| 1 | Green fluorescent protein (GFP) transgenic pig produced by somatic cell nuclear transfer显示文摘Transgenic somatic cell nuclear transfer is a very promising route for producing transgenic farm ani-mals. Research on GFP transgenic pigs can provide useful information for breeding transgenic pigs, human disease models and human organ xenotransplantation. In this study, a liposomal transfection system was screened and transgenic embryos were reconstructed by nuclear transfer of GFP positive cells into enucleated in vitro matured oocytes. The development of reconstructed embryos both in vitro and in vivo was observed, and GFP expression was determined. The results showed that porcine fe-tal-derived fibroblast cells cultured with 4.0 μL/mL liposome and 1.6 μg/mL plasmid DNA for 6 h re-sulted in the highest transfection rate (3.6%). The percentage of GFP reconstructed embryos that de-veloped in vitro to the blastocyst stage was 10%. Of those the GFP positive percentage was 48%. Re-constructed transgenic embryos were transferred to 10 recipients. 5 of them were pregnant, and 3 de-livered 6 cloned piglets in which 4 piglets were transgenic for the GFP as verified by both GFP protein expression and GFP DNA sequence analysis. The percentage of reconstructed embryos that resulted in cloned piglets was 1.0%; while the percentage of piglets that were transgenic was 0.7%. | 2 | Hydrogen generation from decomposition of hydrous hydrazine over Ni-Ir/CeO_2 catalyst显示文摘The synthesis of highly active and selective catalysts is the central issue in the development of hydrous hydrazine(N_2H_4·H_2O) as a viable hydrogen carrier. Herein, we report the synthesis of bimetallic Ni-Ir nanocatalyts supported on CeO_2 using a one-pot coprecipitation method. A combination of XRD, HRTEM and XPS analyses indicate that the Ni-Ir/CeO_2 catalyst is composed of tiny Ni-Ir alloy nanoparticles with an average size of around 4 nm and crystalline CeO_2 matrix. The Ni-Ir/CeO_2 catalyst exhibits high catalytic activity and excellent selectivity towards hydrogen generation from N_2H_4·H_2O at mild temperatures. | 3 | Effect of river plume on phytoplankton community structure in Zhujiang River estuary显示文摘To examine the phytoplankton assemblages and the effect of diluted waters on them,a research cruise was conducted from July 19 to August 7,2015 in the Zhujiang(Pearl)River estuary in the northern South China Sea(21°N–23.5°N,111°E–117°E).Samples were collected from 65 stations including one for time-series sampling.A total of 212 phytoplankton taxa were identified from 61 genera belonging to 4 phyla.Among them,122 species identified from 42 genera of Bacillariophyta and 83 species from 15 genera of Pyrrophyta.Chain-forming diatoms dominated the phytoplankton community where Pseudonitzschia delicatissima,Guinardia striata,Thalassionema nitzschioides,and P.pungens comprised about 52%of the total abundance.However,higher cell abundances concentrated on both sides of the estuary,because of low salinity and high nutrients brought by diluted water.In addition,Canonical Correspondence Analysis revealed that salinity and dissolved inorganic nitrogen shaped the species composition in the study area.Furthermore,the Jaccard similarity index showed prevailing high similarity in the distribution of species in low-salinity diluted waters,and the Bray-Curtis similarity depicted distinguished grouping for phytoplankton assemblages along the salinity gradient.However,phytoplankton diel vertical cycles showed maximum abundance occurred at 2:00 am,which was mainly contributed by benthic phytoplankton species Pseudo-nitzschia spp.and T.nitzschioides. | Qi ZHONG Bing XUE Md Abu NOMAN Yuqiu WEI Haijiao LIU Hongbin LIU Liping ZHENG Hongmei JING Jun SUN | 2021 | Journal of Oceanology and Limnology2021,39,2: | 2 |
| 4 | Optimal time for subarachnoid transplantation of neural progenitor cells in the treatment of contusive spinal cord injury显示文摘This study aimed to identify the optimal neural progenitor cell transplantation time for spinal cord injury in rats via the subarachnoid space. Cultured neural progenitor cells from 14-day embryonic rats, constitutively expressing enhanced green fluorescence protein, or media alone, were injected into the subarachnoid space of adult rats at 1 hour (acute stage), 7 days (subacute stage) and 28 days (chronic stage) after contusive spinal cord injury. Results showed that grafted neural progenitor cells migrated and aggregated around the blood vessels of the injured region, and infiltrated the spinal cord parenchyma along the tissue spaces in the acute stage transplantation group. However, this was not observed in subacute and chronic stage transplantation groups. O4- and glial fibrillary acidic protein-positive cells, representing oligodendrocytes and astrocytes respectively, were detected in the core of the grafted cluster attached to the cauda equina pia surface in the chronic stage transplantation group 8 weeks after transplantation. Both acute and subacute stage transplantation groups were negative for O4 and glial fibrillary acidic protein cells. Basso, Beattie and Bresnahan scale score comparisons indicated that rat hind limb locomotor activity showed better recovery after acute stage transplantation than after subacute and chronic transplantation. Our experimental findings suggest that the subarachnoid route could be useful for transplantation of neural progenitor cells at the acute stage of spinal cord injury. Although grafted cells survived only for a short time and did not differentiate into astrocytes or neurons, they were able to reach the parenchyma of the injured spinal cord and improve neurological function in rats. | 16 | Biliary candidiasis mimicking IgG4-related disease in a patient with normal immunity显示文摘Purpose:To report a rare case of biliary candidiasis in a host with normal immunity.Methods:Laboratory tests indicated an elevated G level of 1000 pg/ml.Microscopic examination of pathological sections demonstrated multiple,narrow base,budding yeast cells following Grocott's methenamine silver staining and periodic acid-Schiff staining.Subsequent sequence analysis supported the diagnosis of biliary candidiasis.Results:A 31-year-old previously healthy man developed obstructive jaundice under no obvious inducement.He experienced cholecystectomy and biliary drainage.Surgical specimens revealed granulomatous chronic inflammation in the gallbladder,liver,bile ducts and mesenteric lymph nodes.He was misdiagnosed with tuberculosis and received antituberculosis treatment.However,his condition did not improve.He was finally diagnosed with biliary candidiasis and administered the antifungal treatment with fluconazole.His symptoms improved after such treatment.Conclusions:We reported a case of biliary candidiasis that mimicked IgG4-RD in a host with normal immunity.Our findings highlighted the need for pathological diagnosis in patients with symptoms resembling IgG4-RD. | Wenjie Bian Yuchao Zhong Xia Zhang Hongbin Chen Ruie Feng Xinyu Zhang Danhua Shen Yanying Liu | 2022 | Rheumatology & Autoimmunity2022,2,3: | 0 |
