4Mining,characterization,and exploitation of EST-derived microsatellites in Gossypium barbadense显示文摘Simple sequence repeats(SSRs) have been widely applied as molecular markers in genetic studies.However,the number of expressed sequence tags(ESTs) and SSR markers from Gossypium barbadense is fewer than those from other cotton species.In this study,EST-SSR distribution from G.barbadense was characterized and new G.barbadense-derived EST-SSR markers were determined on the basis of the ESTs obtained by randomly sequencing 2 cDNA libraries associated with fiber development in G.barbadense.By mining 9697 non-redundant ESTs,a total of 638 SSR loci derived from 595 ESTs were observed.In G.barbadense,the frequency of ESTs containing SSRs was 6.13%,with an average of 1 SSR in every 10.4 kb of EST sequence.Furthermore,trinucleotide was found to be the most abundant repeat type among 2-6-nucleotide repeat types.It accounted for 26.6% of the total,followed by the hexanucleotide(26.0%) and pentanucleotide repeats(25.9%).Among all the repeat motifs,(AAG)n accounted for the highest proportion.EST-SSR primer pairs were developed using the Primer3 program,and the redundant primers were removed using the virtual PCR approach.As a result,380 non-redundant EST-SSR primer pairs were developed and used to detect polymorphisms between the mapping parents G.hirsutum 'TM-1' and G.barbadense 'Hai7124' for constructing linkage groups in cultivated allotetraploid cotton.Out of these,98(25.8%) primer pairs detected polymorphisms.Finally,95 polymorphic loci from 82 primer pairs were integrated into the backbone genetic map;of these,42 were mapped into the A subgenome and 53 into the D subgenome.The present work provided the foundation for constructing saturated genetic maps and conducting comparative genomic studies on different cotton species.LU YuanDa CAI CaiPing WANG Lei LIN ShaoYan ZHAO Liang TIAN LiangLiang LU JunHong ZHANG TianZhen GUO WangZhen 2010Chinese Science Bulletin2010,55,18:5
5Late Holocene hydroclimatic variations and possible forcing mechanisms over the eastern Central Asia显示文摘Hydroclimatic variations over the eastern Central Asia are highly sensitive to changes in hemispheric-scale atmospheric circulation systems. To fully understand the long-term variability and relationship between hydroclimate and atmospheric circulation system, we present a high-resolution lascustrine record of late Holocene hydroclimate from Lake Sayram, Central Tianshan Mountains, China, based on the total organic carbon, total nitrogen, and carbonate contents, carbon/nitrogen ratios, and grain size. Our results reveal four periods of substantially increased precipitation at the interval of 4000–3780, 3590–3210, 2800–2160, and 890–280 cal yr BP, and one period of slightly increased precipitation from 1700–1370 cal yr BP. These wetter periods broadly coincide with those identified in other records from the mid-latitude Westerlies-dominated eastern Central Asia, including the northern Tibetan Plateau. As such, a similar hydroclimatic pattern existed over this entire region during the late Holocene. Based on a close similarity of our record with reconstruction of North Atlantic Oscillation indices and solar irradiance, we propose that decreased solar irradiance and southern migration of the entire circum-North Atlantic circulation system, particularly the main pathway of the mid-latitude Westerlies, significantly influenced hydroclimate in eastern Central Asia during the late Holocene. 6Energetic particles’fluxes and dose in the Radiation Gene Box measured by space radiation detector onboard SJ-10 satellite显示文摘Background To evaluate the hazard of space radiation posing to the tissues,it is important to obtain exact fluxes of different radiation particles.The Radiation Gene Box(RGB)onboard SJ-10 spacecraft was an instrument designed to investigate the effects of space environment on the mESCs and drosophila.To derive the dose received by the tissues inside the RGB,the Space Radiation Detector(SRD)was installed inside it.Purpose The SRD was designed to derive the fluxes of electron,proton,hellion and gamma rays around it.If the type of the particles,the energies,the fluxes and the conversion coefficients are known,the dose received by the tissues could be