3Compound Zhebei granules combined with chemotherapy for the treatment of refractory acute leukemia: a randomized clinical trial显示文摘OBJECTIVE: To observe the effect of compound Zhebei granules(CZBG) with chemotherapy in the treatment of refractory acute leukemia.METHODS: In this multicenter, double-blind, placebo-controlled clinical trial, we used a central(online) randomization system to assign 235 patients to two treatment groups. A total of 118 patients received chemotherapy combined with CZBG(4 g,twice daily) and 117 patients received chemotherapy plus placebo. The clinical efficacy was evaluated at the end of one chemotherapeutic cycle.RESULTS: In the full analysis set, in which deaths due to disease progression were regarded as inefficacy, the rates of complete remission(CR) and partial remission(CR + PR) were 32.35% and 50.00%,respectively, for the chemotherapy combined with CZBG group, and 23.08% and 35.58%, respectively,for the chemotherapy plus placebo group. There was a statistically significant difference between the two groups according to a ysis set(PPχ2test(P < 0.05). 4Enhanced Expression of miR-425 Promotes Esophageal Squamous Cell Carcinoma Tumorigenesis by Targeting SMAD2显示文摘Esophageal squamous cell carcinoma(ESCC) is one of the most common and deadly cancers in the world. Currently, clinical therapy of ESCC remains limited and the five-year survival rate is poor. The function of miR-425 has been reported in multiple human cancers.However, the tumorigenic role and clinical significance of miR-425 in ESCC remains unclear. We found that enhanced expression of miR-425 in ESCC cell lines not only promoted cell proliferation and colony formation, but also increased cellular metastasis. Furthermore, we revealed the mechanism that miR-425 inhibited the expression of SMAD2 by targeting the second binding site in the 30-untranslated region(30-UTR) in ESCC. This mode of action influenced not only SMAD2 mRNA expression but also protein expression. In addition, we detected the expression of miR-425 in ESCC tissues and plasma. Moreover, we analyzed the relationship between miR-425 expression and SMAD2 m RNA expression. We found that miR-425 was overexpressed in ESCC tissues and the plasma relative to adjacent normal tissues and plasma of healthy individuals. Furthermore, there was a negative correlation between miR-425 expression and SMAD2. 7Diabetes fuels periodontal lesions via GLUT1-driven macrophage inflammaging显示文摘Hyperglycemia induces chronic low-grade inflammation(inflammaging),which is a newly identified contributor to diabetes-related tissue lesions,including the inflammatory bone loss in periodontitis.It is also a secondary senescent pattern mediated by an increased burden of senescent cells and senescence-associated secretory phenotype(SASP).Macrophage is a key SASPspreading cell and may contribute to the maintenance of SASP response in the periodontal microenvironment.Using a transgenic diabetic model(BLKS/J-Lepr^(db)/lepr^(db)mice)we identified striking senescence of the periodontium in young(18-wk)-diabetic mice accompanied by amassed p16^(+)-macrophages and enhanced early SASP response.Exposed to high glucose in vitro,bone marrow-derived macrophage(BMDM)revealed a strong GLUT1 m RNA response driving the elevated-glucose uptake.GLUT1 is a representative and facilitative glucose transporter in macrophages with potential roles in hyperglycemia-induced inflammation.In this study,both GLUT1 and the downstream GTPase Rheb expression upregulated in the gingiva of diabetic mice with impaired condition.Furthermore,SASP release and p16/p21 signaling were proven to be triggered by m TOR phosphorylation in BMDM and antagonized by restricting glucose uptake in GLUT1^(-/-)BMDM.Taken together,our findings suggest that elevated-GLUT1 sensor responded to high glucose is important for macrophage senescence and SASP response,generated as a result of hyperglycemia,and it is a potential molecular mechanism for the exacerbation of periodontitis in diabetes.Qian Wang Lulingxiao Nie Pengfei Zhao Xinyi Zhou Yi Ding Qianming Chen Qi Wang 2021International Journal of Oral Science2021,13,3:5
