1Immunomodulatory effect of human amniotic epithelial cells on restoration of ovarian function in mice with autoimmune ovarian disease显示文摘Autoimmune ovarian disease(AOD)is considered to be a major cause of premature ovarian failure(POF).The immunomodulatory properties of human amniotic epithelial cells(hAECs)have been studied in many disease models.We previously reported that hAECs restored ovarian function in chemotherapy-induced POF mice,but the immunomodulatory mechanism of hAECs is still unclear.To investigate the effect of hAECs on recipient mice,especially on regulatory Treg cells,hAECs and hAEC-conditioned medium(hAEC-CM)were intravenously injected into AOD mice immunized with zona pellucida protein 3 peptides(pZP3).Ovarian function was evaluated through estrous cycle,hormone secretion,follicle development,and cell apoptosis analysis.Immune cells including CD3,CD4,CD8 and Treg cells in the spleens were tested by flow cytometry.To elucidate the effect of hAEC-CM on macrophage function,inflammation model in vitrowas established in RAW264.7 cells induced by lipopolysaccharide(LPS).hAECs and hAEC-CM regulated estrous cycles,promoted follicle development,ameliorated cell apoptosis and fibrosis in ovaries of AOD mice.In addition,hAECs significantly reversed the decrease of pZP3-induced Treg cells in the spleens.In vitro,hAEC-CM significantly inhibited the inflammatory reaction induced by LPS in RAW264.7 cells via up-regulating the expression of M2 macrophage genes.Further study demonstrated that hAECsecreted transforming growth factor-beta and macrophage inhibitory factor played important roles in the macrophage polarization and migration under inflammatory stimulation.Taken together,hAECs restored ovarian function by up-regulating Treg cells in the spleens and reduced the inflammatory reaction via modulating the activated macrophage function in a paracrine manner in the ovaries of AOD mice.Qiuwan Zhang Yating Huang Junyan Sun Tingting Gu Xiaoyan Shao Dongmei Lai 2019Acta Biochimica et Biophysica Sinica2019,51,8:14
2The Genesis Mechanism of the Mantle Fluid Action and Evolution in the Ore-Forming Process: A Case Study of the Laowangzhai Gold Deposit in Yunnan, China显示文摘Based on petrological studies of the wall rocks, mineralizing rocks, ores and veins from the Laowangzhai gold deposit, it is discovered that along with the development of silication, carbonation and sulfidation, a kind of black opaque ultra-microlite material runs through the spaces between grains, fissures and cleavages. Under observations of the electron microprobe, scanning electronic microscopy and energy spectrum, this kind of ultra-microlite material is confirmed to consist of ultra microcrystalline quartz, silicate, sulfides and carbonates, as well as rutile, scheelite and specularite (magnetite), showing characters of liquation by the analyses of SEM and energy spectrum. The coexistence of immiscibility and precipitating co-crystallization strongly suggests that the mineralizing fluid changed from the melt to the hydrothermal fluid. Combined with the element geochemical researches, it is realized that the ultra-microlite aggregate is the direct relics of the mantle fluid behaving like a melt and supercritical fluid, which goes along with the mantle-derived magma and will escape from the magma body at a proper time. 3The effect of Fe-Mn minerals and seawater interface and enrich-ment mechanism of ore-forming elements of polymetallic crusts and nodules from the South China Sea显示文摘Ferromanganese crusts and nodules are important submarine mineral resources that contain various metal elements with significant economic value. In this study, polymetallic crusts and nodules obtained from the South China Sea(SCS) were determined by using X-ray power diffraction(XRD), Raman spectroscopy(RS), Fourier transform infrared spectroscopy(FTIR), and X-ray photoelectron spectroscopy(XPS) to systematically investigate and analyze the mineralogical and spectral characteristics of the Fe-Mn minerals. XRD measurements revealed that the SCS polymetallic crusts and nodules were composed of vernadite, quartz, and plagioclase. The nodules also contained todorokite. The Fe-phase minerals of the SCS crusts and nodules were composed of amorphous Fe oxide/hydroxide, and the Mn-and Fe-phases minerals exhibited relatively poor degrees of crystallization. FTIR results showed that the Fe-Mn minerals in the crusts and nodules included a large number of surface hydroxyl groups. These surface hydroxyl groups contained protons that could provide reactive sites for complexation of ore-forming elements in seawater. XPS results indicated that the surfaces of the Fe-Mn minerals mainly contained Fe, Mn, and O. Fe was present in the trivalent oxidation state, while Mn, which may contain several bivalent oxidation state, was present