evaluated.Methods The SRDwas designed as a △E-E solid-state telescope.By measuring the energy deposited in the three subdetectors,the particles’type and their energies could be discriminated.The data of SRDwere divided into 15 bins by the types of particles and their energy ranges.Results The gamma ray flux was higher than any other particle flux inside the RGB,and the electron was the most intense charge particle,while the helium ion was the most harmful radiation to the cells inside the RGB.Conclusion The dose rate inside the Radiation Gene Box was much higher than in the ground,but the integral dose of 12 days inside the RGB was about 2.13 mSv.It seemed unlikely to have obvious biological effects on the tissues of mice and drosophila.Xingzhu Cui Yaqing Liu Wenxi Peng Jinzhou Wang Min Gao Dongya Guo Xiaohua Liang Ruirui Fan Huanyu Wang Yunlong Zhang Zhongjian Ma Mingyang Yan Hong Xiao Yuanda Jiang Haiying Hang 2018Radiation Detection Technology and Methods2018,2,2:2
7Interference at the single-photon level based on silica photonics robust against channel disturbance显示文摘Quantum key distribution(QKD) provides a solution for communication of unconditional security. However,the quantum channel disturbance in the field severely increases the quantum bit-error rate, degrading the performance of a QKD system. Here we present a setup comprising silica planar light wave circuits(PLCs), which is robust against the channel polarization disturbance. Our PLCs are based on the asymmetric Mach–Zehnder interferometer(AMZI), integrated with a tunable power splitter and thermo-optic phase modulators. The polarization characteristics of the AMZI PLC are investigated by a novel pulse self-interfering method to determine the operation temperature of implementing polarization insensitivity. Over a 20 km fiber channel with 30 Hz polarization scrambling, our time-bin phase-encoding QKD setup is characterized with an interference fringe visibility of 98.72%. 8Autonomous landing scene recognition based on transfer learning for drones显示文摘In this paper, we study autonomous landing scene recognition with knowledge transfer for drones. Considering the difficulties in aerial remote sensing, especially that some scenes are extremely similar, or the same scene has different representations in different altitudes, we employ a deep convolutional neural network(CNN) based on knowledge transfer and fine-tuning to solve the problem. Then, LandingScenes-7 dataset is established and divided into seven classes. Moreover, there is still a novelty detection problem in the classifier, and we address this by excluding other landing scenes using the approach of thresholding in the prediction stage. We employ the transfer learning method based on ResNeXt-50 backbone with the adaptive momentum(ADAM) optimization algorithm. We also compare ResNet-50 backbone and the momentum stochastic gradient descent(SGD) optimizer. Experiment results show that ResNeXt-50 based on the ADAM optimization algorithm has better performance. 10Deformation induced structural evolution in bulk metallic glasses显示文摘The structural behavior of binary Cu50Zr50 and ternary Cu50Zr45Ti5 bulk metallic glasses (BMGs) under applied stress was investigated by means of in-situ high energy X-ray synchrotron diffraction. The components of the strain tensors were determined from the shifts of the maxima of the atomic pair correlation functions (PDF) in real space. The anisotropic atomic reorientation in the first-nearest-neighbor shell versus stress suggests structural rearrangements in short-range order. Within the plastic deformation range the overall strain of the metallic glass is equal to the yield strain. After unloading,the atomic structure returns to the stress-free state,and the short-range order is identical to that of the undeformed state. Plastic deformation,however,leads to localized shear bands whose contribution to the volume averaged diffraction pattern is too weak to be detected. A concordant region evidenced by the anisotropic component is activated to counterbalance the stress change due to the atomic bond reorientation in the first-nearest-neighbor shell. 