8V2X-Communication Assisted Interference Minimization for Automotive Radars显示文摘With the development of automated driving vehicles, more and more vehicles will be fitted with more than one automotive radars, and the radar mutual interference will become very significant. Vehicle to everything (V2X) communication is a potential way for coordinating automotive radars and reduce the mutual interference. In this paper, we analyze the positional relation of the two radars that interfere with each other, and evaluate the mutual interference for different types of automotive radars based on Poisson point process (PPP). We also propose a centralized framework and the corresponding algorithm, which relies on V2X communication systems to allocate the spectrum resources for automotive radars to minimize the interference. The minimum spectrum resources required for zero-interference are analyzed for different cases. 10Auxin regulation involved in gynoecium morphogenesis of papaya flowers显示文摘The morphogenesis of gynoecium is crucial for propagation and productivity of fruit crops.For trioecious papaya(Carica papaya),highly differentiated morphology of gynoecium in flowers of different sex types is controlled by gene networks and influenced by environmental factors,but the regulatory mechanism in gynoecium morphogenesis is unclear.Gynodioecious and dioecious papaya varieties were used for analysis of differentially expressed genes followed by experiments using auxin and an auxin transporter inhibitor.We first compared differential gene expression in functional and rudimentary gynoecium at early stage of their development and detected significant difference in phytohormone modulating and transduction processes,particularly auxin.Enhanced auxin signal transduction in rudimentary gynoecium was observed.To determine the role auxin plays in the papaya gynoecium,auxin transport inhibitor(N-1-Naphthylphthalamic acid,NPA)and synthetic auxin analogs with different concentrations gradient were sprayed to the trunk apex of male and female plants of dioecious papaya.Weakening of auxin transport by 10 mg/L NPA treatment resulted in female fertility restoration in male flowers,while female flowers did not show changes.NPA treatment with higher concentration(30 and 50 mg/L)caused deformed flowers in both male and female plants.We hypothesize that the occurrence of rudimentary gynoecium patterning might associate with auxin homeostasis alteration.Proper auxin concentration and auxin homeostasis might be crucial for functional gynoecium morphogenesis in papaya flowers.These results will lead to further investigation on the auxin homeostasis and gynoecium morphogenesis in papaya.Ping Zhou Mahpara Fatima Xinyi Ma Juan Liu Ray Ming 2019Horticulture Research2019,6,1:3
11Effect of aging treatment on microstructure and corrosion behavior of a Fe-18Cr-15Mn-0.66N stainless steel显示文摘The effect of aging treatment on the microstructure and corrosion behavior of a Fe-18Cr-15Mn-0.66N high-nitrogen stainless steel(HNSS) in 3.5 wt.% Na Cl solution was investigated using a series of electrochemical tests, scanning electronic microscopy(SEM), X-ray diffraction(XRD) and X-ray photoelectron spectroscopy(XPS). The results showed that the aging treatment led to the precipitation of CrN particles along the grain boundaries and their morphologies changed from dispersive particles to continuous network as the aging time increased up to 60 min. Aging time had minor effects on the corrosion potential and corrosion current density, but resulted in the sharp decrease in the pitting corrosion potential.The passive film behaved as a n-type semiconductor, and the donor density of the passive film increased with the aging time. Meanwhile, the fraction of stable oxide(CrO) in the passive film decreased with the aging time. 12Flotillin1 promotes EMT of human small cell lung cancer via TGF-β signaling pathway显示文摘Objective:Small cell lung carcinoma(SCLC)is considered one of the most aggressive types of lung cancer due to its rapid growth and early metastasis.No tumor markers or therapeutic targets have been demonstrated to be specific or effective in SCLC to date.This study aims to evaluate the potential of Flotillin1(Flot1)as a target of SCLC treatment.Methods:Flot1 expression level in the tissue of SCLC and other tissue of lung disease was detected using immunohistochemical staining.Transwell and Matrigel assays were employed to examine migration and invasion of cancer cells.Flow cytometry and xCELLigence system were used to evaluate cell apoptosis and cell viability,respectively.Expression levels of Flot1,epithelialmesenchymal transition(EMT)marker E-cadherin,vimentin,cyclinD1,TGF-β-Smad2/3,and p-AKT were examined using Western blot.Furthermore,xenograft tumor in nude mice was used to evaluate the growth and metastasis of NCI-H446 cells in vivo.Results:Our results demonstrated that Flot1 is highly expressed in SCLC samples and that its expression correlates strongly with clinical