1Serum semaphorin 4C as a diagnostic biomarker in breast cancer:A multicenter retrospective study显示文摘Background:To date,there is no approved blood-based biomarker for breast cancer detection.Herein,we aimed to assess semaphorin 4C(SEMA4C),a pivotal protein involved in breast cancer progression,as a serum diagnostic biomarker.Methods:We included 6,213 consecutive inpatients from Tongji Hospital,Qilu Hospital,and Hubei Cancer Hospital.Training cohort and two validation cohorts were introduced for diagnostic exploration and validation.A pan-cancer cohort was used to independently explore the diagnostic potential of SEMA4C among solid tumors.Breast cancer patients who underwent mass excision prior to modified radical mastectomy were also analyzed.We hypothesized that increased pretreatment serum SEMA4C levels,measured using optimized in-house enzymelinked immunosorbent assay kits,could detect breast cancer.The endpoints were diagnostic performance,including area under the receiver operating characteristic curve(AUC),sensitivity,and specificity.Post-surgery pathological diagnosis was the reference standard and breast cancer staging followed the TNM classification.There was no restriction on disease stage for eligibilities.Results:We included 2667 inpatients with breast lesions,2378 patients with other solid tumors,and 1168 healthy participants.Specifically,118 patients with breast cancer were diagnosed with stage 0(5.71%),620 with stage I(30.00%),966 with stage II(46.73%),217 with stage III(10.50%),and 8 with stage IV(0.39%).Patients with breast cancer had significantly higher serum SEMA4C levels than benign breast tumor patients and normal controls(P<0.001).Elevated serum SEMA4C levels had AUC of 0.920(95%confidence interval[CI]:0.900–0.941)and 0.932(95%CI:0.911–0.953)for breast cancer detection in the two validation cohorts.The AUCs for detecting early-stage breast cancer(n=366)and ductal carcinoma in situ(n=85)were 0.931(95%CI:0.916–0.946)and 0.879(95%CI:0.832–0.925),respectively.Serum SEMA4C levels significantly decreased after surgery,and the reduction was more striking after modified radical mastectomy,compared with mass excision(P<0.001).The positive rate of enhanced serum SEMA4C levels was 84.77%for breast cancer and below 20.75%for the other 14 solid tumors.Conclusions:Serum SEMA4C demonstrated promising potential as a candidate biomarker for breast cancer diagnosis.However,validation in prospective settings and by other study groups is warranted.Ya Wang Long Qiao Jie Yang Xiong Li Yaqi Duan Jiahao Liu Shaoqi Chen Huayi Li Dan Liu Tian Fang Jingjing Ma Xiaoting Li Fei Ye Junxiang Wan Juncheng Wei Qin Xu Ensong Guo Ping Jin Mingfu Wu Lin Zhang Yun Xia Yaqun Wu Jun Shao Yaojun Feng Qing Zhang Zongyuan Yang Gang Chen Qinghua Zhang Xingrui Li Shixuan Wang Junbo Hu Xiaoyun Wang Mona P.Tan Kazuaki Takabe Beihua Kong Qifeng Yang Ding Ma Qinglei Gao 2021Cancer Communications2021,41,12:3
2Determination of time-and size-dependent fine particle emission with varied oil heating in an experimental kitchen显示文摘Particulate matter(PM) from cooking has caused seriously indoor air pollutant and aroused risk to human health.It is urged to get deep knowledge of their spatial-temporal distribution of source emission characteristics,especially ultrafine particles(UFP < 100 nm) and accumulation mode particles(AMP 100-555 nm).Four commercial cooking oils are auto dipped water to simulate cooking fume under heating to 255℃ to investigate PM emission and decay features between 0.03 and 10 μm size dimension by electrical low pressure impactor(ELPI) without ventilation.Rapeseed and sunflower produced high PM_(2.5) around5.1 mg/m^3,in comparison with those of soybean and corn(5.87 and 4.55 mg/m^3,respectively)at peak emission time between 340 and 450 sec since heating oil,but with the same level of particle numbers 6-9 × 10~5/cm^3.Mean values of PM_(1.0)/PM_(2.5) and PM_(2.5)/PM_(10) at peak emission time are around 0.51-0.55 and 0.23-0.29.After 15 min naturally deposition,decay rates of PM_(1.0),PM_(2.5) and PM_(10) are 13.3%-29.8%,20.1%-33.9%and 41.2%-54.7%,which manifest that PM_(1.0) is quite hard to decay than larger particles,PM_(2.5) and PM_(1.0).The majority of the particle emission locates at 43 nm with the largest decay rate at 75%,and shifts to a larger size between137 and 555 nm after 15 min decay.The decay rates of the particles are sensitive to the oil type.Shuangde Li Jiajia Gao Yiqing He Liuxu Cao Ang Li Shengpeng Mo Yunfa Chen Yaqun Cao 2017Journal of Environmental Sciences2017,29,1:3
3The oxidative DNA damage response: A review of research undertaken with Tsinghua and Xiangya students at the University of Pittsburgh显示文摘Endogenous stress and exogenous toxicants(chemicals and UV light) alter genetic information either directly or indirectly through the production of reactive oxygen species(ROS), thereby driving genomic instability in cells and promoting tumorigenesis. All living cells try to faithfully preserve and transmit their genomic information from one generation to the next using DNA repair mechanisms to repair oxidative DNA damage to prevent cancer or premature aging. Oxidative DNA damage comprises a mixture of DNA lesions including base damage, DNA single strand breaks(SSBs), and DNA double strand breaks(DSBs). This review summarizes some of the studies on DNA damage response at a defined genome locus that are performed by students from the Tsinghua University School of Medicine and the School of Medicine of Central South University(Xiangya Hospital) at the University of Pittsburgh School of Medicine. 