2Pharmacological effects of Astragaloside IV: a literature review显示文摘OBJECTIVE: To review the pharmacological effects and mechanisms of action of Astragaloside IV in Huangqi (Radix Astragali Mongolici). METHODS: Aticles focusing on Astragaloside IV in English and Chinese in databases were collected and reviewed in order to summarize the latest extraction separation, pharmacokinetics, and the pharmacological effects of astrageloside IV. RESULTS: Protective effects of Astrageloside IV on the cardiovascular system, immune, digestive, nervous system were identified, and the action mechanisms were associated with regulation of the calcium balance, anti-oxydant, antiapoptosis, antivirus, and so on. 4Andrographolide sulfonate ameliorates lipopolysaccharide-induced acute lung injury in mice by down-regulating MAPK and NF-κB pathways显示文摘Acute lung injury(ALI) or acute respiratory distress syndrome(ARDS) is a severe, lifethreatening medical condition characterized by widespread inflammation in the lungs, and is a significant source of morbidity and mortality in the patient population. New therapies for the treatment of ALI are desperately needed. In the present study, we examined the effect of andrographolide sulfonate, a water-soluble form of andrographolide(trade name: Xi-Yan-Ping Injection), on lipopolysaccharide(LPS)-induced ALI and inflammation. Andrographolide sulfonate was administered by intraperitoneal injection to mice with LPS-induced ALI. LPS-induced airway inflammatory cell recruitment and lung histological alterations were significantly ameliorated by andrographolide sulfonate. Protein levels of pro-inflammatory cytokines in bronchoalveolar lavage fluid(BALF) and serum were reduced by andrographolide sulfonate administration. m RNA levels of pro-inflammatory cytokines in lung tissue were also suppressed. Moreover, andrographolide sulfonate markedly suppressed the activation of mitogen-activated protein kinase(MAPK) as well as p65 subunit of nuclear factor-κB(NF-κB). In summary, these results suggest that andrographolide sulfonate ameliorated LPS-induced ALI in mice by inhibiting NF-κB and MAPK-mediated inflammatory responses. 5The FTO/miR-181b-3p/ARL5B signaling pathway regulates cell migration and invasion in breast cancer显示文摘Background:N6-methyladenosine(m6A)RNA modification has been demonstrated to be a significant regulatory process in the progression of various tumors,including breast cancer.Fat mass and obesity-associated(FTO)enzyme,initially known as the obesity-related protein,is the first identified m6A demethylase.However,the relationship between FTO and breast cancer remains controversial.In this study,we aimed to elucidate the role and clinical significance of FTO in breast cancer and to explore the underlying mechanism.Methods:We first investigated the expression of FTO in breast cancer cell lines and tissues by quantitative reverse transcription-PCR(qRT-PCR),Western blotting,and immunohistochemistry.Wound healing assay and Transwell assay were performed to determine the migration and invasion abilities of SKBR3 and MDAMB453 cells with either knockdown or overexpression of FTO.RNA sequencing(RNA-seq)was conducted to decipher the downstream targets of FTO.qRT-PCR,luciferase reporter assay,and Western blotting were employed to confirm the existence of the FTO/miR-181b-3p/ARL5B axis.The biological function of ADP ribosylation factor like GTPase 5B(ARL5B)in breast cancer cells was evaluated by wound healing assay and Transwell invasion assay.Results:High FTO expression was observed in human epidermal growth factor receptor 2(HER2)-positive breast cancer,predicting advanced progression(tumor size[P<0.001],nuclear grade[P=0.001],peritumoral lymphovascular invasion[P<0.001),lymph node metastasis[P=0.002],and TNM stage[P=0.001])and poor prognosis.Moreover,FTO promoted cell invasion and migration in vitro.Mechanistically,RNA-seq and further confirmation studies suggested that FTO up-regulated ARL5B by inhibiting miR-181b-3p.We further verified that ARL5B also displayed carcinogenic activity in breast cancer cells.Conclusion:Our work demonstrated the carcinogenic activity of FTO in promoting the invasion and migration of breast cancer cells via the FTO/miR-181b-3p/ARL5B signaling pathway.Yuanyuan Xu Shuang Ye Nan Zhang Shuhui Zheng Huatao Liu Kewen Zhou Ling Wang Yue Cao Peng Sun Tinghuai Wang 2020Cancer Communications2020,40,10:24