11Dynamic mechanical behavior of a Zr-based bulk metallic glass during glass transition and crystallization显示文摘The dynamic mechanical behaviors of the Zr41Ti14Cu12.5Ni8Be22.5Fe2 bulk metallic glass (BMG) during continuous heating at a constant rate were investigated. The glass transition and crystallization of the Zr-based BMG were thus characterized by the measurements of storage modulus E′ and internal friction Q-1. It was found that the variations of these dynamic mechanical quantities with temperature were interre-lated and were well in agreement with the DSC trace obtained at the same heating rate. The origin of the first peak in the internal friction curve was closely related to the dynamic glass transition and subsequent primary crystallization. Moreover, it can be well described by a physical model, which can characterize atomic mobility and mechanical response of disordered condense materials. 12Nano-structure of Iron Formed by Mechanical Grinding显示文摘Nano-crystalline elemental iron powder was successfully formed by high-energy ball grinding.The x-ray diffraction,transmission electron microscopy and Mdssbauer spectroscopy were employed to follow the structure changes in the process of mechanical grinding with milling time.The thermal stability of nanocrystalline iron powder was also studied by differential scanning calorimetry.The results showed that the grain size reduces drastically at the initial stage of the milling and reaches 6nm after 40h milling.The broadening of Mdssbauer spectrum is attributed to the large fraction of iron atoms existing at the interfaces for the powder milled.YANG Yuanzheng MA Xueming DONG Yuanda HE Yizhen WANG Gengmiao 1992Chinese Physics Letters1992,9,5:1
13Recapitulation of growth factor-enriched microenvironment via BMP receptor activating hydrogel显示文摘Exposure to a growth factor abundant milieu has remarkable regenerative and rejuvenating effects on organ diseases,tissue damage,and regeneration,including skeletal system defects and bone regeneration.Although the introduction of candidate growth factors into relevant fields has been reported,their regenerative effects remain unsatisfactory,mainly because of the experimental challenges with limited types of growth factors,elusive dosage adjustment,and asynchronous stem cell activation with cytokine secretion.Here,an innovative hydrogel recapitulating a growth factor-enriched microenvironment(GEM)for regenerative advantage,is reported.This sulfated hydrogel includes bone morphogenetic protein-2(BMP-2),an essential growth factor in osteogenesis,to direct mesenchymal stem cell(MSC)differentiation,stimulate cell proliferation,and improve bone formation.The semi-synthetic hydrogel,sulfonated gelatin(S-Gelatin),can amplify BMP-2 signaling in mouse MSCs by enhancing the binding between BMP-2 and BMP-2 type II receptors(BMPR2),which are located on MSC nuclei and activated by the hydrogel.Importantly,the dramatically improved cytokine secretion of MSCs throughout regeneration confirms the growth factor-acquiring potential of S-Gelatin/rhBMP-2 hydrogel,leading to the vascularization enhancement.These findings provide a new strategy to achieve an in situ GEM and accelerated bone regeneration by amplifying the regenerative capacity of rhBMP-2 and capturing endogenous growth factors.Qinghao Zhang Yuanda Liu Jie Li Jing Wang Changsheng Liu 2023Bioactive Materials2023,,2:0
14Influence of extraction solvents on antioxidant and antimicrobial activities of the pulp and seed of Anisophyllea laurina R.Br.ex Sabine fruits显示文摘Objective:To evaluate the influence of extraction solvents on antioxidant and antimicrobial activities of the pulp and seed of Anisophyllea laurina R.Br.ex Sabine fruits.Methods:The antibacterial activities of pulp and seed extracts were tested by using disk diffusion method against eight bacterial strains and three fungal strains.Total phenolic,flavonoid,monomeric anthocyanin and tannin contents,and antioxidant activities were determined by spectrometric methods.Results:The antioxidant analysis of pulp extract revealed the strong radical scavenging capacity and total phenolic content(4 329.66 mg of gallic acid/100 g),while seed extract showed the high antioxidant activity and total tannin content(5 326.78 mg catechin equivalent/100 g).Antibacterial and antifungal activities of methanol and ethanol extracts exhibited potent growth inhibitory activity against Aeromonas