| 4 | Sequential fate-switches in stem-like cells drive the tumorigenic trajectory from human neural stem cells to malignant glioma显示文摘Glioblastoma(GBM)is an incurable and highly heterogeneous brain tumor,originating from human neural stem/progenitor cells(hNSCs/hNPCs)years ahead of diagnosis.Despite extensive efforts to characterize hNSCs and end-stage GBM at bulk and single-cell levels,the de novo gliomagenic path from hNSCs is largely unknown due to technical difficulties in early-stage sampling and preclinical modeling.Here,we established two highly penetrant hNSC-derived malignant glioma models,which resemble the histopathology and transcriptional heterogeneity of human GBM.Integrating time-series analyses of whole-exome sequencing,bulk and single-cell RNA-seq,we reconstructed gliomagenic trajectories,and identified a persistent NSC-like population at all stages of tumorigenesis.Through trajectory analyses and lineage tracing,we showed that tumor progression is primarily driven by multi-step transcriptional reprogramming and fate-switches in the NSC-like cells,which sequentially generate malignant heterogeneity and induce tumor phenotype transitions.We further uncovered stage-specific oncogenic cascades,and among the candidate genes we functionally validated C1QL1 as a new glioma-promoting factor.Importantly,the neurogenic-to-gliogenic switch in NSC-like cells marks an early stage characterized by a burst of oncogenic alterations,during which transient AP-1 inhibition is sufficient to inhibit gliomagenesis.Together,our results reveal previously undercharacterized molecular dynamics and fate choices driving de novo gliomagenesis from hNSCs,and provide a blueprint for potential early-stage treatment/diagnosis for GBM. | Xiaofei Wang Ran Zhou Yanzhen Xiong Lingling Zhou Xiang Yan Manli Wang Fan Li Chuanxing Xie Yiming Zhang Zongyao Huang Chaoqiong Ding Kaidou Shi Weida Li Yu Liu Zhongwei Cao Zhen-Ning Zhang Shengtao Zhou Chong Chen Yan Zhang Lu Chen Yuan Wang | 2021 | Cell Research2021,31,6: | 8 |
| 5 | Focal adhesion protein Kindlin-2 regulates bone homeostasis in mice显示文摘Our recent studies demonstrate that the focal adhesion protein Kindlin-2 is critical for chondrogenesis and early skeletal development. Here, we show that deleting Kindlin-2 from osteoblasts using the 2.3-kb mouse Col1 a1-Cre transgene minimally impacts bone mass in mice, but deleting Kindlin-2 using the 10-kb mouse Dmp1-Cre transgene, which targets osteocytes and mature osteoblasts, results in striking osteopenia in mice. Kindlin-2 loss reduces the osteoblastic population but increases the osteoclastic and adipocytic populations in the bone microenvironment. Kindlin-2 loss upregulates sclerostin in osteocytes,downregulates β-catenin in osteoblasts, and inhibits osteoblast formation and differentiation in vitro and in vivo. Upregulation ofβ-catenin in the mutant cells reverses the osteopenia induced by Kindlin-2 deficiency. Kindlin-2 loss additionally increases the expression of RANKL in osteocytes and increases osteoclast formation and bone resorption. Kindlin-2 deletion in osteocytes promotes osteoclast formation in osteocyte/bone marrow monocyte cocultures, which is significantly blocked by an anti-RANKLneutralizing antibody. | 7 | Enhanced gas separation performance of mixed matrix hollow fiber membranes containing post-functionalized S-MIL-53显示文摘Mixed matrix hollow fiber membranes(MMHFMs)filled with metal-organic frameworks(MOFs)have great potential for energy-efficient gas separation processes,but the major hurdle is polymer/MOFs interfacial defects and membrane plasticization.Herein,lab-synthesized MIL-53 was post-functionalized by aminosilane grafting and subsequently incorporated into Ultem~?1000 polymer matrix to fabricate high performance MMHFMs.SEM,DLS,XRD and TGA were performed to characterize silane-modified MIL-53(S-MIL-53)and prepared MMHFMs.Moreover,the effect of MOFs loading was systematically investigated first;then gas separation performance of MMHFMs for pure and mixed gas was evaluated under different pressures.MMHFMs containing post-functionalized S-MIL-53 achieved remarkable gas permeation properties which was better than model predictions.Compared to pure HFMs,CO_2permeance of MMHFM loaded with 15%S-MIL-53 increased by 157%accompanying with 40%increase for CO_2/N_2selectivity,which outperformed the MMHFM filled with naked MIL-53.The pure and mixed gas permeation measurements with elevated feed pressure indicated that incorporation of S-MIL-53 also increased the resistance against CO_2plasticization.This work reveals that post-modified MOFs embedded in MMHFMs facilitate the improvement of gas separation performance and suppression of membrane plasticization. | Haitao Zhu Xingming Jie Lina Wang Guodong Kang Dandan Liu Yiming Cao | 2018 | Journal of Energy Chemistry2018,27,3: | 6 |
| 8 | Characteristics of deformation and stress distribution of small coa pillars under leading abutment pressure显示文摘Based on the engineering project on a small coal pillar of 12,521 working face roadway in Xieqiao Coalmine, data regarding surface displacements of the coal pillar, deep displacements and mining stress have been collected and analyzed. The results show that macroscopic transverse fractures of the inner coal pillar are developed within 2–4 m of the roadway surface, which is located outside the coal pillar anchorage zone. There is a displacement of 530 mm at the monitoring point in the 6 m deep zone of the pillar. Transfer of the fracture zone is found in a small coal pillar and the fractures within 3–4 m of the coal-rock zone from the roadway surface undergo propagation and closure of cracks which means this fracture zone is transferred from 3–4 m outside the roadway to only 2–3 m from the roadway surface. In the monitoring zone, vertical and horizontal stresses increase with a feature that shows that acceleration in the deep zone of the pillar is greater than