8Additive and Over-dominant Effects Resulting from Epistatic Loci Are the Primary Genetic Basis of Heterosis in Rice显示文摘A set of 148 F9 recombinant inbred lines(RILs) was developed from the cross of an indica cultivar 93-11 and japonica cultivar DT713,showing strong F1 heterosis.Subsequently,two backcross F1(BCF1) populations were constructed by backcrossing these 148 RILs to two parents,93-11 and DT713.These three related populations(281BCF1 lines,148 RILs) were phenotyped for six yield-related traits in two locations.Significant inbreeding depression was detected in the population of RILS and a high level of heterosis was observed in the two BCF1 populations.A total of 42 main-effect quantitative trait loci(M-QTLs) and 109 epistatic effect QTL pairs(E-QTLs) were detected in the three related populations using the mixed model approach.By comparing the genetic effects of these QTLs detected in the RILs,BCF1 performance and mid-parental heterosis(HMP),we found that,in both BCF1 populations,the QTLs detected could be classified into two predominant types:additive and over-dominant loci,which indicated that the additive and over-dominant effect were more important than complete or partially dominance for M-QTLs and E-QTLs.Further,we found that the E-QTLs detected collectively explained a larger portion of the total phenotypic variation than the M-QTLs in both RILs and BCF1 populations.All of these results suggest that additive and over-dominance resulting from epistatic loci might be the primary genetic basis of heterosis in rice.Xiaojin Luo Yongcai Fu Peijiang Zhang Shuang Wu Feng Tian Jiayong Liu Zuofeng Zhu Jinshui Yang Chuanqing Sun 2009Journal of Integrative Plant Biology2009,51,4:20

10Evaluation of vehicle-track-bridge interacted system for the continuous CRTS-II non-ballast track slab显示文摘Considering the CRTS-II track slab,which is commonly used in the Chinese high-speed railway system,a vehicle-track-bridge dynamic analysis method is proposed in which the vehicle subsystem equations are established by the rigid body dynamics method,the track subsystem and the bridge subsystem equations are established by the FEM,the wheel-rail contact relation is defined by the corresponding assumption in vertical direction and the Kalker linear creep theory in lateral direction.The in-span spring element is derived to model the track-bridge interaction;the equal-band-width storage is adopted to fit the track structure with multilayer uniform section beam;and the dynamic equilibrium equations are solved by the inter-history iteration method.As a case study,the response of a CRH2 high-speed train transverses a simply-supported bridge with successive 31.5m double bound pre-stress beams is simulated.The result shows that using the vehicle-track-bridge interaction model instead of the vehicle-bridge interaction model helps predict the rotation angle at beam ends and choose an economic beam vertical stiffness.ZHANG Nan ZHOU Shuang XIA He SUN Lu 2014Science China(Technological Sciences)2014,57,10:14

14GSA:Genome Sequence Archive显示文摘With the rapid development of sequencing technologies towards higher throughput and lower cost, sequence data are generated at an unprecedentedly explosive rate. To provide an efficient and easy-to-use platform for managing huge sequence data, here we present Genome Sequence Archive(GSA; http://gffzzdbc7b6aaae734bddhbwx0wcnnfok966n0.ffgz.tsg.suse.edu.cn/gsa or http://gffzz4eaf941a184140e3hbwx0wcnnfok966n0.ffgz.tsg.suse.edu.cn), a data repository for archiving raw sequence data. In compliance with data standards and structures of the International Nucleotide Sequence Database Collaboration(INSDC), GSA adopts four data objects(Bio Project, Bio Sample,Experiment, and Run) for data organization, accepts raw sequence reads produced by a variety of sequencing platforms, stores both sequence reads and metadata submitted from all over the world,and makes all these data publicly available to worldwide scientific communities. 16Metamorphic Characteristics and Tectonic Implications of the Kadui Blueschist in the Central Yarlung Zangbo Suture Zone,Southern Tibet显示文摘The Kadui blueschist is located in the central section of Yarlung Zangbo suture zone(YZSZ), southern Tibet, and has been subjected to the subduction of the Neo-Tethyan Ocean below the Asian Plate and provides important clues for better understanding the evolution of the India-Asia convergence zone. In this paper, the systematical petrographic and mineral chemical studies were carried out on the Kadui blueschist, which reveal a mineral assemblage of sodic amphibole, chlorite, epidote, albite and quartz with accessory minerals of titanite, calcite and zircon. Electron microprobe analyses demonstrate that amphibole shows zoned from actinolite core to ferrowinchite/riebeckite rim composition indicating that the sodic amphibole has formed during a prograde metamorphic event. The protolith of the blueschist is an intermediate-basic pyroclastic rock. The calculated pseudosection indicates a clockwise P-T path and constrains peak metamorphic conditions of about 5.9 kbar at 345 ℃. This condition is transitional between pumpellyite-actinolite, greenschist and blueschist facies with a burial depth of 20–22 km and a thermal gradient of 15–16 ℃/km. This thermal gradient belongs to high pressure intermediate P/T facies series and is possibly related to a warm subduction setting of young oceanic slabs. 