1Construction and external validation of a nomogram that predicts lymph node metastasis in early gastric cancer patients using preoperative parameters显示文摘Objective: To create a nomogram to predict the incidence of lymph node metastasis(LNM) in early gastric cancer(EGC) patients and to externally validate the nomogram.Methods: To construct the nomogram,we retrospectively analyzed a primary cohort of 272 EGC patients.Univariate analysis and a binary logistic regression were performed.A nomogram predicting the incidence of LNM in EGC patients was created.The discrimination ability of the nomogram was measured using the concordance index(c-index),and the nomogram was also calibrated.Then,another prospective cohort of 81 patients was analyzed to validate the nomogram.Results: In the primary cohort,LNM was pathologically confirmed in 37(13.6%) patients.In multivariate analysis,the presence of an ulcer,the maximum lesion diameter observed via gastroscopy,the thickness of the lesion observed via endoscopic ultrasonography,and the presence of enlarged lymph nodes on computed tomography(CT) were independent risk factors for LNM.A nomogram was then created based on the regression model with the c-index of 0.905,and the calibration curve of the nomogram fell approximately on the ideal 45-degree line.The cut-off score of the nomogram was 110,and the sensitivity,specificity,positive predictive and negative predictive values of the nomogram in the primary cohort were 81.1%,86.0%,47.6% and 96.7%,respectively,and in the prospective validation cohort were 75.0%,91.0%,60.0% and 95.5%,respectively.The calibration curve of the external validation cohort was almost on the 45-degree line.Conclusions: We developed an effective nomogram predicting the incidence of LNM for EGC patients.Yinan Zhang Yiqiang Liu Ji Zhang Xiaojiang Wu Xin Ji Tao Fu Ziyu Li Qi Wu Zhaode Bu Jiafu Ji 2018Chinese Journal of Cancer Research2018,30,6:12
2Highly stable lead-free Cs3Bi2I9 perovskite nanoplates for photodetection applications显示文摘Lead halide perovskites have received tremendous attentions recently for their excellent properties such as high light absorption coefficient and long charge carrier diffusion length. However, the stability issues and the existence of toxic lead cations have largely limited their applications in optoelectronic area. Herein, we report the synthesis and investigation of highly stable and lead-free Cs3Bi2I9 perovskite nanoplates for visible light photodetection applications. The Cs3Bi2I9 nanoplates were synthesized through a facile solution-processed method, which is also applicable to various substrates. The achieved nanoplates present very good crystal quality and exhibit excellent long-term stability even exposed in moist air for several months. Photodetectors were constructed based on these high-quality perovskite nanoplates for the first time, and display a maximum photoresponsivity of 33.1 mA/W under the illumination of 450 nm laser, which is six times higher than the solution-synthesized CH3NH3PbI3 nanowire photodetectors. The specific detectivity of these devices can reach up to 10^10 Jones. Additionally, the devices exhibit fast rise and decay time of 10.2 and 37.2 ms, respectively, and highly stable photoswitching behavior with their photoresponse well retaining under alternating light and darkness. 3Durability of neutralizing antibodies and T-cell response post SARS-CoV-2 infection显示文摘The ongoing pandemic of coronavirus disease 19(COVID-19)is caused by a newly discoveredβcoronavirus named severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).How long the adaptive immunity triggered by SARS-CoV-2 can last is of critical clinical relevance in assessing the probability of second infection and efficacy of vaccination.Here we examined,using ELISA,the IgG antibodies in serum specimens collected from 17 COVID-19 patients at 6–7 months after diagnosis and the results were compared to those from cases investigated 