2Somatic Embryogenesis Receptor Kinases Control Root Development Mainly via Brassinosteroid-Independent Actions in Arabidopsis thaliana显示文摘Brassinosteroids(BRs),a group of plant steroidal hormones,play critical roles in many aspects of plant growth and development.Previous studies showed that BRI1-mediated BR signaling regulates cell division and differentiation during Arabidopsis root development via interplaying with auxin and other phytohormones.Arabidopsis somatic embryogenesis receptor-like kinases(SERKs),as co-receptors of BRI1,were found to play a fundamental role in an early activation step of BR signaling pathway.Here we report a novel function of SERKs in regulating Arabidopsis root development.Genetic analyses indicated that SERKs control root growth mainly via a BR-independent pathway.Although BR signaling pathway is completely disrupted in the serk1 bak1 bkk1 triple mutant,the root growth of the triple mutant is much severely damaged than the BR deficiency or signaling null mutants.More detailed analyses indicated that the triple mutant exhibited drastically reduced expression of a number of genes critical to polar auxin transport,cell cycle,endodermis development and root meristem differentiation,which were not observed in null BR biosynthesis mutant cpd and null BR signaling mutant bri1-701.Junbo Du Hongju Yin Shasha Zhang Zhuoyun Wei Baolin Zhao Jinghua Zhang Xiaoping Gou Honghui Lin Jia Li 2012Journal of Integrative Plant Biology2012,54,6:22
3A highly alkaline-stable metal oxide@metal-organic framework composite for high-performance electrochemical energy storage显示文摘Most metal-organic frameworks(MOFs) hardly maintain their physical and chemical properties after exposure to alkaline aqueous solutions,thus precluding their use as potential electrode materials for electrochemical energy storage devices.Here,we present the design and synthesis of a highly alkaline-stable metal oxide@MOF composite,Co3O4 nanocube@Co-MOF(Co3O4@Co-MOF),via a controllable and facile one-pot hydrothermal method under highly alkaline conditions.The obtained composite possesses exceptional alkaline stability,retaining its original structure in 3.0 M KOH for at least 15 days.Benefitting from the exceptional alkaline stability,unique structure,and larger surface area,the Co3O4@Co-MOF composite shows a specific capacitance as high as 1020 F g-1 at 0.5 A g-1 and a high cycling stability with only 3.3% decay after 5000 cycles at 5 A g-1.The as-constructed solid-state flexible device exhibits a maximum energy density of 21.6 mWh cm-3.Shasha Zheng Qing Li Huaiguo Xue Huan Pang Qiang Xu 2020National Science Review2020,7,2:16
4Nonhuman primate models of focal cerebral ischemia显示文摘Rodents have been widely used in the production of cerebral ischemia models. However, successful therapies have been proven on experimental rodent stroke model, and they have often failed to be effective when tested clinically. Therefore, nonhuman primates were recommended as the ideal alternatives, owing to their similarities with the human cerebrovascular system, brain metabolism, grey to white matter ratio and even their rich behavioral repertoire. The present review is a thorough summary of ten methods that establish nonhuman primate models of focal cerebral ischemia; electrocoagulation, endothelin-1-induced occlusion, microvascular clip occlusion, autologous blood clot embolization, balloon inflation, microcatheter embolization, coil embolization, surgical suture embolization, suture, and photochemical induction methods. This review addresses the advantages and disadvantages of each method, as well as precautions for each model, compared nonhuman primates with rodents, different species of nonhuman primates and different modeling methods. Finally it discusses various factors that need to be considered when modelling and the method of evaluation after modelling. 