2Deciphering suppressive effects of Lianhua Qingwen Capsule on COVID-19 and synergistic effects of its major botanical drug pairs显示文摘The COVID-19 pandemic has resulted in excess deaths worldwide.Conventional antiviral medicines have been used to relieve the symptoms,with limited therapeutic effect.In contrast,Lianhua Qingwen Capsule is reported to exert remarkable anti-COVID-19 effect.The current review aims to:1)uncover the main pharmacological actions of Lianhua Qingwen Capsule for managing COVID-19;2)verify the bioactive ingredients and pharmacological actions of Lianhua Qingwen Capsule by network analysis;3)investigate the compatibility effect of major botanical drug pairs in Lianhua Qingwen Capsule;and 4)clarify the clinical evidence and safety of the combined therapy of Lianhua Qingwen Capsule and conventional drugs.Numerous bioactive ingredients in Lianhu Qingwen,such as quercetin,naringenin,β-sitosterol,luteolin,and stigmasterol,were identified to target host cytokines,and to regulate the immune defence in response to COVID-19.Genes including androgen receptor(AR),myeloperoxidase(MPO),epidermal growth factor receptor(EGFR),insulin(INS),and aryl hydrocarbon receptor(AHR)were found to be significantly involved in the pharmacological actions of Lianhua Qingwen Capsule against COVID-19.Four botanical drug pairs in Lianhua Qingwen Capsule were shown to have synergistic effect for the treatment of COVID-19.Clinical studies demonstrated the medicinal effect of the combined use of Lianhua Qingwen Capsule and conventional drugs against COVID-19.In conclusion,the four main pharmacological mechanisms of Lianhua Qingwen Capsule for managing COVID-19 are revealed.Therapeutic effect has been noted against COVID-19 in Lianhua Qingwen Capsule.CHEN Yuanyuan ZHANG Cheng WANG Ning FENG Yibin 2023Chinese Journal of Natural Medicines2023,21,5:2
3PIWIL1 governs the crosstalk of cancer cell metabolism and immunosuppressive microenvironment in hepatocellular carcinoma显示文摘Altered energy metabolism of cancer cells shapes the immune cell response in the tumor microenvironment that faclitates tumorprogression.Herein,we reported the novel of tumor cell-expressed Piwi Like RNA-Mediated Gene Silencing 1(PWL1)in mediatingthe crosstlk of fatty acid metabolism and immune response of human hepatocellular carcinoma(HCC).PlWIL1 expression in HCCwas increased compared to normal hepatic tissues and was positively correlated with the proliferation rate of HCC cell ines.PIWL1overexpression accelerated in vitro proliferation and in vivo growth of HCC tumors,while PIWL1 knockdown showed opositeeffects.PIWL1 increased oxygen consumption and energy production via fatty acid metabolism without altering aerobic glycolysis.lnhibition of fatty acid metablism abolished PWIL1-induced HCC prolferation and growth.RNA-seq analysis revealed that immunesystem regulation might be involved,which was echoed by the experimental observation that PIWL1-overexpressing HCC cellsattracted myeloid-derived suppressor cells(MDSCs)into the tumor microenvironment.MDSCs depletion reduced the prolferationand growth of PlWIL1-overexpressing HCC tumors.Complement C3,whose secretion was induced by PIWL1 in HCC cells,mediatesthe interaction of HCC cells with MDSCs by activated p38 MAPK signaling in MDSCs,which in turn initiated expression of immunosuppressive cytokine L10.Neutralizing lL10 secretion reduced the immunosuppressive activity of MDSCs in the microenvironment of PIWL1-overexpressing HCC.Taken together,our study unraveled the critical role of PIWL1 in initiating theinteraction of cancer cell metabolism and immune cell response in HCC.Tumor cell-expressed PlIWL1 may be a potential target forthe devellopment of novel HCC treatment.Ning Wang Hor-Yue Tan Yuanjun Lu Yau-Tuen Chan Di Wang Wei Guo Yu Xu Cheng Zhang Feiyu Chen Guoyi Tang Yibin Feng 2021Signal Transduction and Targeted Therapy2021,6,3:2

