| 1 | Genetic and biological characteristics of the globally circulating H5N8 avian influenza viruses and the protective efficacy offered by the poultry vaccine currently used in China显示文摘The H5N8 avian influenza viruses have been widely circulating in wild birds and are responsible for the loss of over 33 million domestic poultry in Europe, Russia, Middle East, and Asia since January 2020. To monitor the invasion and spread of the H5N8 virus in China, we performed active surveillance by analyzing 317 wild bird samples and swab samples collected from 41,172 poultry all over the country. We isolated 22 H5N8 viruses from wild birds and 14 H5N8 viruses from waterfowls. Genetic analysis indicated that the 36 viruses formed two different genotypes: one genotype viruses were widely detected from different wild birds and domestic waterfowls;the other genotype was isolated from a whopper swan. We further revealed the origin and spatiotemporal spread of these two distinct H5N8 virus genotypes in 2020 and 2021. Animal studies indicated that the H5N8 isolates are highly pathogenic to chickens, mildly pathogenic in ducks, but have distinct pathotypes in mice. Moreover, we found that vaccinated poultry in China could be completely protected against H5N8 virus challenge. | 2 | RNA-seq analysis identified hormone-related genes associated with prognosis of triple negative breast cancer显示文摘Triple negative breast cancer(TNBC) is an aggressive subtype of breast cancer that currently lacks effective biomarkers and therapeutic targets required to investigate the diagnosis and treatment of TNBC. Here we performed a comprehensive differential analysis of 165 TNBC samples by integrating RNA-seq data of breast tumor tissues and adjacent normal tissues from both our cohort and The Cancer Genome Atlas(TCGA). Pathway enrichment analysis was conducted to evaluate the biological function of TNBC-specific expressed genes. Further multivariate Cox proportional hazard regression was performed to evaluate the effect of these genes on TNBC prognosis. In this report, we identified a total of 148 TNBC-specific expressed genes that were primarily enriched in mammary gland morphogenesis and hormone levels related pathways, suggesting that mammary gland morphogenesis might play a unique role in TNBC patients differing from other breast cancer types. Further survival analysis revealed that nine genes(FSIP1, ADCY5, FSD1, HMSD, CMTM5, AFF3, CYP2 A7, ATP1 A2,and C11 orf86) were significantly associated with the prognosis of TNBC patients, while three of them(ADCY5,CYP2 A7, and ATP1 A2) were involved in the hormone-related pathways. | 4 | Low power and high uniformity of HfO_x-based RRAM via tip-enhanced electric fields显示文摘In this paper, the HfO_x-based resistive random access memory(RRAM) devices with sub-100 nm pyramid-type electrodes were fabricated. With the help of tip-enhanced electric field around the pyramid-type electrodes, it was experimentally demonstrated that the novel devices have better cycle-to-cycle variation control, lower forming/set voltage(1.97/0.7V) and faster switching speed(≤30 ns under 0.9V pulse) as well as better endurance reliability than conventional flat electrode resistive memory devices. In addition,the novel RRAM is experimentally demonstrated with independence of electrode number for the first time to present great potential in scaling down. The SDC and BCY powders were mixed in the weight ratio of 95:5, 85:15, and 75:25, respectively(named as BS95, BS85, and BS75). Because of the composite effect between the SDC and BCY phases, the BS95 and BS85 exhibit improved conductivity compared with the pure SDC and BCY. The conductivity of BS95 is higher than that of BS85, indicating that the composite effect of BS95 is greater than that of BS85. Nevertheless, the composite effect in BS75 does not exist. Hence, we conclude that the composite effect in the BCY-SDC composites will decrease with the increase of the amount of BCY and even disappear when the amount of BCY exceeds a certain value. In our case, the optimum composition of the composite electrolyte is 95 wt% SDC and 5 wt% BCY. The BS95 has the highest conductivity(σ1t=0.07808 S cm-1, at 800 °C) and the fuel cell based on the BS95 shows the best performance(the maximum power density reaches as high as 526 mw cm-2 at 750 °C). The geometries of reactions, transition state and products were completely optimized by B3LYP/6-31G(d). All the energy of the species was obtained at the CCSD(T)/6-31G(d,p) level. The calculated results indicated that all the major pathways of the reaction were obtained on the singlet potential energy surface. The singlet CCl2 can not only insert the Cα—H (reaction