1An EcoCity model for regulating urban land cover structure and thermal environment: Taking Beijing as an example显示文摘Urban land-use/cover changes and their effects on the eco-environment have long been an active research topic in the urbanization field. However, the characteristics of urban inner spatial heterogeneity and its quantitative relationship with thermal environment are still poorly understood, resulting in ineffective application in urban ecological planning and management.Through the integration of 'spatial structure theory' in urban geography and 'surface energy balance' in urban climatology, we proposed a new concept of urban surface structure and thermal environment regulation to reveal the mechanism between urban spatial structure and surface thermal environment. We developed the EcoCity model for regulating urban land cover structure and thermal environment, and established the eco-regulation thresholds of urban surface thermal environments. Based on the comprehensive analysis of experimental observation, remotely sensed and meteorological data, we examined the spatial patterns of urban habitation, industrial, infrastructure service, and ecological spaces. We examined the impacts of internal land-cover components(e.g., urban impervious surfaces, greenness, and water) on surface radiation and heat flux. This research indicated that difference of thermal environments among urban functional areas is closely related to the proportions of the land-cover components.The highly dense impervious surface areas in commercial and residential zones significantly increased land surface temperature through increasing sensible heat flux, while greenness and water decrease land surface temperature through increasing latent heat flux. We also found that different functional zones due to various proportions of green spaces have various heat dissipation roles and ecological thresholds. Urban greening projects in highly dense impervious surfaces areas such as commercial, transportation, and residential zones are especially effective in promoting latent heat dissipation efficiency of vegetation, leading to strongly cooling effect of unit vegetation coverage. This research indicates that the EcoCity model provides the fundamentals to understand the coupled mechanism between urban land use structure and surface flux and the analysis of their spatiotemporal characteristics. 2Analysis and construction status of SG-Ⅱ 5PW laser facility显示文摘We present a recent progress of the SG-II 5 PW facility, which designed a multi-petawatt ultrashort pulse laser based on optical parametric chirped-pulse amplification(OPCPA). The prior two optical parametric amplifiers have been accomplished and chirped pulses with an energy of 49.7 J and a full-width-at-half-maximum(FWHM) spectrum bandwidth of 85 nm have been achieved. In the PW-scale optical parametric amplification(OPA), with the pump pulse that has an energy of 118 J from the second harmonic generation of the SG-II 7 th beam, the pump-to-signal conversion efficiency is up to 41.9%, which to the best of our knowledge is the highest among all of the reported values for OPCPA systems. The compressed pulse is higher than 37 J in 21 fs(1.76 PW), and the focal spot is ~10 μm after the closed-loop corrections by the adaptive optics. Limited by the repetition of the pump laser, the SG-II 5 PW facility operates one shot per hour. 4Soil Microbial Community and Enzyme Activity Responses to Herbaceous Plant Expansion in the Changbai Mountains Tundra, China显示文摘As one of the most sensitive regions to global climate change, alpine tundra in many places around the world has been undergoing dramatic changes in vegetation communities over the past few decades.Herbaceous plant species in the Changbai Mountains area have significantly expanded into tundra shrub communities over the past 30 yr.Soil microbial communities, enzyme activities, and soil nutrients are intertwined with this expansion process.In order to understand the responses of the soil microbial communities to such an expansion, we analyzed soil microbial community structures and enzyme activities in shrub tundra as well as areas with three different levels of herbaceous plant expansion.Our investigation was based on phospholipid fatty acid(PLFA) analysis and 96-well microtiter plates.The results showed that herbs have expanded greatly in the tundra, and they have become the dominant species in herbaceous plant expansion areas.There were differences for community composition and appearance among the shrub tundra and the mild expansion, moderate expansion, and severe expansion areas.Except for soil organic matter, soil nutrients were increased in herbaceous plant expansion areas, and the total nitrogen(TN), total phosphorus(TP), available nitrogen(AN), and available phosphorus(AP) were greatest in moderate expansion areas(MOE), while soil organic matter levels were highest in the non-expanded areas(CK).The total soil PLFAs in the three levels of herbaceous plant expansion areas were significantly higher than those in the non-expanded areas, and total soil PLFAs were highest in the moderately expanded area and lowest in the severely