5Pancreatic melatonin enhances anti-tumor immunity in pancreatic adenocarcinoma through regulating tumor-associated neutrophils infiltration and NETosis显示文摘Tumor microenvironment contributes to poor prognosis of pancreatic adenocarcinoma(PAAD)patients.Proper regulation could improve survival.Melatonin is an endogenous hormone that delivers multiple bioactivities.Here we showed that pancreatic melatonin level is associated with patients'survival.In PAAD mice models,melatonin supplementation suppressed tumor growth,while blockade of melatonin pathway exacerbated tumor progression.This anti-tumor effect was independent of cytotoxicity but associated with tumor-associated neutrophils(TANs),and TANs depletion reversed effects of melatonin.Melatonin induced TANs infiltration and activation,therefore induced cell apoptosis of PAAD cells.Cytokine arrays revealed that melatonin had minimal impact on neutrophils but induced secretion of Cxcl2 from tumor cells.Knockdown of Cxcl2 in tumor cells abolished neutrophil migration and activation.Melatonin-induced neutrophils presented an N1-like anti-tumor phenotype,with increased neutrophil extracellular traps(NETs)causing tumor cell apoptosis through cell-to-cell contact.Proteomics analysis revealed that this reactive oxygen species(ROS)-mediated inhibition was fueled by fatty acid oxidation(FAO)in neutrophils,while FAO inhibitor abolished the anti-tumor effect.Analysis of PAAD patient specimens revealed that CXCL2 expression was associated with neutrophil infiltration.CXCL2,or TANs,combined with NET marker,can better predict patients'prognosis.Collectively,we discovered an anti-tumor mechanism of melatonin through recruiting N1-neutrophils and beneficial NET formation.Yau-tuen Chan Hor-yue Tan Yuanjun Lu Cheng Zhang Chien-shan Cheng Junyu Wu Ning Wang Yibin Feng 2023Acta Pharmaceutica Sinica B2023,13,4:1

9Modifications in aroma characteristics of'Merlot'dry red wines aged in American,French and Slovakian oak barrels with different toasting degrees显示文摘Aging in oak barrels is widely used in enology which could bring flavor changes and aromatic complexity to wines.In the present study,the aroma compounds were analyzed from the‘Merlot’dry red wines,which were fermented in two types of fermenters(stainless steel tank and rotated oak barrel)and aged in six types of oak barrels(three geographic origins×two toasting degrees)for different time(0,3,6 and 9 months,respectively).Results showed that 30 volatiles were associated with barrels and increased during oak aging.The fermenters could influence the intensities of the toast,leathery,smoky,fruity,floral and caramel aromas.The concentration of whisky lactone,eugenol,cis-isoeugenol,and the intensities of the toast and spicy aromas were highest in the wines aged in American oak and were lowest in the wines aged in French oak barrels.The concentrations of guaiacol,syringol,trans-isoeugenol,furfural alcohol,vanilla,cis-whisky lactone enabled the medium toasting barrels to be distinguished from the light toasting ones.The compounds originating from the barrels could be used to distinguish the types of different barrels,but the other general grape-derived and fermentation-derived volatiles could not.The fermenters,oak species and toasting degrees of the barrels all had significant effects on the aroma profiles of the aged‘Merlot’dry red wines,but the influence of the geographic origin was not obvious.Haocheng Lu Binhao Cheng Yibin Lan Changqing Duan Fei He 2024Food Science and Human Wellness2024,13,1:0