5Lactic acid in tumor microenvironments causes dysfunction of NKT cells by interfering with mTOR signaling显示文摘Cellular metabolism has been shown to regulate differentiation and function of immune cells.Tumor associated immune cells undergo phenotypic and functional alterations due to the change of cellular metabolism in tumor microenvironments.NKT cells are good candidates for immunotherapies against tumors and have been used in several clinical trials.However,the influences of tumor microenvironments on NKT cell functions remain unclear.In our studies,lactic acid in tumor microenvironments inhibited IFNγ and IL4 productions from NKT cells,and more profound influence on IFNγ was observed.By adjusting the pH of culture medium we further showed that,dysfunction of NKT cells could simply be induced by low extracellular pH.Moreover,low extracellular pH inhibited NKT cell functions by inhibiting mammalian target of rapamycin(mTOR) signaling and nuclear translocation of promyelocytic leukemia zinc-finger(PLZF).Together,our results suggest that tumor acidic microenvironments could interfere with NKT cell functions through metabolic controls.Di Xie Shasha Zhu Li Bai 2016Science China(Life Sciences)2016,59,12:13
6Tissue-specific transcription reprogramming promotes liver metastasis of colorectal cancer显示文摘Metastasis,the development of secondary malignant growths at a distance from a primary tumor,is the cause of death for 90%of cancer patients,but little is known about how metastatic cancer cells adapt to and colonize new tissue environments.Here,using clinical samples,patient-derived xenograft(PDX)samples,PDX cells,and primary/metastatic cell lines,we discovered that liver metastatic colorectal cancer(CRC)cells lose their colon-specific gene transcription program yet gain a liver-specific gene transcription program.We showed that this transcription reprogramming is driven by a reshaped epigenetic landscape of both typical enhancers and super-enhancers.Further,we identified that the liver-specific transcription factors FOXA2 and HNF1A can bind to the gained enhancers and activate the liver-specific gene transcription,thereby driving CRC liver metastasis.Importantly,similar transcription reprogramming can be observed in multiple cancer types.Our data suggest that reprogrammed tissue-specific transcription promotes metastasis and should be targeted therapeutically.Shuaishuai Teng Yang Eric Li Ming Yang Rui Qi Yiming Huang Qianyu Wang Yanmei Zhang Shanwen Chen Shasha Li Kequan Lin Yang Cao Qunsheng Ji Qingyang Gu Yujing Cheng Zai Chang Wei Guo Pengyuan Wang Ivan Garcia-Bassets Zhi John Lu Dong Wang 2020Cell Research2020,30,1:11

8Characteristics of ozone pollution and the sensitivity to precursors during early summer in central plain, China显示文摘In this study,we conducted an observation experiment from May 1 to June 30,2018 in Zhengzhou,a major city in central China,where ground ozone(O3)pollution has become serious in recent years.The concentrations of O3 and its precursors,as well as H2O_(2) and meteorological data were obtained from the urban site(Yanchang,YC),suburban(Zhengzhou University,ZZU)and background sites(Ganglishuiku,GLSK).Result showed that the rates of O3 concentration exceeded Chinese National Air Quality Standard GradeⅡ(93.3 ppbv)were 59.0%,52.5%,and 55.7%at the above three sites with good consistency,respectively,indicating that O3 pollution is a regional problem in Zhengzhou.The daily peak O3 appeared at 15:00-16:00,which was opposite to VOCs,NOx,and CO and consistent with H2O_(2).The exhaustive statistical analysis of meteorological factors and chemical effects on O3 formation at YC was advanced.The high concentration of precursors,high temperature,low relative humidity,and moderately high wind speed together with the wind direction dominated by south and southeast wind contribute to urban O3 episodes in Zhengzhou.O3 formation analysis showed that reactive alkenes such as isoprene and cis-2-butene contributed most to O3 formation.The VOCs/NOx ratio and smog production model were used to determine O3-VOC-NOx sensitivity.The O3 formation in Zhengzhou during early summer was mainly under VOC-limited and transition regions alternately,which implies that the simultaneous emission reduction of alkenes and NOx is effective in reducing O3 pollution in Zhengzhou.Yasong Li Shasha Yin Shijie YU Ling Bai Xudong Wang Xuan Lu Shuangliang Ma 2021Journal of Environmental Sciences2021,33,1:10

