| 1 | Transplantation of collagen scaffold with autologous bone marrow mononuclear cells promotes functional endometrium reconstruction via downregulating ΔNp63 expression in Asherman's syndrome显示文摘Asherman's syndrome(AS) is a common disease that presents endometrial regeneration disorder. However, little is known about its molecular features of this aregenerative endometrium in AS and how to reconstruct the functioning endometrium for the patients with AS. Here, we report that ΔNp63 is significantly upregulated in residual epithelial cells of the impaired endometrium in AS; the upregulated-ΔNp63 induces endometrial quiescence and alteration of stemness. Importantly, we demonstrate that engrafting high density of autologous bone marrow mononuclear cells(BMNCs) loaded in collagen scaffold onto the uterine lining of patients with AS downregulates ΔNp63 expression, reverses ΔNp63-induced pathological changes, normalizes the stemness alterations and restores endometrial regeneration. Finally, five patients achieved successful pregnancies and live births. Therefore, we conclude that ΔNp63 is a crucial therapeutic target for AS. | 3 | Development of a Haploid-Inducer Mediated Genome Editing System for Accelerating Maize Breeding显示文摘Crop breeding aims to generate pure in bred lines with multiple desired traits. Doubled haploid (DH) and genome editing using CRISPR/Cas9 are two powerful game-changing technologies in crop breeding. However, both of them still fall short for rapid generation of pure elite lines with integrated favorable traits. Here, we report the development of a Haploid-Inducer Mediated Genome Editing (IMGE) approach, which utilizes a maize haploid inducer line carrying a CRISPR/Cas9 cassette targeting for a desired agronomic trait to pollinate an elite maize in bred line and to generate genome-edited haploids in the elite maize background. | 5 | Does CMIP6 Inspire More Confidence in Simulating Climate Extremes over China?显示文摘Based on climate extreme indices calculated from a high-resolution daily observational dataset in China during 1961–2005,the performance of 12 climate models from phase 6 of the Coupled Model Intercomparison Project(CMIP6),and 30 models from phase 5 of CMIP(CMIP5),are assessed in terms of spatial distribution and interannual variability.The CMIP6 multi-model ensemble mean(CMIP6-MME)can simulate well the spatial pattern of annual mean temperature,maximum daily maximum temperature,and minimum daily minimum temperature.However,CMIP6-MME has difficulties in reproducing cold nights and warm days,and has large cold biases over the Tibetan Plateau.Its performance in simulating extreme precipitation indices is generally lower than in simulating temperature indices.Compared to CMIP5,CMIP6 models show improvements in the simulation of climate indices over China.This is particularly true for precipitation indices for both the climatological pattern and the interannual variation,except for the consecutive dry days.The arealmean bias for total precipitation has been reduced from 127%(CMIP5-MME)to 79%(CMIP6-MME).The most striking feature is that the dry biases in southern China,very persistent and general in CMIP5-MME,are largely reduced in CMIP6-MME.Stronger ascent together with more abundant moisture can explain this reduction in dry biases.Wet biases for total precipitation,heavy precipitation,and precipitation intensity in the eastern Tibetan Plateau are still present in CMIP6-MME,but smaller,compared to CMIP5-MME. | Huanhuan ZHU Zhihong JIANG Juan LI Wei LI Cenxiao SUN and Laurent LI | 2020 | Advances in Atmospheric Sciences2020,37,10: | 27 |
| 6 | Changes in precipitation and extreme precipitation in a warming environment in China显示文摘This study analyses the decadal changes in winter precipitation and extreme precipitation in a warming environment in China. The results show that, together with a trend of winter warming in China, winter precipitation and extreme precipitation in the region are also increasing. In addition, concurrent with the decadal warming shift that occurred in the mid-1980s, precipitation and extreme precipitation both increased significantly. Quantitative analysis shows that precipitation and extreme precipitation increased at rates of 9.7% and 22.6% per 1℃ of surface warming in China. This rate of precipitation increase is greater than the global mean, which indicates that precipitation in China is highly sensitive to climate warming and further highlights the importance of studying regional responses to climate warming. | 10 | Achieving nitritation at low temperatures using free ammonia inhibition on Nitrobacter and real-time control in an SBR treating landfill leachate显示文摘Free ammonia(FA) inhibition on nitrite-oxidized bacteria(NOB) and real-time control are used to achieve nitrogen removal from landfill leachate via nitrite pathway at low temperatures in sequencing batch reactor. The inhibition of FA on NOB activity during the aerobic period was prolonged using real-time control. The degree of nitrite accumulation was monitored along with variations of the ammonia-oxidizing bacteria and NOB population using fluorescence in situ hybridization techniques. It is demonstrated that the end-point of ammonia oxidization is detected from the on-line measured dissolved oxygen,oxidization–reduction potential, and p H signals, which could avoid the loss