| 1 | Corrosion and biocompatibility improvement of magnesium-based alloys as bone implant materials:a review显示文摘Magnesium(Mg)or its alloys are widely tested as potential orthopedic implants,particularly as biodegradable alloys for fracture fixation due to their mechanical properties are close to those of bone.Currently,available Mg or its alloys are confronted with challenges in passing regulatory biosafety tests prior to clinical trials due to its fast degradation and associated degradation products.The degradation of Mg is accompanied by the release of Mg ions,the rise of pH and osmolality in surrounding environments.According to the standard of ISO 10993 Part 13,the pH value shall be appropriate to the site of intended use maintaining in an appropriate range.Approaches to overcome these challenges include the selection of adequate alloying elements,proper surface treatment techniques and control of the degradation rate of Mg or its alloys developed as orthopedic implants.To date,Mg or its alloy-based bone implants have not yet been widely used in clinical applications as medical implants.This review critically summarized published methods to improve the corrosion resistance of Mg and its alloys.The current progress on in vitro cytotoxicity and in vivo biocompatibility properties of these metals was also reviewed.This review aimed to provide a reference for further research and development(R&D)of biodegradable Mg and its alloys with regard to the evaluation of their corrosion process and biocompatibility and facilitation of their translation to clinical applications. | Long Li Ming Zhang Ye Li Jie Zhao Ling Qin Yuxiao Lai | 2017 | Regenerative Biomaterials2017,4,2: | 9 |
| 2 | Porphyromonas gingivalis infection promotes mitochondrial dysfunction through Drp1-dependent mitochondrial fission in endothelial cells显示文摘Porphyromonas gingivalis(P.gingivalis),a key pathogen in periodontitis,has been shown to accelerate the progression of atherosclerosis(AS).However,the definite mechanisms remain elusive.Emerging evidence supports an association between mitochondrial dysfunction and AS.In our study,the impact of P.gingivalis on mitochondrial dysfunction and the potential mechanism were investigated.The mitochondrial morphology of EA.hy926 cells infected with P.gingivalis was assessed by transmission electron microscopy,mitochondrial staining,and quantitative analysis of the mitochondrial network.Fluorescence staining and flow cytometry analysis were performed to determine mitochondrial reactive oxygen species(mtROS)and mitochondrial membrane potential(MMP)levels.Cellular ATP production was examined by a luminescence assay kit.The expression of key fusion and fission proteins was evaluated by western blot and immunofluorescence.Mdivi-1,a specific Drp1 inhibitor,was used to elucidate the role of Drp1 in mitochondrial dysfunction.Our findings showed that P.gingivalis infection induced mitochondrial fragmentation,increased the mtROS levels,and decreased the MMP and ATP concentration in vascular endothelial cells.We observed upregulation of Drp1(Ser616)phosphorylation and translocation of Drp1 to mitochondria.Mdivi-1 blocked the mitochondrial fragmentation and dysfunction induced by P.gingivalis.Collectively,these results revealed that P.gingivalis infection promoted mitochondrial fragmentation and dysfunction,which was dependent on Drp1.Mitochondrial dysfunction may represent the mechanism by which P.gingivalis exacerbates atherosclerotic lesions. | Tong Xu Qin Dong Yuxiao Luo Yanqing Liu Liang Gao Yaping Pan Dongmei Zhang | 2021 | International Journal of Oral Science2021,13,3: | 7 |

| 4 | Tofu-inspired microcarriers from droplet microfluidics for drug delivery显示文摘Microcarriers have attracted increasing interests in drug delivery. In order to develop this technique, it is prone to focus on the generation of functional particles through using simple approaches and novel but accessible materials. Here, inspired by the formation mechanism of tofu that through the mixing of soymilk and brine for cross-linking soybean proteins, we present novel soybean protein microcarriers by using microfluidic generation approach for drug delivery. Since the soybean protein droplets are generated by microfluidic emulsification method, the tofu microparticles present highly monodisperse and homogeneous morphologies. Because of the excellent biocompatibility of the soybean protein and the interconnected porous structures throughout the whole microparticles after freeze-drying, various kinds of drugs and active molecules could be absorbed and loaded in the microcarriers, which makes them versatile for drug delivery. | 5 | Network pharmacological prediction and molecular docking analysis of the combination of Atractylodes macrocephala Koidz.and Paeonia lactiflora Pall.in the treatment of functional constipation and its verification显示文摘Background:We aimed to reveal the mechanism of functional constipation in the treatment of Atractylodes macrocephala Koidz.