Durability of neutralizing antibodies and T-cell response post SARS-CoV-2 infection显示文摘The ongoing pandemic of coronavirus disease 19(COVID-19)is caused by a newly discoveredβcoronavirus named severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).How long the adaptive immunity triggered by SARS-CoV-2 can last is of critical clinical relevance in assessing the probability of second infection and efficacy of vaccination. Here we examined, using ELISA,the IgG antibodies in serum specimens collected from 17 COVID-19 patients at 6–7 months after diagnosis and the results were compared to those from cases investigated 2 weeks to 2 months post-infection.All samples were positive for IgGs against the S-and N-proteins of SARS-CoV-2.Notably,14 samples available at 6–7 months post-infection all showed significant neutralizing activities in a pseudovirus assay, with no difference in blocking the cell-entry of the 614D and 614G variants of SARS-CoV-2.Furthermore,in 10 blood samples from cases at 6–7 months post-infection used for memory T-cell tests,we found that interferonγ-producing CD4+and CD8+cells were increased upon SARS-CoV-2 antigen stimulation.Together, these results indicate that durable anti-SARS-CoV-2 immunity is common in convalescent population, and vaccines developed from 614D variant may offer protection from the currently predominant 614D variant of SARS-CoV-2.Yun Tan Feng Liu Xiaoguang Xu Yun Ling Weijin Huang Zhaoqin Zhu Mingquan Guo Yixiao Lin Ziyu Fu Dongguo Liang Tengfei Zhang Jian Fan Miao Xu Hongzhou Lu Saijuan Chen 2020Frontiers of Medicine2020,14,6:6
COVID-19 vaccination intention and vaccine characteristics infuencing vaccination acceptance: a global survey of 17 countries显示文摘Background:The availability of various types of COVID-19 vaccines and diverse characteristics of the vaccines pre‑sent a dilemma in vaccination choices, which may result in individuals refusing a particular COVID-19 vaccine ofered, hence presenting a threat to immunisation coverage and reaching herd immunity. The study aimed to assess global COVID-19 vaccination intention, vaccine characteristics infuencing vaccination acceptance and desirable vaccine characteristics infuencing the choice of vaccines. Methods:An anonymous cross-sectional survey was conducted between 4 January and 5 March 2021 in 17 coun‑tries worldwide.Proportions and the corresponding 95%confdence intervals(CI)of COVID-19 vaccine acceptance and vaccine characteristics infuencing vaccination acceptance were generated and compared across countries and regions.Multivariable logistic regression analysis was used to determine the factors associated with COVID-19 vaccine hesitancy. Results:Of the 19,714 responses received,90.4%(95%CI 81.8–95.3)reported likely or extremely likely to receive COVID-19 vaccine.A high proportion of likely or extremely likely to receive the COVID-19 vaccine was reported in Australia(96.4%),China(95.3%)and Norway(95.3%),while a high proportion reported being unlikely or extremely unlikely to receive the vaccine in Japan(34.6%),the U.S.(29.4%)and Iran(27.9%).Males,those with a lower educational level and those of older age expressed a higher level of COVID-19 vaccine hesitancy.Less than two-thirds(59.7%;95%CI 58.4–61.0)reported only being willing to accept a vaccine with an efectiveness of more than 90%,and 74.5%(95%CI 73.4–75.5)said they would accept a COVID-19 vaccine with minor adverse reactions.A total of 21.0%(95%CI 20.0–22.0)reported not accepting an mRNA vaccine and 51.8%(95%CI 50.3–53.1)reported that they would only accept a COVID-19 vaccine from a specifc country‐of‐origin.Countries from the Southeast Asia region reported the highest proportion of not accepting mRNA technology. The highest proportion from Europe and the Americas would only accept a vaccine produced by certain countries. The foremost important vaccine characteristic infuencing vaccine choice is adverse reactions(40.6%;95%CI 39.3–41.9)of a vaccine and efectiveness threshold(35.1%;95%CI 33.9–36.4).Conclusions:The inter-regional and individual country disparities in COVID-19 vaccine hesitancy highlight the importance of designing an efcient plan for the delivery of interventions dynamically tailored to the local population.Li Ping Wong Haridah Alias Mahmoud Danaee Jamil Ahmed Abhishek Lachyan Carla Zi Cai Yulan Lin Zhijian Hu Si Ying Tan Yixiao Lu Guoxi Cai Di Khanh Nguyen Farhana Nishat Seheli Fatma Alhammadi Milkar D.Madhale Muditha Atapattu Tasmi Quazi‑Bodhanya Samira Mohajer Gregory D.Zimet Qinjian Zhao 2021Infectious Diseases of Poverty2021,10,5:0
An implantable,wireless,battery-free system for tactile pressure sensing显示文摘The sense of touch is critical to dexterous use of the hands and thus an essential component of efforts to restore hand function after amputation or paralysis.Prosthetic systems have addressed this goal with wearable tactile sensors. However, such wearable sensors are suboptimal for neuroprosthetic systems designed to reanimate a patient’s own paralyzed hand. Here,we developed an implantable tactile sensing system intended for subdermal placement. The system is composed of a microfabricated capacitive pressure sensor,a custom integrated circuit supporting wireless powering and data transmission, and a laser-fused hermetic silica package. The miniature device was validated through simulations, benchtop assessment, and testing in a primate hand. The sensor implanted in the fingertip accurately measured applied skin forces with a resolution of 4.3 mN.The output from this novel sensor could be encoded in the brain with microstimulation to provide tactile feedback.More broadly, the materials, system design, and fabrication approach establish new foundational capabilities for various applications of implantable sensing systems.Lin Du Han Hao Yixiao Ding Andrew Gabros Thomas C.E.Mier Jan Van der Spiegel Timothy H.Lucas Firooz Aflatouni Andrew G.Richardson Mark G.Allen 2023Microsystems & Nanoengineering2023,9,5:0
