1A Domestication-Associated Gene GmPRR3b Regulates the Circadian Clock and Flowering Time in Soybean显示文摘Improved soybean cultivars have been adapted to grow at a wide range of latitudes,enabling expansion of cultivation worldwide.However,the genetic basis of this broad adaptation is still not clear.Here,we report the identification of GmPRR3b as a major flowering time regulatory gene that has been selected during domestication and genetic improvement for geographic expansion.Through a genome-wide association study of a diverse soybean landrace panel consisting of 279 accessions,we identified 16 candidate quantitative loci associated with flowering time and maturity time.The strongest signal resides in the known flowering gene E2,verifying the effectiveness of our approach.We detected strong signals associated with both flowering and maturity time in a genomic region containing GmPRR3b.Haplotype analysis revealed that GmPRR3bH6 is the major form of GmPRR3b that has been utilized during recent breeding of modern cultivars.mRNA profiling analysis showed that GmPRR3bH6 displays rhythmic and photoperiod-dependent expression and is preferentially induced under long-day conditions.Overexpression of GmPRR3bH6 increased main stem node number and yield,while knockout of GmPRR3bH6 using CRISPR/Cas9 technology delayed growth and the floral transition.GmPRR3bH6 appears to act as a transcriptional repressor of multiple predicted circadian clock genes,including GmCCAIa,which directly upregulates J/GmELF3a to modulate flowering time.The causal SNP(Chr12:5520945)likely endows GmPRR3bH6 a moderate but appropriate level of activity,leading to early flowering and vigorous growth traits preferentially selected during broad adaptation of landraces and improvement of cultivars.Cong Li Ying-hui Li Yanfei Li Hongfeng Lu Huilong Hong Yu Tian Hongyu Li Tao Zhao Xiaowei Zhou Jun Liu Xinan Zhou Scott A.Jackson Bin Liu Li-juan Qiu 2020Molecular Plant2020,13,5:11
2Electrochemical and spectroscopic characteristics of dissolved organic matter in a forest soil profile显示文摘Dissolved organic matter(DOM) represents one of the most mobile and reactive organic compounds in ecosystem and plays an important role in the fate and transport of soil organic pollutants, nutrient cycling and more importantly global climate change.Electrochemical methods were first employed to evaluate DOM redox properties, and spectroscopic approaches were utilized to obtain information concerning its composition and structure. DOM was extracted from a forest soil profile with five horizons. Diferential pulse voltammetry indicated that there were more redox-active moieties in the DOM from upper horizons than in that from lower horizons.Cyclic voltammetry further showed that these moieties were reversible in electron transfer. Chronoamperometry was employed to quantify the electron transfer capacity of DOM, including electron acceptor capacity and electron donor capacity, both of which decreased sharply with increasing depth. FT-IR, UV-Vis and fluorescence spectra results suggested that DOM from the upper horizons was enriched with aromatic and humic structures while that from the lower horizons was rich in aliphatic carbon, which supported the findings obtained by electrochemical approaches. Electrochemical approaches combined with spectroscopic methods were applied to evaluate the characteristics of DOM extracted along a forest soil profile. 3Development and utilization of a new chemically-induced soybean library with a high mutation density显示文摘Mutagenized populations have provided important materials for introducing variation and identifying gene function in plants. In this study, an ethyl methanesulfonate(EMS)-induced soybean(Glycine max) population,consisting of 21,600 independent M_2 lines, was developed.Over 1,000 M_(4(5))families, with diverse abnormal phenotypes for seed composition, seed shape, plant morphology and maturity that are stably expressed across different environments and generations were identified. Phenotypic analysis of the population led to the identification of a yellow pigmentation mutant, gyl, that displayed significantly decreased chlorophyll(Chl) content and abnormal chloroplast development. Sequence analysis showed that gyl is allelic to Minn Gold, where a different single nucleotide polymorphism variation in the Mg-chelatase subunit gene(ChlI1a) results in golden yellow leaves. A cleaved amplified polymorphic sequence marker was developed and may be applied to marker-assisted selection for the golden yellow phenotype in soybean breeding. 