13Chemical genomics reveals inhibition of breast cancer lung metastasis by Ponatinib via c-Jun显示文摘Metastasis is the leading cause of human cancer deaths.Unfortunately,no approved drugs are available for antimetastatic treatment.In our study,high-throughput sequencing-based high-throughput screening(HTS^2)and a breast cancer lung metastasis(BCLM)-associated gene signature were combined to discover anti-metastatic drugs.After screening of thousands of compounds,we identified Ponatinib as a BCLM inhibitor.Ponatinib significantly inhibited the migration and mammosphere formation of breast cancer cells in vitro and blocked BCLM in multiple mouse models.Mechanistically,Ponatinib represses the expression of BCLM-associated genes mainly through the ERK/c-Jun signaling pathway by inhibiting the transcription of JUN and accelerating the degradation of c-Jun protein.Notably,JUN expression levels were positively correlated with BCLM-associated gene expression and lung metastases in breast cancer patients.Collectively,we established a novel approach for the discovery of anti-metastatic drugs,identified Ponatinib as a new drug to inhibit BCLM and revealed c-Jun as a crucial factor and potential drug target for BCLM.Our study may facilitate the therapeutic treatment of BCLM as well as other metastases.Wei Shao Shasha Li Lu Li Kequan Lin Xinhong Liu Haiyan Wang Huili Wang Dong Wang 2019Protein & Cell2019,10,3:7
14Performance of the Autism Spectrum Rating Scale and Social Responsiveness Scale in Identifying Autism Spectrum Disorder Among Cases of Intellectual Disability显示文摘The Autism Spectrum Rating Scale(ASRS) and the Social Responsiveness Scale(SRS) have been widely used for screening autism spectrum disorder(ASD) in the general population during epidemiological studies, but studies of individuals with intellectual disability(ID) are quite limited. Therefore, we recruited the parents/caregivers of 204 ASD cases, 71 ID cases aged 6–18 years from special education schools, and 402 typically developing(TD) children in the same age span from a communitybased population to complete the ASRS and SRS. The results showed that the ID group scored significantly lower on total and subscale scores than the ASD group on both scales(P \ 0.05) but higher than TD children(P \ 0.05).Receiver operating characteristic analyses demonstrated a similar fair performance in discriminating ASD from ID with the ASRS(area under the curve(AUC) = 0.709,sensitivity = 77.0%, specificity = 52.1%, positive predictive value(PPV) = 82.2%) and the SRS(AUC = 0.742,sensitivity = 59.8%, specificity = 77.5%, PPV = 88.4%).The results showed that individuals with ID had clear autistic traits and discriminating ASD from ID cases was quite challenging, while assessment tools such as ASRS and SRS, help to some degree.Chunpei Li Hao Zhou Tianqi Wang Shasha Long Xiaonan Du Xiu Xu Weili Yan Yi Wang 2018Neuroscience Bulletin2018,34,6:5
15Phytosterol content and the campesterol:sitosterol ratio influence cotton fiber development: role of phytosterols in cell elongation显示文摘Phytosterols play an important role in plant growth and development,including cell division,cell elongation,embryogenesis,cellulose biosynthesis,and cell wall formation.Cotton fiber,which undergoes synchronous cell elongation and a large amount of cellulose synthesis,is an ideal model for the study of plant cell elongation and cell wall biogenesis.The role of phytosterols in fiber growth was investigated by treating the fibers with tridemorph,a sterol biosynthetic inhibitor.The inhibition of phytosterol biosynthesis resulted in an apparent suppression of fiber elongation in vitro or in planta.The determination of phytosterol quantity indicated that sitosterol and campesterol were the major phytosterols in cotton fibers;moreover,higher concentrations of these phytosterols were observed during the period of rapid elongation of fibers.Furthermore,the decrease and increase in campesterol:sitosterol ratio was associated with the increase and decease in speed of elongation,respectively,during the elongation stage.The increase in the ratio was associated with the transition from cell elongation to secondary cell wall synthesis.In addition,a number of phytosterol biosynthetic genes were down-regulated in the short fibers of ligon lintless-1mutant,compared to its near-isogenic wild-type TM-1.These results demonstrated that phytosterols play a crucial role in cotton fiber development,and particularly in fiber elongation.Shasha Deng Ting Wei Kunling Tan Mingyu Hu Fang Li Yunlan Zhai Shue Ye Yuehua Xiao Lei Hou Yan Pei Ming Luo 2016Science China(Life Sciences)2016,59,2:5

