1Clinical value of preoperative methylated septin 9 in Chinese colorectal cancer patients显示文摘BACKGROUND The methylated septin 9(mSEPT9) assay was the first blood-based test approved by the United States Food and Drug Administration as a colorectal screening test.However, the diagnostic and prognostic role of preoperative mSEPT9 for colorectal cancer(CRC) in Chinese patients is still unknown.AIM To improve the understanding of diagnostic and prognostic factors, serum m SEPT9 was detected in Chinese CRC patients.METHODS A retrospective analysis of 354 cases, of which 300 had CRC and 54 were normal,was performed in China. Patients' characteristics, treatments, and laboratory data, including age, the date of surgery, Union for International Cancer Control(UICC) stages, distant metastasis(M), and so on, were collected. Methylation levels of SEPT9 were quantified by quantitative, methylation-specific polymerase chain reaction before surgery. In addition, the effects of mSEPT9 on the occurrence and prognosis of 330 CRC cases from The Cancer Genome Atlas(TCGA) database were evaluated using bioinformatics analyses. Potential prognostic factors for overall survival(OS) and progression-free survival(PFS)were evaluated by Kaplan-Meier univariate analysis.RESULTSIn Chinese CRC patients, positive mSEPT9 was strongly associated with advanced UICC stages, deeper invasion by the primary tumor, and more distant metastasis. Methylation levels of SEPT9 were stage-dependent and showed a stepwise increase in UICC stages(I–IV), primary tumor categories(T1–T4),regional node categories(N0–N2), and distant metastasis categories(M0–M1).The patients with positive mSEPT9 showed a tendency toward lower PFS. After analyzing TCGA clinical data, the high mSEPT9 group was found to be obviously correlated only with more distant metastasis. The patients with high mSEPT9 levels showed a tendency toward lower OS. Besides, nine meaningful mSEPT9 sites were found to provide guidance for the follow-up studies.CONCLUSION MSEPT9 analysis may add valuable information to current tumor staging. 5Functional and numerical responses of Cyrtorhinus lividipennis to eggs of Nilaparvata lugens are not affected by genetically modified herbicide-tolerant rice显示文摘To safely and sustainably utilize genetic breeding techniques for crop production, greater understanding of the potential effects of genetically modified herbicide-tolerant(GMHT) crops on the ecological functions of predators is required. In the laboratory, we examined the functional and numerical responses of Cyrtorhinus lividipennis Reuter to eggs of brown planthopper(BPH), Nilaparvata lugens(St?l), which were reared on GMHT rice Bar68-1; the untransformed parental cultivar, D68; or a BPH-susceptive rice variety, Taichung Native 1. All stages of nymphs and female adult of C. lividipennis, either on GMHT rice or control plants, exhibited typical type II functional responses when fed on BPH eggs; the attacking rate and handling time of C. lividipennis on GMHT rice Bar68-1 was not significantly different from that on D68. The numerical responses of C. lividipennis on GMHT rice or controls fit Beddington's model; there were no significant differences in the parameters of numerical responses between GMHT rice Bar68-1 and D68. 6Drug-resistant tuberculosis control in China: progress and challenges显示文摘Background:China has the second highest caseload of multidrug-resistant tuberculosis(MDR-TB)in the world.In 2009,the Chinese government agreed to draw up a plan for MDR-TB prevention and control in the context of a comprehensive health system reform launched in the same year.Discussion:China is facing high prevalence rates of drug-resistant TB and MDR-TB.MDR-TB disproportionally affects the poor rural population and the highest rates are in less developed regions largely due to interrupted and/or inappropriate TB treatment.Most households with an affected member suffer a heavy financial burden because of a combination of treatment and other related costs.The influential Global Fund programme for MDR-TB control in China provides technical and financial support for MDR-TB diagnosis and treatment.However,this programme has a fixed timeline and cannot provide a long term solution.In 2009,the Bill and Melinda Gates Foundation,in cooperation with the National Health and Family Planning Commission of China,started to develop