4An experimental study of phase transformations in olivine under pressure and temperature conditions corresponding to the mantle transition zone显示文摘High-pressure polymorphs of olivine (wadsleyite and ringwoodite) are major minerals in the mantle transition zone (MTZ).Phase transformations in olivine are important for a series of geodynamic problems such as the mineralogical and evolutionary history of the mantle,mantle convection patterns,and deep focus earthquakes in subduction zones.In this study,we examine phase transformations in olivine with two compositions,namely Mg 2 SiO 4 (Fo 100) and (Mg 0.9 Fe 0.1) 2 SiO 4 (Fo 90),at pressures between 14.1 and 20 GPa and a constant temperature of 1400°C,using the newly installed multi-anvil system at the Laboratory for Studies of the Earth's Deep Interior (SEDI),China University of Geosciences (Wuhan).At 14.1 GPa,Fo 90 transformed completely into the wadsleyite structure (β),while Fo 100 remained as olivine (α).Between 14.8 and 15.6 GPa,both Fo 100 and Fo 90 transformed into the wadsleyite structure.Wadsleyite crystals were identified by two characteristic Raman peaks between 722 and 723 and 917 and 919 cm 1.They exhibit a bimodal grain size distribution:large-crystals with average grain sizes greater than 100 μm and microcrystals less than 10 μm.The population of microcrystals increased with pressure,apparently due to the increase in over-pressure (the difference between the experimental pressure condition and the equilibrium transformation pressure at 1400°C),which promotes nucleation and retards grain growth.All run charges contained large numbers of wadsleyite microcrystals,because of the low activation energy of the nucleation process.The experimentally observed microstructure may shed light on the morphology of wadsleyite observed in shocked meteorites.At 19.5 GPa,wadsleyite coexisted with ringwoodite (γ) in Fo 100,but was absent in Fo 90.At 20 GPa,both samples transformed completely into ringwoodite,which was characterized by the 798 and 840 cm 1 Raman lines.Ringwoodite crystals are euhedral grains (average grain size 10-20 μm),with well-developed triple junctions.The complex upper mantle structure in eastern China determined from seismological studies cannot be explained by the simple transformation sequence of the olivine system alone.Phase transformations in other pyroxene-normative components (including pyroxenes and garnets) and the interaction of these components with olivine may be responsible for the complex structure.High-pressure and high-temperature experimental studies on complex systems (e.g.olivine-pyroxene),combined with data from geophysical exploration,may help in establishing a more realistic geological-petrological model for eastern China and further our understanding of the possible physical mechanisms that are responsible for the complex structure.Such studies will have profound implications for understanding the dynamic processes in the deep Earth interior.WU Yao WANG YanBin ZHANG YanFei JIN ZhenMin WANG Chao ZHOU ChunYin 2012Chinese Science Bulletin2012,57,8:5
5Homologous and heterologous interactions between catalytic and regulatory subunits of Escherichia coli acetohydroxyacid synthaseⅠand Ⅲ显示文摘Homologous and heterologous interactions between acetohydroxyacid synthase (AHAS) I and Ⅲ from E. coli have been studied by surface plasmon resonance (SPR). The catalytic and regulatory subunits association for AHAS I (KD = 1.13 × 10-7 M) was stronger than that for AHAS Ⅲ (KD = 5.29 × 10-7 M). A strong heterologous association between regulatory and catalytic subunits and heterologous activation of catalytic subunits were observed. SPR results combined with enzyme kinetics indicate that the reconstituted heterologous enzymes had similar kinetic properties as homologous enzymes, implying that the regulatory subunit of AHAS I could be replaced by the regulatory subunit of AHAS Ⅲ and vice versa. 7Effects of Epichloë endophyte on antioxidant enzymes activities, photosynthesis and growth of three ecotypes of Elymus dahuricus显示文摘Fungal endophytes of some cultivated grasses can increase plant performance and competitive abilities,especially under stress. Far less is known about the influence of Epichlo? infections in wild populations of wild grasses. In this study, plants of three Elymus dahuricus ecotypes(WLS, QY and WTS) either infected with Epichlo? endophyte(E+) or uninfected(E–) were grown in the field. The activities of the antioxidant enzymes ascorbate peroxidase, catalase, peroxidase and superoxide dismutase, and concentrations of H_2O_2 and malondialdehyde were examined in the leaves of E+ and E– plants. We also determined photosynthesis parameters,leaf blade and sheath carbohydrate concentration and plant growth parameters of both E+ and E– plants. E+ plants from the WLS and QY populations had significantly higher antioxidant enzyme activities and photosynthetic capability(P < 0.05), superior growth characteristics including more abundant carbohydrate concentration than E– plants. In contrast, in plants from the WTS population,the endophyte had no significant effect on reactive oxygen species scavenging capability and growth performance(P > 0.05), and even displayed some negative effects on plant photosynthetic capability. 