2 weeks to 2 months post-infection.All samples were positive for IgGs against the S-and N-proteins of SARS-CoV-2.Notably,14 samples available at 6–7 months post-infection all showed significant neutralizing activities in a pseudovirus assay,with no difference in blocking the cell-entry of the 614D and 614G variants of SARS-CoV-2.Furthermore,in 10 blood samples from cases at 6–7 months post-infection used for memory T-cell tests,we found that interferonγ-producing CD4+and CD8+cells were increased upon SARS-CoV-2 antigen stimulation.Together,these results indicate that durable anti-SARS-CoV-2 immunity is common in convalescent population,and vaccines developed from 614D variant may offer protection from the currently predominant 614D variant of SARS-CoV-2.Yun Tan Feng Liu Xiaoguang Xu Yun Ling Weijin Huang Zhaoqin Zhu Mingquan Guo Yixiao Lin Ziyu Fu Dongguo Liang Tengfei Zhang Jian Fan Miao Xu Hongzhou Lu Saijuan Chen 2020Frontiers of Medicine2020,14,6:6
4Anatomical variation of infra-pyloric artery origination: A prospective multicenter observational study (IPA-Origin)显示文摘Objective: Infra-pyloric artery(IPA) is an important anatomical landmark in treatment of gastric cancer and is the key vessel for pylorus-preserving gastrectomy and subgroup of infra-pyloric lymph nodes. However, its anatomical variation is not thoroughly understood. Our study aimed to clarify the origination of the IPA.Methods: We did this prospective, multicenter, open-label, observational study at gastric surgery departments of34 hospitals in China. Gastric cancer patients aged 18 years or older and scheduled to undergo elective total or distal gastrectomy were assigned. During the surgery, IPA dissecting and exposing the origination point with photographs or video clips were required. The primary outcome was the origination of the IPA. Analysis of variance, χ~2 tests and Fisher's tests were used to analyze the differences between groups. The study is registered at Clinicaltrials.gov(No. NCT03071237).Results: Between May 8 and July 31, 2017, 429 patients were assigned for the study, and 419(97.7%) patients had the IPA dissected and recorded through photograph or video and were included in the primary outcome analysis. The median age was 62 years old, and 73.7% were male. Among the patients, 78.5% received laparoscopic surgery. Single IPA origination was identified in 398(95.0%) patients, including gastroduodenal artery(GDA) in154(36.8%) patients, anterior superior pancreaticoduodenal artery(ASPDA) in 130(31.0%) patients, and right gastroepiploic artery(RGEA) in 114(27.2%) patients. Fifteen(3.6%) patients were identified with multiple IPA and 6(1.4%) patients were identified as IPA absence. The differences in the distribution of surgical approach(P=0.003) and geographic area(P=0.030) were statistically significant. 7Host protection against Omicron BA.2.2 sublineages by prior vaccination in spring 2022 COVID-19 outbreak in Shanghai显示文摘The Omicron family of SARS-CoV-2 variants are currently driving the COVID-19 pandemic.Here we analyzed the clinical laboratory test results of 9911 Omicron BA.2.2 sublineages-infected symptomatic patients without earlier infection histories during a SARS-CoV-2 outbreak in Shanghai in spring 2022.Compared to an earlier patient cohort infected by SARS-CoV-2 prototype strains in 2020,BA.2.2 infection led to distinct fluctuations of pathophysiological markers in the peripheral blood.In particular,severe/critical cases of COVID19 post BA.2.2 infection were associated with less pro-inflammatory macrophage activation and stronger interferon alpha response in the bronchoalveolar microenvironment.Importantly,the abnormal biomarkers were significantly subdued in individuals who had been immunized by 2 or 3 doses of SARS-CoV-2 prototypeinactivated vaccines,supporting the estimation of an overall 96.02% of protection rate against severe/critical disease in the 4854 cases in our BA.2.2 patient cohort with