(AMK)and Paeonia lactiflora Pall.(PLP).Methods:The main active ingredients of AMK and PLP were screened by the Traditional Chinese Medicine Systems Pharmacology(TCMSP)platform.A database of functional constipation targets was established by GeneCard and OMIM.An“ingredient-target”network map was constructed with Cytoscape software(version 3.7.1),and molecular docking analysis was performed on the components and genes with the highest scores.The rats in the normal group were given saline,and those in the other groups were given 10 mg/kg diphenoxylate once a day for 14 days.The serum and intestinal tissue levels of adenosine monophosphate(cAMP),protein kinase A(PKA),and adenylyl cyclase(AC)of the rats and aquaporin(AQP)1,AQP3,and AQP8 were measured.Results:AMK and PLP had a significant role in the regulation of targets in the treatment of functional constipation.After treatment with AMK,PLP,or mosapride,the serum and intestinal tissue levels of AC,cAMP,and PKA were significantly downregulated.Groups receiving AMK and PLP or mosapride exhibited a reduction in the level of AQP1,AQP3,and AQP8 to varying degrees.Conclusion:Molecular docking analysis revealed that AMK and PLP had a significant role in the regulation of targets in the treatment of functional constipation.Studies have confirmed that AMK and PLP can also affect AC,cAMP,and PKA.AC,cAMP,and PKA in model rats were significantly downregulated.AQP expression is closely related to AC,cAMP,and PKA.AMK and PLP can reduce the expression of AQP1,AQP3,and AQP9 in the colon of constipated rats. | Yuxiao Meng Xiaojun Li Xiaoting Wang Lu Zhang Jiaqi Guan | 2022 | Animal Models and Experimental Medicine2022,5,2: | 4 |

| 8 | Evaluation of coronary plaque and stent deployment by intravascular optical coherence tomography in elderly patients with unstable angina and non-ST-elevation myocardial infarction显示文摘Objective To evaluate the feasibility and efficacy of intravascular optical coherence tomography (OCT) in the assessment of plaque characteristics and drug eluting stent deployment quality in the elderly patients with unstable angina (UA) and non-ST segment elevation myocardial infarction (NSTEMI). Methods OCT was used in elderly patients undergoing percutaneous coronary interventions. Fifteen patients, 9 males and 6 females with mean age of 72.6±5.3 years (range 67-92 years) were enrolled in the study. Images were obtained before initial balloon dilatation and following stent deployment. The plaque characteristics before dilation, vessel dissection, tissue prolapse, stent apposition and strut distribution after stent implantation were evaluated. Results Fifteen lesions were selected from 32 angiographic lesions as study lesions for OCT imaging after diagnostic coronary angiography. There were 7 lesions in the left anterior descending artery, 5 lesions in the right coronary artery and 3 lesions in the left circumflex coronary artery. Among them, 12 (80.0%) were lipid-rich plaques, and 10 (66.7%) were vulnerable plaques with fibrous cap thickness 54.2±7.3 μm. Seven ruptured culprit plaques (46.7%) were found; 4 in UA patients and 3 in NSTEMI patients. Tissue prolapse was observed in 11 lesions (73.3%). Irregular stent strut distribution was detected in 8 lesions (53.3%). Vessel dissections were found in 5 lesions (33.3%). Incomplete stent apposition was observed in 3 stents (20%) with mean spacing between the struts and the vessel wall 172±96 mm (range 117-436 mm). | 11 | Identification and Characterization of Peptides Mimicking the Epitopes of Metalloprotease of Schistosoma Japonicum显示文摘In an attempt to isolate and characterize peptides mimicking epitopes of metalloprotease and explore their immunological protection against Schistosoma japonicum (S. japonicum), polyclonal anti-metalloprotease sera was prepared to screen a 12-mer random peptide library to isolate phages binding specially to antisera IgG. Then,phage ELISA, animal immunization, DNA sequencing, Western blotting and enzymatic activity neutralizing analysis were used to characterize the selected phage clones. All of ten randomly picked clones were shown to be positive. Five peptides of different amino acid sequences deduced from DNA sequences were obtained and two of them (peptides 2 and 3) could induce significant reduction (31.0% and 31.8%, respectively) in worm burden and high reduction (52.6% and 54.9%, respectively) in liver eggs per gram (LEPG), while, unexpectedly, others (peptides 1, 4 and 5) could not elicit enough protection against infection of S. japonicum. Peptides 2 and 3 could be recognized by S. japonicum infected mouse sera (IMS) and could elicit neutralizing Abs. The results show that peptides 2 and 3 are antigenic and immunogenic. | 15 | Six-month results of a biodegradable polymer and rapamycin-coating stent for coronary artery disease显示文摘Objective To assess the safety and efficacy