| 1 | A novel m6A reader Prrc2a controls oligodendroglial specification and myelination显示文摘While N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, is linked to cell differentiation and tissue development, the biological significance of m6A modification in mammalian glial development remains unknown. Here, we identify a novel m6A reader, Prrc2a (Proline rich coiled-coil 2A), which controls oligodendrocyte specification and myelination. Nestin-Cre-mediated knockout of Prrc2a induces significant hypomyelination, decreased lifespan, as well as locomotive and cognitive defects in a mouse model. Further analyses reveal that Prrc2a is involved in oligodendrocyte progenitor cells (OPCs) proliferation and oligodendrocyte fate determination. Accordingly, oligodendroglial-lineage specific deletion of Prrc2a causes a similar phenotype of Nestin-Cre-mediated deletion. Combining transcriptome-wide RNA-seq, m6A-RIP-seq and Prrc2a RIP-seq analysis, we find that Olig2 is a critical downstream target gene of Prrc2a in oligodendrocyte development. Furthermore, Prrc2a stabilizes Olig2 mRNA through binding to a consensus GGACU motif in the Olig2 CDS (coding sequence) in an m6A-dependent manner. Interestingly, we also find that the m6A demethylase, Fto, erases the m6A modification of Olig2 mRNA and promotes its degradation. Together, our results indicate that Prrc2a plays an important role in oligodendrocyte specification through functioning as a novel m6A reader. | 2 | Adjuvant chemotherapy with paclitaxel and cisplatin in lymph node-positive thoracic esophageal squamous cell carcinoma显示文摘Objective:No standard postoperative adjuvant chemotherapy has ever been established in node-positive esophageal squamous cell carcinoma(ESCC).This is a study to explore the effect of postoperative paclitaxel(PTX)and cisplatin(DDP)in lymph node-positive,completely resected thoracic ESCC patients.Methods:We conducted a prospective phase II trial.Patients had pathologically node-positive thoracic ESCC with negative margins.Outcomes of disease-free survival(DFS)and overall survival(OS)were compared with a matched historical control cohort.The postoperative chemotherapy regimen consisted of 4 to 6 cycles of PTX 150mg/m2 administered intravenously on d 1 followed by DDP 50 mg/m2 on d 2 every 14 d.Results:Forty-three patients were accrued from December 2007 to May 2012 at Cancer Hospital of Chinese Academy of Medical Sciences for adjuvant chemotherapy.The historical control group consisted of 80 patients who received complete resection but no adjuvant chemotherapy during the same period of time.Of the 43 patients with adjuvant chemotherapy,37(86.0%)patients completed 4 to 6 cycles of chemotherapy.The 3-year DFS rates were56.3%in the adjuvant group and 34.6%in the control group(P=0.006).The 3-year OS rates were 55.0%in the adjuvant group and 37.5%in the control group(P=0.013).Multivariate analysis revealed that postoperative chemotherapy was the significant predictor for improved OS(P=0.005).Conclusions:Biweekly adjuvant PTX and DDP might improve 3-year DFS and OS in lymph node-positive,curatively resected thoracic ESCC patients.These conclusions warrant further study in randomized phase III clinical trials. | Liangze Zhang Weiwei Li Xiao Lyu Yan Song Yousheng Mao Shaoming Wang Jing Huang | 2017 | Chinese Journal of Cancer Research2017,29,2: | 15 |

| 4 | Serum levels of perfluorinated compounds in the general population in Shenzhen, China显示文摘Perfluorinated compounds (PFCs) have been detected in many environmental matrices, biota, and nonoccupationally exposed populations in China recently. However, little is known about the distribution and levels of various PFCs in the general population living in areas where there is PFC exposure. In the present study, the levels and prevalence of ten target PFCs were determined in 227 serum samples from a population of nonoccupationally exposed individuals in Shenzhen, China. Results indicated that human exposure to PFCs was prevalent in Shenzhen. Perfluorooctanoate (PFOA) was the dominant PFC contaminant in the serum samples, with a median concentration of 6.72 ng/mL, followed by perfluorooctane sulfonate (PFOS) with a median concentration of 2.07 ng/mL. Other PFCs were detected at much lower concentrations, with median concentrations ranging from 0.02 to 0.87 ng/mL. Statistically, no significant (P>0.05) gender differences were observed for any of the PFCs. Significant (P<0.01) positive correlations were found between age and serum concentrations of the target PFCs, except for perfluorobutane sulfonate (R = 0.16, P = 0.01), perfluorohexanoic acid (R = 0.08, P = 0.22), and perfluoroheptanoic acid (R = 0.11, P = 0.10). | 5 | Peripheral Lymphocyte Subsets as a Marker of Parkinson's Disease in a Chinese Population显示文摘In this study, we conducted a clinical analysis of lymphocyte subtypes in 268 patients with Parkinson's disease(PD) to assess their clinical impact as a potential marker of advanced PD in Chinese patients. The participants comprised 268 sporadic PD patients and 268 