traceable vaccination records.Furthermore,even though age was a critical risk factor of the severity of COVID-19 post BA.2.2 infection,vaccination-elicited protection against severe/critical COVID-19 reached 90.15% in patients aged≥60 years old.Together,our study delineates the pathophysiological features of Omicron BA.2.2 sublineages and demonstrates significant protection conferred by prior prototype-based inactivated vaccines.Ziyu Fu Dongguo Liang Wei Zhang Dongling Shi Yuhua Ma Dong Wei Junxiang Xi Sizhe Yang Xiaoguang Xu Di Tian Zhaoqing Zhu Mingquan Guo Lu Jiang Shuting Yu Shuai Wang Fangyin Jiang Yun Ling Shengyue Wang Saijuan Chen Feng Liu Yun Tan Xiaohong Fan 2023Frontiers of Medicine2023,17,3:1
8Enhanced ethanol sensing performance of N-doped ZnO derived from ZIF-8显示文摘Metal oxides derived from metal-organic framework(MOF) have attracted considerable attention due to its excellent performance and unique structure. Doping is considered as an effective method to improve gas-sensing performance. However, nonmetal doped metal oxides derived from MOF as gas-sensing materials have not been reported. Within this work, N atoms were successfully doped into the lattice of ZnO nanoparticles using ZIF-8 as a self-sacrificial template through a thermal treatment process with the assistant of urea. The obtained N-ZnO exhibited competitive ethanol-sensing performance, in which the response value of N-ZnO-5 to 100 ppm ethanol reached 115 at 190 ℃ with a satisfactory selectivity. It was found that the N-doping in ZnO facilitated the formation of oxygen vacancy that promoted the generation of adsorbed oxygen species to achieve the enhanced gas-sensing performance. 10CONCENTRATIONS AND FLUXES OF DISSOLVED NUTRIENTS IN THE YANGTZE RIVER: LONG-TERM TRENDS AND ECOLOGICAL IMPACTS显示文摘Intensifying human activity in the Yangtze River Basin(YRB) has substantially increased nutrient concentrations in the Yangtze River Estuary,leading to degradation of the coastal environment.Analysis of nutrient determinations published over the past 50 years reveals a gradual decreasing trend in the concentrations and fluxes of dissolved silicate(DSi).However,both dissolved inorganic nitrogen(DIN) and dissolved inorganic phosphate(DIP) concentrations have increased significantly since the 1970 s.The frequency and area covered by red tide outbreaks have increased greatly during this period,mainly due to changes in nutrient supply ratios [i.e.,N/P(DIN/DIP),N/Si(DIN/DSi),P/Si(DIP/DSi)].A strong correlation was found between the riverine DIN fluxes and the estimated DIN inputs from the major N sources,particularly fertilizers and atmospheric deposition.The data provide a comprehensive assessment of nutrients in the YRB and their ecological impacts and indicate a potentially significant influence of atmospheric deposition on DIN loadings and fluxes.Yandan FU Jiahui KANG Ziyue LI Xuejun LIU Wen XU 2021Frontiers of Agricultural Science and Engineering2021,8,4:0
11PathogenTrack and Yeskit:tools for identifying intracellular pathogens from single-cell RNA-sequencing datasets as illustrated by application to COVID-19显示文摘Pathogenic microbes can induce cellular dysfunction,immune response,and cause infectious disease and other diseases including cancers.However,the cellular distributions of pathogens and their impact on host cells remain rarely explored due to the limited methods.Taking advantage of single-cell RNA-sequencing(scRNA-seq)analysis,we can assess the transcriptomic features at the single-cell level.Still,the tools used to interpret pathogens(such as viruses,bacteria,and fungi)at the single-cell level remain to be explored.Here,we introduced PathogenTrack,a python-based computational pipeline that uses unmapped scRNA-seq data to identify intracellular pathogens at the single-cell level.In addition,we established an R package named Yeskit to import,integrate,analyze,and interpret pathogen abundance and transcriptomic features in host