that in the shallow zone. | 9 | Spatio-temporal Variations in Drought with Remote Sensing from the Mongolian Plateau During 1982–2018显示文摘The Mongolian Plateau is one of the regions most sensitive to climate change,the more obvious increase of temperature in 21 st century here has been considered as one of the important causes of drought and desertification.It is very important to understand the multi-year variation and occurrence characteristics of drought in the Mongolian Plateau to explore the ecological environment and the response mechanism of surface materials to climate change.This study examines the spatio-temporal variations in drought and its frequency of occurrence in the Mongolian Plateau based on the Advanced Very High Resolution Radiometer(AVHRR)Normalized Difference Vegetation Index(NDVI)(1982–1999)and the Moderate-resolution Imaging Spectroradiometer(MODIS)(2000–2018)datasets;the Temperature Vegetation Dryness Index(TVDI)was used as a drought evaluation index.The results indicate that drought was widespread across the Mongolian Plateau between1982 and 2018,and aridification incremented in the 21 st century.Between 1982 and 2018,an area of 164.38×10^4 km^2/yr suffered from drought,accounting for approximately 55.28%of the total study area.An area of approximately 150.06×10^4 km^2(51.43%)was subject to more than 160 droughts during 259 months of the growing seasons between 1982 and 2018.We observed variable frequencies of drought occurrence depending on land cover/land use types.Drought predominantly occurred in bare land and grassland,both of which accounting for approximately 79.47%of the total study area.These terrains were characterized by low vegetation and scarce precipitation,which led to frequent and extreme drought events.We also noted significant differences between the areal distribution of drought,drought frequency,and degree of drought depending on the seasons.In spring,droughts were widespread,occurred with a high frequency,and were severe;in autumn,they were localized,frequent,and severe;whereas,in summer,droughts were the most widespread and frequent,but less severe.The increase in temperature,decrease in precipitation,continuous depletion of snow cover,and intensification of human activities have resulted in a water deficit.More severe droughts and aridification have affected the distribution and functioning of terrestrial ecosystems,causing changes in the composition and distribution of plants,animals,microorganisms,conversion between carbon sinks and carbon sources,and biodiversity.We conclude that regional drought events have to be accurately monitored,whereas their occurrence mechanisms need further exploration,taking into account nature,climate,society and other influencing factors. | CAO Xiaoming Feng Yiming SHI Zhongjie | 2020 | Chinese Geographical Science2020,30,6: | 4 |
| 10 | Monte Carlo Simulation of in situ Gamma-Spectra Recorded by Na I(Tl) Detector in the Marine Environment显示文摘To develop a Na I(Tl) detector for in situ radioactivity monitoring in the marine environment and enhance the confidence of the probability of the gamma-spectrum analysis, Monte Carlo simulations using the Monte Carlo N-Particle( MNCP) code were performed to provide the response spectra of some interested radionuclides and the background spectra originating from the natural radionuclides in seawater recorded by a Na I(Tl) detector. A newly developed 75 mm × 75 mm Na I(Tl) detector was calibrated using four reference radioactive sources 137 Cs, 60 Co, 40 K and 54 Mn in the laboratory before the field measurements in seawater. A simulation model was established for the detector immersed in seawater. The simulated spectra were all broadened with Gaussian pulses to reflect the statistical fluctuations and electrical noise in the real measurement. The simulated spectra show that the single-energy photons into the detector are mostly scattering low-energy photons and the high background in the low energy region mainly originates from the Compton effect of the high energy γ-rays of natural radionuclides in seawater. The simulated background spectrum was compared with the experimental one recorded in field measurement and they seem to be in good agreement. | 15 | Hydrocarbon Generation, Migration, and Accumulation in the Eocene Niubao Formation in the Lunpola Basin, Tibet, China: Insights from Basin Modeling and Fluid Inclusion Analysis显示文摘The Eocene Niubao Formation is the primary research target of oil exploration in the Lunpola Basin.Crude oil was extracted from Well Z1 on the northern margin of the basin in 1993.In this study,an integrated evaluation of the source rock geothermal,and maturity histories and the fluid inclusion and fluid potential distributions was performed to aid in predicting areas of hydrocarbon accumulation.Due to the abundance of organic matter,the kerogen types,maturity,and oil-sources correlate with the geochemical data.The middle submember of the second member of the Niubao Formation(E2n^2-2)is the most favorable source rock based on the amount of oil produced from the E2n^2-3and E2n^3-1reservoirs.One-and twodimensional basin modeling,using BasinMod software,shows that the E2n^2-2source rock started to generate hydrocarbon at 35-30 Ma,reached a maturity of Ro=0.7%at 25-20 Ma,and at present,it has reached the peak oil generation stage with a thermal maturity of Ro=0.8%to less than Ro=1.0%.By using fluid inclusion petrography,fluorescence spectroscopy,and microthermometry,two major periods of oil charging have been revealed at 26.1-17.5 and 32.4-24.6 Ma.The oil accumulation modeling results,conducted by using the Trinity software,show a good fit of the oil shows in the wells and predict that the structural highs and lithologic transitions within the Jiangriaco and Paco sags are potential oil traps. | Yiming Liu Jiaren Ye Qiang Cao Baolin Yang Zhongrong Liu | 2020 | Journal of Earth Science2020,31,1: | 2 |
