1SlMYB75,an MYB-type transcription factor,promotes anthocyanin accumulation and enhances volatile aroma production in tomato fruits显示文摘Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality.However,the use of a single gene to impact several metabolic pathways is difficult.Here,we show that overexpression of the single gene SlMYB75(SlMYB75-OE)is effective at improving multiple fruit quality traits.In these engineered fruits,the anthocyanin content reached 1.86mg g−1 fresh weight at the red-ripe stage,and these SlMYB75-OE tomatoes displayed a series of physiological changes,including delayed ripening and increased ethylene production.In addition to anthocyanin,the total contents of phenolics,flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6,4,and 1.2 times,respectively,compared to those of wild-type(WT)fruits.Interestingly,a number of aroma volatiles,such as aldehyde,phenylpropanoid-derived and terpene volatiles,were significantly increased in SlMYB75-OE fruits,with some terpene volatiles showing more than 10 times higher levels than those in WT fruits.Consistent with the metabolic assessment,transcriptomic profiling indicated that the genes involved in the ethylene signaling,phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits.Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC,AADC2 and TPS genes.The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality.Wei Jian Haohao Cao Shu Yuan Yudong Liu Juanfang Lu Wang Lu Ning Li Jianhui Wang Jian Zou Ning Tang Chan Xu Yulin Cheng Yanqiang Gao Wanpeng Xi Mondher Bouzayen Zhengguo Li 2019Horticulture Research2019,6,1:39
2Impending HRT wave precursors to the Wenchuan M_s8.0 earthquake and methods of earthquake impending prediction by using HRT wave显示文摘We deployed four geo-electric monitoring stations in Sichuan and Yunnan provinces from 2004, using the new generation of equipment (PS-100) and technologies to capture the HRT wave earthquake precursor. Before the Wenchuan Ms8.0 earthquake, we recorded the HRT wave precursor at the only operating station in Hongge (HG, Δ=465 km) and found that significant impending signal had been recorded at the station in the early morning ( 0―5 am) of 12th of May, 2008. The precursor for this earthquake is consistent with precursors recorded for other strong earthquakes. The measured physical properties (geo-resistivity and telluric-current) show tidal wave period oscillations from several days to several months before the earthquakes and the amplitude of such HT oscillation increases significantly towards the occurrence of an earthquake. These HT and RT waves from the epicenter have a causal relationship with the earthquakes that happened several days later. The arrival time of two RT waves is proportional to the distance from the station to the epicenter. The estimated natural decay of the amplitude is correlated with the natural period (T0) of the earthquake fault, which is proportional to the fault length. From this relationship, we can predict the earthquake magnitude. For magnitude 6―9 earthquakes, the natural period is about 1―6 hours. Such oscillation comes from the epicenter area and they can propagate several thousand kilometers in the Earth's crust. Before a strong earthquake in the shallow crust, the conductive pore fluid will experience major changes before the fault rapture. Such fluid change will emit an oscillation in the pore fluid pressure. This is the mechanism for the HRT wave generation. Since the China Earthquake Administration funded the HRT wave short-term earth-quake prediction project in 2003, the first record of HRT precursor wave has been recorded from the 2004-12-26 Sumatra Mw9.0 earthquake with the largest epicentre distance Δ=2900 km. Thereafter, we have captured HRT waves from more than twenty strong earthquakes, which are well-matched and show repeatability, consistency and regularity. All our observation with the HRT waves demonstrate that HRT wave precursors to earthquakes indeed exist. Strong earthquakes can be predicted and short-term and impending earthquake prediction is achievable in the very near future. 3Neural substrates of data-driven scientific discovery:An fMRI study during performance of number series completion task显示文摘Although much has been known about how humans psychologically perform data-driven scientific discovery,less has been known about its brain mechanism.The number series completion is a typical data-driven scientific discovery task,and has been demonstrated to possess the priming effect,which is attributed to the regularity identification and its subsequent extrapolation.In order to reduce the heterogeneities and make the experimental task proper for a brain imaging study,the number magnitude and arithmetic operation involved in number series completion tasks are further restricted.Behavioral performance in Experiment 1 shows the reliable priming effect for targets