stage,distant metastasis,and poor survival.The knockdown of Flot1 decreased the growth,migration,and invasiveness of SCLC cells and reversed EMT phenotype in vitro and in vivo,while enhanced Flot1 expression exhibited the opposite behavior.Gene expression profile analysis demonstrated that Flot1-regulated genes frequently mapped to the AKT and TGF-β-Smad2/3pathways.Our results further revealed that Flot1 affected the progression of SCLC via regulation of EMT progression.Conclusions:These findings indicated an oncogenic role of Flot1 via promoting EMT in SCLC and suggested its potential as a tumor marker and prognostic indicator.Lianmei Zhao Jie Li Yueping Liu Wei Zhou Yanan Shan Xinyi Fan Xinliang Zhou Baoen Shan Yongmei Song Qimin Zhan 2018Cancer Biology & Medicine2018,15,4:3

14Low-density lipoprotein receptor-related protein 1 is a CROPs-associated receptor for Clostridioides infection toxin B显示文摘As the leading cause of worldwide hospital-acquired infection,Clostridioides difficile(C.difficile)infection has caused heavy economic and hospitalized burden,while its pathogenesis is not fully understood.Toxin B(Tcd B)is one of the major virulent factors of C.difficile.Recently,CSPG4 and FZD2 were reported to be the receptors that mediate Tcd B cellular entry.However,genetic ablation of genes encoding these receptors failed to completely block Tcd B entry,implicating the existence of alternative receptor(s)for this toxin.Here,by employing the CRISPR-Cas9 screen in CSPG4-deficient He La cells,we identified LDL receptor-related protein-1(LRP1)as a novel receptor for Tcd B.Knockout of LRP1 in both CSPG4-deficient He La cells and colonic epithelium Caco2 cells conferred cells with increased Tcd B resistance,while LRP1 overexpression sensitized cells to Tcd B at a low concentration.Co-immunoprecipitation assay showed that LRP1 interacts with full-length Tcd B.Moreover,CROPs domain,which is dispensable for Tcd B’s interaction with CSPG4 and FZD2,is sufficient for binding to LRP1.As such,our study provided evidence for a novel mechanism of Tcd B entry and suggested potential therapeutic targets for treating C.difficile infection.Shengjie Guo Yiou Chen Jingze Liu Xinyi Zhang Zhiheng Liu Zhuo Zhou Wensheng Wei 2022Science China(Life Sciences)2022,65,1:2

16Soy Protein Isolate Non-Isocyanates Polyurethanes(NIPU)Wood Adhesives显示文摘Soy-protein isolate(SPI)was used to prepare non-isocyanate polyurethane(NIPU)thermosetting adhesives for wood panels by reacting it with dimethyl carbonate(DMC)and hexamethylene diamine.Both linear as well as branched oligomers were obtained and identified,indicating how such oligomer structures could further cross-link to form a hardened network.Unusual structures were observed,namely carbamic acid-derived urethane linkages coupled with lactam structures.The curing of the adhesive was followed by thermomechanical analysis(TMA).It appeared to follow a two stages process:First,at a lower temperature(maximum 130℃),the growth of linear oligomers occurred,finally forming a physically entangled network.This appeared to collapse and disentangle,causing a decrease of MOE,as the temperature increases.This appears to be due to the ever more marked Brownian movements of the linear oligomer chains with the increase of the temperature.Second,chemical cross-linking of the chains appeared to ensue,forming a hardened network.This was shown by the thermomechanical analysis(TMA)showing two distinct MOE maxima peaks,one around 130℃ and the other around 220℃,with a very marked MOE decrease between the two.Plywood panels were prepared and bonded with the SPI-NIPU wood adhesive and the results obtained are presented.The adhesive appeared to pass comfortably the requirements for dry strength of relevant standards,showing to be suitable for interior grade plywood panels.It did not pass the requirements for wet tests.However,addition of 15%of glycerol diglycidyl ether improved the wet tests results but still not enough to satisfy the standards requirements.Xinyi Chen Antonio Pizzi Xuedong Xi Xiaojian Zhou Emmanuel Fredon Christine Gerardin 2021Journal of Renewable Materials2021,9,6:2