healthy controls. The numbers of natural killer(NK) cells and CD3+, CD3+CD4+, CD3+CD8+, and CD19+ lymphocytes from peripheral blood were determined by immunostaining and flow cytometric analysis and the percentages of these CD+ T cells were calculated. The ratio of regulatory T(Treg)/helper T 17(Th17) lymphocytes from 64 PD patients and 46 controls was determined by flow cytometric analysis.The results showed that the percentage of NK cells was higher in advanced PD patients than in controls(22.92% ±10.08% versus 19.76% ± 10.09%, P = 0.006), while CD3+ T cells are decreased(62.93% ± 9.27% versus65.75% ± 9.13%, P = 0.005). The percentage of CD19+B cells in male patients was lower(P = 0.021) than in female patients, whereas NK cells were increased(P \ 0.0001). The scores on the Unified Parkinson's Disease Rating Scale(UPDRS) and the Non-Motor Symptoms Scale in late-onset PD patients were significantly higher than those in earlyonset patients(P = 0.024 and P = 0.007, respectively). The percentage of CD19+ B cells in patients with UPDRS scores[24 was lower than in those with scores \24(10.17% ±4.19% versus 12.22% ± 5.39%, P = 0.009). In addition, the Treg/Th17 ratio in female patients was higher than that in female controls(13.88 ± 6.32 versus 9.94 ± 4.06, P =0.042). | 6 | Immunodiagnostic efficacy of detection of Schistosoma japonicum human infections in China:a meta analysis显示文摘Objective:To assess the diagnostic efficacy of the currently most widely used indirect hemagglutination assay(IHA) and enzyme-linked immunosorbent assay(ELISA) for detection of Schistosoma japonicum human infections.Methods:A comprehensive search was undertaken from China National Knowledge Infrastructure,Wanfang Database.VIP Database,PubMed. Cochrane Library,Science Citation Index Expanded.Proquest,and the inclusion and exclusion criteria were strictly settled.The funnel plot was used to assess the publication bias.Cochran’s Q test was employed to measure the homogeneity between studies,a summary receiver operating characteristic(SROC) curve was used to compare the diagnostic accuracy between the IHA and ELISA qualitatively by means of the Weighted Least Square method,the Ordinary Least Square method and the Robust regression method,and the diagnostic odds ratio(DOR) was drawn to compare the accuracy quantitatively.Results:Out of 785 publications,19 papers were eventually selected for analysis.Literature finality assessment indicated that minor publication bias existed in studies pertaining IHA test,but no bias was found in literatures regarding ELISA test.The heterogeneity test showed a heterogeneity between studies was present(χ~2 =466.07 and 34.67. both P values<0.0001).The areas under the SROC curves of IHA were all higher than that of ELISA test using the three methods(Weighted Least Square method:0.766 vs.0.695.Ordinary Least Square method:0.826 vs.0.741.Robust regression:0.815 vs.0.715).The TPR* values for IHA and EUSA were 0.710.0.759.0.749.and 0.650.0.686 and 0.666.respectively,and OR values were 5.997.9.937.8.893.and 3.432.4.784 and 3.959.respectively.The DOR of IHA was 9.41(95% CI:4.88-18.18).and 4.78(95%CI:3.21-7.13) for ELISA.Conclusions:All above results revealed that the diagnostic performance of IHA is better than that of ELISA.However,taking into account their unsatisfactory diagnostic value in areas with low infection intensity,a search for a better diagnostic test that can be applied in field situations in China should be given high priority. | Wei Wang Youzi Li Hongjun Li Yuntian Xing Guoli Qu Jianrong Dai Yousheng Liang | 2012 | Asian Pacific Journal of Tropical Medicine2012,5,1: | 4 |
| 7 | Demonstration of biophoton-driven DNA replication via gold nanoparticle-distance modulated yield oscillation显示文摘Biologically,there exist two kinds of syntheses:photosynthesis and ATP-driven biosynthesis.The light harvesting of photosynthesis is known to achieve an efficiency of〜95%by the quantum energy transfer of photons.However,how the ATP-driven biosynthesis reaches its high efficiency still remains unknown.Deoxynucleotide triphosphates(dNTPs)in polymerase chain reaction(PCR)adopt the identical way of ATP to release their energy,and thus can be employed to explore the ATP energy process.Here,using a gold nanoparticle(AuNP)enhanced PCR(AuNP-PCR),we demonstrate that the energy released by phosphoanhydride-bond(PB)hydrolysis of dNTPs is in form of photons(PB-photons)to drive DNA replication,by modulating their resonance with the average inter-AuNP distance(D).The experimental results show that both the efficiency and yield of PCR periodically oscillate with D increasing,indicating a quantized process,but not simply a thermal one.The PB-photon wavelength is further determined to 8.4 pm.All these results support that the release,transfer and utilization of bioenergy are in the form of photons.Our findings of ATP-energy quantum conversion will open a new avenue to the studies of high-efficiency bioenergy utilization,biochemistry,biological quantum physics,and even brain sciences. | Na Li Daoling Peng Xianjing Zhang Yousheng Shu Feng Zhang Lei Jiang Bo Song | 2021 | Nano Research2021,14,1: | 4 |