I) but also can react with Cβ—H (reaction Ⅱ). There are three main existing pathways and the products are P1 (CH3COHCCl2), P2 (CH2COHCHCl2) and P4[CHCl2CHCHOH] respectively. Reaction Ⅱ happens more easily according to the energy changes and the barrier in rate-controlling step. In addi-tion, the important geometries in domain pathways have been studied by AIM theory. | 17 | Identification of A-to-I RNA editing profiles and their clinical relevance in lung adenocarcinoma显示文摘Adenosine-to-inosine(A-to-I)RNA editing is a widespread posttranscriptional modification that has been shown to play an important role in tumorigenesis.Here,we evaluated a total of 19,316 RNA editing sites in the tissues of 80 lung adenocarcinoma(LUAD)patients from our Nanjing Lung Cancer Cohort(NJLCC)and 486 LUAD patients from the TCGA database.The global RNA editing level was significantly increased in tumor tissues and was highly heterogeneous across patients.The high RNA editing level in tumors was attributed to both RNA(ADAR1 expression)and DNA alterations(mutation load).Consensus clustering on RNA editing sites revealed a new molecular subtype(EC3)that was associated with the poorest prognosis of LUAD patients.Importantly,the new classification was independent of classic molecular subtypes based on gene expression or DNA methylation.We further proposed a simplified model including eight RNA editing sites to accurately distinguish the EC3 subtype in our patients.The model was further validated in the TCGA dataset and had an area under the curve(AUC)of the receiver operating characteristic curve of 0.93(95%CI:0.91-0.95).In addition,we found that LUAD cell lines with the EC3 subtype were sensitive to four chemotherapy drugs.These findings highlighted the importance of RNA editing events in the tumorigenesis of LUAD and provided insight into the application of RNA editing in the molecular subtyping and clinical treatment of cancer. | Cheng Wang Mingtao Huang Congcong Chen Yuancheng Li Na Qin Zijian Ma Jingyi Fan Linnan Gong Hui Zeng Liu Yang Xianfeng Xu Jun Zhou Juncheng Dai Guangfu Jin Zhibin Hu Hongxia Ma Fengwei Tan Hongbing Shen | 2022 | Science China(Life Sciences)2022,65,1: | 1 |

However, due to the inherent correlation between spectral bands and within-class variability in the data, the data processing of HHR is a challenging work. In this paper, based on spectral mixture analysis theory, a new stack of parts description features were extracted, and then incorporated with a stack of morphology based spatial features. Partially supervised constrained energy minimization (CEM) and unsupervised nonnegative matrix factorization (NMF) were used to extract the part-features. The joint features were then integrated by SVM classifier. The advantages of this method are the representation of physical composition of the urban area by the parts-features and the show of multi-scale structure information by morphology profiles. Experiments with an airborne hyper-spectral data flightline over the Washington DC Mall were performed, and the performance of the proposed algorithm was evaluated in comparison with well-known nonparametric weighted feature extraction (NWFE) and feature selection method. | 20 | Theoretical study of the charge carrier mobilities of the molecular materials tetrathiafulvalene (TTF) and 2,5-bis(1,3-dithiolan-2-ylidene)-1,3,4,6-tetrathiapentalene (BDH-TTP)显示文摘Tetrathiafulvalene (TTF) is a kind of fused ring aromatic compound containing four sulfur atoms in one molecule, which is well known as a charge transport material. In order to calculate the charge mobility of this semiconductor, Marcus electron transfer theory and the embedded model, which can give small intramolecular reorganization energies, were employed. The calculated results were in good agreement with the experimental values, so the above computing model is appropriate to assess the electrical property of TTF. On this basis, we predicted the charge mobility of 2,5-bis(1,3-dithiolan-2-ylidene)-1,3,4,6-tetrathiapentalene (BDH-TTP) crystals, for which the molecular structure is similar to TTF. The calculated results indicated that BDH-TTP is a p-type material, which has a better performance than TTF in hole transfer due to larger hole coupling and the smaller hole injection barrier. In addition, the direct coupling (DC) and the site energy correction (SEC) methods were used to calculate the charge transfer integrals. Although the results were slightly different, the qualitative trends were the same. Furthermore we took into account the anisotropic transfer properties of TTF and BDH-TTF, since obviously the mobilities along one dimension are larger than those along three dimensions.