as expected.Then,a factorial design (the priming effect:prime vs.target;the period length:simple vs.complex) of event-related functional magnetic resonance imaging (fMRI) is used in Experiment 2 to examine the neural basis of data-driven scientific discovery.The fMRI results reveal a double dissociation of the left DLPFC (dorsolateral prefrontal cortex) and the left APFC (anterior prefrontal cortex) between the simple (period length=1) and the complex (period length=2) number series completion task.The priming effect in the left DLPFC is more significant for the simple task than for the complex task,while the priming effect in the left APFC is more significant for the complex task than for the simple task.The reliable double dissociation may suggest the different roles of the left DLPFC and left APFC in data-driven scientific discovery.The left DLPFC (BA 46) may play a crucial role in rule identification,while the left APFC (BA 10) may be related to mental set maintenance needed during rule identification and extrapolation.ZHONG Ning LIANG PeiPeng QIN YuLin LU ShengFu YANG YanHui LI KunCheng 2011Science China(Life Sciences)2011,54,5:8
4Carbon materials for stable Li metal anodes: Challenges, solutions, and outlook显示文摘Lithium(Li)metal is regarded as the ultimate anode for next-generation Li-ion batteries due to its highest specific capacity and lowest electrochemical potential.However,the Li metal anode has limitations,including virtually infinite volume change,nonuniform Li deposition,and an unstable electrode-electrolyte interface,which lead to rapid capacity degradation and poor cycling stability,significantly hindering its practical application.To address these issues,intensive efforts have been devoted toward accommodating and guiding Li deposition as well as stabilizing the interface using various carbon materials,which have demonstrated excellent effectiveness,benefiting from their vast variety and excellent tunability of the structure-property relationship.This review is intended as a guide through the fundamental challenges of Li metal anodes to the corresponding solutions utilizing carbon materials.The specific functionalities and mechanisms of carbon materials for stabilizing Li metal anodes in these solutions are discussed in detail.Apart from the stabilization of the Li metal anode in liquid electrolytes,attention has also been paid to the review of anode-free Li metal batteries and solid-state batteries enabled by strategies based on carbon materials.Furthermore,we have reviewed the unresolved challenges and presented our outlook on the implementation of carbon materials for stabilizing Li metal anodes in practical applications.Qiongqiong Lu Yulin Jie Xiangqi Meng Ahmad Omar Daria Mikhailova Ruiguo Cao Shuhong Jiao Yan Lu Yaolin Xu 2021Carbon Energy2021,3,6:6
5Zanthoxylum-specific whole genome duplication and recent activity of transposable elements in the highly repetitive paleotetraploid Z.bungeanum genome显示文摘Zanthoxylum bungeanum is an important spice and medicinal plant that is unique for its accumulation of abundant secondary metabolites,which create a characteristic aroma and tingling sensation in the mouth.Owing to the high proportion of repetitive sequences,high heterozygosity,and increased chromosome number of Z.bungeanum,the assembly of its chromosomal pseudomolecules is extremely challenging.Here,we present a genome sequence for Z.bungeanum,with a dramatically expanded size of 4.23 Gb,assembled into 68 chromosomes.This genome is approximately tenfold larger than that of its close relative Citrus sinensis.After the divergence of Zanthoxylum and Citrus,the lineage-specific whole-genome duplication event q-WGD approximately 26.8 million years ago(MYA)and the recent transposable element(TE)burst~6.41 MYA account for the substantial genome expansion in Z.bungeanum.The independent Zanthoxylum-specific WGD event was followed by numerous fusion/fission events that shaped the genomic architecture.Integrative genomic and transcriptomic analyses suggested that prominent speciesspecific gene family expansions and changes in gene expression have shaped the biosynthesis of sanshools,terpenoids,and anthocyanins,which contribute to the special flavor and appearance of Z.bungeanum.In summary,the reference genome provides a valuable model for studying the impact of WGDs with recent TE activity on gene gain and loss and genome reconstruction and provides resources to accelerate Zanthoxylum improvement.Shijing Feng Zhenshan Liu Jian Cheng Zihe Li Lu Tian Min Liu Tuxi Yang Yulin Liu Yonghong Liu He Dai Zujun Yang Qing Zhang Gang Wang Jisen Zhang Huifeng Jiang Anzhi Wei 2021Horticulture Research2021,8,1:5

