2A Teosinte-derived Allele of a MYB Transcription Repressor Confers Multiple Disease Resistance in Maize.显示文摘Natural alleles controlling multiple disease resistances (MDR) are valuable for crop breeding. However, only one MDR gene have been cloned in maize, and molecular mechanisms of MDR are not clear. By map-based cloning, we have cloned a teosinte-derived allele of a resistance gene, Mexicana lesion mimic 1 (ZmMM1), which has a lesion mimic phenotype and confers resistance to northern leaf blight (NLB), gray leaf spot (GLS) and southern corn rust (SCR). Strong MDR conferred by the teosinte allele is linked with the polymorphisms in the 3' untranslated region of the ZmMM1 gene that cause increased accumulation of ZmMM1 protein. ZmMM1 acts as a transcription repressor and negatively regulates transcription of specific target genes including ZmMM1-target gene 3 (ZmMT3), which functions as a negative regulator of plant immunity and associated cell death. 3A Hg(Ⅱ)-specific probe for imaging application in living systems and quantitative analysis in environmental/food samples显示文摘Mercury ions are highly toxic and can accumulate along food chains in water,soil,crops and animals.Effective detection of mercury ions in various media is of great significance for maintaining the ecological environment and protecting people's health.In this work,a mercury ions specific fluorescent probe was developed by a simple one-step reaction of commercial substrates of 4-chloro-7-nitro-2,1,3-benzoxadiazole and 1-(2-aminoethyl)-4-methylpiperazine.Investigation on sensing behavior showed that this probe had high sensitivity and selectivity towards mercury ions.Furthermore,this probe could be used as a tool to track the level of mercury ions in living system.In living cells,the probe with green emission emitted a bright red fluorescence when it was bound to mercury ions.In Arabidopsis thaliana,similar red emission could be detected from the root tip and stalk when A.thaliana was grown in culture medium containing mercury ions.The imaging in zebrafish showed that mercury ions were mainly concentrated in the stomach and head of zebrafish.Especially,this probe could be applied in quantitative analysis of mercury ions in tap water,green tea,sea shrimp and soil.This work provided a practical tool for the detection of mercury ions in living systems and quantitative analysis in real samples.Guangjin Li Jiali Wang Dongyang Li Shenghua Liu Jun Yin Zhibing Lai Guangfu Yang 2021Chinese Chemical Letters2021,32,4:2

7Significant histological changes are not rare in indeterminate‐phase chronic hepatitis B patients with hepatitis B e antigen‐negative and normal alanine aminotransferase levels显示文摘Background&Objectives:The degree of liver injury in indeterminate chronic hepatitis B(CHB)infection patients with Hepatitis B e antigen(HBeAg)‐negative and persistently normal alanine aminotransferase(PNALT)levels is yet unclear.Therefore,we aimed to assess liver histological changes in such patients by liver biopsy and explore possible predictors.Methods:Overall,711 HBeAg‐negative CHB patients with PNALT levels who underwent liver biopsy from January 2017 to June 2022 were included in this retrospective study.The relationships between histological changes and predictors were assessed by smooth curve fitting and multivariate logistic regression analysis models.Data were also analyzed using American Association for the Study of Liver Disease(AASLD)modified alanine aminotransferase(ALT)criteria.Results:The proportion of significant histological changes in the indeterminate phase was higher than that in the inactive phase(53.97%vs.41.33%).The adjusted odds ratios(aORs)of significant necroinflammation and histological changes for the indeterminate phase were 2.05 and 1.43,respectively,when compared with the inactive phase by multivariate logistic regression analyses.Significant histological changes in the‐phase were positively associated with age,ALT,Aspartate aminotransferase(AST),Hepatitis B surface antigen(HBsAg),and Hepatitis B virus(HBV)DNA levels but negatively correlated with platelet(PLT)levels.HBV DNA≥5 log10 U/L and PLT<200�109/L were independent predictive factors for assessing histological changes in indeterminate‐phase patients.Similar analysis findings were obtained using the two sets of modified ALT criteria.Conclusions:Significant histological changes are not rare in indeterminatephase CHB patients and are higher than in the inactive phase,regardless of the ALT criteria.Histological investigation is strongly suggested for intermediate stage patients with HBV DNA>5 log10 U/L or PLT<200�109/L and antiviral therapy should be considered for such patients.Deliang Huang Guangde Zhou Jinghan Peng Qinxian Cai Guojun Li Hong Yu Zhibing Zhu Yuanyuan Chen Huiyi Lai Jinyan Jiang Hong Yu Jun Chen 2023iLABMED2023,1,3:0
8High-quality assembled and annotated genomes of Nicotiana tabacum and Nicotiana benthamiana reveal chromosome evolution and changes in defense arsenals显示文摘Nicotiana tabacum and Nicotiana benthamiana are widely used models in plant biology research.However,genomic studies of these species have lagged.Here we report the chromosome-level reference genome assemblies for N.benthamiana and N.tabacum with an estimated 99.5%and 99.8%completeness,respec-tively.Sensitive transcription start and termination site sequencing methods were developed and used for accurate gene annotation in N.tabacum.Comparative analyses revealed evidence for the parental origins and chromosome structural changes,leading to hybrid genome formation of each species.Interestingly,theantiviral silencinggenesRDR1,RDR6,DCL2,DCL3,andAGO2were lost from one or both subgenomes in N.benthamiana,while both homeologs were kept in N.tabacum.Furthermore,the N.benthamiana genome encodes fewer immune receptors and signaling components than that of N.tabacum.These find-ings uncover possible reasons underlying the hypersusceptible nature of N.benthamiana.We developed the user-friendly Nicomics(http:/lifenglab.hzau.edu.cn/Nicomics/)web server to facilitate better use of Nicotiana genomic resources as well as gene structure and expression analyses.Jubin Wang Qingling Zhang Jeffrey Tung Xi Zhang Dan Liu Yingtian Deng Zhendong Tian Huilan Chen Taotao Wang Weixiao Yin Bo Lij Zhibing Lai Savithramma P.Dinesh-Kumar Barbara Baker Feng Li 2024Molecular Plant2024,17,3:0