innovative approaches to TB/MDR-TB management and case-based payment mechanisms for treatment,alongside increased health insurance benefits for patients,in order to contain medical costs and reduce financial barriers to treatment.Although these efforts appear to be in the right direction,they may not be sufficient unless(a)domestic sources are mobilized to raise funding for TB/MDR-TB prevention and control and(b)appropriate incentives are given to both health facilities and their care providers.Summary:Along with the on-going Chinese health system reform,sustained government financing and social health protection schemes will be critical to ensure universal access to appropriate TB treatment in order to reduce risk of developing MDR-TB and systematic MDR-TB treatment and management.Qian Long Yan Qu Henry Lucas 2016Infectious Diseases of Poverty2016,5,1:3

8An improved defect recognition framework for casting based on DETR algorithm显示文摘The current casting surface defect detection algorithms suffer from poor small target defect recognition and imbalance between detection performance and detection time.An improved algorithmic framework for casting defect detection was proposed based on the DEtection TRansformer(DETR)algorithm.The algorithm takes ResNet with an efficient channel attention(ECA)-Net module as the backbone network.In addition,based on the original algorithm architecture,dynamic anchor boxes,improved multi-scale deformable attention module,and SIoU loss function are introduced to improve the sensitivity of transformer structure to input location information and scale size,and the small target defect detection performance is effectively improved.The recognition performance of the algorithm in a self-built casting defect dataset was studied.The improved DETR algorithm has 97.561% accuracy in recognizing two defects,namely sandinclusion and notch,with the detection rate being improved by 65.854% and 17.073% compared with the original DETR and you only look once(Yolo)-V5,respectively.This algorithm verifies the applicability of the transformer architecture target detection algorithm for casting defect detection tasks and provides new ideas for detecting other similar application scenarios.Long Zhang Sai-fei Yan Jun Hong Qian Xie Fei Zhou Song-lin Ran 2023Journal of Iron and Steel Research(International)2023,30,5:1
9Hybrid-panel-based new design idea for large reflector antenna显示文摘Large reflector antennas are widely used as radio telescopes and active main reflectors are generally applied to improve the surface accuracy. Considering that the high cost has been one important problem in engineering, it is worth discussing whether it is necessary to install actuators on all the panels. Thus, in this paper, a hybrid-panel-based new design idea for large reflector antenna is proposed. Assuming that the actuators are installed only in the region of the reflector with large deformations and there are no actuators in other region to reduce the actuator number, the surface accuracies and the corresponding electromagnetic(EM) performances calculated by three different panel adjustment strategies are compared. The most effective method is that the deformed reflector should be first preadjusted to reduce the gravity deformation and then the panels equipped with actuators should be adjusted to the locations determined by the best fitting reflector(BFR) derived by the deformed reflector with no actuators. A 35 m reflector antenna is adopted as an example to calculate the surface accuracy and EM performance when parts of the panels are equipped with actuators. The simulation results show that there is no need to install actuators on all panels and the presented method can greatly reduce the number of actuators with guaranteed surface accuracy. Thus, during the antenna structural design phase, once the surface accuracy requirement is given, the number of actuators can be minimized to reduce the manufacturing and maintenance costs as much as possible. 