Antimicrobial resistance characteristics and phylogenetic relationships of pleuromutilin-resistant Enterococcus isolates from different environmental samples along a laying hen production chain显示文摘Antimicrobial resistance in the laying hen production industry has become a serious public health problem. The antimicrobial resistance and phylogenetic relationships of the common conditional pathogen Enterococcus along the laying hen production chain have not been systematically clarified.105 Enterococcus isolates were obtained from 115 environmental samples (air, dust, feces, fies, sewage, and soil) collected along the laying hen production chain (breeding chicken, chick, young chicken, and commercial laying hen). These Enterococcus isolates exhibited resistance to some clinically relevant antibiotics, such as tetracycline (92.4%),streptomycin (92.4%),and erythromycin (91.4%),and all strains had multidrug resistance phenotypes.Whole genome sequencing characterized 29 acquired antibiotic resistance genes (ARGs) that conferred resistance to 11 classes of antibiotics in 51pleuromutilin-resistant Enterococcus isolates, and lsa(E),which mediates resistance to pleuromutilins, always co-occurred with lnu(B).Alignments with the Mobile Genetic Elements database identified four transposons (Tn554,Tn558,Tn6261,and Tn6674) with several ARGs(erm(A),ant(9)-la,fex(A),and optrA) that mediated resistance to many clinically important antibiotics. Moreover,we identified two new transposons that carried ARGs in the Tn554 family designated as Tn7508 and Tn7492.A complementary approach based on conventional multi-locus sequence typing and whole genome single nucleotide polymorphism analysis showed that phylogenetically related pleuromutilin-resistant Enterococcus isolates were widely distributed in various environments on different production farms. Our results indicate that environmental contamination by antimicrobial-resistant Enterococcus requires greater attention, and they highlight the risk of pleuromutilin-resistant Enterococcus and ARGs disseminating along the laying hen production chain, thereby warranting effective disinfection.Cong Lin Yuxuan Feng Xianjun Xie Haoyu Zhang Jie Wu Yixiao Zhu Jing Yu Jingyi Feng Wen Su Shanming Lai Anyun Zhang 2024Journal of Environmental Sciences2024,,3:0
CO_(2)electrolysis:Advances and challenges in electrocatalyst engineering and reactor design显示文摘Electrochemical reduction of CO_(2)(CO_(2)RR)coupled with renewable electrical energy is an attractive way of upgrading CO_(2)to value-added chemicals and closing the carbon cycle.However,CO_(2)RR electrocatalysts still suffer from high overpotential, and the complex reaction pathways of CO_(2)RR often lead to mixed products.Early research focuses on tuning the binding of reaction intermediates on electrocatalysts, and recent efforts have revealed that the design of electrolysis reactors is equally important for efficient and selective CO_(2)RR.In this review,we present an overview of recent advances and challenges toward achieving high activity and high selectivity in CO_(2)RR at ambient conditions, with a particular focus on the progress of CO_(2)RR electrocatalyst engineering and reactor design. Our discussion begins with three types of electrocatalysts for CO_(2)RR(noble metalbased,none-noble metal-based,and metal-free electrocatalysts),and then we examine systems-level strategies toward engineering specific components of the electrolyzer, including gas diffusion electrodes, electrolytes, and polymer electrolyte membranes. We close with future perspectives on catalyst development,in-situ/operando characterization, and electrolyzer performance evaluation in CO_(2)RR studies.Jiayi Lin Yixiao Zhang Pengtao Xu Liwei Chen 2023Materials Reports(Energy)2023,3,2:0
A cell wall invertase modulates resistance to fusarium crown rot and sharp eyespot in common wheat显示文摘Fusarium crown rot(FCR) and sharp eyespot(SE)are serious soil-borne diseases in wheat and its relatives that have been reported to cause wheat yield losses in many areas. In this study, the expression of a cell wall invertase gene, TaCWI-B1,was identified to be associated with FCR resistance through a combination of bulk segregant RNA sequencing and genome resequencing in a recombinant inbred line population. Two biparental populations were developed to further verify TaCWI-B1 association with FCR resistance.Overexpression lines and ethyl methanesulfonate(EMS) mutants revealed TaCWI-B1 positively regulating FCR resistance. Determination of cell wall thickness and components showed that the TaCWI-B1-overexpression lines exhibited considerably increased thickness and pectin and cellulose contents. Furthermore, we found that TaCWI-B1 directly interacted with an alphagalactosidase(TaGAL). EMS mutants showed that TaGAL negatively modulated FCR resistance. The expression of TaGAL is negatively correlated with TaCWI-B1 levels, thus may reduce mannan degradation in the cell wall, consequently leading to thickening of the cell wall. Additionally, TaCWI-B1-overexpression lines and TaGAL mutants showed higher resistance to SE;however, TaCWI-B1 mutants were more susceptible to SE than controls. This study provides insights into a FCR and SE resistance gene to combat soil-borne diseases in common wheat.Guoguo Lv Yixiao Zhang Lin Ma Xiangning Yan Mingjie Yuan Jianhui Chen Yongzhen Cheng Xi Yang Qi Qiao Leilei Zhang Mohsin Niaz Xiaonan Sun Qijun Zhang Shaobin Zhong Feng Chen 2023Journal of Integrative Plant Biology2023,65,7:0