hydrophila,Bacillus subtilis,Escherichia coli O157:H7,Pseudomonas aeruginosa,Salmonella Typhimurium and Staphylococcus aureus ATCC 6538 with minimum inhibitory concentration values ranged from 125 to 250 μg/mL.However,seed extract had the strongest potential activity against Aspergillus niger and Candida albicans with minimum inhibitory concentration value of 500 μg/mL compared to pulp extract.Conclusions:Our results therefore demonstrated that ethanol and methanol extractions were more efficient in extracting antioxidants and bioactive compound in pulp and seed.These results support that these plant extracts can be used for the treatment of bacterial infections.Gbago Onivogui Rebaone Letsididi Mohamed Diaby Liping Wang Yuanda Song 2016Asian Pacific Journal of Tropical Biomedicine2016,6,1:0
15Oncolytic adenoviruses expressing checkpoint inhibitors for cancer therapy显示文摘Despite the remarkable success of immune checkpoint inhibitors(ICIs),primary resistance to ICIs causes only subsets of patients to achieve durable responses due to the complex tumor microenvironment(TME).Oncolytic viruses(OVs)can overcome the immunosuppressive TME and promote systemic antitumor immunity in hosts.Engineered OVs armed with ICIs would likely have improved effectiveness as a cancer therapy.According to the diverse immune cell landscapes among different types of tumors,we rationally and precisely generated three recombinant oncolytic adenoviruses(OAds):OAd-SIRPα-Fc,OAd-Siglec10-Fc and OAd-TIGIT-Fc.These viruses were designed to locally deliver SIRPα-Fc,Siglec10-Fc or TIGIT-Fc fusion proteins recognizing CD47,CD24 or CD155,respectively,in the TME to achieve enhanced antitumor effects.Our results suggested that OAd-SIRPα-Fc and OAd-Siglec10-Fc both showed outstanding efficacy in tumor suppression of macrophage-dominated tumors,while OAd-TIGIT-Fc showed the best antitumor immunity in CD8+T-cell-dominated tumors.Importantly,the recombinant OAds activated an inflammatory immune response and generated long-term antitumor memory.In addition,the combination of OAd-Siglec10-Fc with anti-PD-1 significantly enhanced the antitumor effect in a 4T1 tumor model by remodeling the TME.In summary,rationally designed OAds expressing ICIs tailored to the immune cell landscape in the TME can precisely achieve tumor-specific immunotherapy of cancer.Daoyuan Xie Yaomei Tian Die Hu Yuanda Wang Yuling Yang Bailing Zhou Rui Zhang Zhixiang Ren Mohan Liu Jie Xu Chunyan Dong Binyan Zhao Li Yang 2023Signal Transduction and Targeted Therapy2023,8,12:0
16Angiopoietin-1 mRNA expression in estradiol-treated ovariectomized rats with focal cerebral ischemia after reperfusion显示文摘BACKGROUND: Epidemiologic studies have indicated that the incidence of stroke in premenopausal females is lower than in males at the same age, but it significantly rises in postmenopausal females. Estrogen is used clinically to alleviate injury caused by cerebral ischemia. It has been hypothesized that the neuroprotective role of estrogen relates to angiopoietin (Angpt), which plays an important role in vascularization, vascular remodeling and maturation. OBJECTIVE: To observe and validate the effect of estradiol on angiopoietin-1 (Angpt1) mRNA expression in ovariectomized rats with focal cerebral ischemia after reperfusion, so as to explore the molecular mechanisms of estradiol-mediated protection from cerebral ischemic damage. DESIGN, TIME AND SETTING: Randomized, controlled, molecular biology, prospective animal study. The experiment was performed at the Central Laboratory of Chongqing Medical University from September to December 2005. MATERIALS: Fifty healthy female wild type (WT) rats aged 6 months and fifty female rats aged 6 months with knockout of the estrogen-alpha receptor gene (ERKO). METHODS: WT rats and ERKO rats were divided into estradiol and control groups (n = 25), and injected in- tramuscularly with estradiol benzoate (100 μg/kg per day) or corn oil (1 mL/kg per day) for 7 days, 30 days after bilateral ovariectomy. Rat models of cerebral ischemia/reperfusion were established with the middle cerebral artery occlusion method. After 30 minutes of middle cerebral artery occlusion, rats from the estradiol and control groups were injected intramuscularly with estradiol benzoate or corn oil