2Association between Vitamin D Insufficiency and the Risk for Gestational Diabetes Mellitus in Pregnant Chinese Women显示文摘Objective To investigate the association between vitamin D deficiency and risk of gestational diabetes mellitus(GDM) in pregnant Chinese women.Methods A nested case-control study was conducted.Clinical and biochemical data were analyzed for 200 subjects with GDM and 200 subjects with normal glucose tolerance(NGT).Results The median(interquartile range) serum 25-hydroxyvitamin D(25OHD) levels were 22.39(17.67,29.38) and 25.86(19.09,34.88) nmol/L in the GDM and NGT groups,respectively.Rates of 25OHD deficiency or insufficiency were significantly higher in the GDM group than in the NGT group.Subjects with 25OHD levels <25 nmol/L had a 1.8-fold higher risk of GDM compared with subjects with higher vitamin D levels.In the GDM group,serum 25OHD was independently associated with HbA1c and insulin resistance after adjusting for confounding factors.In the NGT group,serum 25OHD was independently associated with fasting plasma glucose and systolic blood pressure after adjusting for maternal age and other confounding factors.Conclusion 25OHD insufficiency is very common in Chinese women.Low 25OHD status may be associated with insulin resistance and act as a risk factor for GDM.WANG Ou NIE Min HU Ying Ying ZHANG Kui LI Wei PING Fan LIU Jun Tao CHEN Li Meng XING Xiao Ping 2012Biomedical and Environmental Sciences2012,25,4:21

4Generation of a monkey with MECP2 mutations by TALEN-based gene targeting显示文摘Gene editing in model organisms has provided critical insights into brain development and diseases. Here, we report the generation of a cynomolgus monkey(Macaca fascicularis) carrying MECP2 mutations using transcription activator-like effector nucleases(TALENs)-mediated gene targeting. After injecting TALENs mRNA into monkey zygotes achieved by in vitro fertilization and embryo transplantation into surrogate monkeys, we obtained one male newborn monkey with an MECP2 deletion caused by frameshifting mutation in various tissues. The monkey carrying the MECP2 mutation failed to survive after birth, due to either the toxicity of TALENs or the critical requirement of MECP2 for neural development. The level of MeCP2 protein was essentially depleted in the monkey's brain. 7Prediction of Primary Climate Variability Modes at the Beijing Climate Center显示文摘Climate variability modes, usually known as primary climate phenomena, are well recognized as the most important predictability sources in subseasonal–interannual climate prediction. This paper begins by reviewing the research and development carried out, and the recent progress made, at the Beijing Climate Center(BCC) in predicting some primary climate variability modes. These include the El Ni?o–Southern Oscillation(ENSO), Madden–Julian Oscillation(MJO), and Arctic Oscillation(AO), on global scales, as well as the sea surface temperature(SST) modes in the Indian Ocean and North Atlantic, western Pacific subtropical high(WPSH), and the East Asian winter and summer monsoons(EAWM and EASM, respectively), on regional scales. Based on its latest climate and statistical models, the BCC has established a climate phenomenon prediction system(CPPS) and completed a hindcast experiment for the period 1991–2014. The performance of the CPPS in predicting such climate variability modes is systematically evaluated. The results show that skillful predictions have been made for ENSO, MJO, the Indian Ocean basin mode, the WPSH, and partly for the EASM, whereas less skillful predictions were made for the Indian Ocean Dipole(IOD) and North Atlantic SST Tripole, and no clear skill at all for the AO, subtropical IOD, and EAWM. 8Molecular Typing of Leptospira interrogans Strains Isolated from Rattus tanezumi in Guizhou Province,Southwest of China显示文摘Objective To identify and type three leptospires isolated from Rattus tanezumi in Guizhou Province by using three molecular techniques (PFGE, MLVA, and MLST), reveal the molecular characteristic of causative agents of local leptospirosis and evaluate these three molecular methods based on their detection resolution and efficiency. Methods Three Leptospira strains were isolated from the kidney of Rattus tanezumi and cultured with EMJH medium. PFGE, MLVA, and MLST assays were applied to type the three strains isolated from Rattus tanezumi in Guizhou Province. Results PFGE, MLVA, and MLST typing showed that the three leptospiral isolates matched with leptospiral serogroup Icterohaemorrhagiae serovar Lai. The findings of the genotyping methods were consistent. MLVA and MLST defined genotypes, whereas PFGE allowed the recognition of additional subgroups within the genotypes, and the findings of molecular typing were also consistent with those of traditional techniques. 