2TPX2 knockdown suppressed hepatocellular carcinoma cell invasion via inactivating AKT signaling and inhibiting MMP2 and MMP9 expression显示文摘Objective: Targeting protein for Xenopus kinesin-like protein 2(TPX2) is a nuclear proliferation-related protein that plays a critical role in the formation of mitotic spindle. High expression of TPX2 has been observed in several types of tumors. However, the role of TPX2 in hepatocellular carcinoma(HCC) remains unclear. Our study aimed to investigate the effect of TPX2 on HCC cell invasion.Methods: The immortalized normal human liver cell line L02 and six HCC cell lines including SMMC-7721, BEL-7402, Huh-7, HepG2, Hep3 B and SKHep1 were subjected to qRT-PCR and western blot for TPX2 mRNA and protein, respectively. Furthermore, TPX2 small interfering RNA(siRNA) was used to knock down TPX2 expression in SMMC-7721 and HepG2 cells. Cell proliferation and invasion were determined by MTT and transwell assays. Otherwise, expression of p-AKT, MMP2 and MMP9 were evaluated by western blot in SMMC-7721 cells.Results: The expression of TPX2 in HCC cell lines was markedly higher than that in normal human liver cell line. TPX2 knockdown using a specific TPX2-siRNA reduced the number of invaded cells and inhibited cell proliferation in SMMC-7721 and HepG2 cells. Furthermore, TPX2 knockdown resulted in inactivation of AKT signaling and down-regulation of MMP2 and MMP9 expression in SMMC-7721 cells.Conclusions: Our study identified that TPX2 might contribute to tumor cell invasion through activating AKT signaling and subsequently increasing MMP2 and MMP9 in HCC.Qingquan Liu Pinghua Yang Kangsheng Tu Hongyong Zhang Xin Zheng Yingmin Yao Qingguang Liu 2014Chinese Journal of Cancer Research2014,26,4:11
3Prognostic significance of combining high mobility group Box-1 and OV-6 expression in hepatocellular carcinoma显示文摘The inflammatory environment and existence of cancer stem cells are critical for progression and intrahepatic recurrence of hepatocellular carcinoma(HCC) after curative resections. Here, we investigated the prognostic significance of combining high mobility group box 1(HMGB1) expression and hepatic progenitor marker OV6 in hepatocellular carcinoma. Expression of HMGB1 and OV6 was evaluated using immunohistochemistry profiling in tissue microarrays containing samples from 208 HCC patients. Invasive clinical or pathological factors were found in patients with high expression of HMGB1 or OV6. Higher HMGB1 was associated with poorer clinical outcomes, and independently related to elevated 5-year recurrence incidence(85.5% vs. 62.4%, P<0.001). We also found that more OV6 positive staining was correlated with poor prognosis of HCC patients(P<0.001). Notably, expression of HMGB1 was positively correlated with OV6 in density(R2=0.032, P<0.001) and reversely related to HCC outcomes. Abnormal expression of HMGB1 in combination with positive staining of OV6 displayed poorer prognostic performance than single biomarker alone(area under curve(AUC) survival=0.696). 14Liver resection versus liver transplantation for hepatocellular carcinoma within the Milan criteria based on estimated microvascular invasion risks显示文摘Background:Preoperative prediction of microvascular invasion(MVI)in hepatocellular carcinoma(HCC)may optimize individualized treatment decision-making.This study aimed to investigate the prognostic differences between HCC patients undergoing liver resection(LR)and liver transplantation(LT)based on predicted MVI risks.Methods:We analysed 905 patients who underwent LR,including 524 who underwent anatomical resection(AR)and 117 who underwent LT for HCC within the Milan criteria using propensity score matching.A nomogram model was used to predict preoperative MVI risk.Results:The concordance indices of the nomogram for predicting MVI were 0.809 and 0.838 in patients undergoing LR and LT,respectively.Based on an optimal cut-off value of 200 points,the nomogram defined patients as high-or low-risk MVI groups.LT resulted in a lower 5-year recurrence rate and higher 5-year overall survival(OS)rate than LR among the high-risk patients(23.6%vs 73.2%,P<0.001;87.8%vs 48.1%,P<0.001)and low-risk patients(19.0%vs 45.7%,P<0.001;86.5%vs 70.0%,P=0.002).The hazard ratios(HRs)of LT vs LR for recurrence and OS were 0.18(95%confidence interval[CI],0.09–0.37)and 0.12(95%CI,0.04–0.37)among the high-risk patients and 0.37(95%CI,0.21–0.66)and 0.36(95%CI,0.17–0.78)among the low-risk patients.LT also provided a lower 5-year recurrence rate and higher 5-year OS rate than AR among the high-risk patients(24.8%vs 63.5%,P=0.001;86.7%vs 65.7%,P=0.004),with HRs of LT vs AR for recurrence and OS being 0.24(95%CI,0.11–0.53)and 0.17(95%CI,0.06–0.52),respectively.The 5-year recurrence and OS rates between patients undergoing LT and AR were not significantly different in the low-risk patients(19.4%vs 28.3%,P=0.129;85.7%vs 77.8%,P=0.161).Conclusions:LT was superior to LR for patients with HCC within the Milan criteria with a predicted high or low risk of MVI.No significant differences in prognosis were found between LT and AR in patients with a low risk of MVI.Pinghua Yang Fei Teng Shilei Bai Yong Xia Zhihao Xie Zhangjun Cheng Jun Li Zhengqing Lei Kui Wang Baohua Zhang Tian Yang Xuying Wan Hao Yin Hao Shen Timothy M Pawlik Wan Yee Lau Zhiren Fu Feng Shen 2023Gastroenterology Report2023,11,1:1
15Effect of High Temperature Curing on the Frost Resistance of Recycled Aggregate Concrete and the Physical Properties of Second-Generation Recycled Coarse Aggregate under Freeze-Thaw Cycles显示文摘With the emphasis on environmental issues,the recycling of waste concrete,even recycled concrete,has become a hot spot in the field of architecture.But the repeated recycling of waste concrete used in harsh environments is still a complex problem.This paper discusses the durability and recyclability of recycled aggregate concrete(RAC)as a prefabricated material in the harsh environment,the effect of high-temperature curing(60℃,80℃,and 100℃)on the frost resistance of RAC and physical properties of the second generation recycled coarse aggregate(RCA_(2))of RAC after 300 freeze-thaw cycles were studied.The frost resistance of RAC was characterized by compressive strength,relative dynamic elastic modulus,and mass loss.As the physical properties of RCA_(2),the apparent density,water absorption,and crushing value were measured.And the SEM images of RAC after 300 freeze-thaw cycles were shown.The results indicated that the frost resistance of RAC cured at 80℃ for 7 days was comparable to that cured in the standard condition(cured for 28 days at 20℃±2℃ and 95%humidity),and the RAC cured at 100℃ was slightly worse.However,the frost resistance of RAC cured at 60℃ deteriorated seriously.The RAC cured at 80℃ for 7 days is the best.Whether after the freeze-thaw cycle or not,the RCA that curd at 60℃,80℃,and 100℃ for 7 days can also meet the requirements of Grade III RCA and be used as the aggregate of non-bearing part of prefabricated concrete components.RCA_(2) which is cured at 80℃ for 7 days had the best physical properties.Xintong Chen Pinghua Zhu Xiancui Yan Lei Yang Huayu Wang 2023Journal of Renewable Materials2023,11,6:1
