| 2 | Anlotinib has good efficacy and low toxicity: a phase Ⅱ study of anlotinib in pre-treated HER-2 negative metastatic breast cancer显示文摘Objective:Anlotinib is a novel tyrosine kinase inhibitor blocking angiogenesis.This study was performed to assess the efficacy and safety of anlotinib in patients with metastatic breast cancer.Methods:Patients with HER2-negative breast cancer,who were pre-treated with anthracycline or taxanes in a neoadjuvant,adjuvant,or metastatic setting,and had treatment failure after at least one prior chemotherapy regimen in the metastatic setting were enrolled.Anlotinib was administered at 12 mg daily for 14 days in a 21-day cycle until disease progression or unacceptable toxicity occurred.Simultaneously,5–10 m L of venous blood was collected to perform circulating tumor DNA(ct DNA)testing every 2 treatment cycles.The primary endpoint was the objective response rate(ORR).Secondary endpoints included the disease control rate(DCR),progression-free survival(PFS),overall survival,safety,and biomarkers.Results:Twenty-six eligible patients were enrolled,with a median age of 56(30–75)years.The median follow-up time was 10.5 months.The ORR was 15.4%,the DCR was 80.8%,and the median PFS was 5.22 months(95%confidence interval 2.86–6.24).Fourteen(53.8%)patients survived for more than 10 months.The changes in the detectable ct DNA variant allele frequency were consistent with the tumor response.The most common treatment-related adverse events were hypertension(57.7%),thyroidstimulating hormone elevation(34.6%),and hand-foot syndrome(23.1%).Conclusion:Anlotinib showed objective efficacy with tolerable toxicity in heavily pre-treated,metastatic HER2-negative breast cancer.The dynamic changes in the ct DNA variant allele fraction may be predictive of the tumor response. | Nanlin Hu Yiran Si Jian Yue Tingting Sun Xue Wang Zhuqing Jia Songlin Gao Qiao Li Yang Shao Jiayu Wang Yang Luo Fei Ma Binghe Xu Peng Yuan | 2021 | Cancer Biology & Medicine2021,18,3: | 11 |
| 3 | Recursive super-convergence computation for multi-dimensional problems via one-dimensional element energy pro jection technique显示文摘This paper presents a strategy for computation of super-convergent solutions of multi-dimensional problems in the finite element method(FEM) by recursive application of the one-dimensional(1D) element energy projection(EEP) technique. The main idea is to conceptually treat multi-dimensional problems as generalized 1D problems,based on which the concepts of generalized 1D FEM and its consequent EEP formulae have been developed in a unified manner. Equipped with these concepts, multi-dimensional problems can be recursively discretized in one dimension at each step, until a fully discretized standard finite element(FE) model is reached. This conceptual dimension-bydimension(D-by-D) discretization procedure is entirely equivalent to a full FE discretization. As a reverseD-by-D recovery procedure, by using the unified EEP formulae together with proper extraction of the generalized nodal solutions, super-convergent displacements and first derivatives for two-dimensional(2D) and three-dimensional(3D) problems can be obtained over the domain. | 4 | Spinal Cord Stimulation Frequency Influences the Hemodynamic Response in Patients with Disorders of Consciousness显示文摘Spinal cord stimulation(SCS) is a promising technique for treating disorders of consciousness(DOCs).However, differences in the spatio-temporal responsiveness of the brain under varied SCS parameters remain unclear. In this pilot study, functional near-infrared spectroscopy was used to measure the hemodynamic responses of 10 DOC patients to different SCS frequencies(5 Hz, 10 Hz, 50 Hz, 70 Hz, and 100 Hz). In the prefrontal cortex, a key area in consciousness circuits, we found significantly increased hemodynamic responses at 70 Hz and 100 Hz, and significantly different hemodynamic responses between 50 Hz and 70 Hz/100 Hz. In addition, the functional connectivity between prefrontal and occipital areas was significantly improved with SCS at 70 Hz. | 5 | The Status and Associated Factors of Successful Aging among Older Adults Residing in Longevity Areas in China显示文摘Objective This study aims to assess the status of successful aging(SA) in longevity areas in China and explore multiple factors associated with SA among the young-old and oldest-old. Methods A total of 2296 elderly people aged 65 and older were interviewed in the longevity areas sub-sample of the Chinese