in the tetravalent and trivalent oxidation states. The SCS polymetallic crusts and nodules were compared with Pacific seamount crusts, and results showed that the surface hydroxyl(-OH) groups of the SCS crusts and nodules numbered more than the lattice oxygen(O^(2-)). But the lattice oxygen of Pacific seamount crusts numbered more than the surface hydroxyl groups. This characteristic indicated that the degree of crystallization of Fe-Mn minerals from the Pacific Ocean was higher than that of minerals from the South China Sea. Comprehensive studies showed that ore-forming elements in the interface between seawater and the Fe-Mn minerals in the submarine ferromanganese crusts and nodules employed the following enrichment mechanisms:(1) the metal ion complexed with the surface hydroxyl of Fe-Mn minerals to form hydroxyl complexes, which were connected by coordination bonds or stable inner-sphere complexes that exchanged protons on the mineral surfaces;(2) the charged surfaces of the minerals and metal cations formed outer-sphere complexes, which made up the electrostatic double layer, through electrostatic adsorption; and(3) the metal cations isomorphously exchanged the Mn and Fe ions of the mineral lattice structure.GUAN Yao SUN Xiaoming IANG Xiaodong SA Rina ZHOU Li HUANG Yi LIU Yating LI Xiaojie LU Rongfei WANG Chi 2017Acta Oceanologica Sinica2017,36,6:4
4Probing particle removal in brush scrubber cleaning with fluorescence technique显示文摘Brush scrubber cleaning is widely used for post chemical mechanical polishing(CMP)cleaning in semiconductor manufacturing.In this study,an experimental system based on fluorescence technique and particle-tracking velocimetry(PTV)technique was employed to characterize the particle removal displacement and velocity in the interface between a transparent copper film and a porous polyvinyl alcohol(PVA)brush during the cleaning process.Several different cleaning conditions including rotation speeds,loading pressure and cleaning agent were examined and the particle removal rate was compared.Elastic and friction removal,hydrodynamic removal and mixed-type removal are the three types of particle removal.Particles with an arc trace and uniform velocity curves were removed by friction and elastic force which were related to the brush load.Particles with a random trace and fluctuant velocity curves were removed by hydrodynamic force which was determined by the brush rotation speed.The increase of particle removal rate(PRR)with brush rotation speed is a logistic function.It is easier to improve PRR by increasing the brush load or by adding surfactant than by increasing the brush rotation speed.HUANG YaTing LI Yang GUO Dan MENG ChunLing 2013Science China(Technological Sciences)2013,56,12:3
5Heavy soil drying during mid-to-late grain filling stage of the main crop to reduce yield loss of the ratoon crop in a mechanized rice ratooning system显示文摘Yield loss(Y_(Loss)) in the ratoon crop due to crushing damage to left stubble from mechanical harvesting of the main crop is a constraint for wide adoption of mechanized rice ratooning technology.Soil drying before the harvest of the main crop has been proposed to overcome this problem.The objective of this study was to determine the effect of soil drying during the mid-to-late grain filling stage of the main crop on grain yield of the ratoon crop in a mechanized rice ratooning system.Field experiments were conducted to compare Y_(Loss) between light(LD) and heavy(HD) soil drying treatments in Hubei province,central China in 2017 and 2018.Y_(Loss) was calculated as the percentage of yield reduction in the ratoon crop with the main crop harvested mechanically,relative to the grain yield of the ratoon crop with the main crop harvested manually.In comparison with LD,soil hardness was increased by 42.8%-84.7% in HD at the 5-20 cm soil depth at maturity of the main crop.Soil hardness at 5 and 10 cm depths reached respectively 4.05 and 7.07 kg cm^(-2) in HD.Soil drying treatment did not significantly affect the grain yield of the main crop.Under mechanical harvesting of the main crop,HD increased the grain yield of the ratoon crop by 9.4% relative to LD.Consequently,Y_(Loss) was only 3.4% in HD,in contrast to 16.3% in LD.The differences in grain yield and Y_(Loos) between the two soil drying treatments were explained mainly by panicles m^(-2),which was increased significantly by HD in the track zone of the ratoon crop compared with LD.These results suggest that heavy soil drying practice during the mid-to-late grain filling stage of the main crop is effective for reducing Y_(Loss) of the ratoon crop in a mechanized rice ratooning system.Chang Zheng Yuechao Wang Shen Yuan Sen Xiao Yating Sun Jianliang Huang Shaobing Peng 2022The Crop Journal2022,10,1:3