at the above dose. MAIN OUTCOME MEASURES: We used radio-immunity analysis and laser-Doppler flowmetry to measure plasma estradiol levels and changes in cerebral blood flow. We used immunohistochemical staining of CD34 epitopes to measure changes in the capillary density in brain following cerebral ischemia/reperfusion, and quantitative RT-PCR analysis to assess mRNA expression levels of Angpt1, Angpt2, Tie2, vascular endothelial growth factor (Vegf), VegfR1, and VegfR2. RESULTS: In WT ovariectomized rats treated with estradiol, the change in cerebral blood flow following cerebral ischemia/reperfusion, capillary density in the basal nuclei and parietal lobe cortex and Angpt1 mRNA level were significantly higher than in the control group (P < 0.01). We did not identify any such changes in ERKO rats treated with estradiol. In addition, the plasma estradiol levels in WT and ERKO ovariectomized rats treated with estradiol were remarkably higher than in their corresponding control groups (P < 0.01). CONCLUSION: Angpt1 is a critical factor in many processes during the repair of cerebral ischemia/reperfusion injury. For example, it confers estrogen-mediated protection, restoration of cerebral blood flow and increases in brain capillary density. 17Angiopoietin-1 mRNA and Bcl-2 expression following estradiol treatment in ovariectomized rats with focal cerebral ischemia/reperfusion injury显示文摘BACKGROUND:Estrogen has been clinically demonstrated to attenuate ischemic brain injury. However,the precise mechanisms remain controversial. OBJECTIVE:To investigate the effects of estradiol on angiopoietin-1 mRNA and Bcl-2 expression, as well as apoptosis and cerebral blood flow,in ovariectomized rats with focal cerebral ischemia following reperfusion. DESIGN,TIME AND SETTING:Randomized,controlled,animal experiment.The study was performed at the Central Laboratory,Chongqing Medical University from September to December 2005. MATERIALS:Estradiol benzoate was purchased from Shanghai Ninth Pharmaceutical Factory; com oil was purchased from Walmart Supercenter;TUNEL kit,rabbit anti-rat Bcl-2 polyclonal antibody,and biotin-labeled goat anti-rabbit antibody were purchased from Wuhan Boster,China. METHODS:Healthy,female,6-month-old Wistar rats-wild-type and estrogen alpha receptor gene knockout(ERKO)-were randomly divided into estradiol and control groups with 25 animals in each group.The rats were intramuscularly injected with estradiol benzoate(100μg/kg per day) at 30 days following bilateral ovariectomy or corn oil(1 mL/kg per day) for seven consecutive days.Following administration,cerebral ischemia/reperfusion models were established using the right middle cerebral artery occlusion(MCAO) method.After 30 minutes of MCAO,estradiol and control groups were separately injected with estradiol benzoate and corn oil with the above-mentioned doses. MAIN OUTCOME MEASURES:Cell apoptosis was determined by TUNEL;angiopoietin-1 mRNA and Bcl-2 gene expression was determined,respectively,by immunohistochemical staining and RT-PCR.In addition,changes in cerebral blood flow were measured by laser Doppler flowmetry. RESULTS:Changes in angiopoietin-1 mRNA and cerebral blood flow in estradiol-treated,wild-type, MCAO rats following ischemia/reperfusion were greater than in control rats(P<0.01 or 0.05). However,no significant difference was observed between estradiol-treated ERKO MCAO rats and control rats.In addition,estradiol-treated wild-type and ERKO MCAO rats exhibited significantly increased Bcl-2 expression(P<0.05) and decreased number of apoptotic cells in brain tissues compared with control groups(P<0.05). 18Venovenous extracorporeal membrane oxygenation for COVID-19 and influenza H1N1 associated acute respiratory distress syndrome:A comparative cohort study in China显示文摘Background Venovenous extracorporeal membrane oxygenation(VV-ECMO)has been demonstrated to be effective in treating patients with virus-induced acute respiratory distress syndrome(ARDS).However,whether the management of ECMO is different in treating H1N1 influenza and coronavirus disease 2019(COVID-19)-associated ARDS patients remains unknown.Methods This is a retrospective cohort study.We included 12 VV-ECMO-supported COVID-19 patients admitted to The First Affiliated Hospital of Guangzhou