9Development of a supercritical and an ultra-supercritical circulating fluidized bed boiler显示文摘The supercritical circulating fluidized bed(CFB)boiler,which combines the advantages of CFB combustion with low cost emission control and supercritical steam cycle with high efficiency of coal energy,is believed to be the future of CFB combustion technology.It is also of greatest importance for low rank coal utilization in China.Different from the supercritical pulverized coal boiler that has been developed more than 50 years,the supercritical CFB boiler is still a new one which requires further investigation.Without any precedentor engineering reference,Chinese researchers have con ducted fundamental research,development,design of the supercritical CFB boilers independently.The design theory and key technology for supercritical CFB boiler were proposed.Key components and novel structures were invented.The first 600 MWe supercritical CFB boiler and its auxiliaries were successfully developed and demonstrated in Baima Power Plant,Shenhua Group as well as the simulator,control technology,installation technology,commissioning technology,system integration and operation technology.Compared with the 460 MWe supercritical CFB in Poland,developed in the same period and the only other supercritical one of commercial running in the word beside Baima,the 600 MWe one in Baima has a better performance.Besides,supercritical CFB boilers of 350 MWe have been developed and widely commercialized in China.In this paper,the updated progress of 660 MWe ultra-supercritical CFB boilers under development is introduced.Junfu LYU Hairui YANG Wen LING Li NIE Guangxi YUE Ruixin LI Ying CHEN Shilong WANG 2019Frontiers in Energy2019,13,1:11
10TaqMan Real-time RT-PCR Assay for Detecting and Differentiating Japanese Encephalitis Virus显示文摘Objective To detect Japanese encephalitis virus(JEV) rapidly and distinguish its genotypes, a TaqMan-based reverse transcriptase quantitative polymerase chain reaction(RT-PCR) detection system was developed.Methods By aligning the full-length sequences of JEV(G1-G5), six sets of highly specific TaqMan real-time RT-PCR primers and probes were designed based on the highly conserved NS1, NS2, and M genes of JEV, which included one set for non-specific JEV detection and five sets for the detection of specific JEV genotypes. Twenty batches of mosquito samples were used to evaluate our quantitative PCR assay.Results With the specific assay, no other flavivirus were detected. The lower limits of detection of the system were 1 pfu/mL for JEV titers and 100 RNA copies/μL. The coefficients of variation of this real-time RT-PCR were all < 2.8%. 11Clinical applications of MARSALA for preimplantation genetic diagnosis of spinal muscular atrophy显示文摘Conventional PCR methods combined with linkage analysis based on short tandem repeats(STRs) or Karyomapping with single nucleotide polymorphism(SNP) arrays, have been applied to preimplantation genetic diagnosis(PGD) for spinal muscular atrophy(SMA), an autosome recessive disorder. However, it has limitations in SMA diagnosis by Karyomapping, and these methods are unable to distinguish wildtype embryos with carriers effectively. Mutated allele revealed by sequencing with aneuploidy and linkage analyses(MARSALA) is a new method allowing embryo selection by a one-step next-generation sequencing(NGS) procedure, which has been applied in PGD for both autosome dominant and X-linked diseases in our group previously. In this study, we carried out PGD based on MARSALA for two carrier families with SMA affected children. As a result, one of the couples has given birth to a healthy baby free of mutations in SMA-causing gene. It is the first time that MARSALA was applied to PGD for SMA, and we can distinguish the embryos with heterozygous deletion(carriers) from the wild-type(normal) ones accurately through this NGS-based method. In addition, direct mutation detection allows us to identify the affected embryos(homozygous deletion), which can be regarded as probands for linkage analysis, in case that the affected family member is absent. 