7A new electroplated Ir/Ir(OH)_x pH electrode and its application in the coastal areas of Newport Harbor, California显示文摘Resulting from the rising levels of atmospheric carbon, ocean acidification has become a global problem. It has significant impacts on the development, survival, growth and physiology of marine organisms. Therefore, a highprecision sensor is urgently needed to measure the pH of sea-water. Iridium wire with a diameter of 0.25 mm is used as the substrate, and an Ir/Ir(OH)_x pH electrode is prepared by a one-step electrochemical method in a Li OH solution at the room temperature. A scanning electron microscope(SEM) observation reveals that it is coated with nanoscale particles. In laboratory tests, the electrode exhibits a very promising pH response, with an ideal Nernst slope(56.14–59.52), fast response, good stability and long life-span in tested pH buffer solutions. For a sea trial,four pH electrodes and one Ag/Ag Cl reference electrode are integrated with a self-made chemical sensor, and a profile detection of nearly 70 m is implemented near Newport Harbor, California on August 3, 2015. The results reflect that the pH value measured by the sensor is very close to the data given by Sea-Bird 911 plus CTD, with a difference value ranging from 0.000 075 to 0.064 719. And the sensor shows a better data matching degree in 0–40m water depth. 8Inhibitory mechanism of angiotensin-converting enzyme inhibitory peptides from black tea显示文摘The aim of this work is to discover the inhibitory mechanism of tea peptides and to analyse the affinities between the peptides and the angiotensin-converting enzyme(ACE)as well as the stability of the complexes using in vitro and in silico methods.Four peptide sequences identified from tea,namely peptides I,II,III,and IV,were used to examine ACE inhibition and kinetics.The half maximal inhibitory concentration(IC_(50))values of the four peptides were(210.03±18.29),(178.91±5.18),(196.31±2.87),and(121.11±3.38)μmol/L,respectively.The results of Lineweaver-Burk plots showed that peptides I,II,and IV inhibited ACE activity in an uncompetitive manner,which requires the presence of substrate.Peptide III inhibited ACE in a noncompetitive manner,for which the presence of substrate is not necessary.The docking simulations showed that the four peptides did not bind to the active sites of ACE,indicating that the four peptides are allosteric inhibitors.The binding free energies calculated from molecular dynamic(MD)simulation were-72.47,-42.20,-52.10,and-67.14 kcal/mol(1 kcal=4.186 kJ),r espectively.The lower IC_(50)value of peptide IV may be attributed to its stability when docking with ACE and changes in the flexibility and unfolding of ACE.These four bioactive peptides with ACE inhibitory ability can be incorporated into novel functional ingredients of black tea.Yating LU Yu WANG Danyi HUANG Zhuang BIAN Peng LU Dongmei FAN Xiaochang WANG 2021Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2021,22,7:2

18Single-cell-resolution transcriptome map revealed novel genes involved in testicular germ cell progression and somatic cells specification in Chinese tongue sole with sex reversal显示文摘Female-to-male sex reversals(pseudomales)are common in lower vertebrates and have been found in natural populations,which is a concern under rapid changes in environmental conditions.Pseudomales can exhibit altered spermatogenesis.However,the regulatory mechanisms underlying pseudomale spermatogenesis remain unclear.Here,we characterized spermatogenesis in Chinese tongue sole(Cynoglossus semilaevis),a species with genetic and environmental sex determination,based on a highresolution single-cell RNA-seq atlas of cells derived from the testes of genotypic males and pseudomales.We identified five germ cell types and six somatic cell types and obtained a single-cell atlas of dynamic changes in gene expression during spermatogenesis in Chinese tongue sole,including alterations in pseudomales.We detected decreased levels of Ca^(2+)signaling pathway-related genes in spermatogonia,insufficient meiotic initiation in spermatocytes,and a malfunction of somatic niche cells in pseudomales.However,a cluster of CaSR genes and MAPK signaling factors were upregulated in undifferentiated spermatogonia of pseudomales.Additionally,we revealed that Z chromosome-specific genes,such as piwil2,dhx37,and ehmt1,were important for spermatogenesis.These results improve our understanding of reproduction after female-to-male sex-reversal and provide new insights into the adaptability of reproductive strategies in lower vertebrates.Hong-Yan Wang Xiang Liu Jian-Yang Chen Yingyi Huang Yifang Lu Fujian Tan Qun Liu Mingming Yang Shuo Li Xianghui Zhang Yating Qin Wenxiu Ma Yingming Yang Liang Meng Kaiqiang Liu Qian Wang Guangyi Fan Rafael H.Nóbrega Shanshan Liu Francesc Piferrer Changwei Shao 2023Science China(Life Sciences)2023,66,5:1