11血供丰富的乏脂肪肾脏错构瘤与肾透明细胞癌的CT鉴别诊断(英文)显示文摘Objective: The aim of our study was to investigate the feature of minimal fat renal angiomyolipoma with sufficient blood supply using CT scans and improve the diagnosis accuracy required to differentiate it from clear cell renal carcinoma.Methods: Retrospective analysis of 24 cases of post-surgery confirmed angiomyolipoma with sufficient blood supply (total of 25 tumors) in our hospital that were used for a pathological comparison study.Results: Among the 24 patients diagnosed with angiomyolipoma,nobody had bloody urine.Of the 96 patients diagnosed with clear cell renal cancer,14 had bloody urine (14.6%).In our studied group,the size of angiomyolipomas with sufficient blood supply was between 1.5 cm × 2.0 cm to 8.0 cm × 10.0 cm.During CT scan analysis,twenty tumors had similar density,and five of them had higher density.Only one tumor had a few dots of calcification (4%).Adipose tissue was not visible in 9 tumors,while 16 tumors had visible dots of adipose tissue,as visualized by CT scan.Intensive scanning indicated that all of the tumors showed a strong enhancement in the renal corticomedullary phase.Twenty tumors had significant heterogeneous enhancement in the early phase,while another set of five cases had homogenous prolonged enhancement.Nineteen patients had surgery to remove the angiomyolipomas,while six patients had single side kidney removal due to misdiagnosis for renal cancer in cases where the tumor severely compromised the renal parenchyma and sinus.All 25 cases were classified as renal angiomyolipoma by pathological analysis.Within the 96 cases of clear cell renal cancer,64 tumors had relatively low density,29 tumors had equal density,and 3 cases had relatively higher density.Fourteen of the tumors had calcification (14.6%),and none of them had visualized adipose tissue.Enhanced CT scans indicated that 69 cases of renal cancer showed significant enhancement in the renal corticomedullary phase,which had the abnormal pattern of 'fast-in-and-fast-out'.Additionally,27 cases had slow and prolonged enhancement.Conclusion: Similar to clear cell renal carcinomas,angiomyolipomas with sufficient blood supply also appear to exhibit abnormal enhancement with a pattern of 'fast-in-and-fast-out' during the early phase,which is easily misdiagnosed as renal cancer.It is difficult to differentiate them merely through CT scans;the key to differentiating them is to identify the adipose tissue within the tumor.Therefore,it is helpful to use thin-layer CT scans to locate the adipose tissue.Yan Guo Qian Peng Zhuo Wang Mingjuan Liu Xufeng Yang Teng Long 2011The Chinese-German Journal of Clinical Oncology2011,10,1:1

14FTO is a transcriptional repressor to auto-regulate its own gene and potentially associated with homeostasis of body weight显示文摘Fat mass and obesity-associated (FTO) protein is a ferrous ion (Fe^2+)/2-oxoglutarate (2-OG)-dependent demethylase preferentially catalyzing m^6A sites in RNA. The FTO gene is highly expressed in the hypothalamus with fluctuation in response to various nutritional conditions, which is believed to be involved in the control of whole body metabolism. However, the underlying mechanism in response to different nutritional cues remains poorly understood. Here we show that ketogenic diet-derived ketone body β-hydroxybutyrate (BHB) transiently increases FTO expression in both mouse hypothalamus and cultured cells. Interestingly, the FTO protein represses Fto promoter activity, which can be offset by BHB. We then demonstrate that FTO binds to its own gene promoter, and Fe^2+, but not 2-OG, impedes this binding and increases FTO expression. The BHB-induced occupancy of the promoter by FTO influences the assembly of the basal transcriptional machinery. Importantly, a loss-of-function FTO mutant (I367F), which induces a lean phenotype in FTO^I367F mice, exhibits augmented binding and elevated potency to repress the promoter. Furthermore, FTO fails to bind to its own promoter that promotes FTO expression in the hypothalamus of high-fat diet-induced obese and 48-h fasting mice, suggesting a disruption of the stable expression of this gene. 16Pomalidomide improves the function of CD133-or HER2-specific CAR T cells显示文摘Chimeric antigen receptor(CAR)T-cell therapy is mostly limited to hematological malignancies and has a poor effect on solid tumors.CAR T cells as a kind of immune cell may be affected by some immunomodulatory drugs such as pomalidomide,so the use of pomalidomide may improve the effect of CAR T cells on solid tumors.In this study,CD133-or HER2-specific CAR T cells were chosen to investigate whether pomalidomide can regulate the function of CAR T cells in vitro.We found that pomalidomide can significantly enhance the ability of CD133-CAR T cells and HER2-CAR T cells to kill tumor cells and increase the cytokine secretion of CD133-CAR T cells and HER2-CAR T cells.Also,pomalidomide was shown to induce down-regulation of protein levels of IL-2 transcriptional repressors Aiolos and Ikaros in CAR T cells.This study suggests that the combination of pomalidomide and CAR T cells may be a new strategy for the treatment of solid tumors.ZHIXIONG WANG NA RISU JIAYU FU HUI LIU GUOMIN ZHOU QIAN LIU YAN ZOU JIAXING TANG LONG LI XUEKAI ZHU 2021BIOCELL2021,45,1:0