| 8 | Laminar Distribution of Neurochemically-Identified Interneurons and Cellular Co-expression of Molecular Markers in Epileptic Human Cortex显示文摘Inhibitory GABAergic interneurons are fundamental elements of cortical circuits and play critical roles in shaping network activity. Dysfunction of interneurons can lead to various brain disorders, including epilepsy,schizophrenia, and anxiety. Based on the electrophysiological properties, cell morphology, and molecular identity,interneurons could be classified into various subgroups. In this study, we investigated the density and laminar distribution of different interneuron types and the coexpression of molecular markers in epileptic human cortex.We found that parvalbumin(PV) and somatostatin(SST)neurons were distributed in all cortical layers except layer I, while tyrosine hydroxylase(TH) and neuropeptide Y(NPY) were abundant in the deep layers and white matter.Cholecystokinin(CCK) neurons showed a high density in layers IV and VI. Neurons with these markers constituted*7.2%(PV), 2.6%(SST), 0.5%(TH), 0.5%(NPY), and4.4%(CCK) of the gray-matter neuron population. Doubleand triple-labeling revealed that NPY neurons were also SST-immunoreactive(97.7%), and TH neurons were more likely to express SST(34.2%) than PV(14.6%). A subpopulation of CCK neurons(28.0%) also expressed PV, but none contained SST. | 11 | Adhesion Wear on Tool Rake and Flank Faces in Dry Cutting of Ti-6Al-4V显示文摘Titanium alloys are very chemically reactive and,therefore,have a tendency to weld to the cutting tool during machining.The deterioration in the tool life caused by adhesion is a serious problem when titanium alloys are cut using carbide tools.The chemical reactivity of titanium alloys with carbide tool materials and their consequent welding by adhesion onto the cutting tool during dry cutting leads to excessive chipping,premature tool failure,and poor surface finish.In the present study,dry turning and milling tests were carried out on Ti-6Al-4V alloys with WC?Co carbide tools.The adhesion on the tool rake and flank face was explored,the adhesive joint interface between the workpiece materials and tools were observed.SEM observation showed that adhesion can be observed both on the rake and the flank face,and was more pronounced in rake face than in flank face.There was evidence of element diffusion from the tool rake face to the adhering layer(vice versa) through the adhesive joint interface,which leads to the tool element loss and microstructure change.While the adhering materials at the flank face can be easily separated from the joint interface owing to the lower temperature and less pressure at the flank face,the adhesive wear attack results in an abrasive wear in the flank face.Moreover,adhesion is more notable in turning than in milling.The proposed research provides references for studying the adhesion between the workpiece materials and the tools,the adhesion mechanisms and their effect on the tool wear. | DENG Jianxin LI Yousheng ZHANG Hui | 2011 | Chinese Journal of Mechanical Engineering2011,24,6: | 2 |
| 12 | Effect of Metallic Zn Powder on Oxidation Resistance of Al_2O_3-C Refractories显示文摘Al2O3-C specimens were prepared by using tabular corundum, flaky graphite, Al, Si, and Zn powder as starting materials and phenolic formaldehyde resin as binder. Keeping the mass ratios of tabular corundum, flaky graphite, Al and Si constant, different contents of Zn (0, 0.5%, 1%, 2% and 3%) were added respectively. The specimens were heat treated at 120-220℃ for 24h, and then fired at 1400℃ for 3h in air atmosphere. The mass change rate, thickness of decarburization layer, apparent porosity, bulk density and microstructure of the products were determined and observed by means of SEM and EDX. The results show that the specimen with 2% Zn was characterized by minimum in thickness of decarburization layer and in apparent porosity and maximum in bulk density. | 18 | Development of a Fast ELISA for the Specific Detection of both Leucomalachite Green and Malachite Green显示文摘Malachite green(MG), a dye, is an antifungal agent that has been used to treat and prevent fish diseases. It is metabolized into reduced leucomalachite green forms(LMG) that may reside in fish muscles for a long period, thus being harmful to human health. The aim of this study was to develop a competitive and direct enzyme-linked immunosorbent assay(ELISA) to detect MG and LMG specifically. The monoclonal antibody(m Ab) to LMG was generated using a hybridoma technique. The obtained m Ab showed good cross-reactivity(CR) to malachite green(MG), but not to crystal violet(CV) and Brilliant Green(BG). The m Ab was used to develop a fast detecting ELISA of MG and LMG in fish. By introducing the conjugation LMG-HRP, the detection capability was 0.37 ng m L-1 for MG and LMG. The mean recovery from spiked grass carp tissues ranged from 76.2% to 82.9% and the coefficients of variation varied between 1.8% and 7.5%.