of a novel biodegradable polymer and rapamycin-coating stent, the EXCEL stent, in the treatment of coronary artery disease (CAD), as compared with the CypherTM stent. Methods In this prospective, non-randomized study, 60 consecutive patients with symptomatic CAD received either an EXCEL stent (n=32), or a CypherTM stent(n=28),according to their respective treatment intention. Follow-up angiography was performed at a mean of 180±40 days. The primary endpoint of the study was the occurrence of a major adverse cardiac event (MACE), including death, myocardial infarction, or target-vessel revascularization during the 6 months after stenting. The secondary end points included the in-stent late luminal loss (LLL), percentage of in-stent stenosis of the luminal diameter, and the rate of restenosis (luminal narrowing of 50 percent or more) at 6 months. Results There were no significant differences between the two groups in baseline characteristics, including the distribution of target vessel and lesion types. During the follow up period of 6 months, there were no occurrences of MACE in either group. Twenty-seven patients(84%) in the EXCEL group and 10 (36 %) in the CypherTM group underwent quantitative coronary angiography at 6 months. For these patients, no restenosis occurred, and there were no differences in the in-stent stenosis of the luminal diameter (5.98±5.52% vs 5.21 ±6.3%,P>0.05) and the LLL (-0.02±0.09 mm vs -0.01±0.07 mm, P>0.05). | 17 | C-reactive protein as a predictor for cardiac events in Chinese elderly patients with coronary heart disease显示文摘Background and objective To assess the predictive value of C-reactive protein(CRP) for major adverse cardiac events and the association between CRP level and the coronary lesion morphology and extent in patients with coronary heart disease (CHD).Methods CRP was measured on admission in 177 consecutive elderly (age≥60 years) patients with CHD who underwent coronary angiography. Patients were divided into high CRP group (CRP≥3mg/L) and normal CRP group (CRP <3mg/L). The association between CRP levels and the coronary lesion features, including severity of stenosis (mild, moderate, severe), extent of lesion (diffused or nondiffused), eccentricity of the plaque (eccentric or non-eccentric) were analyzed. Patients were followed up for a mean of 8 months for the occurrences of major adverse cardiac events (MACE). Results Compared with patients in normal CRP group, patients in high CRP group were more frequently to have unstable angina, multi-vessel, diffuse, eccentric lesions, positive remodeling, and non-smooth plaques (P<0.01). Kaplan-Meier analysis showed patients in high CRP group had a significantly lower MACE-free survival rate than patients in normal CRP group (Log-rank = 12.0, P<0.01); Cox regression analysis indicated CRP level as an independent predictor for the occurrence of MACE (OR=3.16, P<0.05) Conclusions High CRP level is associated with more extend, severe and eccentric coronary lesions and is an independent predictor for MACE in elderly patients with CHD. | Guangyong HUANG Caiyi LU Xingli WU Yuxiao ZHANG | 2006 | Journal of Geriatric Cardiology2006,3,2: | 1 |
| 18 | A Vaccine Based on the Receptor-Binding Domain of the Spike Protein Expressed in Glycoengineered Pichia pastoris Targeting SARS-CoV-2 Stimulates Neutralizing and Protective Antibody Responses显示文摘In 2020 and 2021,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),a novel coronavirus,caused a global pandemic.Vaccines are expected to reduce the pressure of prevention and control,and have become the most effective strategy to solve the pandemic crisis.SARS-CoV-2 infects the host by binding to the cellular receptor angiotensin converting enzyme 2(ACE2)via the receptor-binding domain(RBD)of the surface spike(S)glycoprotein.In this study,a candidate vaccine based on a RBD recombinant subunit was prepared by means of a novel glycoengineered yeast Pichia pastoris expression system with characteristics of glycosylation modification similar to those of mammalian cells.The candidate vaccine effectively stimulated mice to produce high-titer anti-RBD specific antibody.Furthermore,the specific antibody titer and virus-neutralizing antibody(NAb)titer induced by the vaccine were increased significantly by the combination of the double adjuvants Al(OH)_(3) and CpG.Our results showed that the virus-NAb lasted for more than six months in mice.To summarize,we have obtained a SARS-CoV-2 vaccine based on the RBD of the S glycoprotein expressed in glycoengineered Pichia pastoris,which stimulates neutralizing and protective antibody responses.A technical route for fucose-free complex-type N-glycosylation modified recombinant subunit vaccine preparation has been established. | Bo Liu Ying Yin Yuxiao Liu Tiantian Wang Peng Sun Yangqin Ou Xin Gong Xuchen Hou Jun Zhang Hongguang Ren Shiqiang Luo Qian Ke Yongming Yao Junjie Xu Jun Wu | 2022 | Engineering2022,8,6: | 1 |
