target for the restoration or regeneration of SG function in vivo. | Xingyu Yuan Xianlan Duan Zhao Li Bin Yao Enhejirigala Wei Song Yi Kong Yuzhen Wang Fanliang Zhang Liting Liang Shijun Zhu Mengde Zhang Chao Zhang Sha Huang Xiaobing Fu | 2022 | Burns & Trauma2022,10,1: | 0 |