cells.Robustness of these tools has been tested on various real and simulated scRNA-seq datasets.PathogenTrack is competitive to the state-of-the-art tools such as Viral-Track,and the first tools for identifying bacteria at the single-cell level.Using the raw data of bronchoalveolar lavage fluid samples(BALF)from COVID-19 patients in the SRA database,we found the SARS-CoV-2 virus exists in multiple cell types including epithelial cells and macrophages.SARS-CoV-2-positive neutrophils showed increased expression of genes related to type I interferon pathway and antigen presenting module.Additionally,we observed the Haemophilus parahaemolyticus in some macrophage and epithelial cells,indicating a co-infection of the bacterium in some severe cases of COVID-19.The PathogenTrack pipeline and the Yeskit package are publicly available at GitHub.Wei Zhang Xiaoguang Xu Ziyu Fu Jian Chen Saijuan Chen Yun Tan 2022Frontiers of Medicine2022,16,2:0
12Integrated analysis of gut microbiome and host immune responses in COVID-19显示文摘Emerging evidence indicates that the gut microbiome contributes to the host immune response to infectious diseases.Here,to explore the role of the gut microbiome in the host immune responses in COVID-19,we conducted shotgun metagenomic sequencing and immune profiling of 14 severe/critical and 24 mild/moderate COVID-19 cases as well as 31 healthy control samples.We found that the diversity of the gut microbiome was reduced in severe/critical COVID-19 cases compared to mild/moderate ones.We identified the abundance of some gut microbes altered post-SARS-CoV-2 infection and related to disease severity,such as Enterococcus faecium,Coprococcus comes,Roseburia intestinalis,Akkermansia ntuciniphila,Bacteroides cellulosilyticus and Blautia obeum.We further analyzed the correlation between the abundance of gut microbes and host responses,and obtained a correlation map between clinical features of COVID-19 and 16 severity-related gut microbe,including Coprococcus comes that was positively correlated with CD3^(+)/CD4^(+)/CD8^(+)lymphocyte counts.In addition,an integrative analysis of gut microbiome and the transcriptome of peripheral blood mononuclear cells(PBMCs)showed that genes related to viral transcription and apoptosis were up-regulated in Coprococcus comes low samples.Moreover,a number of metabolic pathways in gut microbes were also found to be differentially enriched in severe/critical or mild/moderate COVID-19 cases,including the superpathways of polyamine biosynthesis II and sulfur oxidation that were suppressed in severe/critical COVID-19.Together,our study highlighted a potential regulatory role of severity related gut microbes in the immune response of host.Xiaoguang Xu Wei Zhang Mingquan Guo Chenlu Xiao Ziyu Fu Shuting Yu Lu Jiang Shengyue Wang Yun Ling Feng Liu Yun Tan Saijuan Chen 2022Frontiers of Medicine2022,16,2:0

14Prevalence and risk factors for minimal hepatic encephalopathy in cirrhotic patients with different etiologies显示文摘Aims:Minimal hepatic encephalopathy(MHE)significantly affects the prognosis of patients with cirrhosis.This study was performed to determine whether there is a difference in the prevalence of MHE among patients with cirrhosis of different etiologies and whether the etiology directly influences the occurrence of MHE.Methods:This multicenter,cross-sectional study enrolled 1879 patients with confirmed cirrhosis at 40 hospitals from October 25,2021,to January 10,2023(Trial registration:http://gffzzd3f38a9638994581sub9k5bxbfpnf65nb.ffgz.tsg.suse.edu.cn/[NCT05140837]).The patients'demographics,etiologies of cirrhosis,and laboratory test results were collected.The psychometric hepatic encephalopathy score(PHES)was determined in all patients to screen for MHE.Multivariate logistic analyses were performed to identify the risk factors for MHE.Results:In total,736 patients with cirrhosis were analyzed.The prevalence of MHE was 42.0%(n=309).The primary etiology among all patients