8Identification of an E3 ligase-encoding gene RFWD3 in non-small cell lung cancer显示文摘In order to unveil ubiquitin pathway genes (UPGs) that are essential for non-small cell lung cancer (NSCLC) cell proliferation,we recently conducted a siRNA screening experiment to knockdown the expression of 696 UPGs found in the human genome in A549 and H1975 NSCLC cells.We found that silencing of one of the candidates,RFWD3 that encodes an E3 ubiquitin ligase essential for the repair of DNA interstrand cross-links inresponse to DNA damage,led to dramatic inhibition of NSCLC cell proliferation with significant Z-scores.Knockdown of RFWD3 suppressed colony forming activity of NSCLC cells.We further evaluated the significance of RFWD3 in NSCLCs and found that this gene was more elevated in tumor samples than in paired normal lung tissues and was inversely associated with the clinical outcome of patients with NSCLC.Moreover,RFWD3 expression was significantly higher in smokers than in non-smokers.These results show for the first time that RFWD3 is required for NSCLC cell proliferation and may have an important role in lung carcinogenesis.Yanfei Zhang Xinchun Zhao Yongchun Zhou Min Wang Guangbiao Zhou 2020Frontiers of Medicine2020,14,3:3
9Synergetic Improvement of Interlaminar Fracture Toughness in Carbon Fiber/Epoxy Composites Interleaved with PES/PEK‑C Hybrid Nanofiber Veils显示文摘In this study,two types of soluble thermoplastic resins were added to epoxy resin at a fixed weight ratio to prepare a three-phase cast body.The cast was then manufactured into hybrid nanofiber as interleaves for interlaminar toughening of carbon fiber/epoxy resin(CF/EP)composites using a co-solvent method.The results revealed that when the hybrid components reached 15 wt%,Polyethersulfone(PES)and polyaryletherketone cardo(PEK-C)exhibited the best synergistic toughening effect,and the fracture toughness increased by 99.8%and 39.8%,respectively,compared with the reference or the same proportion of the single PES toughened sample.We used PES/PEK-C hybrid nanofibers with an areal density of 19.2 g per square meter(gsm)as composite toughening layers.Apart from the lack of significant influence of PES nanofiber on CF/EP composites,the interlaminar fracture toughness of mode I and mode II layers increased by 88.3%and 46.9%,respectively,compared to the reference sample.Scanning Electron Microscopy of the fracture surface and cross-section micromorphology of the laminate displayed that the thermoplastic microspheres of different sizes contribute differently to crack resistance:PEK-C consumes more energy due to the debonding and extraction of microspheres and resin,whereas the presence of the PES phase can induce more plastic deformation and crack deflection.Jinli Zhou Chenyu Zhang Chao Cheng Ming Wang Zhihui Yang Yanfei Yang Hongying Yang Muhuo Yu 2022Advanced Fiber Materials2022,4,5:3
10SAR motion compensation based on the correction of residual attitude errors显示文摘As residual attitude errors are likely to affect the synthetic aperture radar (SAR) imaging, this paper presents a SAR motioncompensation algorithm based on the correction of residual attitude errors. The existing methods all use the antenna stableplatform to correct the attitude errors, and then compensate the trajectory deviations in the following imaging process. Besidescompensating the trajectory deviations, the modified method of this paper also considers the influence of residual attitude er-rors on the SAR imaging, and can compensate both the trajectory deviations and the residual attitude errors. Compared withthe existing methods, the modified method in this paper can more precisely compensate the imperfect motion on the SAR im-aging, especially good for the SAR system with a small platform, near operating distance and a narrow antenna beam. Such asystem causes severe residual attitude errors and needs to consider the influence of antenna beam pointing errors on the imag-ing. 12A super-resolution algorithm for synthetic aperture radar based on modified spatially variant apodization显示文摘The existing spatially variant apodizations(SVAs) either cannot depress the sidelobes effectively or reduce the energy of the mainlobe.To improve this,a modified SVA(MSVA) is put forward in this paper,which expands the traditional filter from 3-taps to 5-taps and sets relevant parameters according to different sampling rates to get the excellent result that satisfies constrained optimization theory.A modified super-SVA is also presented,which compares the result after the iteration with the original signal and makes the one whose amplitude is smaller as the initial value of the next iteration.This method can eliminate the sidelobes produced by the intermediate operation,so that the following bandwidth extrapolation is more available.Super-MSVA is presented based on the modified SVA and modified super-SVA,which is suitable for any Nyquist sampling rate,can extrapolate the signal bandwidth many times through iteration with a commensurate improvement in resolution,as demonstrated by the result of the experiment.NI Chong WANG YanFei XU XiangHui ZHOU ChangYi CUI PengFei 2011Science China(Physics,Mechanics & Astronomy)2011,54,2:2