17The complete and fully-phased diploid genome of a male Han Chinese显示文摘Since the release of the complete human genome,the priority of human genomic study has now been shifting towards closing gaps in ethnic diversity.Here,we present a fully phased and well-annotated diploid human genome from a Han Chinese male individual(CN1),in which the assemblies of both haploids achieve the telomere-to-telomere(T2T)level.Comparison of this diploid genome with the CHM13 haploid T2T genome revealed significant variations in the centromere.Outside the centromere,we discovered 11,413 structural variations,including numerous novel ones.We also detected thousands of CN1 alleles that have accumulated high substitution rates and a few that have been under positive selection in the East Asian population.Further,we found that CN1 outperforms CHM13 as a reference genome in mapping and variant calling for the East Asian population owing to the distinct structural variants of the two references.Comparison of SNP calling for a large cohort of 8869 Chinese genomes using CN1 and CHM13 as reference respectively showed that the reference bias profoundly impacts rare SNP calling,with nearly 2 million rare SNPs miss-called with different reference genomes.Finally,applying the CN1 as a reference,we discovered 5.80 Mb and 4.21 Mb putative introgression sequences from Neanderthal and Denisovan,respectively,including many East Asian specific ones undetected using CHM13 as the reference.Our analyses reveal the advances of using CN1 as a reference for population genomic studies and paleo-genomic studies.This complete genome will serve as an alternative reference for future genomic studies on the East Asian population.Chentao Yang Yang Zhou Yanni Song Dongya Wu Yan Zeng Lei Nie Panhong Liu Shilong Zhang Guangji Chen Jinjin Xu Hongling Zhou Long Zhou Xiaobo Qian Chenlu Liu Shangjin Tan Chengran Zhou Wei Dai Mengyang Xu Yanwei Qi Xiaobo Wang Lidong Guo Guangyi Fan Aijun Wang Yuan Deng Yong Zhang Jiazheng Jin Yunqiu He Chunxue Guo Guoji Guo Qing Zhou Xun Xu Huanming Yang Jian Wang Shuhua Xu Yafei Mao Xin Jin Jue Ruan Guojie Zhang 2023Cell Research2023,33,10:0

19Fabrication, bacteriostasis and osteointegration properties researches of the additively-manufactured porous tantalum scaffolds loading vancomycin显示文摘Infected bone defects(IBDs)remains a challenging problem for orthopedists.Clinically,routine management for IBDs has two stages:debridement and systematic antibiotics administration to control infection,and secondary grafting to repair bone defects.Whereas the efficacy is not satisfactory,because the overuse of antibiotics may lead to systemic toxicity,and the emergence of drug-resistant bacteria,as well as the secondary surgery would cause additional trauma and economic burden to the patients.Therefore,it is imperative to develop a novel scaffold for one-stage repair of IBDs.In this study,vancomycin(Van)was encapsulated into poly(lactic co-glycolic acid)(PLGA)microspheres through the double emulsion method,which were then loaded into the additively-manufactured porous tantalum(AM-Ta)through gelatin methacryloyl(GelMA)hydrogel to produce the composite Ta/GelMA hydrogel(Gel)/PLGA/vancomycin(Van)scaffolds for repairing IBDs.Physiochemical characterization of the newly-developed scaffold indicated that the releasing duration of Van was over 2 weeks.Biological experiments indicated good biocompatibility of the composite scaffold,as well as bacteriostasis and osteointegration properties,which showed great potential for clinical application.The construction of this novel scaffold would provide new sight into the development of orthopaedic implants,shedding a novel light on the treatment of IBDs.Hu Qian Ting Lei Long Hua Yu Zhang Dongyu Wang Jiangyu Nan Wenbin Liu Yan Sun Yihe Hu Pengfei Lei 2023Bioactive Materials2023,,6:0