17One-pot synthesis of graphene oxide and Ni-Al layered double hydroxides nanocomposites for the efficient removal of U(VI) from wastewater显示文摘Graphene oxide and Ni-Al layered double hydroxides(GO@LDH) nanocomposites were synthesized via a one-pot hydrothermal process,and characterized by X-ray diffraction(XRD),Fourier transformed infrared spectroscopy(FTIR),scanning electron microscopy(SEM),transmission electron microscopy(TEM),X-ray photoelectron spectroscopy(XPS) and Raman spectroscopy in detail.The exploration of U(VI) sorption on GO@LDH surface was performed as a function of ionic strength,solution pH,contact time,U(VI) initial concentrations and temperature.Results of Langmuir isotherms showed that the sorption capacity of GO@LDH(160 mg/g) was much higher than those of LDH(69 mg/g) and GO(92 mg/g).The formed surface complexes between surface oxygen-containing functional groups of GO@LDH and U(VI) turned out to be the interaction mechanism of U(VI) with GO@LDH.According to the thermodynamic studies results,the sorption interaction was actually a spontaneous and endothermic chemical process.The sorption isotherms were better fitted with the Langmuir model compared with other models,which suggested the interaction was mainly dominated by mono layer coverage.The GO@LDH nanocomposites provide potential applications as adsorbents in the enrichment of radionuclides from wastewater in nuclear waste management and environmental remediation.Shujun Yu Jian Wang Shuang Song Kunyu Sun Jun Li Xiangxue Wang Zhongshan Chen Xiangke Wang 2017Science China Chemistry2017,60,3:12
18Triptolide prolonged allogeneic islet graft survival in chemically induced and spontaneously diabetic mice without impairment of islet function显示文摘BACKGROUND:Triptolide(TPT)is a diterpenoid triepoxide extracted from the Chinese herb Tripterygium wilfordii Hook.F.It exhibits potent immunosuppressive and anti-inflammatory properties.This study was undertaken to investigate its effects on prolongation of islet allograft survival in rodents.Additionally,we investigated whether TPT would be toxic to islet function in vivo.METHODS:We transplanted BALB/c islets to either chemically induced diabetic C57BL/6 mice or spontaneously diabetic nonobese diabetic(NOD)mice.TPT was injected within 2 weeks or continuously,until rejection,in the two combinations.Then, we evaluated the toxicity of TPT on islet function by daily injection to naive BALB/c or diabetic BALB/c that was cured by syngeneic islet transplantation under the kidney capsule.Mice injected with cyclosporine A(CsA)or vehicle served as controls.Intraperitoneal glucose tolerance tests(IPGTTs)performed at 4 and 8 weeks in the na?ve BALB/c group,and at 2,4,6,and 8 weeks in the syngeneic transplanted group.RESULTS:The medium survival time of islets allograft from TPT treated C57BL/6 and NOD recipients were 28.5 days(range 24-30 days,n=10)and 33.0 days(range 15-47 days,n=6), respectively,and they were significantly different from those of the vehicle treated controls,which were 14.0 days(range 13-16 days,n=6)and 5.0 days(range 4-10 days,n=6),respectively(all P<0.0001).The IPGTT demonstrated that there was no difference between the TPT treated and vehicle treated groups, either in the normal or syngeneic transplanted islet BALB/c mice.However,CsA injection impaired islet function in both normal and syngeneic transplanted mice as early as 4 weeks.CONCLUSION:TPT prolonged islets allograft survival in a chemically induced diabetic or an autoimmune diabetic murine model without impairment of islet function.Xin, Ming-Jun Cui, Shi-Hua Liu, Shuang Sun, Hai-Chen Li, Fei Sun, Jia-Bang Luo, Bin 2010Hepatobiliary & Pancreatic Diseases International2010,9,3:11

20Protective effects of extracts of Schisandra chinensis stems against acetaminophen-induced hepatotoxicity via regulation of MAPK and caspase-3 signaling pathways显示文摘The present study was designed to evaluate protective activity of an ethanol extract of the stems of Schisandra chinensis(SCE) and explore its possible molecular mechanisms on acetaminophen(APAP) induced hepatotoxicity in a mouse model. The results of HPLC analysis showed that the main components of SCE included schisandrol A, schisandrol B, deoxyschisandrin, schisan--drin B, and schisandrin C and their contents were 5.83, 7.11, 2.13, 4.86, 0.42 mg·g^(-1), respectively. SCE extract was given for 7 con--secutive days before a single hepatotoxic dose of APAP(250 mg·kg^(-1)) was injected to mice. Our results showed that SCE pretreatment ameliorated liver dysfunction and oxidative stress, which was evidenced by significant decreases in aspartate transaminase(AST), alanine aminotransferase(ALT), malondialdehyde(MDA) contents and elevations in reduced glutathione(GSH) and superoxide dismutase(SOD) levels. These findings were associated with the result that the SCE pretreatment significantly decreased expression levels of 4-hydroxynonenal(4-HNE) and 3-nitrotyrosine(3-NT). SCE also significantly decreased the expression levels of Bax, mitogen-activated protein kinase(MAPK), and cleaved caspase-3 by APAP exposure. Furthermore, supplementation with SCE suppressed the expression levels of inducible nitric oxide synthase(iNOS) and cyclooxygenase-2(COX-2), suggesting alleviation of inflammatory response.