13Grass phasiRNAs and male fertility显示文摘Recent studies have indicated that a special type of small noncoding RNAs, phased small-interfering RNAs(phasiRNAs) play crucial roles in many cellular processes of plant development. PhasiRNAs are generated from long RNA precursors at intervals of 21 or 24 nt in plants, and they are produced from both protein-coding gene and long noncoding RNA genes. Different from those in eudicots, grass phasiRNAs include a special class of small RNAs that are specifically expressed in reproductive organs.These grass phasiRNAs are associated with gametogenesis, especially with anther development and male fertility. 14Abemaciclib induces apoptosis in cardiomyocytes by activating the Hippo signaling pathway显示文摘Abemaciclib is the newest cyclin-dependent kinase 4/6 inhibitor that has received approval from the US Food and Drug Administration for using in patients with advanced breast cancer. However, its potential adverse effects on cardiomyocytes remain unknown. In this study, we used the cell counting kit-8 assay, western blot analysis, flow cytometry, immunostaining, and quantitative polymerase chain reaction to investigate the role of abemaciclib in inducing apoptosis and in inhibiting the viability and proliferation of AC16 human cardiomyocyte cells. The results revealed that abemaciclib induced apoptosis and inhibited cell proliferation by activating the Hippo signaling pathway. 15Immobilization of RGD Peptide onto the Surface of Apatite-Wollastonite Ceramic for Enhanced Osteoblast Adhesion and Bone Regeneration显示文摘The arginine-glycine-aspartic(RGD) acid peptide was grafted to the surface of apatitewollastonite(AW) ceramic in an effort to improve its cell adhesion, proliferation and osteoinduction. RGD peptide was covalently immobilized onto the surface of AW ceramic via the synthetic cross linker AAPTS-E and 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide(EDC). The modified surfaces were characterized by attenuated total reflectance Fourier transform infrared spectroscopy(ATR-FTIR) and X-ray photoelectron spectroscopy(XPS). The chemical analysis indicated that RGD peptide had been immobilized onto the AW surface successfully. The growth of osteoblast-like cells(MG63) showed that modifying the AW surface with RGD peptide enhanced the cell adhesion and proliferation. 16Identification of a potential tumor suppressor gene, UBL3, in non-small cell lung cancer显示文摘Objective: Oncogenes have been shown to be drivers of non-small cell lung cancer(NSCLC), yet the tumor suppressing genes involved in lung carcinogenesis remain to be systematically investigated. This study aimed to identify tumor suppressing ubiquitin pathway genes(UPGs) that were critical to lung tumorigenesis.Methods: The 696 UPGs were silenced by an siRNA screening in NSCLC cells;the potential tumor suppressing UPGs were analyzed, and their clinical significance was investigated.Results: We reported that silencing of 11 UPGs resulted in enhanced proliferation of NSCLC cells, and four UPGs(UBL3, TRIM22, UBE2 G2, and MARCH1) were significantly downregulated in tumor samples compared to that in normal lung tissues and their expression levels were positively associated with overall survival(OS) of NSCLC patients. Among these genes, UBL3 was the most significant one. UBL3 expression was decreased in tumor samples compared to that in paired normal lung tissues in 59/86(68.6%) NSCLCs, was correlated with TNM stage and sex of NSCLC patients, and was significantly higher in non-smoking patients than in smoking patients. 17EMC10 governs male fertility via maintaining sperm ion balance显示文摘Infertility is a severe public health problem worldwide that prevails up to 15% in reproductive-age couples,and male infertility accounts for half of total infertility.Studies on genetically modified animal models have identifed lots of genes involved in the pathogenesis of male infertility.The underlying causes,however,remain largely unclear.In this study,we provide evidence that EMCIO,one subunit of endoplasmic reticulum (ER)membrane protein complex (EMC),is required for male fertility.EMC10 is significantly decreased in spermatozoa from patients with asthenozoospermia and positively associated with human sperm motility. Male mice lacking Emc10 gene are completely sterile.Emc10-null spermatozoa exhibit multiple defects including abnormal morphology,decreased motility,impaired capacitation,and impotency of acrosome reaction,thereby which are incapable of fertilizing intact or ZP-free oocytes.However,intracytoplasmic sperm injection could rescue this defect caused by EMC10 deletion.