expanded area(SEE).Bacteria increased significantly more than fungi and actinomycetes with herbaceous plant expansion.Soil hydrolase activities(β-1,4-glucosidase(βG) activity, β-1, 4-N-acetylglucosaminidase(NAG) activity, and acid phosphatase(aP) activity) were highest in MOE and lowest in the CK treatment.Soil oxidase activities(polyphenol oxidase(PPO) activities and peroxidase(PER) activities) were also highest in MOE, but they were lowest in the SEE treatment.The variations in total soil PLFAs with herbaceous plant expansion were mostly correlated with soil organic matter and available phosphorus concentrations, while soil enzyme activities were mostly correlated with the total soil nitrogen concentration.Our results suggest that herbaceous plant expansion increase the total soil PLFAs and soil enzyme activities and improved soil nutrients.However, soil microorganisms, enzyme activity, and nutrients responded differently to levels of herbaceous plant expansion.The soil conditions in mild and moderate expansion areas are more favorable than those in severe expansion areas.JIN Yinghua ZHANG Yingjie XU Zhiwei GU Xiaonan XU Jiawei TAO Yan HE Hongshi WANG Ailin LIU Yuxia NIU Liping 2019Chinese Geographical Science2019,29,6:4
5Damage of hippocampal neurons in rats with chronic alcoholism显示文摘Chronic alcoholism can damage the cytoskeleton and aggravate neurological deficits. However, the effect of chronic alcoholism on hippocampal neurons remains unclear. In this study, a model of chronic alcoholism was established in rats that were fed with 6% alcohol for 42 days. Endogenous hydrogen sulfide content and cystathionine-beta-synthase activity in the hippocampus of rats with chronic alcoholism were significantly increased, while F-actin expression was decreased. Hippocampal neurons in rats with chronic alcoholism appeared to have a fuzzy nuclear membrane, mitochondrial edema, and ruptured mitochondrial crista. 6Comparative Assessment of Tundra Vegetation Changes Between North and Southwest Slopes of Changbai Mountains, China, in Response to Global Warming显示文摘Vegetation in high altitude areas normally exhibits the strongest response to global warming. We investigated the tundra vegetation on the Changbai Mountains and revealed the similarities and differences between the north and the southwest slopes of the Changbai Mountains in response to global warming. Our results were as follows: 1) The average temperatures in the growing season have increased from 1981 to 2015, the climate tendency rate was 0.38℃/10 yr, and there was no obvious change in precipitation observed. 2) The tundra vegetation of the Changbai Mountains has changed significantly over the last 30 years. Specifically, herbaceous plants have invaded into the tundra zone, and the proportion of herbaceous plants was larger than that of shrubs. Shrub tundra was transforming into shrub-grass tundra. 3) The tundra vegetation in the north and southwest slopes of the Changbai Mountains responded differently to global warming. The southwest slope showed a significantly higher degree of invasion from herbaceous plants and exhibited greater vegetation change than the north slope. 4) The species diversity of plant communities on the tundra zone of the north slope changed unimodally with altitude, while that on the tundra zone of the southwest slope decreased monotonously with altitude. Differences in the degree of invasion from herbaceous plants resulted in differences in species diversity patterns between the north and southwest slopes. 7Determination of Cd-MT in Pteria penguin with Indirect Non-Competitive ELISA显示文摘Cadmium-metallothionein (Cd-MT) of Pteria penguin was used to immunize domestic rabbit in order to obtain polyclonal antibody against Cd-MT. Using the anti-Cd-MT antibody, a method of indirect non-competitive enzyme-linked immunosorbent assay (non-competitive ELISA) was established to detect shellfish Cd-MT. In this case, the minimum detectable concentration of P. penguin Cd-MT was (4.563±0.051) ng/mL. The linear range of detection was 9.75-2.0×104 ng/mL. Intra-assay relative standard deviation (RSD) was less than 11.6%, and inter-assay RSD less than 6.7%. The recovery rates ranged from 83.7% to 119.0%. The polyclonal antibody against Cd-MT of P. penguin can also be used to detect MT of other four types of shellfish. 8Characterization of novel microsatellite markers of the Emei Shan Liocichla using restriction site-associated DNA sequencing显示文摘Background:The Emei Shan Liocichla(Liocichla omeiensis) is an endemic bird species to southwestern China with a small geographic range. However, little was known about the genetic status of this threatened species.Methods:We applied restriction-site-associated DNA sequencing(RAD-Seq) for rapid mass identification of microsatellite markers of the Emei Shan Liocichla.Results:A total of 11,564 microsatellite sequences were obtained, 600 random loci were designed for screening and 24 polymorphic microsatellite loci were selected for further validation. The average allele number, average observed heterozygosity and average expected heterozygosity were relatively low in our samples, which were 6.08, 0.6618 and 0.7048, respectively, indicating that the Emei Shan Liocichla might have lost some genetic diversity. Further analyses suggested that the populations distributed on two mountains(Daxiangling and Xiaoliangshan) showed a modest degree of genetic differentiation.Conclusions:These novel microsatellite markers provided valuable preliminary knowledge regarding the genetic status of the Emei Shan Liocichla and can be useful in further studies, as well as in the management and conservation of this species.Ailin Yang De Chen Pengcheng Wang Yiqiang Fu Zhengwang Zhang 2017Avian Research2017,8,2:2

