1Pharmacometabolomic prediction of individual differences of gastrointestinal toxicity complicating myelosuppression in rats induced by irinotecan显示文摘Pharmacometabolomics has been already successfully used in toxicity prediction for one specific adverse effect. However in clinical practice, two or more different toxicities are always accompanied with each other, which puts forward new challenges for pharmacometabolomics. Gastrointestinal toxicity and myelosuppression are two major adverse effects induced by Irinotecan(CPT-11),and often show large individual differences. In the current study, a pharmacometabolomic study was performed to screen the exclusive biomarkers in predose serums which could predict late-onset diarrhea and myelosuppression of CPT-11 simultaneously. The severity and sensitivity differences in gastrointestinal toxicity and myelosuppression were judged by delayed-onset diarrhea symptoms, histopathology examination, relative cytokines and blood cell counts. Mass spectrometry-based non-targeted and targeted metabolomics were conducted in sequence to dissect metabolite signatures in predose serums. Eventually,two groups of metabolites were screened out as predictors for individual differences in late-onset diarrhea and myelosuppression using binary logistic regression, respectively. This result was compared with existing predictors and validated by another independent external validation set. Our study indicates the prediction of toxicity could be possible upon predose metabolic profile. Pharmacometabolomics can be a potentially useful tool for complicating toxicity prediction. 12A DNA-based nanocarrier for efficient cancer therapy显示文摘The study aimed to achieve enhanced targeted cytotoxicity and cell-internalization of cisplatin-loaded deoxyribonucleic acid-nanothread(CPT-DNA-NT),mediated by scavenger receptors into HeLa cells.DNA-NT was developed with stiff-topology utilizing circular-scaffold to encapsulate CPT.Atomic force microscopy(AFM)characterization of the DNA-NT showed uniformity in the structure with a diameter of 50-150 nm and length of 300-600 nm.The successful fabrication of the DNA-NT was confirmed through native-polyacrylamide gel electrophoresis analysis,as large the molecular-weight(polymeric)DNA-NT did not split into constituting strands under applied current and voltage.The results of cell viability confirmed that blank DNA-NT had the least cytotoxicity at the highest concentration(512 nM)with a viability of 92%as evidence of its biocompatibility for drug delivery.MTT assay showed superior cytotoxicity of CPT-DNA-NT than that of the free CPT due to the depot release of CPT after DNA-NT internalization.The DNA-NT exhibited targeted cell internalizations with the controlled intracellular release of CPT(from DNA-NT),as illustrated in confocal images.Therefore,in vitro cytotoxicity assessment through flow cytometry showed enhanced apoptosis(72.7%)with CPT-DNA-NT(compared to free CPT;64.4%).CPT-DNA-NT,being poly-anionic,showed enhanced endocytosis via scavenger receptors.Muhammad Abbas Mirza Muhammad Faran Ashraf Baig Yaliang Zhang Yu-Shun Yang Songyu Wu Yiqiao Hu Zhong-Chang Wang Hai-Liang Zhu 2021Journal of Pharmaceutical Analysis2021,11,3:0
13Photosynthetic microorganisms coupled photodynamic therapy for enhanced antitumor immune effect显示文摘The ideal photodynamic therapy(PDT)should effectively remove the primary tumor,and produce a stronger immune memory effect to inhibit the tumor recurrence and tumor metastasis.However,limited by the hypoxic and immunosuppressive microenvironment,the PDT efficiency is apparently low.Here,Chlorella(Chl.)is exploited to enhance local effect by producing oxygen to reverse hypoxia,and release adjuvants to reverse immunosuppressive microenvironment to enhance abscopal effect afterwards.Results from different animal models indicated that Chl.could enhance local effect and PDT related immune response.Ultimately,Chl.coupled PDT elicited anti-tumor effects toward established primary tumors(inhibition rate:90%)and abscopal tumors(75%),controlled the challenged tumors(100%)and alleviated metastatic tumors(90%).This Chl.coupled PDT strategy can also produce a stronger anti-tumor immune memory effect.Overall,this Chl.coupled PDT strategy generates enhanced local tumor killing,boosts PDT-induced immune responses and promotes anti-tumor immune memory effect,which may be a great progress for realizing systemic effect of PDT.Haoran Wang Honghui Liu Yunfei Guo Wenjing Zai Xianghui Li Wei Xiong Xiaozhi Zhao Yingfang Yao Yiqiao Hu Zhigang Zou Jinhui Wu 2022Bioactive Materials2022,7,6:0