Medical University,Guangzhou Eighth People's Hospital,and Wuhan Union Hospital West Campus between January 23 and March 31,2020.We retrospectively included VV-ECMO-supported patients with COVID-19 and H1N1 influenza-associated ARDS.Clinical characteristics,respiratory mechanics including plateau pressure,driving pressure,mechanical power,ventilatory ratio(VR)and lung compliance,and outcomes were compared.Results Data from 25 patients with COVID-19(n=12)and H1N1(n=13)associated ARDS who had received ECMO support were analyzed.COVID-19 patients were older than H1N1 influenza patients(P=0.004).The partial pressure of arterial carbon dioxide(PaCO_(2))and VR before ECMO initiation were significantly higher in COVID-19 patients than in H1N1 influenza patients(P<0.001 and P=0.004,respectively).COVID-19 patients showed increased plateau and driving pressure compared with H1N1 subjects(P=0.013 and P=0.018,respectively).Patients with COVID-19 remained longer on ECMO support than did H1N1 influenza patients(P=0.015).COVID-19 patients who required ECMO support also had fewer intensive care unit and ventilator-free days than H1N1.Conclusions Compared with H1N1 influenza patients,COVID-19 patients were older and presented with increased PaCO_(2) and VR values before ECMO initiation.The differences between ARDS patients with COVID-19 and influenza on VV-ECMO detailed herein could be helpful for obtaining a better understanding of COVID-19 and for better clinical management.Yonghao Xu Yin Xi Shuijiang Cai Yuheng Yu Sibei Chen Weijie Guan Weibo Liang Hongkai Wu Weiqun He Xilong Deng Yuanda Xu Rong Zhang Manshu Li Jieyi Pan Zhenting Liang Ya Wang Shaofeng Kong Xiaoqing Liu Zheng Lv Yimin Li 2023Journal of Intensive Medicine2023,3,4:0
19Antioxidant and anti-HepG2 cell activities of a novel bioactive peptide from cowhide collagen in vitro显示文摘In the present study,the antioxidant and anti-human liver cancer(HepG2)cells effects of bioactive peptides from cowhide collagen(BPCC)were evaluated.BPCC exhibited significant scavenging effect on l,1-diphenyl-2-picrylhydrazyl(DPPH)radicals((60.09±3.51)%),2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)radicals((77.40±3.10)%)and hydroxyl radicals((56.00±2.30)%)as well as strong reducing power(0.320士0.025).Meanwhile,BPCC effectively protected biomacromolecules including proteins,lipids and DNA from oxidative damage induced by Cu^(2+)H_(2)O_(2)and 2,2'-azobis(2-methylpropionamidine)dihydrochloride(AAPH).Moreover,BPCC significantly inhibited cell viability of HepG2 cells in a dose-dependent manner with an estimated IC_(50)of 7.61 mg/mL.The results of 4',6-diamidino-2-phenylindole(DAPI)and acridine:orange/ethidium bromide(AO/EB)staining demonstrated the apoptotic morphological changes and cell mediated death in BPCC treated HepG2 cells.In addition,BPCC induced decrease of mitochondrial membrane potential(MMP)in HepG2 cells.Therefore,the present finding proved that BPCC encompasses significant antioxidant activity and anticancer property on HepG2 cells and can be used as alternative food antioxidants for cancer prevention benefits.Zhike Xie Yuhan Zhai Yuqing Zhang Ming He Xuguang Wang Shaoxuan Yu Haifang Xiao Yuanda Song 2024Journal of Future Foods2024,4,3:0
20Scene image recognition with knowledge transfer for drone navigation显示文摘In this paper,we study scene image recognition with knowledge transfer for drone navigation.We divide navigation scenes into three macro-classes,namely outdoor special scenes(OSSs),the space from indoors to outdoors or from outdoors to indoors transitional scenes(TSs),and others.However,there are difficulties in how to recognize the TSs,to this end,we employ deep convolutional neural network(CNN)based on knowledge transfer,techniques for image augmentation,and fine tuning to solve the issue.Moreover,there is still a novelty detection prob-lem in the classifier,and we use global navigation satellite sys-tems(GNSS)to solve it in the prediction stage.Experiment results show our method,with a pre-trained model and fine tun-ing,can achieve 91.3196%top-1 accuracy on Scenes21 dataset,paving the way for drones to learn to understand the scenes around them autonomously.DU Hao WANG Wei WANG Xuerao ZUO Jingqiu WANG Yuanda 2023Journal of Systems Engineering and Electronics2023,34,5:0