12Exploration of the regulatory effect of miR-21 on breast cancer cell line proliferation and invasion as well as the downstream target genes显示文摘Objective: To study the regulatory effects of miR-21 on breast cancer cell line proliferation and invasion as well as the downstream target genes. Methods: Breast cancer cell lines MCF-7 were cultured and transfected with miR-21 mimics and the corresponding negative control mimics(NC mimics), and then MTS kits were used to detect cell viability. Transwell experiment was used to detect cell invasion ability, and fluorescence quantitative PCR was used to detect the expression of proliferation and invasion-related genes in cells. Results: 24 h after transfection of miR-21 mimics and NC mimics, cell OD value and the number of invasive cells of miR-21 group were significantly higher than those of NC group, and m RNA contents of PDCD-4, Fas L, PTEN, Rho B, Maspin, TIMP3 and RECK in cells were significantly lower than those of NC group. 15Innate immune responses to DNA viruses显示文摘The innate immune system acts as the first line of defense against pathogens,which is also essen-tial for initiation of adaptive immunity.Innate immune responses are initiated by pattern-recognition receptors(PRRs),which recognize conserved molecular structures of pathogens called patho-gen-associated molecular patterns(PAMPs).The infection of virus triggers a series of signaling events leading to transcriptional induction of type I interferons(IFNs)and proinflammatory cyto-kines.In recent years,the mechanisms of viral RNA recognition and RNA virus-triggered signaling pathways have been well studied.However,it remains unclear on how DNA virus infection is sensed by host cells and triggers the host antiviral defense.Although ten potential viral DNA sen-sors have been reported,none of them is validated as a generally used sensor for distinct DNA viruses in divergent cell types and animals.Here,we provide a summary and perspective on recent advances in innate immune responses to DNA viruses.Ying Nie Yan-Yi Wang 2013Protein & Cell2013,4,1:6
16Phylogenetic analysis of Toxicodendron(Anacardiaceae) and its biogeographic implications on the evolution of north temperate and tropical intercontinental disjunctions显示文摘Toxicodendron is a genus in the Rhus complex of Anacardiaceae with a disjunct distribution between eastern Asia and North America,extending to southeastern Asia and the neotropics.Nuclear(internal transcribed spacer,external transcribed spacer,and NIA-i3) and chloroplast(ndhF and trnL-F) sequences were used to construct phylogenetic relationships of Toxicodendron.Phylogenetic analysis of these data strongly support Toxicodendron as a monophyletic group distinct from other genera of the Rhus complex,and the phylogeny does not fully corroborate classification at the sectional level.Two temperate disjunct lineages were detected,one from section Toxicodendron and the other between the eastern North American Toxicodendron vernix and the eastern Asian Toxicodendron ver-nicifluum.Their divergence times were estimated to be 13.46(7.95-19.42) and 7.53(2.76-12.86) mya,respectively.The disjunction between section Griffithii(taxa from warm temperate to tropical Asia) and Toxicodendron striatum(from the neotropics) was supported and their divergence time was estimated to be 20.84(11.16-30.52) mya in the early Miocene.Our biogeographic results and the paleontological data support the Bering land bridge as the most likely route to explain the temperate disjunctions,yet the tropical disjunction in Toxicodendron seems to be best explained by the North Atlantic land bridge hypothesis.Ze-Long NIE Hang SUN Ying MENG Jun WEN 2009Journal of Systematics and Evolution2009,47,5:6