13CRISPR/Cas9 screening identifies a kinetochore-microtubule dependent mechanism for Aurora-A inhibitor resistance in breast cancer显示文摘Background:Overexpression of Aurora-A(AURKA)is a feature of breast cancer and associates with adverse prognosis.The selective Aurora-A inhibitor alisertib(MLN8237)has recently demonstrated promising antitumor responses as a single agent in various cancer types but its phase III clinical trial was reported as a failure since MLN8237 did not show an apparent effect in prolonging the survival of patients.Thus,identification of potential targets that could enhance the activity of MLN8237 would provide a rationale for drug combination to achieve better therapeutic outcome.Methods:Here,we conducted a systematic synthetic lethality CRISPR/Cas9 screening of 507 kinases using MLN8237 in breast cancer cells and identified a number of targetable kinases that displayed synthetic lethality interactions with MLN8237.Then,we performed competitive growth assays,colony formation assays,cell viability assays,apoptosis assays,and xenograft murine model to evaluate the synergistic therapeutic effects of Haspin(GSG2)depletion or inhibition with MLN8237.For mechanistic studies,immunofluorescence was used to detect the state of microtubules and the localization of Aurora-B and mitotic centromere-associated kinesin(MCAK).Results:Among the hits,we observed that Haspin depletion or inhibition marginally inhibited breast cancer cell growth but could substantially enhance the killing effects of MLN8237.Mechanistic studies showed that co-treatment with Aurora-A and Haspin inhibitors abolished the recruitment of Aurora-B and mitotic centromere-associated kinesin(MCAK)to centromeres which were associated with excessive microtubule depolymerization,kinetochore-microtubule(KT-MT)attachment failure,and severe mitotic catastrophe.We further showed that the combination of MLN8237 and the Haspin inhibitor CHR-6494 synergistically reduced breast cancer cell viability and significantly inhibited both in vitro and in vivo tumor growth.Conclusions:These findings establish Haspin as a synthetic lethal target and demonstrate CHR-6494 as a potential combinational drug for promoting the therapeutic effects of MLN8237 on breast cancer.Ailin Chen Shijun Wen Fang Liu Zijian Zhang Meiling Liu Yuanzhong Wu Bin He Min Yan Tiebang Kang Eric W-F Lam Zifeng Wang Quentin Liu 2021Cancer Communications2021,41,2:1

15Systematic study of spatiotemporal influences on temporal contrast in the focal region in large-aperture broadband ultrashort petawatt lasers显示文摘Temporal contrast is one of the crucial physical determinants which guarantee the successful performance of laser–matter interaction experiments. We generally reviewed the influences on the temporal contrast in three categories of noises based on the requirement by the physical mechanisms. The spatiotemporal influences on temporal contrast at the focal region of the chromatic aberration and propagation time difference introduced by large-aperture broadband spatial filters, which were spatiotemporally coupled with compression and focusing, were calculated and discussed with a practical case in SG-Ⅱ5 PW ultrashort petawatt laser. 17Analysis and countermeasures of natural gas development in China显示文摘Natural gas has become a transitional bridge from fossil to green and clean energy worldwide.The importance of natural gas utilization in energy structure optimization and green development is no exception for China.However,the supply of China’s natural gas limits the country’s demand with three factors.(1)China’s natural gas resource is relatively rich,but its quality is poor with deep burial depth.Therefore,the annual plateau yield of natural gas cannot be high,with an estimated value of approximately 260-270 x 109 m3.(2)The demand for natural gas in China’s economic development is growing rapidly.The peak demand is estimated to be approximately 550-650 x 109 m3 per year or even higher.The import volume of natural gas will soon exceed that of domestic self-produced gas.(3)Natural gas is a necessity closely related to livelihood.Particularly,a shortage in natural gas supply affects social stability.Therefore,its external dependency should be under 50%and not exceed 60%.In this study,the future situation of China's natural gas is forecasted,and relevant countermeasures and suggestions are proposed in accordance with research on China’s natural gas resource potential,production trend,supplydemand link,and gas supply safety factors.Wenzhi ZHAO Ailin JIA Guosheng ZHANG 2019Frontiers of Engineering Management2019,6,4:1