10Insights into multidimensional transport flux from vertical observation and numerical simulation in two cities in North China显示文摘This study represents the first quantitative evaluation of pollution transport budget within the boundary layer of typical cities in the Beijing-Tianjin-Hebei (BTH) region from the perspective of horizontal and vertical exchanges and further discusses the impact of the atmospheric boundary layer (ABL)-free troposphere (FT) exchange on concentration of fine particulate matter (PM_(2.5)) within the ABL during heavy pollution.From the perspective of the transport flux balance relationship,differences in pollution transport characteristics between the two cities is mainly reflected in the ABL-FT exchange effect.The FT mainly flowed into the ABL in BJ,while in SJZ,the outflow from the ABL to the FT was more intense.Combined with an analysis of vertical wind profile distribution,BJ was found to be more susceptible to the influence of northwest cold high prevailing in winter,while sinking of strong cold air allowed the FT flowing into the ABL influence the vertical exchange over BJ.In addition,we selected a typical pollution event for targeted analysis to understand mechanistic details of the influence of ABL-FT exchange on the pollution event.These results showed that ABL-FT interaction played an important role in PM_(2.5)concentration within the ABL during heavy pollution.Especially in the early stage of heavy pollution,FT transport contributed as much as 82.74% of PM_(2.5)within the ABL.These findings are significant for improving our understanding of pollution transport characteristics within the boundary layer and the effect of ABL-FT exchange on air quality.Zhida Zhang Xiaoqi Wang Shuiyuan Cheng Guiqian Tang Yibin Fu 2023Journal of Environmental Sciences2023,,3:0
11An RNA-RNA crosstalk network involving HMGB1 and RICTOR facilitates hepatocellular carcinoma tumorigenesis by promoting glutamine metabolism and impedes immunotherapy by PD-L1+ exosomes activity显示文摘Hepatocellular carcinoma(HCC)is the global leading cause of cancer-related deaths due to the deficiency of targets for precision therapy.A new modality of epigenetic regulation has emerged involving RNA-RNA crosstalk networks where two or more competing endogenous RNAs(ceRNAs)bind to the same microRNAs.However,the contribution of such mechanisms in HCC has not been well studied.Herein,potential HMGB1-driven RNA-RNA crosstalk networks were evaluated at different HCC stages,identifying the mT0RC2 component RICTOR as a potential HMGB1 ceRNA in HBV^(+)early stage HCC.Indeed,elevated HMGB1 mRNA was found to promote the expressio n of RICTOR mRNA through competitively bin ding with the miR-200 family,especially miR-429.Functio nal assays emplo ying overexpressi on or in terference strategies dem on strated that the HMGB1 and RICTOR 3zuntra nslated regions(UTR)epigenetically promoted the malignant proliferation,self-renewal,and tumorigenesis in HCC cells.Intriguingly,in terference agai nst HMGB1 and RICTOR in HCC cells promoted a stron ger an ti-PD-L1 immuno therapy resp on se,which appeared to associate with the production of PD-L1^(+)exosomes.Mechanistically,the HMGB1-driven RNA-RNA crosstalk network facilitated HCC cell glutamine metabolism via dual mechanisms,activating a positive feedback loop involving mT0RC2-AKT-C-MYC to upregulate glutamine synthetase(GS)expression,and inducing mTORCI signaling to derepress SIRT4 on glutamate dehydrogenase(GDH).Meanwhile,this crosstalk network could impede the efficacy of immunotherapy through mTORCI-P70S6K dependent PD-L1 production and PD-L1^(+)exosomes activity.In conclusion,our study highlights the non-coding regulatory role of HMGB1 with implicatio ns for RNA-based therapeutic targeting together with a predictio n of an ti-PD-L1 immuno therapy in HCC.Yanping Wei Xuewu Tang Yibin Ren Yun Yang Fengliang Song Jingbo Fu Shuowu Liu Miao Yu Jing Chen Suyang Wang Kecheng Zhang Yexiong Tan Zhipeng Han Lixjn Wei Baohua Zhang Zhangjun Cheng Liang Li Hongyang Wang 2022Signal Transduction and Targeted Therapy2022,7,1:0