17Diagenesis and Diagenetic Evolution of Deltaic and Neritic Gas-Bearing Sandstones in the Lower Mingyuefeng Formation of Paleogene, Lishui Sag, East China Sea Shelf Basin: Implications for Depositional Environments and Sequence Stratigraphy Controls显示文摘Gas-bearing deposits in the Lower Mingyuefeng Formation of Paleogene, Lishui Sag, East China Sea Shelf Basin consist of shoreface sandstones of the highstand systems tract(HST) and transgressive systems tract(TST), and deltaic sandstones of the lowstand systems tract(LST) and falling stage systems tract(FSST).Detailed petrographic observations suggest that the diagenetic features and related evolution of these deposits cannot be simply characterized and demonstrated in the depth domain.However, the occurrence of diagenetic minerals systematically depends on the studied interval within the HST, TST, LST, and FSST; therefore, diagenesis in this region can be better constrained when studied in the context of the depositional environments and sequence stratigraphic framework.The eogenetic processes in such settings include:(1) microcrystalline siderite precipitated as concretions in almost all environments and systems tracts, which inhibited further mechanical compaction;(2) grain dissolution and kaolinitization occurred in shoreface HST sandstones and deltaic LST and FSST sandstones;(3) glaucony was locally observed, which did not clearly reflect the controls of facies or sequence stratigraphy; and(4) cementation by pyrite aggregates occurred in the shoreface HST sandstones and deltaic LST sandstones.The mesogenetic diagenesis includes:(1) partial conversion of kaolinite into dickite in deltaic LST sandstones, and minor chlorite cementation in deltaic FSST sandstones;(2) transformation of kaolinite into illite and quartz cementation in deltaic LST and FSST sandstones;(3) frequent precipitation of ankerite and ferroan calcite in shoreface TST sandstones and early HST sandstones, forming baffles and barriers for fluid flow, with common calcite in shoreface HST sandstones as a late diagenetic cement; and(4) formation of dawsonite in the deltaic LST and FSST sandstones, which is interpreted to be a product of the invasion of a CO2-rich fluid, and acts as a good indicator of CO2-bearing reservoirs.This study has thus constructed a reliable conceptual model to describe the spatial and temporal distribution of diagenetic alterations.The results may provide an entirely new conceptual framework and methodology for successful gas exploration in the continental margins of offshore China, thus allowing us to predict and unravel the distribution and quality evolution of clastic reservoirs at a more detailed and reliable scale.XU Fa ZHANG Penghui ZHANG Jinliang LIU Jinshui HOU Guowei ZHANG Ming LI Jingzhe LIU Shasha GUO Jiaqi MENG Ningning 2015Acta Geologica Sinica(English Edition)2015,89,5:5

19Expression of the DNA-repair protein XRCC1 and DNA-repair enzyme Ku70 mRNA in the rat cerebral cortex following global cerebral ischemia/reperfusion Is this correlated with neuroprotective effects of theanine?