| 1 | ^11C-PIBPET和^18F-FDGPET显像诊断阿尔茨海默病与遗忘型轻度认知损害的临床价值显示文摘目的应用11C-PIB PET和18F-FDG PET显像研究阿尔茨海默病和遗忘型轻度认知损害患者β-淀粉样蛋白(Aβ)沉积与葡萄糖代谢之间的关系,联合载脂蛋白E(ApoE)基因型进一步探讨遗忘型轻度认知损害与阿尔茨海默病的相关性。方法利用PET显像对阿尔茨海默病(14例)、遗忘型轻度认知损害(10例)和正常对照者(5例)脑组织Aβ沉积和葡萄糖代谢变化进行分析,采用聚合酶链反应-限制性片段长度多态性方法对ApoE基因型进行分析。结果阿尔茨海默病组患者11C-PIB标准化摄取比值在下顶叶、颞叶外侧、额叶、后扣带回皮质和楔前叶、枕叶和纹状体均高于正常对照组(P<0.05);遗忘型轻度认知损害组患者脑组织11C-PIB结合水平呈双峰形。11C-PIB+aMCI亚组与阿尔茨海默病组、11C-PIB-aMCI亚组与正常对照组之间11C-PIB标准化摄取比值差异均无统计学意义(P>0.05)。18F-FDG PET显像显示,3/5例11C-PIB+aMCI亚组患者双侧顶叶、颞叶和楔前叶代谢减低,其中2例ApoEε4等位基因携带者随访期间进展至阿尔茨海默病;3/5例11C-PIB-aMCI亚组患者双侧额叶和前扣带回代谢减低。结论11C-PIB PET显像是筛查具有阿尔茨海默病病理特点的遗忘型轻度认知损害患者的有效工具。具有阿尔茨海默病病理特征的遗忘型轻度认知损害患者可伴有顶叶、颞叶外侧皮质和楔前叶代谢减低,其中ApoEε4等位基因携带者更易进展至阿尔茨海默病痴呆。 | Shi Zhihong Wang Ying Liu Shuai Liu Shuling Z hou Yuying Wang Jinhuan Cai Li Gao Shuo Ji Yong | 2014 | 中国现代神经疾病杂志2014,14,3: | 8 |
| 2 | The earthquake precursor detected in a granular medium and a proposed model for the propagation of precursive stress-strain signal显示文摘A way to detect the seismic precursor in granular medium is described and a model of propagation for precursive stress-strain signals is proposed.A strain sensor buried in a sandpit is used to measure a seismic precursor signal.The signal has been investigated and confirmed to originate from a specific earthquake.A comparison of simulated and experimental signals indicates that the signal results from the strain in the earth's strata.Based on the behavioral characteristics of granular materials,an analysis of why this method can be so sensitive to the seismic strain signal is undertaken and a model for the propagation of this stress-strain signal is proposed.The Earth's lithosphere is formed of tectonic plates,faults and fault gouges at their boundaries.In the case of the quasi-static mechanics of seismic precursory stress-strain propagation,the crustal lithosphere should be treated as a large-scale granular system.During a seismogenic event,accumulated force generates the stick-slip motion of adjacent tectonic plates and incrementally pushes blocks farther apart through stick-slip shift.The shear force released through this plate displacement causes soil compression deformation.The discrete properties of the sand in the sandpit lead to the sensitive response of the sensor to the deformation signal which enables it to detect the seismic precursor.From the analysis of the mechanism of the stress-strain propagation in the lithosphere,an explanation is found for the lack of signal detection by sensors installed in rocks.The principles and method presented in this paper provide a new technique for investigating seismic precursors to shallow-source earthquakes. | LU KunQuan HOU MeiYing WANG Qiang PENG Zheng SUN Wei SUN XiaoMing WANG YuYing TONG XiaoHui JIANG ZeHui LIU JiXing | 2011 | Chinese Science Bulletin2011,56,11: | 2 |

| 4 | Genome-wide association study of 23 f lowering phenology traits and 4 f loral agronomic traits in tree peony (Paeonia section Moutan DC.) reveals five genes known to regulate f lowering time显示文摘Tree peony is a unique traditional f lower in China,with large,fragrant,and colorful f lowers.However,a relatively short and concentrated f lowering period limits the applications and production of tree peony.A genome-wide association study(GWAS)was conducted to accelerate molecular breeding for the improvement of f lowering phenology traits and ornamental phenotypes in tree peony.A diverse panel of 451 tree peony accessions was phenotyped for 23 f lowering phenology traits and 4 f loral agronomic traits over 3 years.Genotyping by sequencing(GBS)was used to obtain a large number of genome-wide single-nucleotide polymorphisms(SNPs)(107050)for the panel genotypes,and 1047 candidate genes were identified by association mapping.Eighty-two related genes were observed during at least 2 years for f lowering,and seven SNPs repeatedly identified for multiple f lowering phenology traits over multiple years were highly significantly associated with five genes known to regulate f lowering time.We validated the temporal expression profiles of these candidate genes and highlighted their possible roles in the regulation of f lower bud differentiation and f lowering time in tree peony.This study shows that GWAS based on GBS can be used to identify the genetic determinants of complex traits in tree peony.The results expand our understanding of f lowering time control in perennial woody plants.Identification of markers closely related to these f lowering phenology traits can be used in tree peony breeding programs for important agronomic traits. | Yuying Li Lili Guo Zhanying Wang Dehui Zhao Dalong Guo John E.Carlson Weilun Yin Xiaogai Hou | 2023 | Horticulture Research2023,10,2: | 1 |