12Speech Enhancement via Mask-Mapping Based Residual Dense Network显示文摘Masking-based and spectrum mapping-based methods are the two main algorithms of speech enhancement with deep neural network(DNN).But the mapping-based methods only utilizes the phase of noisy speech,which limits the upper bound of speech enhancement performance.Maskingbased methods need to accurately estimate the masking which is still the key problem.Combining the advantages of above two types of methods,this paper proposes the speech enhancement algorithm MM-RDN(maskingmapping residual dense network)based on masking-mapping(MM)and residual dense network(RDN).Using the logarithmic power spectrogram(LPS)of consecutive frames,MM estimates the ideal ratio masking(IRM)matrix of consecutive frames.RDN can make full use of feature maps of all layers.Meanwhile,using the global residual learning to combine the shallow features and deep features,RDN obtains the global dense features from the LPS,thereby improves estimated accuracy of the IRM matrix.Simulations show that the proposed method achieves attractive speech enhancement performance in various acoustic environments.Specifically,in the untrained acoustic test with limited priors,e.g.,unmatched signal-to-noise ratio(SNR)and unmatched noise category,MM-RDN can still outperform the existing convolutional recurrent network(CRN)method in themeasures of perceptual evaluation of speech quality(PESQ)and other evaluation indexes.It indicates that the proposed algorithm is more generalized in untrained conditions.Lin Zhou Xijin Chen Chaoyan Wu Qiuyue Zhong Xu Cheng Yibin Tang 2023Computers, Materials & Continua2023,,1:0

15High uric acid model in Caenorhabditis elegans显示文摘To establish experimental high uric acid model in C.elegans.Hypoxanthine,adenine,xanthine,and uric acid were used to treat C.elegans and then hyperuricemic C.elegans was evaluated by allopurinol.Hyperuricemic C.elegans were obtained after normal worms were treated by xanthine(0.25 mg/mL,18 h).For hyperuricemic worms,there was a statistically significant increase in the uric acid level(p<0.001)and a lower drug damage(p>0.05).Moreover,the model was proved to keep a high uric acid level for up to 12 h.After given allopurinol(0.25 mg/mL,12 h),the uric acid of hyperuricemic C.elegans had a significant reduction by 15%.Furthermore,xanthine oxidase activity in hyperuricemic C.elegans showed a statistically significant increase(p<0.001),which resulted in a raised uric acid content.A high uric acid model with low drug damage and high efficiency and stability was established in C.elegans after simply xanthine treatment.Zhenjing Li Yibin Xue Nifei Wang Jingli Cheng Xiaoying Dong Qingbin Guo Changlu Wang 2019Food Science and Human Wellness2019,8,1:0

20Human skin bacterial microbiota homeostasis:A delicate balance between health and disease显示文摘As the largest organ of the body,the skin acts as a barrier to prevent diseases and harbors a variety of beneficial bacteria.Furthermore,the skin bacterial microbiota plays a vital role in health and disease.Disruption of the barrier or an imbalance between symbionts and pathogens can lead to skin disorders or even systemic diseases.In this review,we first provide an overview of research on skin bacterial microbiota and human health,including the composition of skin bacteria in a healthy state,as well as skin bacterial microbiota educating the immune system and preventing the invasion of pathogens.We then discuss the diseases that result from skin microbial dysbiosis,including atopic dermatitis,common acne,chronic wounds,psoriasis,viral transmission,cutaneous lupus,cutaneous lymphoma,and hidradenitis suppurativa.Finally,we highlight the progress that utilizes skin microorganisms for disease therapeutics,such as bacteriotherapy and skin microbiome transplantation.A deeper knowledge of the interaction between human health and disease and the homeostasis of the skin bacterial microbiota will lead to new insights and strategies for exploiting skin bacteria as a novel therapeutic target.Yibin Zhu Xi Yu Gong Cheng 2023mLife2023,2,2:0