显示文摘BACKGROUND: Numerous studies have shown that apoptosis, caused by cerebral ischemia/reperfusion injury, is a dynamic evolving process that occurs in a time-dependent fashion. Non-apoptotic cells can result in apoptosis, with a prolonged reperfusion period and an accumulation of DNA damage. Recent studies have indicated that theanine has a protective effect on the brain. Nonetheless, there is no research relating to whether theanine is neuroprotective following brain injury. OBJECTIVE: To investigate the effects of theanine on apoptosis in the cerebral cortex and mRNA expression of the DNA repair protein, XRCC1, and DNA repair enzyme, Ku70, at various time points following global cerebral ischemia/reperfusion in rats. DESIGN, TIME AND SETTING: A randomized, controlled animal study was performed at the Pharmacological Laboratory of Animal Experimental Center of Xi'an Jiaotong University, China, from April to October 2007. MATERIALS: Theanine (Wuhan Remote Technology Co., Ltd., China), XRCC1, Ku70 and GAPDH primers (Shanghai Sangon Biological Engineering Technology & Services Co., Ltd., China) were used in this study. METHODS: A total of 108 healthy, male Sprague Dawley rats were randomly divided into sham operation, ischemia/reperfusion and theanine groups. Each group was further divided into 6 sub- groups, with six rats in each, according to reperfusion time (2, 6, 12, 24, 48 and 72 hours). Rat models of global cerebral ischemia/reperfusion were established by the four-vessel occlusion method. Theanine (1 000 mg/kg) was injected into the caudal vein of rats in the theanine group immediately after surgery. Saline (4 mL/kg) was used in the sham operation and ischemia/reperfusion groups. The same dose was administered every 24 hours. MAIN OUTCOME MEASURES: Flow cytometry was employed to determine apoptosis of cerebral cortical neurons. Real-time reverse transcription-polymerase chain reaction was applied to quantify the expression of XRCC1 and Ku70 mRNA in the cerebral cortex. RESULTS: The apoptotic rate in the ischemia/reperfusion group was significantly greater compared with the sham operation group at various time points (P < 0.05). The apoptotic rate in the theanine group was significantly diminished compared with the ischemia/reperfusion group, but significantly increased compared with the sham operation group at 6, 12, 24, 48 and 72 hours (P < 0.05). Ex- pression of XRCC1 and Ku70 mRNA was significantly decreased in the ischemia/reperfusion group compared with the sham operation group at 2, 6, 12, 24, 48 and 72 hours (P < 0.05). The relative expression of XRCC1 mRNA in the theanine group was significantly increased compared with the ischemia/reperfusion group, but significantly reduced compared with the sham operation group at 6, 12, 24, 48 and 72 hours (P < 0.05). The relative expression of Ku70 mRNA was significantly lower in the theanine group compared with the sham operation group at 2, 6 and 12 hours (P < 0.05) and significantly greater than the ischemia/reperfusion group at 6, 12, 24, 48 and 72 hours (P < 0.05). CONCLUSION: The apoptotic rate in rats following global cerebral ischemia/reperfusion injury reaches a peak at 24 hours after reperfusion; however, the levels of DNA repair protein began to decrease after only 2 hours of reperfusion. 20High-throughput sequencing exclusively identified a novel Torque teno virus genotype in serum of a patient with fatal fever显示文摘Torque teno virus(TTV) has been found to be prevalent world-wide in healthy populations and in patients with various diseases, but its etiological role has not yet been determined. Using high-throughput unbiased sequencing to screen for viruses in the serum of a patient with persistent high fever who died of suspected viral infection and prolonged weakness, we identified the complete genome sequence of a TTV(isolate Hebei-1). The genome of TTV-Hebei-1 is 3649 bp in length, encoding four putative open reading frames, and it has a G+C content of 49%. Genomic comparison and a BLASTN search revealed that the assembled genome of TTV-Hebei-1 represented a novel isolate, with a genome sequence that was highly heterologous to the sequences of other reported TTV strains. A phylogenetic tree constructed using the complete genome sequence showed that TTV-Hebei-1 and an uncharacterized Taiwan Residents strain, TW53A37, constitute a new TTV genotype. The patient was strongly suspected of carrying a viral infection and died eventually without any other possible causes being apparent. No virus other than the novel TTV was identified in his serum sample.