| 5 | Inducible CRISPRa screen identifies putative enhancers显示文摘Enhancers are critical cis-regulatory elements that regulate spatiotemporal gene expression and control cell fates.However,the identification of enhancers in native cellular contexts still remains a challenge.Here,we develop an inducible CRISPR activation(CRISPRa)system by transgenic expression of doxycycline(Dox)-inducible dCas9-VPR in mouse embryonic stem cells(iVPR ESC).With this line,a simple introduction of specific guide RNAs targeting promoters or enhancers allows us to realize the effect of CRISPRa in an inducible,reversible,and Dox concentration-dependent manner.Taking advantage of this system,we induce tiled CRISPRa across genomic regions(105 kilobases)surrounding T(Brachyury),one of the key mesodermal development regulator genes.Moreover,we identify several CRISPRa-responsive elements with chromatin features of putative enhancers,including a region the homologous sequence in which humans harbors a body height risk variant.Genetic deletion of this region in ESC does affect subsequent T gene activation and osteogenic differentiation.Therefore,our inducible CRISPRa ESC line provides a convenient platform for high-throughput screens of putative enhancers. | Zhongye Dai Rui Li Yuying Hou Qian Li Ke Zhao Ting Li Mulin Jun Li Xudong Wu | 2021 | Journal of Genetics and Genomics2021,48,10: | 1 |
| 6 | Human umbilical cord mesenchymal stem cell transfusion in immune non-responders with AIDS:a multicenter randomized controlled trial显示文摘We examined the safety and efficacy of human umbilical cord mesenchymal stem cell(hUC-MSC)infusion for immune nonresponder(INR)patients with chronic HIV-1 infection,who represent an unmet medical need even in the era of efficient antiretroviral therapy(ART).Seventy-two INR patients with HIV were enrolled in this phase II randomized,double-blinded,multicenter,placebo-controlled,dose-determination trial(NCT01213186)from May 2013 to March 2016.They were assigned to receive high-dose(1.5 x 106/kg body weight)or low-dose(0.5 x 106/kg body weight)hUC-MSC,or placebo.Their clinical and immunological parameters were monitored during the 96-week follow-up study.We found that hUC-MSC treatment was safe and well-tolerated.Compared with baseline,there was a statistical increase in CD4+T counts in the high-dose(P<0.001)and low-dose(P<0.001)groups after 48-week treatment,but no change was observed in the control group.Kaplan-Meier analysis revealed a higher cumulative probability of achieving an immunological response in the low-dose group compared with the control group(95.8%vs.70.8%,P=0.00A).However,no significant changes in CD4/CD84-T counts and CD4/CD8 ratios were observed among the three groups.In summary,hUC-MSC treatment is safe.However,the therapeutic efficacy of hUC-MSC treatment to improve the immune reconstitution in INR patients still needs to be further investigated in a large cohort study. | Lifeng Wang Zheng Zhang Ruonan Xu Xicheng Wang Zhanjun Shu Xiejie Chen Siyu Wang Jiaye Liu Yuanyuan Li Li Wang Mi Zhang Wei Yang Ying Wang Huihuang Huang Bo Tu Zhiwei Liang Linghua Li Jingxin Li Yuying Hou Ming Shi Fu-Sheng Wang | 2021 | Signal Transduction and Targeted Therapy2021,6,7: | 0 |

| 8 | Optimization of high-pressure processing-assisted extraction of pectic polysaccharides from three berries显示文摘Background and objectives:High-pressure processing(HPP)is a promising assistive method to extract pectic polysaccharides with high rhamnogalacturonan I(RG-I)domain and berries are sources of such pectic polysaccharides.This study extracts pectic polysaccharides from goji berry,raspberry,and cranberry,examines how HPP influences the pectic polysaccharide structure of three berries,and provides a basis for the extraction and modification of pectic polysaccharides with specific structure and bioactivity.Materials and methods:An orthogonal test was performed to optimize the HPP-assisted alkali method to extract the high yield and high RG-I content pectic polysaccharides from three berries.Structural information of pectic polysaccharides extracted by the HPP method and conventional methods were compared from the perspectives of monosaccharide composition,molecular weight,Fourier transform infrared spectroscopy,andnuclearmagneticresonancespectroscopy.Results:For raspberry,the optimal conditions consisted of a pressure of 50o MPa,a pH of 13,and a pressure-holding time of 12 min,while the optimal conditions for goji berry and cranberry were both 40o MPa,pH 13,and 15 min.Under the optimal conditions,the yields for goji berry,raspberry,and cranberry were 10.49%,16.63%,and 17.52%,respectively,and RG-l contents were 81.85%,83.30%,and 63.22%,respectively.HPP showed an effect to degrade homogalacturonan backbones and side chains and increase the RG-I content to some extent.Conclusion:HPP-assisted alkali method was revealed to be an efficient method to extract high RG-I content pectic polysaccharides,especially for cranberry,and was a potential method to modify pectic polysaccharide structure in a certain way. | Xinxin Hu Chengxiao Yu Shokouh Ahmadi Yuying Wang Xingqian Ye Zhiqiang Hou Shiguo Chen | 2023 | Food Quality and Safety2023,7,1: | 0 |