7Simplified full-depth precast concrete deck panel systems for accelerated bridge construction显示文摘A simplified full-depth precast concrete deck panel system for accelerating bridge construction(ABC) is introduced and a finite element analysis(FEA) is conducted to investigate the static and dynamic responses of this conceptual deck system. The FEA results are compared to those of the traditional full-depth precast concrete deck panel system. The comparison results show that the mechanical behavior of the new deck system is different from that of the traditional deck system. The concrete decks in the new system act as two-way slabs, instead of the one-way slab in the traditional system. Meanwhile, the connections in both the longitudinal and transverse directions may need to accommodate the negative moments.Compared to those in the traditional system, the longitudinal nominal stress at middle span increases a lot in the new deck system and the effective flange width varies significantly. 8Effects of urban street trees on human thermal comfort and physiological indices: a case study in Changchun city, China显示文摘Planting trees along urban streets is one of the most important strategies to improve the urban thermal environment.However,the net impacts of urban street trees on human thermal comfort and physiological parameters are still less clear.On three similar east-west orientated streets with different degrees of tree cover-low(13%),medium(35%),and high(75%),urban microclimatic parameters and human physiological indices for six male students were simultaneously measured on three cloudless days in summer 2018.The results show that the differences in tree cover were predominant in influencing urban thermal environment and comfort.The street with the highest tree cover had significantly lower physiological equivalent temperature(PET) and more comfortable than the other two streets.The frequency of strong heat stress(PET> 35℃) was 64%,11 %,and 0%,respectively,for streets with low,medium,and high tree cover.For the six male university students,human physiological indices varied greatly across the three streets with different tree cover.Systolic blood pressure,diastolic blood pressure,and pulse rate increased with decreasing tree cover.The results also suggest that urban thermal environment and comfort had considerable impact on human physiological parameters.Our study provides reasons for urban planners to plant trees along streets to improve the thermal environment and promote urban sustainability.Zhibin Ren Hongbo Zhao Yao Fu Lu Xiao Yulin Dong 2022Journal of Forestry Research2022,33,3:4

10The LTR of endogenous retrovirus ev21 retains promoter activity and exhibits tissue specific transcription in chicken显示文摘Endogenous viruses integrate into the host genome and influence the expression of neighboring genes through their long terminal repeats (LTRs). In this study, we analyzed the promoter, enhancer and transcriptional activities of the chicken endogenous retrovirus ev21 LTR. The LTR was cloned into pGL3-basic and pGL3-promoter luciferase vectors in forward and reverse orientation separately. The luciferase activities were detected respectively in chicken embryonic fibroblast cell, human embryonic kidney cell line, human lung carcinoma cell line, Chinese hamster ovary cell line, and murine melanoma cell line. Relative luciferase activity analysis indicated that the ev21 LTR retained bi-directional promoter activity but no detectable enhancer activity in these cells. The constructs containing the LTR and F1 region show stronger promoter activity than the constructs containing only LTR. The transcriptional pattern of ev21 LTR varied in tissues of late feathering White Leghorn chicken at post-hatch day one. Skin exhibits the highest expression in the tissues examed. 11Clinical Analysis of 240 Patients with HLA-B27 Associated Acute Anterior Uveitis显示文摘Purpose: To analyze the prevalence of HLA-B27 associated acute anterior uveitis and to identify its clinical features. Methods: A total of 240 patients with HLA-B27 associated acute anterior uveitis,who were admitted to Zhejiang Ophthalmologic Hospital between December 2006 and October 2012, were retrospectively analyzed.The age of onset,sex,affected eyes,HLA-B27 antigen detection,recurrence,joint involvement, and surgical complications were investigated. Results:The average age of onset was 37.0 ±12.0 years and the ratio of male to female patients was 2.4:1. Most cases had alternate unilateral or bilateral involvement. Among all participants, 234 cases (97.5%) were HLA-B27 positive, and 124 cases (51.7%) had spondyloarthropathies (SpA), dominated by 108 cases with ankylosing spondylitis. (AS,45.0%),and mostly seen in male subjects(P<0.05).Six patients were HLA-B27 negative(2.5%)and on statistical significance was noted between male and female patients (P >0.05).A total of 193 cases (80.4%) presented with complications, mainly fibrinous exudation, posterior synechia, and vitreous opacity. Conclusion: HLA-B27 that is associated acute anterior uveitis with a relatively high incidence and recurrence presents with more severe clinical features than does idiopathic acute anterior uveitis,and it often accompanies systemic arthritic diseases. 