17Efficacy of rigosertib, a small molecular RAS signaling disrupter for the treatment of KRAS-mutant colorectal cancer显示文摘Objective:Mutant KRAS,the principal isoform of RAS,plays a pivotal role in the oncogenesis of colorectal cancer by constitutively activating the RAF/MEK/ERK and PI3K/AKT pathways.Effective targeted therapies are urgently needed.We investigated whether rigosertib,a benzyl styryl sulfone RAS signaling disruptor,could selectively kill KRAS-mutant colorectal cancer cells.Methods:CCK-8 was used to determine the cell viability.Patient-derived tumor and cancer cell xenograft models were used to detect the inhibitory efficacy of rigosertib.Flow cytometry was used to evaluate the apoptosis and cell cycle progression.Apoptosis and cell cycle arrest markers were detected by Western blot.DCFH-DA was used to determine the reactive oxygen species.Immunohistochemistry staining and Western blot were performed to characterize RAS signaling markers in colorectal cancer tissues and cells.Results:Rigosertib(RGS)exhibited a cytotoxic effect against colorectal cancer cells,which was greater in KRAS-mutant cells.Furthermore,RGS induced mitotic arrest and oxidative stress-dependent apoptosis in KRAS-mutant DLD1 and HCT116 cells.Besides,RGS disrupted RAS signaling,and the inhibition of RAS/MEK/ERK was independent of cellular oxidative stress.Using patient-derived xenograft models,the response and tumor inhibition of RGS were significantly higher in the KRAS-mutant subgroup,while p-MEK,p-ERK,and p-AKT levels of RGS-treated tumors were significantly decreased.Finally,in a KRAS-mutant,chemotherapy-resistant patient-derived xenograft model,RGS showed a stronger therapeutic effect than the combination standard therapy involving fluoropyrimidine+oxaliplatin/irinotecan+bevacizumab.Conclusions:These data showed that targeting RAS signaling using RGS could be a therapeutic treatment for KRAS-mutant colorectal cancer patients.Xinyi Zhou Qian Xiao Dongliang Fu Haochen Zhang Yang Tang Jinjie He Yeting Hu Xiangxing Kong Fei Teng Xiangrui Liu Ying Yuan Kefeng Ding 2022Cancer Biology & Medicine2022,19,2:2

19A Neural Network Method for Monitoring Snowstorm: A Case Study in Southern China显示文摘It has been observed that low temperature, rainfall, snowfall, frost have never occurred over the past 50 years in the southern China, and weather in this area is very complex, so the monitoring equipments are few. Optical and thermal infrared remote sensing is influenced much by clouds, so the passive microwave Advanced Microwave Scanning Radiometer-Earth Observing System(AMSR-E) data are the best choice to monitor and analyze the development of disaster. In order to improve estimation accuracy, the dynamic learning neural network was used to retrieve snow depth. The difference of brightness temperatures of TB18.7V and TB36.5V, TB18.7H and TB36.5H, TB23.8V and TB89 V, TB23.8H and TB89 H are made as four main input nodes and the snow depth is the only one output node of neural network. The mean and the standard deviation of retrieval errors are about 4.8 cm and 6.7 cm relative to the test data of ground measurements. 20DNA methylome and transcriptome landscapes revealed differential characteristics of dioecious flowers in papaya显示文摘Separate sexes in dioecious plants display different morphology and physiological characteristics.The differences between the two sexes lie in their highly differentiated floral characteristics and in sex-related phenotype,which is genetically determined and epigenetically modified.In dioecious papaya(Carica papaya L.),global comparisons of epigenetic DNA methylation and gene expressions were still limited.We conducted bisulfite sequencing of early-stage flowers grown in three seasons(spring,summer and winter)and compared their methylome and transcriptome profiles to investigate the differential characteristics of male and female in papaya.Methylation variances between female and male papaya were conserved among three different seasons.However,combined genome-scale transcriptomic evidence revealed that most methylation variances did not have influence on the expression profiles of neighboring genes,and the differentially expressed genes were most overrepresented in phytohormone signal transduction pathways.Further analyses showed diverse stress-responsive methylation alteration in male and female flowers.Male flower methylation was more responsive to stress whereas female flower methylation varied less under stress.Early flowering of male papaya in spring might be associated with the variation in the transcription of CpSVP and CpAP1 coinciding with their gene-specific hypomethylation.These findings provide insights into the sex-specific DNA methylation and gene expression landscapes of dioecious papaya and a foundation to investigate the correlation between differentiated floral characteristics and their candidate genes.Ping Zhou Xiaodan Zhang Mahpara Fatima Xinyi Ma Hongkun Fang Hansong Yan Ray Ming 2020Horticulture Research2020,7,1:2