the FA inhibition on NOB caused by excess aeration. At low temperature(13.0–17.6°C), the level of nitrite pathway rapidly increased from 19.8% to 90%, suggesting that nitritation was successfully started up at low temperature by applying syntrophic association of the FA inhibition and real-time control, and then this high level of nitrite pathway was stably maintained for as long as 233 days. Mechanism analysis shows that the establishment of nitritation was primarily the result of predominant ammonia-oxidizing bacteria developed in the nitrifying bacteria population compared to NOB. | 12 | Intercropping with Solanum nigrum and Solanum photeinocarpum from Two Ecoclimatic Regions Promotes Growth and Reduces Cadmium Uptake of Eggplant Seedlings显示文摘Soil contamination by heavy metals is a serious environmental problem worldwide, and reduction of heavy metal accumulation in vegetables grown on contaminated land is a matter of urgency. A pot experiment was conducted to study the effects of intercropping with the Cd hyperaccumulators Solanum nigrum and Solanum photeinocarpum from two ecoclimatic regions, Ya'an and Chengdu,Sichuan Province, China, on the growth and cadmium(Cd) uptake of eggplant(Solanum melongena L.). The biomass, photosynthetic pigment contents, and activities of antioxidant enzymes of eggplant were enhanced by intercropping. The biomass of eggplant was the highest after intercropping with S. photeinocarpum from Ya'an, but did not differ significantly from that after intercropping with S. nigrum from Chengdu. The shoot Cd content of eggplant was significantly reduced by intercropping with the hyperaccumulators,which ranked as follows: S. nigrum from Chengdu > S. nigrum from Ya'an > S. photeinocarpum from Chengdu > S. photeinocarpum from Ya'an, with the decreases being 19.60%, 14.36%, 9.66%, and 6.42%, respectively, as compared with the control. The lowest shoot Cd content and translocation factor of eggplant were attained after intercropping with S. nigrum from Chengdu. | 14 | Antioxidant and Anti-inflammatory Capacity of Ferulic Acid Released from Wheat Bran by Solid-state Fermentation of Aspergillus niger显示文摘Objective A strain of Aspergillus niger(A. niger), capable of releasing bound phenolic acids from wheat bran, was isolated. This strain was identified by gene sequence identification. The antioxidant and anti-inflammatory capacity of ferulic acid released from wheat bran by this A. niger strain(FA-WB) were evaluated. Methods Molecular identification techniques based on PCR analysis of specific genomic sequences were conducted; antioxidant ability was examined using oxygen radical absorbance capacity(ORAC), cellular antioxidant activity(CAA) assays, and erythrocyte hemolysis assays. RAW264.7 cells were used as a model to detect anti-inflammatory activity. Results The filamentous fungal isolate was identified to be A. niger. ORAC and CAA assay showed that FA-WB had better antioxidant activity than that of the ferulic acid standard. The erythrocyte hemolysis assay results suggested that FA-WB could attenuate AAPH-induced oxidative stress through inhibition of reactive oxy gen species(ROS) generation. FA-WB could significantly restore the AAPH-induced increase in intracellular antioxidant enzyme activities to normal levels as well as inhibit the intracellular malondialdehyde formation. TNF-?, IL-6, and NO levels indicated that FA-WB can inhibit the inflammation induced by lipopolysaccharide(LPS). | 16 | Aloe-emodin exerts cholesterol-lowering effects by inhibiting proprotein convertase subtilisin/kexin type 9 in hyperlipidemic rats显示文摘Hyperlipidemia(HPL)characterized by metabolic disorder of lipids and cholesterol is one of the important risk factors for cardiovascular diseases.Proprotein convertase subtilsin/kexin type 9(PCSK9)is a potent circulating regulator of LDL through its ability to induce degradation of the low-density lipoprotein cholesterol receptor(LDLR)in the lysosome of hepatocytes.Aloe-emodin(AE)is one of potentially bioactive components of Chinese traditionalmedicine Daming capsule.In this study we evaluated the HPL-lowering efficacy of AE in both in vivo and in vitro HPL models.High-fat diet-induced rats were treated with AE(100 mg/kg per day,ig)for 6 weeks.We found that AE administration significantly decreased the levels of total cholesterol(TC)and LDL in the serum and liver tissues.Moreover,AE administration ameliorated HPL-induced hepaticlipid aggregation.But AE administration did not significantly inhibit HMG-CoA reductase activity in the liver of HPL rats.A cellular model of HPL was established in human hepatoma(HepG2)cells treated with cholesterol(20 μg/mL)and 25-hydroxycholesterol(2 μg/mL),which exhibited markedly elevated cholesterol levels.The increased cholesterol levels could be reversed by subsequent treatment with AE(30 μM).In both the in vivo and in vitro HPL models,we revealed that AE selectively suppressed the stero-regulatory element-binding protein-2(SREBP-2)and hepatocyte nuclear factor(HNF)1α-mediated PCSK9 signaling,which in turn upregulated LDL receptor(LDLR)and promoted LDL uptake.This study demonstrates that AE reduces cholesterol content in HPL rats by inhibiting the hepatic PCSK9/LDLR pathway. | Zhen-li Su Peng-zhou Hang Juan Hu Yu-yang Zheng Han-qi Sun Jing Guo Ke-yu Liu Zhi-min Du | 2020 | Acta Pharmacologica Sinica2020,41,8: | 14 |