17Ketoconazole Associated Hepatotoxicity: A Systematic Review and Metaanalysis显示文摘Objective To evaluate the incidence of Ketoconazole associated hepatotoxicity and related factors. Methods Literature retrieval was conducted by using multi-databases for meta-analysis on Ketoconazole associated hepatotoxicity. The data were collected with a standardized form. Overall estimation of incidence of hepatotoxicity for specific study type was calculated by using a DerSimonian-Laird random-effects model owing to the substantial differences among the studies. Results Totally 204 eligible studies were included in the analysis. The incidence of Ketoconazole associated hepatotoxicity was 3.6%-4.2%. The dosage and duration specific subgroup analyses did not show any significant difference among groups, while the age specific subgroup analysis showed the incidence in children and people aged >60 years was 1.4% (95% CI: 0.5%-4.2%) and 3.2% (95% CI: 1.1%-8.7%) respectively. Additionally, the incidence of the hepatotoxicity was higher in people who had oral administration of ketoconazole beyond the provisions of the usage instructions, and the incidence was 5.7% (95% CI: 4.5%-7.2%). Conclusion Ketoconazole associated hepatotoxicity was common. Off-label use might increase the risk of liver damage. 19Estimation of Methane Emissions from Municipal Solid Waste Landfills in China Based on Point Emission Sources显示文摘The methane(CH4) emissions from municipal solid waste(MSW) landfills in China in 2007 were estimated based on database of the three-dimensional emission factors matrix and point sources, by an IPCC recommended FOD(firstorder decay) model. The location, capacity and age of landfills constitute the three dimensions of the emission factors matrix, which were obtained by laboratory analysis and in situ investigation. Key parameters such as waste composition,degradable organic carbon ratio, CH4 correction factor, oxidation factor and recovery rate, were carefully analyzed in terms of these three dimensions. The point sources database consists of 2,107 MSW landfills in cities and towns of China in 2007. The results show that the CH4 emissions from MSW landfills were 1.186 Mt in 2007. Compared with the CH4 emissions of 2.20 Mt in 2005, the significant discrepancy mainly comes from statistical data of landfills, e.g., number of landfills and amount of waste disposed in landfills. CH4 emissions were lower than 700 t for most of the landfills, whereas there were 279 landfills with emissions larger than 1,000 t, and only 10 landfills with emissions larger than 10,000 t.Jiangsu province ranks the largest emitter with 98,700 t while Tibet is the smallest emitter with 2,100 t. 20MoS_(2)-Decorated/Integrated Carbon Fiber:Phase Engineering Well-Regulated Microwave Absorber显示文摘Phase engineering is an important strategy to modulate the electronic structure of molybdenum disulfide(MoS_(2)).MoS_(2)-based composites are usually used for the electromagnetic wave(EMW)absorber,but the effect of different phases on the EMW absorbing performance,such as 1T and 2H phase,is still not studied.In this work,micro-1T/2H MoS_(2) is achieved via a facile one-step hydrother-mal route,in which the 1T phase is induced by the intercalation of guest molecules and ions.The EMW absorption mechanism of single MoS_(2) is revealed by presenting a comparative study between 1T/2H MoS_(2) and 2H MoS_(2).As a result,1T/2H MoS_(2) with the matrix loading of 15%exhibits excellent microwave absorption property than 2H MoS_(2).Furthermore,taking the advantage of 1T/2H MoS_(2),a flexible EMW absorbers that ultrathin 1T/2H MoS_(2)grown on the carbon fiber also performs outstanding performance only with the matrix loading of 5%.This work offers necessary reference to improve microwave absorption performance by phase engineering and design a new type of flexible electromagnetic wave absorption material to apply for the portable microwave absorption electronic devices.Jing Yan Ying Huang Xiangyong Zhang Xin Gong Chen Chen Guangdi Nie Xudong Liu Panbo Liu 2021Nano-Micro Letters2021,13,7:5