Longitudinal Healthy Longevity Survey(CLHLS) in 2012. Baseline assessments included a researcher-administered questionnaire, physical examination, and laboratory testing. A logistic regression model was used to identify factors associated with SA. Results The prevalence of SA was 38.81% in the CLHLS in 2012. There were significant differences between ages groups, with SA compromising 56.85% among ≥65 years group and 20.31% among ≥100 years group(χtrend2=126.73, P<0.01). The prevalence of SA among females was 33.59%, which was significantly lower than that among males(45.58%)(χ2gender=33.65, P<0.05). In the regression analysis, having anemia(OR=0.744, 95% CI: 0.609-0.910), poor lifestyle(OR=0.697, 95% CI: 0.568-0.854), poor sleep quality(OR=0.558, 95% CI: 0.456-0.682), and central obesity(OR=0.684, 95% CI: 0.556-0.841) were the main factors associated with SA. The promoting SA rate decreased as age increased, and the group of 65-79 years had higher odds than the other age group. | 13 | Short-term Chronic Intermittent Hypobaric Hypoxia Alters Gut Microbiota Composition in Rats显示文摘Chronic intermittent hypobaric hypoxia (CIHH) is a treatment of moderate hypoxia that simulates high altitude interrupted by normoxia. Growing evidence shows that CIHH has multiple beneficial effects on the body[1]. Our previous studies showed that CIHH (simulating 5, 000-m altitude for 28 days, 6 hours daily) protects the heart against ischemic/reperfusion-induced arrhythmias and damage through increased coronary flow and myocardial capillary angiogenesis, enhanced cardiac anti-oxidation, induction of heat shock protein 70, inhibition of endoplasmic reticulum stress, activation of ATP-sensitive potassium channels, and inhibition of mitochondrial permeability transition pores[2]. In addition, CIHH decreases high blood pressure by facilitating the baroreceptor reflex and enhancing arterial relaxation in renal vascular hypertensive rats. CIHH also improves metabolic dysfunction in rats with diabetes and metabolic syndrome. | 14 | Factors associated with upstaging in patients preoperatively diagnosed with ductal carcinoma in situ by core needle biopsy显示文摘Objective: Patients preoperatively diagnosed with ductal carcinoma in situ(DCIS) by core needle biopsy(CNB) exhibit a significant risk for upstaging on final pathology, which leads to major concerns of whether axillary staging is required at the primary operation. The present study aimed to identify clinicopathological factors associated with upstaging in patients preoperatively diagnosed with DCIS by CNB.Methods: The present study enrolled 604 patients(cN0 M0) with a preoperative diagnosis of pure DCIS by CNB, who underwent axillary staging between August 2006 and December 2015, at Fudan University Shanghai Cancer Center(Shanghai, China).Predictive factors of upstaging were analyzed retrospectively.Results: Of the 604 patients, 20.03%(n = 121) and 31.95%(n = 193) were upstaged to DCIS with microinvasion(DCISM) and invasive breast cancer(IBC) on final pathology, respectively. Larger tumor size on ultrasonography(> 2 cm) was independently associated with upstaging [odds ratio(OR) 1.558, P = 0.014]. Additionally, patients in lower breast imaging reporting and data system(BI-RADS) categories were less likely to be upstaged(4 B vs. 5: OR 0.435, P = 0.002;4 C vs. 5: OR 0.502, P = 0.001). Overall,axillary metastasis occurred in 6.79%(n = 41) of patients. Among patients with axillary metastasis, 1.38%(4/290), 3.31%(4/121)and 17.10%(33/193) were in the DCIS, DCISM, and IBC groups, respectively.Conclusions: For patients initially diagnosed with DCIS by CNB, larger tumor size on ultrasonography(> 2 cm) and higher BIRADS category were independent predictive factors of upstaging on final pathology. | 17 | Trend and seasonal variations of atmospheric CH_4 in Beijing显示文摘The atmospheric CH\-4 in Beijing is still increasing, even though its increasing rate has significantly decreased from 1.76%/a during 1985—1989 to 0.50%/a during 1990—1997. The seasonal variation of CH\-4 concentration showed a double\|peak pattern, one peak appearing in winter and the other in summer. It is evident that the annually seasonal variations of atmospheric CH\-4 in Beijing are different. From 1986 to 1997, the atmospheric CH\-4 increased by 185 ppbv, 37% and 21% of which were due to the increase in winter and in summer, respectively. After 1993, the annually seasonal increasing rate of CH\-4 concentration in summer (due to emission from