4Removal of hydrophobic volatile organic compounds with sodium hypochlorite and surfactant in a co-current rotating packed bed显示文摘A co-current flow rotating packed bed was applied to remove volatile organic compounds(VOCs) by sodium hypochlorite(Na Cl O) and surfactant(sodium dodecyl benzene sulfonate,SDBS) from air stream. Xylene was used as a model VOC herein. The effect of p H,concentration of Na Cl O and SDBS solution, liquid flow rate, gas flow rate and rotational speed on xylene removal efficiency and overall mass transfer coefficient(KGa) were discussed. 5Genomic diversity and post-admixture adaptation in the Uyghurs显示文摘Population admixture results in genome-wide combinations of genetic variants derived from different ancestral populations of distinct ancestry, thus providing a unique opportunity for understanding the genetic determinants of phenotypic variation in humans. Here, we used whole-genome sequencing of 92 individuals with high coverage(30–60×) to systematically investigate genomic diversity in the Uyghurs living in Xinjiang, China(XJU), an admixed population of both European-like and East-Asian-like ancestry.The XJU population shows greater genetic diversity, especially a higher proportion of rare variants,compared with their ancestral source populations, corresponding to greater phenotypic diversity of XJU.Admixture-induced functional variants in EDAR were associated with the diversity of facial morphology in XJU. Interestingly, the interaction of functional variants between SLC24A5 and OCA2 likely influences the diversity of skin pigmentation. Notably, selection has seemingly been relaxed or canceled in several genes with significantly biased ancestry, such as HERC2–OCA2. Moreover, signatures of post-admixture adaptation in XJU were identified, including genes related to metabolism(e.g. CYP2D6), digestion(e.g.COL11A1), olfactory perception(e.g. ANO2) and immunity(e.g. HLA). 8Activation of G-protein-coupled receptor 39 reduces neuropathic pain in a rat model显示文摘Activated G-protein-coupled receptor 39(GPR39)has been shown to attenuate inflammation by interacting with sirtuin 1(SIRT1)and peroxisome proliferator-activated receptor-γcoactivator 1α(PGC-1α).However,whether GPR39 attenuates neuropathic pain remains unclear.In this study,we established a Sprague-Dawley rat model of spared nerve injury-induced neuropathic pain and found that GPR39 expression was significantly decreased in neurons and microglia in the spinal dorsal horn compared with sham-operated rats.Intrathecal injection of TC-G 1008,a specific agonist of GPR39,significantly alleviated mechanical allodynia in the rats with spared nerve injury,improved spinal cord mitochondrial biogenesis,and alleviated neuroinflammation.These changes were abolished by GPR39 small interfering RNA(siRNA),Ex-527(SIRT1 inhibitor),and PGC-1αsiRNA.Taken together,these findings show that GPR39 activation ameliorates mechanical allodynia by activating the SIRT1/PGC-1αpathway in rats with spared nerve injury.Longqing Zhang Xi Tan Fanhe Song Danyang Li Jiayi Wu Shaojie Gao Jia Sun Daiqiang Liu Yaqun Zhou Wei Mei 2024Neural Regeneration Research2024,19,3:0
9Experimental Study of Dissolution-Alteration of Amphibole in a Hydrothermal Environment显示文摘Amphibole is a rock-forming mineral widely existing on the earth.It is easily dissolved and altered during the later stage of diagenesis and mineralization,and often forms chloritization,which is an important indicator for prospecting.To explore amphibole's dissolution process and alteration mechanism,dissolution experiments were carried out under acidic conditions using pargasite-rich amphibole as raw material,and the effects of temperature,p H,and experiment duration on amphibole alteration were investigated.Experimental samples and products were analyzed using X-ray diffractometer,field emission scanning electron microscope,electron probe micro analyzer,and transmission electron microscopy.It was found that many pores and erosion edges are produced after amphibole dissolution,and there is a clear interface between the dissolved residual portion and the parent.The dissolved residual portion remains in the amphibole phase,but as the temperature and time increase,the intensity of the diffraction peak of the phase in the product decreases,and the peak position shifts to a small angle.Many clay minerals such as chlorite and griffithite formed on the amphibole surface.In an environment with strong acidity(p H=3),the amount of chamosite increases with temperature(180℃→210℃→240℃),whereas clinochlore is only increased in a 150–210℃environment.Griffithite growth was observed in the acidic(p H=6)and low temperature(<180℃)environments.Based on this analysis,large radius Cl–enters the amphibole lattice or cracks to promote dissolution.The Al-poor and Ca-and Fe-rich regions between the edge and core of the amphibole are caused by dynamic equilibrium in amphibole dissolution and alteration process,which is an essential indicator for the beginning of amphibole dissolution-alteration.Diffusion and the coupled dissolution-reprecipitation mechanism accomplishes the process of dissolution and alteration to form clay minerals.The energy of the system determined by temperature and p H is the key to controlling the rate of growth and nucleation of clay minerals.High temperature and strong acidity will dissolve more iron from amphibole,which is conducive to chlorite growth.Compared to chlorite,griffithite is more sensitive to temperature.Griffithite attaches on the amphibole surface with a star-like in a weak acid and low-temperature environment.The results of this study can provide a mineralogical basis for the analysis of hydrothermal alteration processes and the division of alteration zones.LI Yongli HUANG Fei GAO Wenyuan TANG Xu REN Yaqun MENG Lin ZHANG Zhibin 2019Acta Geologica Sinica(English Edition)2019,93,6:0