12Gold nanoparticle-directed autophagy intervention for antitumor immunotherapy via inhibiting tumor-associated macrophage M2 polarization显示文摘Tumor-associated macrophages(TAMs),one of the dominating constituents of tumor microenvironment,are important contributors to cancer progression and treatment resistance.Therefore,regulation of TAMs polarization from M2 phenotype towards M1 phenotype has emerged as a new strategy for tumor immunotherapy.Herein,we successfully initiated antitumor immunotherapy by inhibiting TAMs M2 polarization via autophagy intervention with polyethylene glycol-conjugated gold nanoparticles(PEG-Au NPs).PEG-Au NPs suppressed TAMs M2 polarization in both in vitro and in vivo models,elicited antitumor immunotherapy and inhibited subcutaneous tumor growth in mice.As demonstrated by the m RFP-GFP-LC3 assay and analyzing the autophagy-related proteins(LC3,beclin1 and P62),PEGAu NPs induced autophagic flux inhibition in TAMs,which is attributed to the PEG-Au NPs induced lysosome alkalization and membrane permeabilization.Besides,TAMs were prone to polarize towards M2phenotype following autophagy activation,whereas inhibition of autophagic flux could reduce the M2polarization of TAMs.Our results revealed a mechanism underlying PEG-Au NPs induced antitumor immunotherapy,where PEG-Au NPs reduce TAMs M2 polarization via induction of lysosome dysfunction and autophagic flux inhibition.This study elucidated the biological effects of nanomaterials on TAMs polarization and provided insight into harnessing the intrinsic immunomodulation capacity of nanomaterials for effective cancer treatment.Siyue Zhang Fangyuan Xie Kaichun Li He Zhang You Yin Yuan Yu Guangzhao Lu Shihao Zhang Yan Wei Ke Xu Yan Wu Hong Jin Lan Xiao Leilei Bao Can Xu Yulin Li Ying Lu Jie Gao 2022Acta Pharmaceutica Sinica B2022,12,7:2

14Comprehensive metabolic profiling of Alismatis Rhizoma triterpenes in rats based on characteristic ions and a triterpene database显示文摘Alismatis Rhizoma(AR)is widely used in Chinese medicine,and its major bioactive components,triterpenes,reportedly possess various pharmacological activities.Therefore,it is very important to study the metabolism of triterpenes in vivo.However,the metabolism of AR triterpene extract has not been comprehensively elucidated due to its complex chemical components and metabolic pathways.In this study,an ultra-performance liquid chromatography quadrupole time-of-flight mass spectrometry method,which was based on the characteristic ions from an established database of known triterpenes,was used to analyze the major metabolites in rats following the oral administration of Alismatis Rhizoma extracts(ARE).As a result,a total of 233 constituents,with 85 prototype compounds and 148 metabolites,were identified for the first time.Hydrogenation,oxidation,sulfate and glucuronidation conjugation were the major metabolic pathways for triterpenes in AR.In addition,the mutual in vivo transformation of known ARE triterpenes was discovered and confirmed for the first time.Those results provide comprehensive insights into the metabolism of AR in vivo,which will be useful for future studies on its pharmacodynamics and pharmacokinetics.Moreover,this established strategy may be useful in metabolic studies of similar compounds.Lu Wang Sen Li Jiaxin Li Zhongzhe Cheng Yulin Feng Hui Ouyang Zhifeng Du Hongliang Jiang 2021Journal of Pharmaceutical Analysis2021,11,1:2

18Development of a mobile application for identification of grapevine(Vitis vinifera L.)cultivars via deep learning显示文摘Traditional vine variety identification methods usually rely on the sampling of vine leaves followed by physical,physiological,biochemical and molecular measurement,which are destructive,time-consuming,labor-intensive and require experienced grape phenotype analysts.To mitigate these problems,this study aimed to develop an application(App)running on Android client to identify the wine grape automatically and in real-time,which can help the growers to quickly obtain the variety information.Experimental results showed that all Convolutional Neural Network(CNN)classification algorithms could achieve an accuracy of over 94%for twenty-one categories on validation data,which proves the feasibility of using transfer deep learning to identify grape species in field environments.In particular,the classification model with the highest average accuracy was GoogLeNet(99.91%)with a learning rate of 0.001,mini-batch size of 32,and maximum number of epochs in 80.Testing results of the App on Android devices also confirmed these results.Yixue Liu Jinya Su Lei Shen Nan Lu Yulin Fang Fei Liu Yuyang Song Baofeng Su 2021International Journal of Agricultural and Biological Engineering2021,14,5:1