biogenic sources) is negative while the increasing rate in winter (due to emission from non\|biogenic sources) is positive about 25 ppbv/a. As a result, the increase of CH\-4 emission from non\|biogenic sources in winter is the major reason that caused the annually seasonal increasing rate from 1993 to 1997. | 19 | Construction of Salmonella Pullorum ghost by co-expression of lysis gene E and the antimicrobial peptide SMAP29 and evaluation of its immune efficacy in specific-pathogen-free chicks显示文摘In this study,a safety enhanced Salmonella Pullorum(S.Pullorum)ghost was constructed using an antimicrobial peptide gene,and evaluated for its potential as a Pullorum disease(PD)vaccine candidate.The antimicrobial peptide SMAP29 was co-expressed with lysis gene E to generate S.Pullorum ghosts.No viable bacteria were detectable either in the fermentation culture after induction of gene E-and SMAP29-mediated lysis for 24 h or in the lyophilized ghost products.Specific-pathogenfree(SPF)chicks were intraperitoneally immunized with ghosts at day 7 of age and no mortality,clinical symptoms or signs of PD such as anorexia,depression and diarrhea were observed.On challenge with a virulent S.Pullorum strain at 4 wk post-immunization,a comparatively higher level of protection was observed in the S.Pullorum ghost immunized chickens with a minimum of pathological lesions and bacterial loads compared to the birds in inactivated vaccine groups.In addition,immunization with the S.Pullorum ghosts induced a potent systemic Ig G response and was associated with significantly increased levels of cytokine IFN-γand IL-4 and relative percentages of CD4^+ and CD8^+ T lymphocytes.Our results indicate that SMAP29 can be employed as a new secondary lethal protein to enhance the safety of bacterial ghosts,and to prepare a non-living bacterial vaccine candidate that can prevent PD in chickens. | TIAN Qiu-feng ZHOU Wei SI Wei YI Fei HUA Xin YUE Min CHEN Li-ping LIU Si-guo YU Shen-ye | 2018 | Journal of Integrative Agriculture2018,17,1: | 2 |
| 20 | A Novel ATM Antisense Transcript ATM-AS Positively Regulates ATM Expression in Normal and Breast Cancer Cells显示文摘Objective:The ataxia telangiectasia mutated(ATM)gene is a master regulator in cellular DNA damage response.The dysregulation of ATM expression is frequent in breast cancer,and is known to be involved in the carcinogenesis and prognosis of cancer.However,the underlying mechanism remains unclear.The bioinformatic analysis predicted a potential antisense transcript ATM-antisense(AS)from the opposite strand of the ATM gene.The purpose of this study was to identify ATM-AS and investigate the possible effect of ATM-AS on the ATM gene regulation.Methods:Single strand-specific RT-PCR was performed to verify the predicted antisense transcript ATM-AS within the ATM gene locus.qRT-PCR and Western blotting were used to detect the expression levels of ATM-AS and ATM in normal and breast cancer cell lines as well as in tissue samples.Luciferase reporter gene assays,biological mass spectrometry,ChIP-qPCR and RIP were used to explore the function of ATM-AS in regulating the ATM expression.Immunofluorescence and host-cell reactivation(HCR)assay were performed to evaluate the biological significance of ATM-AS in ATM-mediated DNA damage repair.Breast cancer tissue samples were used for evaluating the correlation of the ATM-AS level with the ATM expression as well as prognosis of the patients.Results:The ATM-AS significantly upregulated the ATM gene activity by recruiting KAT5 histone acetyltransferase to the gene promoter.The reduced ATM-AS level led to the abnormal downregulation of ATM expression,and impaired the ATM-mediated DNA damage repair in normal breast cells in vitro.The ATM-AS level was positively correlated with the ATM expression in the examined breast cancer tissue samples,and the patient prognosis.Conclusion:The present study demonstrated that ATM-AS,an antisense transcript located within the ATM gene body,is an essential positive regulator of ATM expression,and functions by mediating the binding of KAT5 to the ATM promoter.These findings uncover the novel mechanism underlying the dysregulation of the ATM gene in breast cancer,and enrich our understanding of how an antisense transcript regulates its host gene. | He CHENG Er-shao ZHANG Xiao SHI Ping-ping CAO Bei-jing PAN Xin-xin SI Yue LIU Nan YANG Ying CHU Xu-chun WANG Xiao HAN Zhi-hong ZHANG Yu-jie SUN | 2022 | Current Medical Science2022,42,4: | 2 |