was hepatitis B virus(HBV)-related cirrhosis(71.9%[529/736]).The prevalence of MHE was significantly higher in patients with alcoholic cirrhosis(57.1%[40/70])than in those with HBV-related cirrhosis(40.6%[215/529],p=0.009)or hepatitis C virus(HCV)-related cirrhosis(38.2%[26/68],p=0.026).Age(odds ratio[OR],1.042;95%confidence interval[CI],1.024-1.059;p<0.001),duration of education(OR,0.935;95%CI,0.899-0.971;p=0.001),etiology(OR,1.740;95%CI,1.028-2.945;p=0.039),and high MELD-Na scores(OR,1.038;95%CI,1.009-1.067;p=0.009)were independent risk factors for MHE.When patients with cirrhosis of different etiologies were analyzed separately,the results showed that age(OR,1.035;95%CI,1.014-1.057;p=0.001)and duration of education(OR,0.924;95%CI,0.883-0.966;p=0.001)were risk factors for MHE among patients with HBV-related cirrhosis,whereas age(OR,1.138;95%CI,1.033-1.254;p=0.009)and creatinine concentration(OR,16.487;95%CI,1.113-244.160;p=0.042)were risk factors for MHE in patients with HCV-related cirrhosis.No risk factors for MHE were found in patients with autoimmune cirrhosis.For patients with alcoholic cirrhosis,the platelet count(OR,1.014;95%CI,1.000-1.027;p=0.045)was a risk factor for MHE.The PHES subtest results were inconsistent among patients who had MHE with cirrhosis of different etiologies.Patients with HBV-related cirrhosis performed better on Number Connection Test B and the serial dotting test than those with alcoholic cirrhosis(p=0.007 and p<0.001),better on Number Connection Test B than those with HCV-related cirrhosis(p=0.020),and better on the line tracing test than those with autoimmune cirrhosis(p=0.037).Conclusion:The etiology of cirrhosis affected the prevalence of MHE and risk factors for MHE.The domains of major cognitive impairment varied among patients with cirrhosis of different etiologies.Further studies are required to verify these findings.Xiaoyan Li Shanghao Liu Huiling Xiang Qingge Zhang Ying Guo Hongmei Zu Jing Wang Jiaojian Lv Xiaoning Zhang Fanping Meng Jiahuan Li Jie Li Yangzhen Bianba Jia Shang Guo Zhang Fei Liu Zhaowei Tong Chuang Lei Wei Ye Qiaohua Yang Ningning Wang Ying Song Wei Fu Ziyue Li Yanjing Gao Yongping Zhang Jiafang Chen Caiyun Wu Qi Zheng Fang Wang Jiali Yu Lianjie Lin Chuanlong Yang Xiaoting Yang Xiaomin Ye Xiangmei Wang Xuelan Zhao Xiaolong Qi Fusheng Wang Junliang Fu 2023Portal Hypertension & Cirrhosis2023,2,4:0
15Estimations of Land Surface Characteristic Parameters and Turbulent Heat Fluxes over the Tibetan Plateau Based on FY-4A/AGRI Data显示文摘Accurate estimates of land surface characteristic parameters and turbulent heat fluxes play an important role in the understanding of land-atmosphere interaction. In this study, Fengyun-4A (FY-4A) Advanced Geostationary Radiation Imager (AGRI) satellite data and the China Land Data Assimilation System (CLDAS) meteorological forcing dataset CLDAS-V2.0 were applied for the retrieval of broadband albedo, land surface temperature (LST), radiation flux components, and turbulent heat fluxes over the Tibetan Plateau (TP). The FY-4A/AGRI and CLDAS-V2.0 data from 12 March 2018 to 30 April 2018 were first used to estimate the hourly turbulent heat fluxes over the TP. The time series data of in-situ measurements from the Tibetan Observation and Research Platform were divided into two halves-one for developing retrieval algorithms for broadband albedo and LST based on FY-4A, and the other for the cross validation. Results show the root-mean-square errors (RMSEs) of the FY-4A retrieved broadband albedo and LST were 0.0309 and 3.85 K, respectively, which verifies the applicability of the retrieval method. The RMSEs of the downwelling/upwelling shortwave radiation flux and downwelling/upwelling longwave radiation flux were 138.87/32.78 W m^(−2) and 51.55/17.92 W m^(−2), respectively, and the RMSEs of net radiation flux, sensible heat flux, and latent heat flux were 58.88 W m^(−2), 82.56 W m^(−2) and 72.46 W m^(−2), respectively.
