| 3 | Generation of male-sterile soybean lines with the CRISPR/Cas9 system显示文摘Soybean [Glycine max(L.) Merr.] provides a rich source of plant protein and oil worldwide. The commercial use of transgenic technology in soybean has become a classical example of the application of biotechnology to crop improvement. Although genetically modified soybeans have achieved commercial success,hybrid soybean breeding is also a potential way to increase soybean yield. Soybean cytoplasmic malesterile(CMS) lines have been used in three-line hybrid breeding systems, but their application to exploiting soybean heterosis has been limited by rare germplasm resource of sterile lines. The generation of various genetic diversity male-sterile soybean lines will help to overcome the shortcoming. In this study,we used targeted editing of AMS homologs in soybean by CRISPR/Cas9 technology for the first time to generate stable male-sterile lines. Targeted editing of GmAMS1 resulted in a male-sterile phenotype,while editing of GmAMS2 failed to produce male-sterile lines. | 4 | Lower limit of thermal maturity for the carbonization of organic matter in marine shale and its exploration risk显示文摘Based on the drilling data of the Silurian Longmaxi Formation in the Sichuan Basin and periphery, SW China, the Ro lower limits and essential features of the carbonization of organic matter in over-high maturity marine shale were examined using laser Raman, electrical and physical property characterization techniques. Three preliminary conclusions are drawn:(1) The lower limit of Ro for the carbonization of Type I-II1 organic matter in marine shale is 3.5%; when the Ro is less than 3.4%, carbonization of organic matter won't happen in general; when the Ro ranges from 3.4% to 3.5%, non-carbonization and weak carbonization of organic matter may coexist; when the Ro is higher than 3.5%, the carbonization of organic matter is highly likely to take place.(2) Organic-rich shale entering carbonization phase have three basic characteristics: log resistivity curve showing a general 'slender neck' with low-ultralow resistance response, Raman spectra showing a higher graphite peak, and poor physical property(with matrix porosity of only less than 1/2 of the normal level).(3) The quality damage of shale reservoir caused by the carbonization of organic matter is almost fatal, which primarily manifests in depletion of hydrocarbon generation capacity, reduction or disappearance of organic pores and intercrystalline pores of clay minerals, and drop of adsorption capacity to natural gas. Therefore, the lower limit of Ro for the carbonization of Type I-II1 organic matter should be regarded as the theoretically impassable red line of shale gas exploration in the ancient marine shale formations. The organic-rich shale with low-ultralow resistance should be evaluated effectively in area selection to exclude the high risk areas caused by the carbonization of organic matter. | 7 | Stochastic dynamics of conduction failure of action potential along nerve fiber with Hopf bifurcation显示文摘Action potentials can be induced by external electronic impulsive stimulations applied at one end of the unmyelinated fibers(C-fibers), while some action potentials fail to conduct to the other end of the fiber when the stimulation frequency becomes high. Such a phenomenon is called as conduction failure, which was observed in the biological experiments and related to the painful diabetic neuropathy, inflammation, and trauma in the previous studies. On-off firing pattern was recorded from the fiber when conduction failure happened. In the present study, the diffusion Hodgkin-Huxley(HH) model with resting state near a Hopf bifurcation is adopted to simulate the experimental observations. When the periodic electrical pulses with high frequency are applied to one end of the fiber described by the deterministic HH model, conduction failure and the corresponding firing patterns different from the on-off firing pattern are simulated. When noise is introduced to form the stochastic HH model, the firing pattern corresponding to conduction failure becomes the on-off firing pattern, which is characterised by transition behaviors between on-phase(continuous action potentials) and off-phase(a long quiescent state) and large variations in the durations of both phases. Furthermore, the increase of potassium conductance can enhance the conduction failure degree, which closely matches those observed in the experiment and is suggested to be related to the reduction of pain signals. The bottleneck in clinical development of PDEδ inhibitors is the poor antitumor activity of known chemotypes. Here, we identified novel spiro-cyclic PDEδ inhibitors with potent antitumor activity both in vitro and in vivo. | 12 | Sesame oil determination by solid-phase extraction and liquid chromatography-tandem mass spectrometry显示文摘Authenticity of blended oil has drawn increasing attentions. In this study, sesamin was selected as marker of sesame oil to verify authentication of blended oil with sesame oil on label. A simple method based on liquid chromatography-tandem mass spectrometry (LC-MS/MS) combined with solid-phase extraction (SPE) was used to detect sesamin. It has high sensitivity with low limit of detection (LOD) and limit of quantification (LOQ) being 20 ng/mL and 70 ng/mL respectively. Recoveries from 80.16% to 102.50% in 4 different oils indicated it’s reliable to quantify sesamin in edible oil. Four blended oils were used as validation samples and results showed this method could effectively detect whether sesame oil exists. | 15 | Increasing Coal-Fired Power Plant Operational Flexibility by Integrating Solar Thermal Energy and Compressed Air Energy Storage System显示文摘This paper proposed a novel integrated system with solar energy,thermal energy storage(TES),coal-fired power plant(CFPP),and compressed air energy storage(CAES)system to improve the operational flexibility of the CFPP.A portion of the solar energy is adopted for preheating the boiler’s feedwater,and another portion is stored in the TES for the CAES discharging process.Condensate water from the CFPP condenser is used for cooling compressed air during the CAES charging process.The thermodynamic performance of the integrated system under different load conditions is studied.The system operations in a typical day are simulated with EBSILON software.The system enables daily coal saving of 9.88 t and reduces CO_(2)emission by 27.95 t compared with the original CFPP at 100%load.Under partial load conditions,the system enables maximum coal saving of 10.29 t and maximum CO_(2)emission reduction of 29.11 t at 75%load.The system has maximum peak shaving depth of 9.42%under 40%load condition.The potential of the system participating ancillary service is also discussed.It is found that the integration of solar thermal system and CAES system can bring significant ancillary service revenue to a conventional CFPP. | LI Xiaoyu WANG Yumeng ZHANG Xinjing LI Bin XU Yujie CHEN Haisheng SHENG Siqing | 2023 | Journal of Thermal Science2023,32,6: | 1 |