16Novel gold nanoparticles targeting somatostatin receptor subtype two with near-infrared light for neuroendocrine tumour therapy显示文摘Neuroendocrine tumours(NETs)are rare cancers with positive somatostatin receptor 2(SSTR2)expression,and treatment strategies for NETs are not satisfactory.Nanomaterial-mediated therapy targeting SSTR2 in NETs is very promising.This study firstly combined mesoporous silica-coated gold nanorods(AuNRs@mSiO_(2))and targeting-SSTR2 dodecane tetraacetic acidtyrosine3-octreotate(DOTA-TATE)into AuNRs@mSiO_(2)@DOTA-TATE to investigate NETs inhibition under near-infrared light.AuNRs@mSiO_(2)@DOTA-TATE showed good photothermal conversion efficiency.In vitro,under light irradiation,the cell viability significantly decreased with increasing AuNR@mSiO_(2)@DOTA-TATE concentration;in two successfully established neuroendocrine tumour organoids with SSTR2 expression,AuNRs@mSiO_(2)@DOTA-TATE with light inhibited tumours significantly better than AuNRs@mSiO_(2) with light.In vivo,the SSTR2-targeting ability and biodistribution of AuNRs@mSiO_(2)@DOTA-TATE were confirmed with AuNRs@mSiO_(2)@64Cu-DOTA-TATE under micro-positron emission tomography/computed tomography(micro-PET/CT);in the AuNRs@mSiO_(2)@DOTA-TATE with laser group,the tumour surface temperature increased rapidly,with tumour volumes similar to those in the octreotide group and significantly lower than those in other groups.There was no significant difference in mice body weight between the AuNRs@mSiO_(2)@DOTA-TATE with laser group and other groups.No significant inflammatory lesions or cell necrosis was found in the main organs.In summary,we presented a feasible strategy to construct AuNRs@mSiO_(2)@DOTA-TATE with good photothermal conversion efficiency,targetingSSTR2 ability,significant antitumour effects,and good biocompatibility,warranting further explorations of AuNRs@mSiO_(2)@DOTA-TATE for NETs therapy applications.Qichen Chen Zilin Li Jiangyuan Yu Qing Xie Haizhen Lu Yiqiao Deng Jinghua Chen Wenjia Zhu Li Huo Yizhou Zhang Wei Song Jianqiang Lan Jianqiang Cai Zhen Huang Zixi Wang Hong Zhao 2022Nano Research2022,15,10:0
17A New Fiber Detection Method for LAMOST Based on the Front-illuminated Method显示文摘The double revolving fiber positioning technology is one of the key technologies for the success of the Large Sky Area Multi-Object Fiber Spectroscopic Telescope(LAMOST).The accuracy of fiber positioning will directly affect the observation efficiency of LAMOST.To achieve higher fiber positioning accuracy,the original open-loop controlled fiber positioning system urgently needs to be upgraded into a closed-loop control system.The fiber detection is the most important part of the closed-loop controlled fiber positioning system.The back-illuminated detection method is usually used to detect the fiber position by directly detecting the light spot generated at the fiber end in the multi-fiber spectral surveys.In this paper,we introduce a new method to measure the fiber position based on the image of the front-illuminated LAMOST focal plane.The front-illuminated image does not require lighting devices inside the spectrograph,and it could reduce the instability and light pollution in the spectrograph end.Our method measures the fiber position by fitting the profile of the fiber pinhole with a 2D Gaussian function.A series of tests show that the relative position measurement precision of the front-illuminated method is about 012,and the method could have the same accuracy as the back-illuminated method once the system bias is calibrated by a simple radial correction function.The required fiber positioning accuracy of LAMOST is 04,and the new method satisfies the requirement of LAMOST fiber detection accuracy and could be used in the closed-loop fiber control system.Ming Zhou Yong Zhang Guanru Lv Jian Li Zengxiang Zhou Zhigang Liu Jianping Wang Zhongrui Bai Yuan Tian Mengxin Wang Shuqing Wang Hongzhuan Hu Chao Zhai Jiaru Chu Zhijie Han Mingxu Liu Yiqiao Dong Hailong Yuan Yongheng Zhao Yaoquan Chu Haotong Zhang 2022Research in Astronomy and Astrophysics2022,22,6:0

19Vulnerability analysis of the Chinese coupled aviation and high-speed railway network显示文摘The integrated aviation and High-Speed Railway(HSR)transportation system plays a vital role for today’s inter-city transportation services.However,an increasing number of unexpected disruptions(such as operation failures,natural disasters,or intentional attacks)pose a considerable threat to the normal operation of the system,especially on ground transfer,leading to the extensive research on its vulnerability.Previous approaches mainly focus on interruptions within a single transportation mode,neglecting the role of ground transfer which serves as a coupled connection between aviation and High-Speed Railway.This paper proposes a network-based framework for evaluating the vulnerability of the Chinese Coupled Aviation and High-Speed Railway(CAHSR)network from the viewpoint of ground transfer interruption.Taking the end-to-end travel time and passenger flow information into consideration as an evaluation measure and analyzing from the perspective of urban agglomerations,an adaptive method is developed to identify the critical cities and further investigate their failure impacts on the geographic distribution of vulnerability.In addition,the proposed model explores variations of vulnerability under different failure time intervals.Based on the empirical study,some major conclusions are highlighted as follows:(A)Only a few cities show significant impacts on the network’s vulnerability when ground transfer interruptions occurred.(B)The distribution of vulnerability is not proportional to the distance between failure city and influenced city.(C)The vulnerability is more serious in the morning and evening when the ground transfer is disconnected.Our findings may provide new insights for maintenance and optimization of the CAHSR network and other real-world transportation networks.Yiqiao WANG Xianbin CAO Fangyun QIN Lu(Carol)TONG 2022Chinese Journal of Aeronautics2022,35,12:0
